【発明の属する技術分野】
本発明は、試料中のステロイド性生体内微量物質に電子親和性原子団を有する化合物を導入し、負イオンAPCI−MS法(負イオン大気圧化学イオン化質量分析法)により生体内微量物質を測定する方法、その測定に供される反応成績体及びその測定のための試薬に関する。

【従来の技術】
ビタミンD3代謝物(9,10−セコステロイド)を含むステロイドホルモンは、生体内において、極微量にて強力な生理作用を表す。これらのステロイドホルモンの生体内レベルを把握することは、臨床診断及び病態解析上極めて重要であるが、これらの生体内濃度は著しく低い上に、構造類似化合物が共存するため、その分析には極めて高い感度及び特異性が要求される。

従来、これら生体内ステロイドホルモンの測定は、主としてLC(Liquid Chromatography:液体クロマトグラフィ)を用いて行われてきたが、実用的に十分な程度の感度が得られない場合が多かった。

近年、LCとAPCI−MS(Atmospheric Pressure Chemical Ionization-based Mass Spectrometry)を組合せた方法、すなわち、LC/APCI−MSが特異性や多用性の面で有用であることが提案された(非特許文献1参照)。
しかし、LC/APCI−MSを用いた方法においても、ステロイドホルモン化合物自体のイオン化効率が低いために、十分な感度を得ることはできなかった。

さらに、LCとESI(Electro Spray Ionization:エレクトロスプレーイオン化)−MSを組合せる方法も行われているが、本法は極性化合物に有効であるが、多くのステロイドホルモンは極性基を数個しか有していないことから、この方法もあまり実用的ではなかった。

最近になって、正イオン検出APCI−MSを用いた測定において、ステロイド化合物に電子親和性原子団を導入した誘導体が高感度で検出できることが示された(非特許文献2−4参照)。一方、負イオンモードのAPCI−MSは、バックグラウンドノイズが低いことが一般的に知られており、負イオンモードAPCI−MSを用いた測定において、ステロイド化合物に電子親和性原子団を導入すれば、更なる高感度化が期待できると考えられる。
しかしながら、これまで、負イオン検出APCI−MSのための電子親和性原子団の導入については、ペンタフルオロベンジル化の例(非特許文献5参照)が知られているのみで、他の誘導体化についての検討はなされていない。

【発明が解決しようとする課題】
本発明の目的は、負イオンAPCI−MSを用いたステロイド性生体内微量物質の測定において、ステロイド性生体内微量物質に電子親和性原子団を有する化合物を導入することにより、ステロイド性生体内微量物質を高感度に測定する方法を提供することである。また、本発明の目的は、負イオンAPCI−MSを用いたステロイド性生体内微量物質の測定における測定感度を高めるための化合物を含むイオン化試薬、及び当該化合物をステロイド性生体内微量物質に導入した化合物を提供することである。さらに、本発明の目的は、負イオンAPCI−MSを用いたステロイド性生体内微量物質の測定のための試薬を提供することである。

【課題を解決するための手段】
本発明者らは、負イオン検出APCI−MSを用いたステロイド性生体内微量物質の測定において、その測定感度を上昇させるべく鋭意検討したところ、驚くべきことに、特定の電子親和性原子団をステロイド性生体内微量物質に導入すると、その測定感度が数倍〜200倍以上も上昇することを見出した。また、この測定感度はこれまでに知られている電子親和性原子団、すなわち、ペンタフルオロベンジル基を導入した場合に比較しても最大で7倍以上高いものであった。本発明者らは、これらの知見をもとにさらに検討を行い、本発明を完成させた。

加えて、本発明によれば、分析対象が、ステロイド性生体内微量物質である、上記イオン化試薬、並びに、LC/MS分析が、負イオンAPCI−MSである、上記イオン化試薬が提供される。

なお、本発明のイオン化試薬は、APCI法のみならず、ESI法及びCF−FAB(連続フロー高速原子衝撃)法等、他のイオン化方法を用いる質量分析にも用いることができる。

本明細書において用いる語「ステロイド性生体内微量物質」とは、ステロイド化合物及びビタミンD3類縁体を意味する。したがって、ステロイド性生体内微量物質としては、例えば、テストステロン、ジヒドロテストステロン、デヒドロエピアンドロステロン、プレグネノロン、エストロン、2−ヒドロキシエストラジオール、4−ヒドロキシエストラジオール、2−ヒドロキシエストロン、4−ヒドロキシエストロン等のステロイド化合物及び24,25−(ジヒドロキシ)ビタミンD3、1,24,25−(トリヒドロキシ)ビタミンD3等のビタミンD3類縁体を挙げることができる。

本発明における前記式(I)の化合物は、商業的に入手可能な原料より、ヒドラジンによる置換反応によって容易に合成することができる。また、本発明における血清試料は、常法により容易に調製することができる。
すなわち、本発明における血清試料は、採血した全血より血清を分取した後、市販のカートリッジ(例えば、OASIS HLB(登録商標) cartridgeやBond ELUT Si(登録商標) cartridge等)を用いた精製により調製できる。

LC/MS(MS)は、jasco pu-980クロマトグラフに接続したliquid chromatograph-ion trap-mass spectrometer ThermoQuest LCQを用い、APCI条件にて測定した。カラムはJ'sphere ODS H-80(YMC製。4μm,150×4.6mm i.d.。)を用い、Flow Rateは1ml/min.、温度は40℃とした。MS/MS分析においては、ヘリウムをCollision Gasとして用いた。Capillary Voltage及びTube Lens Offset Voltageは、ヒドラジン誘導体についてはそれぞれ−10V及び−15Vとし、ボロン酸誘導体についてはそれぞれ−3V及び−15Vとした。また、両誘導体とも負イオン且つZoomScanモードにて測定した。その他のMS測定条件及び各被検物質の移動相については、表1〜表3に記載した。
