Description
【発明の属する技術分野】
この発明は、酸素添加反応の放射検出方法に関するものである。さらに詳しくは、粗物質系においても微量代謝産物の検出を容易とし、酸素添加反応の機序の解明や生化学的応用等を可能とする新しい放射検出方法に関するものである。

【従来の技術とその課題】
酸素は、生体にとって不可欠の分子であるが、最近になり老化のメカニズムの一つとして、酸化的ストレスの結果がもたらす多くの生化学的変化がクローズアップされ、酸素の生体分子への関与が、大きな問題となってきている。従来、酸素から見た代謝を語る場合、細胞呼吸によりエネルギー(ATP)を生じること、また、酸素添加酵素の働きによりステロイド、プロスタグランジン、カテコールアミン、セロトニン等の多くの生理活性物質の生合成に重要なこと、酸素から生じるラジカルのこと、等が語られてきたが、酸素分子そのものから、これらの代謝を見た研究は、殆ど皆無であった。これは、後にも述べるように、酸素に使い易い放射性同位元素がなかったことによる。

一方、酵素は、生化学的反応に重要な役割を果しているが、その反応過程の解析はそれほど容易でない。たとえば酵素による反応として酸素添加反応が知られており、細胞や生体組織における酸素を用いての反応としてその反応過程が注目されているが、現実的にはその解析手段はほとんど現実的なものとなっていない。これは反応の過程で生じる微量の不安定代謝物の検出が極めて難しいことにその理由がある。

もちろん、これまでにも、酸素添加酵素反応についてその反応過程の検討が行われてきている。酸素添加酵素反応の証明のために、安定同位体である18Oや17Oを用いる方法である(たとえばO.Hayaishi、1957)。
しかしながら、これまでに知られている安定同位体18Oや17Oを用いる方法は、精製酵素に適用するのにはよいが、細胞や組織などの複合系には適用しがたいという問題がある。つまり、より複雑な系において、微量の不安定物質を検知することができないのである。このことは、従来においては酸素原子に利用しやすい放射性同位元素を用いる方法がなかったからである。

このような事情は、酵素反応に限られることなく、酸素が関与する生合成、あるいは一般の化学合成等の反応において、たとえば、金属やその錯体、酸化物等を触媒として用いる場合にもその反応機序の解明が難しく、これら反応技術の発展を大きく制約していたことにも見てとれる。
そこで、この発明は、以上のような従来技術の限界を克服し、より複雑な系においても、酵素反応における微量不安定物質の検出、触媒反応における微量中間体化合物の検出をも可能とする新しい酸素添加反応の検出方法を提供することを目的としている。

【課題を解決するための手段】
この発明は、上記の課題を解決するものとして、メタピロカテカーゼを用いる酸素添加酵素反応において、ポジトロン核種15Oを添加酸素としてピロカテコールに導入することを特徴とする酸素添加酵素反応の放射検出方法を提供する。
そしてまた、この発明は、上記の方法において、安定同位体の17Oおよび/または18Oを併用することや、14N(d,n)15O反応により作製した15Oを酸素−窒素ガスにより導入すること等をその一つの態様として提供する。

【発明の実施の形態】
この発明は、上記の構成によって、様々な基質を対象とする酸素添加反応において、これまでは検出が困難であった微量の不安定代謝物質や反応中間体等の検出を可能とする。このため、反応機序の解明をはじめ、生物細胞やその組織における作用の検討と、様々な医学、生化学的応用、さらには化学合成への応用を可能とする。

15Oは、物理学的半減期は2.07分と比較的短いが、比放射能が高い(理論的には3.36×1021Bq/mol)ことから、微量の不安定代謝物の探索には非常に有利である。
この15Oによる標識では、ポジトロン核種であり、たとえばポジトロンエミッショントモグラフィーに用いられているものを利用することができ、酸素添加反応においてこれを導入すればよい。

酵素反応での反応基質、酵素の種類、触媒反応における原料物質や触媒の種類、さらには生成物の同定手段について特に限定はなく、様々な態様が可能であるが、特にメタピロカテカーゼを用いた酸素添加酵素反応において、基質としてピロカテコールを用いることを対象としている。
また、15Oとともに、17Oや18Oの安定同位体を併用する場合、つまり2重標識法による場合には、15Oで未知代謝物(酸素添加物)等の存在をモニターし、その部分をさらに17Oや18Oの存在によってゆっくりと時間をかけて構造決定できる等の利点がある。このような併用も様々な態様として可能である。

以下、実施例を示し、さらに詳しくこの発明の実施の形態を説明する。

酸素添加酵素反応のモデルとして、
基質:ピロカテコール
酵素:メタピロカテカーゼ
を用いた。

図1は、このモデル反応のための実験系を例示したものである。ベビーサイクロトロンにより定常的に14N(d,n)15O反応によって15Oを作製し、酵素−窒素混合ガスのフローにより1%O2 /99%N2 として250ml/minの速度により実験に供給した。
この反応において、基質のカテコールは、次式のとおり、経時的に放射活性のある単一の産物に変換されたことが確認された。

なお、産物はHPLCおよびLC−MSによって同定した。
図2は、15O2 の反応への関与を示した反応時間と放射能との関係を示したものである。また、図3はHPLCの同定結果を示したものである。なお、HPLCの同定の条件は次のとおりとした。

また、図4はLC−MSのスペクトルを示したもので、最下段は、基質カテコールを示している。また、最上段の最初の放射ピークは、酵素に結合した15Oである。カテコールおよび産物のスペクトルについては、いずれも50〜1000Daでスキャンニングしている。LC−MSの同定条件は次のとおりとした。

もちろん、この発明は以上の例によって何ら限定されるものではない。様々な反応において15Oによる標識化が有効である。

【発明の効果】
以上詳しく説明したとおり、この発明によって、酸素が介在する酵素反応や触媒反応等の化学反応について、酸素添加活性の評価、反応により生成する中間体や産物としての微量の不安定物質の検出、これによる反応機序の解明、さらには18O、17Oという安定同位体による検討との併用等によって、合成化学、生化学、そして細胞、標識における代謝過程、その医学への応用等が可能とされる。

Claims (3)
- メタピロカテカーゼを用いる酸素添加酵素反応において、ポジトロン核種15Oを添加酸素としてピロカテコールに導入することを特徴とする酸素添加酵素反応の放射検出方法。
- 安定同位体の17Oおよび/または18Oを併用する請求項1の放射検出方法。
- 14N(d,n)15O反応により作製した15Oを酸素−窒素混合ガスにより導入する請求項1ないし2のいずれかの放射検出方法。
