【従来の技術】
近年、高齢者、特に閉経後の女性において、骨粗鬆症の発生が問題となっている。その患者数は、高齢化に伴い、既に約1000万人を越えたとも言われ、その発症により、骨折あるいは寝たきりへとつながるため、重大な社会問題のひとつとなっている。
このような骨粗鬆症に関して、現在、治療薬としては、カルシウム製剤、ビタミンD3、ビタミンK2、エストロゲンなどの種々の薬剤が用いられている。しかし、その治療効果は患者の体質によって、それぞれ異なり、これらの薬剤をどのように使い分けるかについては、現在のところ、正確な検討はほとんどされていない。

本発明者らは、上記目的を達成するために種々鋭意検討を重ねた結果、ヒト由来の試料から得たゲノムDNAを、ApoE遺伝子の遺伝子型について分析することにより、得られた遺伝子型に基づいて、ビタミンK2に対して、いかなる感受性をもつヒト由来の検体であるかを予測可能であることを見いだした。すなわち、本発明者らは、ApoE遺伝子の遺伝子多型がE3/4型である場合も、ビタミンK2に対する感受性が高い傾向を示すことを見い出した。このことより、これまで発表されたApoE遺伝子の4型(E2/4、E3/4、E4/4型)を有する場合が、ビタミンK2感受性が低いのではなく、ApoE遺伝子の3型遺伝子を有さない場合(E2/2、E2/4、E4/4型)が、ビタミンK2感受性が低いのであり、ApoE遺伝子の3型(E2/3、E3/3、E3/4型)遺伝子を有する場合が、ビタミンK2感受性が高く、さらに、3型遺伝子多型のホモ・タイプであるE3/3型の場合に、最も感受性が高くなる傾向を示すことを見いだし、本発明に到達した。

また、PCR後に各種のDNA塩基解析装置(シークエンサー)を用いて、増幅DNAの塩基配列を解析し、その結果よりApoE遺伝子の遺伝子多型を分析することもできる。
