【産業上の利用分野】
本発明は、化学分析スライドを用いて血液、尿等の体液に存在する各種成分を定量分析するための化学分析に利用される化学分析スライド用インキユベータに関するものである。

【従来の技術】
体液中に存在する各種成分、例えばグルコース、ビリルビン、尿素、窒素等を定量分析するには、血液等の液体試料を注入した試験管に、分析する成分に応じて調整された液体試薬を投入して行なう方法があった。しかしながら、このような方法では、液体試料である人血等の貴重な体液が多量に必要であること、取扱いが面倒で簡易かつ迅速な測定が困難であること、そして多数の被検液を連続的に測定することが困難であること等の問題があった。このような問題のため、近年、乾式多層フィルム化技術を利用し、分析に必要なすべての試薬を含有させた化学分析スライドを用いて臨床化学検査を行う、乾式法による分析方法が行なわれるようになった。

この化学分析スライドは、血液等の液体試料を点着させるための点着用開口を有しており、このためインキユベータを用いて一定温度で一定時間加熱するインキユベート工程において、化学分析スライドに点着された液体試料中の水分が蒸発し、反応の進行を途中で抑えてしまい反応が完結しないことがあった。そこで、インキユベート中に液体試料から水分が蒸発しないようにしたインキユベータが種々提案されている。

ところで、近年、高齢化社会への急速な移行、高度治療の発達などを背景とした医療費の高騰への対応として、在宅ケアが提唱され、今後の医療制度の1つの核となるものと期待されている。

この在宅ケアの一つとして化学分析スライドを用いた方法があり、この方法は、患者が各家庭において化学分析スライドに血液を点着した後インキユベートし、そしてこのインキユベートされた化学分析スライドを病院に送り、病院が化学分析スライドの分析を行うというものである。

この在宅ケア用の化学分析スライド用インキユベータも、上述した水分の蒸発防止の対策が採られており、例えば、図5に示すような化学分析スライド用インキユベータがあった(実開平3−126499号公報)。

図5において、40は化学分析スライド用インキユベータを示している。この化学分析スライド用インキユベータ40は、ケース41と、このケース41の中に上部が突出して設置されたヒートブロック42と、このヒートブロック42に載置されるカバー43とから成っている。

ヒートブロック42は、上面に上下2段の凹部が穿設され、上段がカバー43と平面視にて略一致する形状を有するカバー支持部44で、下段が化学分析スライドと平面視にて略一致する形状でかつほぼ同じ厚みを有するスライド収納部45である。

【発明が解決しようとする課題】
上記のインキュベータはインキュベート中の水分の蒸発を防止してかなりの精度で分析を行なえるものであったが、化学分析スライドの出入等の操作性に問題があり、これがインキュベート時間のバラツキや操作者が多層分析要素面に手で触れてしまう等を生じ、分析精度及び衛生面の両方で問題となっていた。

本発明の目的は、構造が簡単で安価でありながら、操作が容易で、一定のインキュベート条件を確保でき、かつ多層分析要素面に手を触れることなく確実に収納用ケースに入れられる化学分析スライド用インキュベータを提供することにある。

【課題を解決するための手段】
本発明者らは上記課題を解決すべく鋭意検討の結果、液体試料が供給された化学分析スライドを置く凹所と、該凹所の奥方に連設されたインキュベーション室と、化学分析スライドの前縁を押して、インキュベーション室に押し込む押込部材と、インキュベーション室の上部に装置され化学分析スライドに供給された液体試料の蒸発を防止する蒸発防止蓋と、インキュベーション室に凹所と直角方向に連設された化学分析スライド収納用ケースの設置部と、インキュベーション室から化学分析スライドの側縁を押して化学分析スライド収納用ケースに押し入れる押込部材と、インキュベーション室を加熱する加熱部材と、インキュベーション室の温度を調節する温度調節器からなり、前記蒸発防止蓋がインキュベーション室を閉止する蓋と多層分析要素が露出している化学分析スライドの窓孔を閉止する蓋を有し、両蓋間が上下動自在に連結されている化学分析スライド用インキュベータを案出し、これによって上記目的を達成できることを見出して本発明を完成するに至った。

化学分析スライド収納用ケースは化学分析スライドを収納し、密閉しうるものであればよいが、通常は偏平な箱形でよい。収容する化学分析スライドは1〜5枚程度である。ケース本体は透明なプラスチックが好ましい。ケース本体に取着して密閉する検体は多少弾性を有するものが好ましい。ケース内には乾燥剤を入れておくことが好ましい。乾燥剤は通気性の袋等に入れてケース本体内に入れてもよく、検体を中空にしてそこに入れてもよい。乾燥剤にはシリカゲル、ゼオライト、塩化カルシウム等公知のものを用いることができる。

図1はインキュベータの上板(図示されていない。)を取り去った状態の斜視図であり、インキュベータ本体1の上面中央部に、液体試料が供給された化学分析スライドを置く凹所2とその奥方にインキュベーション室3が連設されている。凹所2の幅と深さはそれぞれ化学分析スライドの全長及び厚さよりやや大きい程度である。インキュベーション室3の幅と深さは凹所と一致している。凹所2とインキュベーション室3の底面は同一平面になっており、両者を合わせた全体の平面形状は全長が化学分析スライドの全幅の2倍より長い長方形になっている。インキュベータ本体1の凹所2の前縁略中央部とインキュベーション室3の左側縁略中央部には外部に連通する浅い角溝4,5が形成され、それぞれ、化学分析スライドの押込部材であるスライド板6,7が摺動自在に遊嵌されている。スライド板6,7の外端縁は押し引き操作を容易にするために上方に折り曲げられている。蒸発防止蓋8はステンレススチール製であり、インキュベーション室2より一回り大きい長方形の板9と閉止コマ10からなっている。閉止コマ10は図1及び図3から理解できるように円盤部とその上面から上方に突出する軸部からなり、円盤部底面外周は浅く巾広に削取されてテーパ面になっている。この底面にはテフロンコーティングが施されている。板9の中央には円孔が設けられ、閉止コマの軸部はこの円孔を遊びをもって貫通して上方に突出している。軸部の上部には周溝が設けられそこにストップリング11が嵌め込まれている。インキュベーション室3の右方にはインキュベーション室3より一回り大きくかつ深い角溝状凹所が外部に連通して形成され、この凹所が化学分析スライド収納用ケースの設置部12になっている。図示されていないインキュベータの上板はインキュベータ本体1の大きさと一致する長方形をしており、中央に2つの長方形を組み合わせた窓穴が設けられている。この窓穴の前部の長方形の位置と大きさは凹所2と一致している。一方、後部の長方形の位置はインキュベーション室3と一致しているが、その大きさは蒸発防止蓋8の板9に略一致しており、従ってインキュベーション室3よりは一回り大きくなっている。その結果、上板とインキュベータ本体1の間には段部が形成され、蒸発防止蓋8はこの窓穴の後部に嵌入されて位置決めされるとともに前記段部によって支持される。このインキュベータ本体1内にはインキュベーション室3を加熱するヒータ13とインキュベーション室3の温度を一定に保つサーモスタット14が取り付けられている。

このインキュベータの使用方法を図2を参照して説明する。まず、電源を入れてインキュベーション室3を恒温に加熱しておくとともに蒸発防止蓋8を装着してインキュベーション室3を閉止しておく。化学分析スライド収納用ケース本体31はその設置部12に挿着しておく。液体試料が点着され、多層分析要素内で展開、滲透の終了した化学分析スライド20を上板の窓穴前部の開口から凹所2に入れ、スライド板6を利用してこれをインキュベーション室3に押し込む。そうすると、化学分析スライド20は進行方向の先端になる後縁が蒸発防止蓋8のコマ10のテーパ面に当接してこれを押し上げながらさらに進行し、この後縁がインキュベーション室3の後面に突当って停止する。コマ10は化学分析スライドの円孔23上方に位置して自重で降下し、テーパ面が円孔23の周縁部に当接してこれを閉止する。そこで、操作者が時計等で測って一定時間インキュベーションを行ない、次いで、スライド板7を押し込んで化学分析スライド20をインキュベーション室3から出してケース本体31に入れる。ケース本体31を設置部12から引き抜き、乾燥剤33を入れて検体32を装着してケースを密閉する。この状態で病院や検査機関等に郵送する。

1)検体の調整
健常者の静脈血20mlを解糖阻止剤(NaF)の所定量を含む真空採血管(テルモ社製)にて採血した。

富士ドライケムGLU−Wスライドを本発明実施例のインキュベータにセットし検体No.1の全血8μlを点着し、直ちに加温ステージに移動させ6分間37℃を重さ2gの板状(30mm×55mm×2mm)のゼオライトと共にプラスチックで形成した化学分析スライド収納用ケース中に封入し、室温に放置してスライドを脱水乾燥させた。同様の操作No.1、2、3それぞれについて3回づつ繰り返した。

本発明のインキュベータにより、在宅患者等の操作者が血液等の液体試料を化学分析スライドに点着し、これをインキュベータの凹所に設置後に一切手を触れることなくインキュベートし、密閉容器に収納して検査機関等へ送付することができる。本発明のインキュベータは構造が簡単で安価に製造でき、操作も容易であることから在宅ケア等に容易に普及させることができるという効果を奏する。
