本発明は、生理活性物質を担持体に固定化させることにより生理活性物質を検出する生理活性物質検出装置及び生理活性物質検出方法に関する。

【従来の技術】
DNAチップは生体試料中に存在する特定のDNA配列を検出する装置であり、遺伝的疾患の検出や薬剤応答性の判定などに使用される。

DNAチップはガラスやシリコンなどの基板上に多数のプローブが固定化された構造をしている。プローブは一本鎖DNAであり、その配列は複数種類搭載されている。そして、どのプローブが試料中の一本鎖DNAとハイブリダイズしたかを検出することにより、試料中のDNA配列を知ることができるようになっている。ハイブリダイズの検出方式には、蛍光検出方式と電流検出方式がある。

【発明が解決しようとする課題】
しかしながら、従来のDNAチップでは、基板上に固定化されたプローブを取り外すことができない。また、電流検出方式のDNAチップでは、ハイブリダイズの検出に用いる挿入剤を再利用できない。従って、DNAチップは再利用することができず、コストが高くなる。また、従来のDNAチップでは、予め定められたDNA配列の検出のみにしか利用できないため、ユーザが特定の配列検出用に自由にプローブを配置して用いることができない。

本発明は上記課題を解決するためになされたもので、その目的とするところは、再利用可能な生理活性物質検出装置及び生理活性物質検出方法を提供することにある。

【課題を解決するための手段】
本発明の一の観点によれば、生理活性物質が固定化された磁気ビーズと、前記磁気ビーズを固定化させる複数の磁気ビーズ固定部位を有する基板と、前記磁気ビーズ固定部位のそれぞれに配置され、前記磁気ビーズ固定部位に前記磁気ビーズを引き寄せる磁界を発生させて前記磁気ビーズを前記磁気ビーズ固定部位に固定化させる複数の第1コイルと、前記複数の第1コイルのそれぞれに接続され、前記第1コイルに流れる電流を切り換える複数の第1スイッチと、前記基板の上部及び下部の少なくとも一方に設けられ、前記第1コイルよりも半径が大きく、前記基板に前記磁気ビーズを引き寄せる磁界を発生させる第2コイルとを具備してなる生理活性物質検出装置が提供される。

このように、基板上の固定部位に、選択的に固定化可能で、また選択的に除去可能な担持体として磁気ビーズを用いる。DNAなどの生理活性物質は、予めこの磁気ビーズに結合させておく。磁気ビーズを送入して選択的に磁気ビーズ固定部位に固定化させることにより、所望の位置に生理活性物質が固定化された生理活性物質検出装置が得られる。各磁気ビーズに異なる種類の生理活性物質を固定化させておくことにより、多様な生理活性物質がプローブとして搭載された検出装置が得られる。

また、第1スイッチにより電流を制御し、電流をオフにしたり、固定化の際と逆向きの電流を流すことにより、固定部位に磁気ビーズを引き寄せる磁界が無くなるため、磁気ビーズを除去できる。この場合、磁気ビーズを基板上から除去した後に新たな磁気ビーズを再度固定化させることができ、検出装置の再利用が可能となる。再利用した場合、検出装置内の特定の位置に固定化される生理活性物質の種類は、再利用の前に固定化させた生理活性物質と同一である必要は無い。従って、一つの検出装置を多用途に用いることができる。また磁気ビーズの除去は第1スイッチを選択的に制御することにより行うことができるので、検出装置内の一部の生理活性物質のみを入れ替えて再利用することもできる。

本発明の一の実施形態によれば、第1スイッチは行列状に配列されてなり、前記第1スイッチの行列の任意の行を選択する行選択手段と、前記第1スイッチの行列の任意の列を選択する列選択手段と、をさらに備える。

【発明の実施の形態】
以下、図面を参照しながら本発明の実施形態を説明する。

(第1実施形態)
図1及び図2は本発明の第1実施形態に係る生理活性物質検出装置の全体構成を示す図であり、図1は装置上面図、図2は断面図である。

図1に示すように、この生理活性物質検出装置1は、送液部3a及び3bを有する上部支持体2を備える。この上部支持体2の中心近傍には、円形のセル4が設けられている。セル4は、試料などの液体を充填する領域である。このセル4には、行列状に複数の固定化ユニットUが複数配置され、磁気ビーズ固定化回路系6をなしている。これら各固定化ユニットUにはそれぞれコイル半径aで、1よりも大きい所定の巻数の円筒状のコイル11が設けられており、このコイル11に電流が流れることにより磁界H1が発生する。この磁界により、各固定化ユニットUに磁気ビーズ12が固定化される。図1は、すべての固定化ユニットUに磁気ビーズ12が固定化された状態を示している。また、磁気ビーズ12にはプローブ13が固定化されている。

磁気ビーズ12とプローブ13との固定化は、周知の技術により、例えば磁気ビーズに付着したアビジンと、プローブ13に付着したビオチンとを結合させればよい。あるいは、磁気ビーズ12とプローブ13を直接付着させてもよい。この磁気ビーズ12とプローブ13を付着させる技術は既に確立されている。

プローブ13は、抗体、酵素などの蛋白質またはデオキシリボ核酸(DNA)などの生理活性物質からなる。例えばDNA検出の場合、所定の塩基配列を有する一本鎖DNAをプローブとして用い、このプローブの塩基配列と相補的な塩基配列を有する物質が試料に存在するか否かをこの生理活性物質検出装置1を用いて検出することができる。すなわち、試料を生理活性物質検出装置1のセル4に送入してハイブリダイズ反応を生じさせ、試料にプローブの塩基配列と相補的な塩基配列を有する物質が含まれているか否かを検出することができる。すなわち、本実施形態の生理活性物質検出装置1はDNAチップとして利用可能である。

図2に示すように、下部支持体7及び上部支持体2にはそれぞれ半径aよりも大きなコイル半径a0の円筒状のコイル14及びコイル15が設けられている。コイル14,15の中心軸は同一軸であるため、これらコイル14及び15に電流が流れると、磁気ビーズ固定化回路系6の上方から下方に向けてほぼ垂直に磁界H2が発生する。この磁界H2は、磁気ビーズ固定化回路系6に磁気ビーズ12を引き寄せる磁界である。また、送液部3a及び送液部3bは、下部支持体7及び上部支持体2間に設けられる。この送液部3a及び3bの一方から試料を送入し、かつ他方から試料を送出することにより、試料を交換することができる。

この生理活性物質検出装置1は、周知のDNAチップなどと同様にDNA断片のハイブリダイズを行うことができる。すなわち、ハイブリダイズの検出は、蛍光検出方式、電流検出方式あるいはそれら以外の方法で行われる。蛍光検出方式の場合は、検出時に下部支持体7から上部支持体2が取り外される。そして、磁気ビーズ固定化回路系6に対してレーザを照射し、蛍光が測定される。

図3は磁気ビーズ固定化回路系6の詳細な構成を示す断面図である。図3には、3つの固定化ユニットU(i,j−1)、U(i,j)、U(i,j+1)が示されている。この磁気ビーズ固定化回路系6は、例えば半導体などにより構成される固定化回路基板20により実現される。この固定化回路基板20のほぼ平坦な表面には各固定化ユニットUに対応して凹状の磁気ビーズ固定部位21が設けられている。この凹状の磁気ビーズ固定部位21は、例えば半導体基板のエッチングにより形成される。このように、固定部位を凹状にすることにより、コイル11による局所磁界H1が極めて強くなる領域を制限することができ、一つの磁気ビーズ固定部位21に所望の数以上の磁気ビーズ13が付着するのを防止することが出来る。

この磁気ビーズ固定部位21の下部にはコイル11が配置されている。このコイル11は、磁気ビーズを磁気ビーズ固定部位21のいずれかに選択的に固定化させるのに用いられる。このコイル11は、例えば固定化回路基板20内に形成されている。コイル11は固定化回路基板20内に形成されていなくても、例えば固定化回路基板20の下部領域に別途形成されていてもよい。コイル11として、カーボンマイクロコイル(Chihiro Kuzuya, Seiji Motojima, Masanobu Kohda and Yukio Hishikawa “Preparation of carbon microcoils with the application of an electromagnetic fields and reaction scheme.” Material Technology, Vol.20, No.1, pp.3-9 (2002))やカーボンナノコイルのような、導電性を有する分子により形成されたコイルを採用してもよい。

このコイル11により、磁気ビーズ固定部位21の近傍に局所的な磁界H1を発生させることができる。この磁界H1は、磁気ビーズ固定部位21に磁気ビーズ12を引き寄せる磁界であり、固定化回路基板20表面に対してほぼ垂直に、かつ固定化回路基板20表面から下面に向けて形成される。コイル11の一端は、各コイル11毎に設けられた電流オンオフ制御用のスイッチSWを介して各コイル11に共通の配線22に接続されている。コイル11の他端は各コイル11に共通の配線23に接続されている。磁気ビーズ固定化の際には、配線22には電圧V1が印加され、配線23には電圧V2(V1>V2)が印加される。

スイッチSWは、各コイル11毎に設けられている。行列状に配列されたコイル11(固定化ユニットU)の行列の行番号をi、列番号をjとすると、固定化ユニットU(i,j)のコイル11には、端子A(i,j)及び端子B(i,j)を有するスイッチSW(i,j)が接続されている。このように、各固定化ユニットU(i,j)に対応してスイッチSWが接続されている。例えばスイッチSW(i,j)を閉じた場合、そのスイッチSW(i,j)が接続された一つのコイル11にのみ電流を流すことができる。従って、その電流が流れるコイル11の上部の磁気ビーズ固定部位21のみに局所的な磁界H1が発生する。他のスイッチSW(i,j−1)やスイッチSW(i,j+1)には電流が流れず、これらのスイッチSW(i,j−1)やスイッチSW(i,j+1)に接続されたコイル11は磁界を発生させない。

このスイッチSWを含めたスイッチ部回路系の詳細な構成を図4に示す。図4に示すように、ロウ選択信号により駆動されるロウデコーダ41にはロウ線i(i=1,2,…)が接続されている。また、カラム選択信号により駆動されるカラムデコーダ42にはカラム線j(j=1,2,…)が接続されている。ロウ線iとカラム線jの交差する領域が固定化ユニットU(i,j)に対応している。この交差領域には、スイッチSW(i,j)が設けられている。具体的には、端子A(i,j)としてMOSトランジスタ43が設けられている。また、端子B(i,j)としてMOSトランジスタ44が設けられている。MOSトランジスタ43のゲートはロウ線iに接続され、MOSトランジスタ43のソース及びドレインの一端はMOSトランジスタ44のソース及びドレインの一端に接続されている。MOSトランジスタ43のソース及びドレインの他端はコイル11に接続される。

なお、コイル11の形状は、図3に示すような円筒状である必要は無い。図9は渦巻き状コイル31の上面図である。図9に示すように、ある平面にコイルが渦巻き状に配置されている。従って、この渦巻き状コイル31の磁界H3は、複数の環電流の合成磁界として表現される。図7から示唆されるように、この環電流の合成磁界H3は、遠方まで到達する磁界で、かつ中心O近傍で十分な強さをもった磁界となる。従って、コイル11としてこのような渦巻き状の形状を採用することにより磁界が遠方まで及ぶ場合には、コイル14やコイル15の一方あるいは双方を無くしてもよい。
