この出願の発明は、ヘテロ−キラル希土類触媒と、これを用いた不斉環状化合物の製造方法に関するものである。

【従来の技術と発明の課題】
従来より、ニトロン類とアルケン類との1,3−付加環化反応は、γ−アミノアルコールへと導くことのできるイソオキサゾリジン類の合成方法として知られている。
しかしながら、γ−アミノアルコール類についてはその光学活性体が、生理活性を持つアルカロイド類、アミノ酸、β−ラクタム、アミノ糖類等の重要な一部を構成し、その合成のためにも注目される物質であるが、ニトロン類とアルケン類との1,3−付加反応をエナンチオ選択的に触媒反応として行うことは、いまだにほとんど検討されてきていない。また、この出願の発明者にとっても、ディールズ・アルダー反応やアザ ディールズ・アルダー反応については触媒を用いての付加反応で不斉合成する方法として、キラルな希土類触媒による不斉付加反応法をこれまでに提供してきているが、ニトロン類の1,3−付加による不斉環化については未検討であった。

そこで、この出願の発明は、以上のとおりの従来技術の限界を超えて、ニトロン類の1,3−付加環化を、触媒反応として、高いジアステレオ選択性と高いエナンチオ選択性のもとに行うことのできる新しい触媒系と、これを用いたイソオキサゾリジン類からなる不斉環状化合物の製造方法を提供することを課題としている。

【発明の実施の形態】
この出願の発明は、上記のとおりの特徴を持つものであるが、ヘテロ−キラル希土類触媒についてさらに説明すると、この触媒は、基本的には、次の成分;
<1>希土類(III) 化合物
<2>フェノール性水酸基を持つ光学活性芳香族化合物
<3>光学活性3級アミン類
とにより構成されている。そして、この発明のヘテロ−キラル触媒では、<2>フェノール性水酸基を持つ光学活性芳香族化合物と<3>光学活性3級アミン類は、相互に異なるキラルティを持ち、いずれか一方がR体であって、他方がS体であることを要件としている。

希土類(III) 化合物は、Sc、Y、ランタニドのうちの各種の希土類元素の化合物であって、たとえばYb、Sc、Sm、Y、Nd、Eu、Er、La等の希土類元素の有機酸エステル塩、アルコラート、有機金属化合物のうちの各種のもが考慮される。
なかでも、その具体例としては、Yb(III) トリフロート(Yb(OTf)3 )、すなわちYb(III) −トリフルオロメタンスルホネートがその好ましいものの一つとして挙げられる。

フェノール性水酸基を持つ光学活性芳香族化合物については、その代表的な例としては、光学活性なビナフトール類がある。このビナフトール類は、ナフタレン環への置換基として、ハロゲン原子、たとえば臭素原子、塩素原子;アルキル基等の炭化水素基、アルコキシ基、ニトロ基、シアノ基等の、この発明の触媒能を損うことがなく、かえって向上させることもできる置換基を適宜に有していてもよい。

もちろん、その種類については、これらのものに限定されることはない。
前記の成分<1><2><3>により構成されるこの発明のヘテロ−キラル希土類触媒については、構造として、希土類(III) 化合物の希土類原子に、フェノール性水酸基を持つ光学活性芳香族化合物の水酸基酸素原子が配位し、水酸基水素原子が、光学活性3級アミン類の窒素原子と水素結合するものが考慮される。

前記成分<1><2><3>については、触媒の調製では溶媒を用いて、所要割合で混合すればよい。この場合の割合は、一般的には、希土類(III) 化合物:フェノール性水酸基:3級アミンの当量比として、1:0.1〜5:0.1〜10程度を目安とすることができる。溶媒については、ハロゲン化炭化水素、芳香族炭化水素等の各種のものが考慮される。

の構造を持つ不斉イソオキサゾリジン化合物を製造することを特徴としている。この合成反応のためのニトロン類およびアルケン類の符号R1 〜R4 の炭化水素基は、鎖状または環状の、そして飽和または不飽和の、脂肪族、脂環式、もしくは芳香族の各種の炭化水素基であってよい。これらはまた、置換基として、反応を阻害することのないものとして、アルキル基、アルケニル基、アリール基、複素環基、ハロゲン原子、水酸基、アルコキシ基、カルボニル基、アミド基、ニトロ基、シアノ基等の各種のものであってよい。R1 〜R4 が複素環基であるものも同様で、各種のものでよい。

反応は、一般的には、ニトロン類に対して、アルケン類を、モル比として1:0.5〜2程度の割合で使用し、ハロゲン化炭化水素、芳香族炭化水素等の溶媒中において、−20〜30℃程度の温度で行うことができる。触媒の使用量は、ニトロン類に対して、0.01〜50モル%、より好ましくは1〜30モル%とするのが適当である。高いジアステレオ選択性、高いエナンチオ選択性のものに優れた化学収率で、不斉環状化合物である不斉イソオキサゾリジン化合物が合成される。

(実施例2)

実施例1と同様にして、各種の反応基質を用いてニトロン類の不斉1,3−付加環化反応を行った。その結果を表3に示した。
広い範囲の各種反応基質に対して優れた触媒作用が得られていることがわかる。イソオキサゾリジン化合物が優れた化学収率と、高いジアステレオ選択性および高いエナンチオ選択性で得られている。

比較のために、(S)−MPEAを用いたところ、化学収率は80%に、endo/exo比は97/3に、endo体eeは35%にまで低下した。
