【発明の属する技術分野】
本発明は次の反応を触媒する新規酵素、その製造方法及び該酵素を生産し得る微生物に関する。

本酵素は、ヌクレオシドホスフォリラーゼによる塩基交換反応を利用したプリンヌクレオシド化合物の製造の効率化に有用である。該プリンヌクレオシド化合物の製造において、本酵素は、ピリミジンヌクレオシドとプリン塩基との塩基交換により生成されるピリミジン塩基を、塩基交換を触媒する酵素(ヌクレオシドホスホリラーゼ)の基質になり得ない化合物に変換する役割を果たす。尚、塩基交換反応による該プリンヌクレオシド化合物の製造方法の詳細は、例えば特開平11−46790に開示されている。また、上記反応から理解できるように、本酵素はバルビツール酸の定量に利用することも可能である。

【従来の技術】
国際生化学連合酵素委員会報告(Enzyme Nomenclature -Recommendations(1978) of the Nomenclature Committee of the International Union of Biochemistry, Academic Press(1979))、同報告の補遺1(NC-IUB, Eur. J. Biochem., 104, 1-4(1979))と補遺2(NC-IUB, Eur. J. Biochem.,116, 423-435(1981))において、バルビツラーゼ(Barubiturase、EC3.5.2.1)は下記式に示すように、バルビツール酸を加水分解してマロン酸と尿素を生成する加水分解酵素(Hydrolase)と定義されている。

バルビツラーゼの起源としては、Mycobacterium属(文献[1]、[2])、Nocardia属(文献[3])、Enterobacter属(文献[4])、及びCorynebacterium属(文献[1]、[5])が報告されている。そして、見いだされたバルビツラーゼの触媒機能と生成物は全て上記の式に合致することが報告されている。しかしながら、これまでバルビツラーゼは精製困難な酵素の一つとされ、途中まで精製されたことはあっても、単離されたことはなかった。

【発明が解決しようとする課題】
本発明の課題は、バルビツール酸の加水分解反応を触媒する安定な酵素、その製造方法及び該酵素を生産し得る微生物を提供することにある。

【課題を解決するための手段】
本発明者らは上述の課題を解決すべく、バルビツール酸加水分解酵素とその生産菌を探索した結果、従来知られているものとは全く性質を異にする新規酵素と該酵素を生産する微生物を見いだし本発明を完成するに至った。

まず本発明の第1は、次の反応を触媒する新規酵素である。

【発明の実施の形態】
以下、本発明を詳細に説明する。

〈新規酵素〉
まず本発明の第1は、次の反応を触媒する新規酵素であり、該酵素の理化学的性質及び酵素学的性質の典型的な例を挙げれば、以下のとおりである。

酵素の理化学的性質及び酵素学的性質
(1)基質特異性:バルビツール酸に対して作用する。

(2)至適pH及びpH安定性:至適pHは8.0付近であり、pH6.0〜7.0において安定である。

(3)至適温度及び熱安定性:至適温度は40℃で、4℃〜50℃で安定である。

(a) SH阻害剤;N−エチルマレイミド[NEM]、5,5'−ジチオビス(2−ニトロ安息香酸)[DTNB]、p−(クロロメルクリ)安息香酸[p-CMB]、N−ブロモスクシンイミド、

〈新規酵素の製造方法〉
次いで、本発明の第2は、上述の新規酵素を生産する能力を有する微生物を培養し、培養物から該酵素を採取することを特徴とする新規酵素の製造方法である。前記微生物は、好ましくはロドコッカス属又はアルスロバクター属に属し、更に好ましくは、ロドコッカス・エリスロポリス JCM 3191、ロドコッカス・エリスロポリス JCM 3132又はアルスロバクター・スピーシーズ YGK 222(FERM BP−5907)である。

本発明で用いる微生物の培養は通常行われる条件で行えばよい。大量に培養するには液体培地を用い、振盪培養又は通気撹拌培養により好気的条件下で行うことが好ましい。培地としては炭素源及び窒素源としてグルコース、グリセリン、ウラシル、チミン、アスパラギン、ペプトン、肉エキス、マルトエキス、イーストエキス、塩化アンモニウムなどを用いることができるが、さらにビタミン類の添加が好ましい。本微生物が生育し、本酵素が十分に生産される条件であれば、培養温度、培養時間、培養のpHは特に制限されない。培養条件によって変動することもありうるが、培養温度は30℃近傍、初発pHは7.0付近で、培養時間は48〜144時間、好ましくは96時間、好気的に培養する。

培養物から本酵素を採取するには、通常用いられる方法に従って行えばよい。例えば、遠心分離などによって集菌した菌体を超音波、あるいはガラスビーズなどで摩砕した後、遠心分離などにより細胞片などの固形物を除き、粗酵素液を得る。次に、硫安、硫酸ナトリウムなどによる塩析法、イオン交換クロマトグラフィー、ゲル濾過クロマトグラフィー、アフィニテイクロマトグラフィーなどによるクロマトグラフ法、ゲル電気泳動法などを用いて精製することができる。この間、本酵素の安定化のためにエチレングリコールを10%程度共存させておくことが好ましい。

〈新規酵素を生産する微生物〉
さらに、本発明の第3は、新規酵素を生産する能力を有するロドコッカス属又はアルスロバクター属に属する微生物を提供することである。好ましくは、ロドコッカス・エリスロポリス(Rhodococcus erythropolis) JCM 3132、ロドコッカス・エリスロポリス(Rhodococcus erythropolis) JCM 3191、又はアルスロバクター・スピーシーズ(Arthrobacter sp.) YGK222(FERM BP−5907)である。最も好ましくは、ロドコッカス・エリスロポリス(Rhodococcus erythropolis) JCM 3132である。

ロドコッカス・エリスロポリス(Rhodococcus erythropolis) JCM 3132とロドコッカス・エリスロポリス(Rhodococcus erythropolis) JCM 3191の菌株は、理化学研究所微生物系統保存施設の保有する菌株であって、JCM菌株カタログ第7版(1999)に掲載されている。アルスロバクター・スピーシーズ(Arthrobacter sp.) YGK 222(FERM BP−5907)は、通商産業省工業技術院生命工学工業技術研究所特許微生物寄託センターに寄託されている。

寄託菌株の菌学的性質を、バージェイス・マニュアル・オブ・デターミナティブ・バクテリオロジー第8版(1975年)、バージェイズ・マニュアル・オブ・システマティック・バクテリオロジー第1版(1984)及び第2版(1986)に準じて検討した結果は次の通りである。なお、実験は主として長谷川武治編著、改訂版「微生物の分類と同定」(学会出版センター、1985年)記載の方法により行った。

【実施例】
以下、実施例により本発明を更に詳しく説明する。

実施例1:本酵素生産菌の探索
本酵素を生産する菌の探索を、2段階の選抜により行った。
第一の選抜として、ウラシル又はチミン2g、リン酸二水素カリウム1g、リン酸水素二カリウム3g、酵母エキス6g及び蒸留水1LよりなるpH7.0の「A培地」に良く生育し、ウラシル又はチミンを著しく資化する微生物を、約1500の土壌及び保有菌株の中から選抜した。培養は各微生物の生育適温で振盪培養により行った。ウラシル及びチミンの資化性はUV検出器を装着した高速液体クロマトグラフィー(HPLC)により、その減少度合いから定量した。

次に、第二の選抜を行った。選抜菌株の培養には、A培地又はグリセリン10g、塩化アンモニウム4g、硫酸マグネシウム・七水塩0.3g、リン酸二水素カリウム1g、リン酸水素二カリウム1g、チアミン塩酸塩1mg、リボフラビン2mg、ニコチン酸2mg、パントテン酸2mg、ピリドキシン塩酸塩2mg、ビオチン 0.1 mg、p−アミノ安息香酸1mg、葉酸 0.1 mg、塩化カルシウム二水和物10mg、塩化マンガン四水和物10mg、塩化ニッケル六水和物10mg、硫酸亜鉛七水和物10mg、塩化カドミウム二水和物10mg、硫酸銅五水和物10mg、硫酸鉄七水和物10mg、硫酸ベリリウム四水和物10mg、塩化ルビジュウム10mg、塩化コバルト(II)・無水10mg及び蒸留水1LからなるpH7.0の「B培地」を用いた。B培地5mLを入れた試験管(16.5 x 160 mm)に上記第一の選抜菌株を接種し、300 rpmで振盪させつつ、28℃で96時間培養し、遠心分離により湿菌体を得た。

バルビツール酸20mM、Hepes(同仁化学研究所製)/NaOH100mM(pH8.0)の水溶液0.1mLに湿菌体10mgを加え、30℃で30〜60分間放置した。遠心分離により上清を得て、上記HPLCによりバルビツール酸の減少と生成物を分析した。

その結果、本酵素を生産する菌株として、ロドコッカス・エリスロポリス JCM 3132、ロドコッカス・エリスロポリス JCM 3191及びアルスロバクター・スピーシーズ YGK 222(FERM BP−5907)が選定された。この中で、ロドコッカス・エリスロポリス JCM 3132が一番活性が強かったので、以下、本菌株が生産する新規酵素について検討した。

実施例2:各種環状アミド関連化合物の新規酵素生産に対する効果
B培地に表1に示す各種環状アミド関連化合物を0.15%(w/v)加えた以外は実施例1と同様の条件でロドコッカス・エリスロポリス JCM 3132を培養し、酵素活性を測定した。なお、酵素活性は湿菌体1gが1分間に分解するバルビツール酸量(μmol)として表示した。化合物を加えなかった場合の活性を100として相対活性を右欄に示した。バルビツール酸、ウラシル及びチミンが最も有効な活性誘導物質であった。

実施例3:本酵素安定化物質
本酵素は不安定な酵素であり、培養菌体を破砕すると、そのままでは急速に失活した。そこで、安定化物質を検索した。湿菌体20gを20mMリン酸カリウムバッファー(pH7.0)20mLに懸濁したものを超音波破砕に供し、遠心分離後得た上清を酵素液として用いた。これに0.2%(w/v)の種々の界面活性剤(Tween20、Tween80、TritonX-100、Span20、Briji35、コール酸ナトリウム、ヘキサデシルピリジニウムクロリド一水和物)、10%(w/v)のグリセロールもしくはエチレングリコールを種々の組み合わせで加え、4℃保存時における安定化効果を検討した。その結果、図1(代表的な結果を示す)に示すように、エチレングリコールがきわめて有効な物質であることを見いだした。

実施例4:本酵素の分離精製
培地Bにチミン0.15%(w/v)を加えた「培地C」500mLを2L坂口フラスコ中に調製し、これにロドコッカス・エリスロポリス JCM 3132を接種し、培養温度28℃、150rpmにて96時間振盪培養した。

以下の操作は全て4℃以下で行った。また、タンパク質はローリー法により定量した。

上記培養の終了後、菌体を遠心分離(10,000g、10分)により集め、得られた湿菌体20gを20mLの10%エチレングリコールを含む10mMリン酸カリウム緩衝液(pH8.0)に懸濁した。該懸濁液を、ダイノ−ミル(DYNO-MILL、ウイリー・エー・バコーフェン社製)によりガラスビーズで破砕し、該破砕液を遠心分離(15,000g、15分)することにより無細胞抽出液を得た。この無細胞抽出液にプロタミン硫酸を添加し、核酸及びミクロゾームを除去した。次いで遠心分離し、得られた上清を、DEAE-Sepharacelカラムを用いたアニオン交換クロマトグラフィーを行い、塩化ナトリウムの濃度勾配溶出により、本酵素活性を有する画分を回収した。このアニオン交換クロマトグラフィーを、塩化ナトリウムの濃度勾配を緩やかにしてもう1度行った。得られた活性画分を、MonoQ HR5/5を用いた強アニオン交換クロマトグラフィーにより精製した。さらに、活性画分をPhenyl-Sepharose CL-4Bを用いた疎水性カラムクロマトグラフイーにより精製した。最後に、Superdex S-200カラムによりゲル濾過クロマトグラフイーを行い、最終精製酵素標品を得た。得られた本酵素酵素標品は、ドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動(SDS-PAGE)において、単一のバンドを示し、その分子量は45,000であった。また、ゲル濾過法による本酵素の分子量は172,000であった。したがって、本酵素は4つの単一なサブユニットからなる、ホモ4量体であることが判明した。

実施例5:本酵素のアミノ酸配列
実施例4で得られた本酵素標品の部分アミノ酸配列を定法により決定した。結果は以下のようであり、部分アミノ酸配列はこれまでに知られているいかなる環状アミド変換酵素とも類似性がない。

実施例6:生成物の同定
精製単離した本酵素をバルビツール酸に作用させ、下記条件の高速液体クロマトグラフィー(HPLC)により反応生成物を分析した。生成物は唯一つであり、分取により単離された。

本生成物に関して、以下のことがHPLCの保持時間の比較、スペクトロメトリーの一致などにより確認された。すなわち、単離した物質はバルビツール酸と異なり250nm付近に吸収を示さなかったことから、環状構造を持たないことが判明した。本物質は加水分解により、尿素とマロン酸を生成した。また、本物質は酸性条件下で自動的に閉環し、元のバルビツール酸に戻ることが、確認された。以上の結果から、本生成物はウレイドマロン酸(ureidomalonate)と同定された。HPLCの結果を図2に示す。(B)は、本酵素によりバルビツール酸から反応生成物が生じたことを示し、(A)は、該反応生成物の加水分解によって尿素とマロン酸が生じたことを示す。

実施例7:温度とpHの影響
(1)温度: バルビツール酸20mM、Hepes/NaOH100mM(pH8.0)に、単離した本酵素を加え、温度を変化させて触媒活性を測定した結果、活性の最適温度は40℃であった。また、温度4℃〜50℃では安定であった。

(2)pH:定法により緩衝液(100mM)のpHを変えて酵素の安定性を調べた結果、活性はpH8.0で最大であったが、pH6.0〜7.0において最も安定であった。

実施例8:反応機構と基質特異性
(1)触媒作用:バルビツール酸20mM、Hepes/NaOH100mM(pH8.0)に、単離した本酵素を40℃で作用させて、反応の経時変化を求めた結果を図3に示す。本反応が平衡に達することを示しており、作用は加水分解と脱水閉環の双方向を触媒し可逆的であることが分かった。

(2)動力学的パラメーター:上記と同一条件で、バルビツール酸加水分解反応における動力学的パラメーターを求めた。最大速度Vmaxは2.5μmol/min/mgであり、ミカエリス定数Kmは1.0mMであった。

(3)基質特異性:上記バルビツール酸の代わりに、ウラシル、チミン、ジヒドロウラシル、ジヒドロチミン、オロチン酸、ジヒドロオロチン酸、アロキサン、パラバン酸、ヒダントイン、スクシンイミド、バルビタール、シクロバルビタール、アロバルビタール、アラントイン、又はイソバルビツール酸を用いて反応を行わせたが、何れも加水分解を受けず、極めて基質特異性が高いことが判明した。

実施例9:反応の阻害
バルビツール酸20mM、Hepes/NaOH 100mM(pH8.0)に単離した本酵素を加え、下記の各種化合物及び金属イオンをそれぞれ添加して、40℃で反応を行わせた結果、

Claims (5)
以下の[1]〜[6]で示される理化学的性質及び酵素学的性質を有し、以下の[7]で示される部分アミノ酸配列を含む新規酵素:
[1]基質特異性:バルビツール酸に対して作用する。
[2]至適 pH 及び pH 安定性:至適 pH は 8.0 付近であり、 pH6.0 〜 7.0 において安定である。
[3]至適温度及び熱安定性:至適温度は 40 ℃で、4℃〜 50 ℃で安定である。
[4]阻害:SH阻害剤、セリンプロテアーゼ阻害剤、ジヒドロオロチン酸、アロキサンにより活性が阻害される。
[5]安定化:エチレングリコールにより安定化される。
[6]分子量:
(a) ゲル濾過法による分子量は 172,000 。
(b) SDS−ポリアクリルアミド電気泳動法によるサブユニットの分子量は 45,000 。

A novel enzyme having the physicochemical and enzymatic properties shown in the following [1] to [6] and comprising a partial amino acid sequence shown in the following [7] :
[1] Substrate specificity: Acts on barbituric acid.
[2] Optimum pH and pH stability: The optimum pH is around 8.0 and is stable at pH 6.0 to 7.0 .
[3] Optimum temperature and thermal stability: The optimum temperature is 40 ° C and is stable at 4 ° C to 50 ° C.
[4] Inhibition: Activity is inhibited by SH inhibitor, serine protease inhibitor, dihydroorotic acid and alloxan.
[5] Stabilization: Stabilized with ethylene glycol.
[6] Molecular weight:
(A) The molecular weight by gel filtration method is 172,000 .
(B) The molecular weight of the subunit by SDS-polyacrylamide electrophoresis is 45,000 .
