本発明は、従来、廃棄処分をしていたエビ、カニの殻等からカロテノイド色素の一種であるアスタキサンチンを抽出する方法及び装置の技術の分野に属する。

【従来の技術】
アスタキサンチンは、カニやエビ等の甲殻動物や、鮭や鱒、おきあみ等の魚に多く含まれている天然色素であり、肉色改良を目的として魚類の飼料添加剤として使用されている化合物である。そして、アスタキサンチンは、一般には、化学的方法によって合成された製品が主流となっており、天然物からの抽出されたものは極わめて少なく、カニやエビの殻からの抽出は殆どなされていないのが現状である。

ところで、キチン、キトサンは白色質のものが要求されるために、エビ、カニ殻からキチン、キトサンを製造する場合、エビ、カニ殻の色素を漂白剤を使用して除去している。キチン・キトサンは、エビやカニの殻を塩酸に浸し、炭酸カルシウムを溶かし出した後、アルカリと煮沸して蛋白質を取り除き、残った沈澱物を洗浄し、乾燥させることによって得られている。

エビ、カニの殻には、カルシウム、タンパク、キチン質、色素であるアスタキサンチンが含有されており、特にキチン質は約30%、アスタキサンチンは約2%含有されている。また、エビ、カニ殻100g中から1〜1.5gのアスタキサンチン抽出液が得られる。アスタキサンチンは、抗酸化剤として食品添加物、着色料、動物の体内で代謝されてビタミンAとなることから貴重な栄養素として、また、アスタキサンチンは、化粧品、医薬品等に広く使用されている。

【発明が解決しようとする課題】
前記特開平5−255241号公報に記載されているものは、ニンジンや藻類等のカロチン含有植物から有機溶媒を用いてカロチンを抽出した抽出液から前記有機溶媒を蒸発させて得られるカロチン含有油脂中のカロチン結晶を一旦溶解させ、結晶溶媒としてカロチン結晶中に取り込まれていた残留有機溶媒を油脂中に溶出させてから不活性ガスを通気しながら減圧蒸発するというものであり、工程が複雑であるためにそのコストが高くなるというものであり、この方法によってはエビ、カニの殻に含有されている色素であるアスタキサンチンを安全にして容易且つ安価に抽出することができるというものではない。

そして、上記公報に記載されているように、従来より、カロチン含有植物を破砕して有機溶媒でカロチンを抽出し、該抽出液から前記有機溶媒を蒸発させて天然色素であるカロチンを製造する方法は、種々行われており、溶媒として低級アルコールを加えてアルコーリシス化することによって脂肪酸低級アルコールエステルを生成させ、減圧蒸留してカロチンを得る方法は、特開昭51ー147532号公報に、また、このアルコーリシス化した溶媒に脂肪酸低級アルキルエステルを主成分とする油層に親水性溶剤及び水を加えて、疎水性の強いカロチンを析出分離する方法は、特開昭61ー115062号公報に記載されている。そして、これらアルコーリシス化によりカロチンを製造する方法は、カロチンを含有する植物油脂をアルコーリシス化することにより親水性のエステルに化学変化させて蒸留したり、或いは、カロチン濃縮層と親水層との液−液分離で取り除いたりしており、いわば化学的な処理により余計な脂肪をエステル化して取り除く方法についてのものである。

本発明に係るエビ、カニの殻からアスタキサンチンを抽出する方法は、エビ、カニ殻を塩酸に浸漬させてエビ、カニ殻中のカルシウムを除去した後、該エビ、カニ殻を水酸化ナトリウム溶液に浸漬させて中和処理するとともにエビ、カニ殻中の蛋白質を除去した後、78℃±5℃の温度の醸造用アルコール20%、水80%からなる溶媒中に浸漬させて、前記溶媒からアスタキサンチンを分離抽出する。

【発明の実施の形態】
