発明の説明
本発明は、ある種の新規ナフトピラン化合物に関する。より詳しくは、本発明は、新規フォトクロミックインデノ縮合ナフトピラン化合物、ならびにそのような新規ナフトピラン化合物を含有する組成物および物品に関する。日光の紫外線照射または水銀ランプの光のような、紫外線を含有する光照射に暴露されたときに、多くのフォトクロミック化合物は可逆的な色の変化を示す。紫外線照射が中止されると、そのようなフォトクロミック化合物は、その元の色または無色状態に戻る。
種々のフォトクロミック化合物が合成され、日光に誘発される可逆的な色の変化または暗色化が所望される用途への使用が提案されている。米国特許第3567605号(Becker)は、ある種のベンゾピランおよびナフトピランを包含する一連のピラン誘導体を開示している。これらの化合物は、クロメンの誘導体として記載され、約−30℃未満の温度における紫外線照射で、例えば無色から黄色−オレンジ色に、変色することが開示されている。その化合物を可視光で照射することによって、または温度を約0℃よりも高い温度に上げたときに、着色状態から無色状態に戻ることが記載されている。
米国特許第5066818号は、種々の3,3−ジアリール−3H−ナフト[2,1−b]ピランが、眼または他の用途に対して、望ましいフォトクロミック特性、即ち高着色性および許容できる退色、を有すると記載している。その米国特許第5066818号において、活性後の退色に許容できない長い期間を必要とする異性2,2−ジアリール−2H−ナフト[1,2−b]ピランも比較例として開示されている。
米国特許第3627690号は、塩基または弱から中の強度の酸のいずれかを少量含有するフォトクロミック2,2−ジ置換−2H−ナフト[1,2−b]ピラン組成物を開示している。ナフトピラン組成物への酸または塩基のいずれかの添加は、着色ナフトピランの退色速度を高め、それによって、サングラスのような眼を保護する用途に有益なものとなると記載している。同特許は、前記添加剤を使用しない2H−ナフト−[1,2−b]ピランの退色速度が、完全に元の色に戻るまでに数時間〜多日数であることを、さらに開示している。米国特許第4818096号は、オルトまたはパラ位に窒素含有置換基を有するフェニル基を、ピラン環の酸素に対してα位に有している紫色/青色着色フォトクロミックベンゾまたはナフトピランを開示している。
本発明は、2、1位がナフトピランのナフト部分の辺fに縮合されている置換または非置換インデノ基、およびピラン環の3位に、ある種の置換基を有する、新規置換ナフトピラン化合物に関する。これらの化合物は、意外にも、フォトクロミック化合物の活性(着色)形態の最大吸収、即ちラムダマックス(lambda max)(Vis)、が発生する可視スペクトル中の波長に対して深色団シフト(batho chromic hift)を示し、それによって、オレンジ色〜青色/灰色の範囲の活性着色を生じることが見い出された。さらに、これらの化合物は、紫外線中における高いモル吸収率(またはモル吸光係数)、酸または塩基不添加での許容される退色速度、高活性強度、および高着色率を示した。
発明の詳細な説明
近年、フォトクロミックプラスチック材料、特に光学的用途のためのプラスチック材料が大きな関心を集めている。特に、フォトクロミック眼用プラスチックレンズがガラスレンズに対して重量的に有益であるために、それらの研究がなされている。さらに、車および飛行機のような乗り物のためのフォトクロミック透明材料が、そのような透明材料によって得られる潜在的安全性によって、関心を集めている。
本発明によって、オレンジ色〜青色/灰色の範囲の活性着色、許容される退色速度、高活性強度、および高着色率を有するある種の新規インデノ[2,1−f]ナフト[1,2−b]ピランを製造し得ることが見い出された。これらの化合物は、ピラン環の3位に、ある種の置換基を有するインデノ縮合[1,2−b]ナフトピランとして記載することができる。ある種の置換基が、化合物の炭素原子の番号5、6、7、8、9、10、11、12または13において存在することもできる。これらの化合物は下記構造式によって示すことができ、式中、a〜nの文字はナフトピラン環の辺を表し、数字はナフトピランの環原子の番号を表す。

反応式Aにおいて、構造式IIIおよびIVによって示される化合物が、二硫化炭素または塩化メチレンのような溶媒中に溶解され、塩化アルミニウムまたは四塩化錫のようなルイス酸の存在下に反応して、構造式V(反応式BのVA、または反応式CのVB)で示される対応する置換ベンゾフェノンを生成する。RおよびR′は、構造式Iに関して前記に記載した可能な置換基を表す。
反応式Bにおいて、構造式Vに示すような、構造式VA[式中、BおよびB′は置換または非置換フェニル以外の基を表してもよい]によって示される置換または非置換ケトンが、無水テトラヒドロフラン(THF)のような適切な溶媒中で、ナトリウムアセチリドと反応して、構造式VIで示される対応するプロパルギルアルコールを生成する。置換または非置換フェニル以外のBまたはB′基を有するプロパルギルアルコールは、商業的入手可能なケトンから、またはハロゲン化アシルと置換または非置換ベンゼン、ナフタレンまたはヘテロ芳香族化合物との反応によって製造されるケトンから、製造することができる。構造式IICで示されるBまたはB′基を有するプロパルギルアルコールは、米国特許第5274132号、第2列、第40〜68行に記載されている方法によって製造することができる。
反応式Cにおいて、構造式VBで示される置換または非置換ベンゾフェノンが、構造式VIIで示される琥珀酸ジメチルのような琥珀酸のエステルと反応する。カリウムt−ブトキシドまたはハロゲン化ナトリウムを塩基として含有する溶媒、例えばトルエンに、反応物を添加することによって、構造式VIIIで示されるストッブ縮合半エステルを生成する。ベンゾフェノン上のR3およびR4が同じでない場合、即ち、構造的に対照でない場合、シスおよびトランス半エステルの混合物が形成され、明瞭な異性体を単離するためにさらに精製が必要となる。半エステル(VIII)が、無水酢酸の存在下にシクロ脱水されて、構造式IXで示されるアセトキシナフタレンを生成する。この生成物を、水酸化ナトリウムのような塩基の水性アルコール溶液中で加水分解し、続いて塩酸(H+)で処理して、構造式Xで示されるカルボキシナフトールを生成する。
反応式Dにおいて、構造式Xで示されるカルボキシナフトールが、燐酸のような酸の存在下に、例えば約160℃〜約220℃に加熱することによって環化されて、構造式XIで示されるヒドロキシ置換ベンズ縮合フルオレノンが生成される。F.C.Baddarらによる、J.Chem.Soc.,p.986.1958の記事を参照。構造式Xlで示される化合物を合成する別の方法が、C.F.Koelschによって、Journal of Organic Chemistry,第26巻,p2590,1961に記載されている。
酸、例えばドデシルベンゼンスルホン酸(DBSA)の触媒量の存在下における、構造式XIで示される化合物と、構造式VIで示されるプロパルギルアルコールとの結合の結果として、構造式IAで示されるインデノ縮合ナフトピランが生成される。ウォルフ−キシュナー還元によって構造式XIで示される化合物を還元し、その結果として、構造式XIAで示される化合物が生成される。構造式XIAで示される化合物と、構造式VIで示されるプロパルギルアルコールとの結合の結果として、構造式IBで示されるインデノ縮合ナフトピランが生成される。構造式IB中の水素の、アルキル基、例えばR1およびR2での置換が、化合物IBとトリアルキルアルミニウムとの反応によって行われて、構造式ICで示される化合物を生成する。
R1およびR2がアルキルの代わりにアルコキシであるかまたは、R1およびR2が一緒になって、酸素原子2個およびスピロ炭素原子を含む炭素原子を3〜6個含むスピロ複素環式基を形成する、構造式ICで示される化合物に類似した化合物を、触媒量の酸の存在下に、構造式IAで示される化合物とアルコールまたはジオールとをそれぞれ反応させることによって製造することができる。
反応式Eにおいて、異なるR1およびR2置換基を有する、構造式Iで示される化合物を製造する別の方法が記載されている。構造式IAで示される化合物で出発し、水酸化リチウムアルミニウム(LAH)で還元して、構造式IDで示される化合物を生成する。カルボニル基を還元する他の方法が、文献The Chemistry of the Carbonyl Gr oup, Chapter 11、Saul Patai,Editor,1966,Intersoience Publishersに開示されている。
構造式IDで示される化合物と、可能な置換基R′を有する塩化アシルとの反応の結果、構造式IEで示される化合物が生成される。構造式IAで示される化合物上に異なるR1およびR2置換基を組み込む別の方法は、化合物(lA)を、可能な置換基Rを有するグリニャールまたはリチウム試薬と反応させて、構造式IFで示される化合物を生成する方法である。それに続いて、塩酸のような酸の存在下に、構造式IFで示される化合物と、可能な置換基R′を有するアルコールとを反応させ、その結果として構造式IGで示される化合物が生成される。
光学レンズ、例えば、視力修正眼用レンズおよび平面レンズ、フェースシールド(face shields)、ゴーグル、まびさし(visors)、カメラレンズ、窓、自動車のフロントガラス、航空機および自動車の透明物、例えば、Tルーフ、サイドライトおよびバックライト、プラスチックフィルムおよびシート、織物および塗料、例えば、ペイントのような塗料、ならびに、真正の認証または証明が所望される銀行券、パスポートおよび自動車免許証のような保証書類上の証明マークのような、有機フォトクロミック物質が使用される用途に、構造式Iで示される化合物を使用することができる。構造式Iで示されるナフトピランは、無色からオレンジ色〜青色/灰色の範囲の着色という色の変化を示す。
本発明の範囲に含まれるナフトピラン化合物の例は下記のものである:

本発明の有機フォトクロミックナフトピランは、単独で使用することもでき、本発明の他のナフトピランとの組み合わせで使用することもでき、または、1種またはそれ以上の他の補足的有機フォトクロミック物質、即ち、約400〜700ナノメートルの範囲内の少なくとも1つの活性吸収極大を有する有機フォトクロミック化合物、またはそれを含有する物質との組み合わせで使用することもでき、フォトクロミック物品を製造するのに使用され、活性化したときに適切な色に着色するポリマー有機ホスト材料中に、組み込む、例えば溶解または分散させることもできる。
作業実施例を除いて、または特に記載のない場合、本明細書において使用されている、波長、成分の量、または反応条件を表す数字は、全ての場合において、「約」という語によって修飾されるものであると理解すべきである。
補足的有機フォトクロミック化合物の例は、他のナフトピラン、ベンゾピラン、フェナントロピラン、スピロ(ベンズインドリン)ナフトピラン、スピロ(インドリン)ベンゾピラン、スピロ(インドリン)ナフトピラン、スピロ(インドリン)キノピラン、スピロ(インドリン)ピラン、スピロ(インドリン)ナフトキサジン、スピロ(インドリン)ピリドペンズオキサジン、スピロ(ベンズインドリン)ピリトベンズオキサジン、スピロ(ベンズインドリン)ナフトオキサジン、スピロ(インドリン)ベンズオキサジン、およびそのようなフォトクロミック化合物の混合物を包含する。
本明細書中に記載されている各フォトクロミック物質は、フォトクロミック化合物または化合物の混合物が被覆または組み込まれている有機ホスト材料が、結果として生ずる所望の色、例えば、未フィルターの日光で活性化されたときに実質的中間色(neutral colour)を示す、即ち、活性化されたフォトクロミック化合物の色が可能な限り中問色に近い色を示すような量(または比率)で使用される。
中間灰色は、400〜700ナノメートルの可視範囲において、比較的等しい吸収を有するスペクトルを示す。中間褐色は、400〜550ナノメートルの範囲における吸収が、550〜700ナノメートルの範囲におけるよりも適度に大きいスペクトルを示す。色を記載する別の方法は色度座標によるものであり、これは、輝度要素に加えた色の質、即ち色度、を記載するものである。CIEシステムにおいては、三刺激値のそれらの合計に対する比、例えばx=X/(X+Y+Z)およびy=Y/(X+Y+Z)を求めることによって、色度座標が得られる。ClEシステムで記載される色を色度グラフにプロットすることができ、通常、色度座標xおよびyのプロットである。F.W.Billmeyer,Jr.,およびMax SaltzmanによるPrinciples of Colo r Technology,Second Edition,John Wiley and Sons,N.Y.(1981)のp.47−52を参照。本明細書で使用されている中間色に近い色とは、色の「x」および「y」の色度座標値が下記範囲内(D65発光体)である色を意味する:太陽輻射(気団1または2)への暴露により40%の視感透過に活性化後、x=0.260〜0.400、y=0.280〜0.400。
ホスト材料に被覆または組み込まれる、フォトクロミック物質またはそれを含有する組成物の量は、活性時に裸眼で識別できるフォトクロミック効果を生じるのに充分な量が使用されることを条件として、限定されない。一般に、そのような量を、フォトクロミック量として記載することができる。特定の使用量は、フォトクロミック物質の照射時に所望される色の強度、およびそのフォトクロミック物質を組み込む、または被覆するのに使用される方法に依存する場合が多い。一般に、より多くのフォトクロミック物質が適用または組み込まれるほど、ある限界までの色の強度がより高くなる。
使用される前記フォトクロミック化合物の相対量は、そのような化合物の活性種の色の相対強度、および所望される最終の色によって、部分的に変化および依存する。一般に、フォトクロミック光学ホスト材料に組み込みまたは被覆される全フォトクロミック物質の量は、フォトクロミック物質が組み込みまたは被覆される表面1平方センチメートルにつき約0.05〜約1.0、例えば、0.1〜約0.45ミリグラムである。
本発明のフォトクロミック物質を、文献に記載されている種々の方法によって、ポリマー有機ホスト材料のようなホスト材料に、被覆または組み込むことができる。そのような方法は、ホスト材料中にフォトクロミック物質を溶解または分散させることを包含し、例えば、重合の前に、モノマーホスト材料にフォトクロミック物質を加えることによって、それを適所に注入すること;フォトクロミック物質の熱溶液にホスト材料を浸漬することによって、または熱移動によって、ホスト材料中にフォトクロミック物質を吸収させること(imbibition);隣接するホスト材料の層の間に、フォトクロミック物質を、分離層として、例えばポリマーフイルムの一部として、配置すること;およびホスト材料の表面上に配置される被膜の一部として、フォトクロミック物質を被覆すること;を包含する。「吸収(imbibition)」または「吸収する(imbibe)」という語は、フォトクロミック物質のみのホスト材料への透過、フォトクロミック物質の、多孔質ポリマーへの溶媒補助移動、蒸気層移動、および他のそのような移動メカニズムを意味し、包含する。
相溶性(化学的および色的に)の色材(tint)、即ち染料を、ホスト材料に被覆して、医学的理由またはファッション的理由に対して、より審美的な結果を生じさせることができる。選択される特定の染料は、前記の要求および達成すべき結果に依存して変化する。1つの態様においては、活性フォトクロミック物質から生じる色を補足するために、例えば、より中間色の色を得るために、または入射光の特定の波長を吸収するために、染料が選択される。別の態様においては、フォトクロミック物質が未活性状態にあるときに、主マトリックスに所望の色合いを与えるように、染料が選択される。
ホスト材料は通常、透明であるが、半透明であってもよく、さらには不透明であってもよい。ホスト材料は、フォトクロミック物質を活性化させる電磁スペクトルの部分、即ち、物質の開放形態(open form)を生じさせる紫外線(UV)の波長に対して、および、そのUV活性形態、即ち開放形態、における物質の吸収極大波長を含む可視スペクトルの部分に対してのみ、透明である必要がある。好ましくは、ホスト材料の色は、フォトクロミック物質の活性形態の色を遮蔽(mask)するような色であってはならず、即ち、色の変化が観察者に用意に認識できるような色でなければならない。より好ましくは、ホスト材料物品は、透明または光学的に透明な固体物質であり、例えば、平面レンズおよび眼用レンズ、窓、自動車の透明物、例えばフロントガラス、航空機透明物、プラスチックシート、ポリマーフィルムなどのような光学的用途に適している物質である。
本明細書に記載のフォトクロミック物質または組成物と共に使用することができるポリマー有機ホスト材料の例は、以下のものを包含する:ポリマー、即ち、ポリオール(アリルカーボネート)モノマー、ジエチレングリコールジメタクリレートモノマー、ジイソプロペニルベンゼンモノマー、エトキシル化ビスフェノールAジメタクリレートモノマー、エチレングリコールビスメタクリレートモノマー、ポリ(エチレングリコール)ビスメタクリレートモノマー、エトキシル化フェノールメタクリレートモノマー、およびアルコキシル化多価アルコールアクリレートモノマー、例えばエトキシル化トリメチロールプロパントリアクリレートモノマーの、ホモポリマーおよびコポリマー;ポリマー、即ち、多官能価、例えば、モノ、ジまたはマルチ官能価の、アクリレートおよび/またはメタクリレートモノマー、ポリ(C1-C12アルキルメタクリレート)のホモポリマー及びコポリマー、例えばポリ(メチルメタクリレート)、ポリ(オキシアルキレンジメタクリレート)、ポリ(アルコキシル化フェノールメタクリレート)、セルロースアセテート、セルローストリアセテート、セルロースアセテートプロピオネート、セルロースアセテートブチレート、ポリ(ビニルアセテート)、ポリ(ビニルアルコール)、ポリ(ビニルクロリド)、ポリ(ビニリデンクロリド)、ポリウレタン、熱可塑性ポリカーボネート、ポリエステル、ポリ(エチレンテレフタレート)、ポリスチレン、ポリ(αメチルスチレン)、コポリ(スチレン−メチルメタクリレート)、コポリ(スチレン−アクリロニトリル)、ポリビニルブチラール;およびポリマー、即ち、ジアリリデンペンタエリトリトールのホモポリマーおよびコポリマー、特に、ポリオール(アリルカーボネート)モノマー、例えばジエチレングリコールビス(アリルカーボネート)、およびアクリレートモノマーとのコポリマー。
透明コポリマーおよび透明ポリマーのブレンドも、ホスト材料として適している。好ましくは、ホスト材料は、商品名LEXANとして市販されている、ビスフェノールAおよびホスゲンから誘導されるカーボネート結合樹脂のような、熱可塑性ポリカーボネート樹脂から製造される光学的に透明な重合有機物質;商品名MYLARとして市販されている物質のようなポリエステル;商品名PLEXIGLASとして市販されている物質のようなポリ(メチルメタクリレート);ポリオール(アリルカーボネート)モノマー、特にジエチレングリコールビス(アリルカーボネート)(このモノマーは商品名CR−39として市販されている)の重合体、および、ポリオール(アリルカーボネート)、例えばジエチレングリコールビス(アリルカーボネート)と、他の共重合性モノマー物質とのコポリマー、例えばビニルアセテートとのコポリマー、例えば、80〜90%ジエチレングリコールビス(アリルカーボネート)と10〜20%ビニルアセテート、特に80〜85%ビス(アリルカーボネート)と15〜20%ビニルアセテートとのコポリマー、および米国特許第4360653号および第4994208号に記載の、末端ジアクリレート官能樋を有するポリウレタンとのコポリマー、および米国特許第5200483号に記載の、その末端部分がアリルまたはアクリリル官能基を有する脂肪族ウレタンとのコポリマーの重合体;ポリ(ビニルアセテート)、ポリビニルブチラール、ポリウレタン、ジエチレングリコールジメタクリレートモノマー、ジイソプロペニルベンゼンモノマー、エトキシル化ビスフェノールAジメタクリレートモノマー、エチレングリコールビスメタクリレートモノマー、ポリ(エチレングリコール)ビスメタクリレートモノマー、エトキシル化フェノールメタクリレートモノマーおよびエトキシル化トリメチロールプロパントリアクリレートモノマーから成る群のモノマーのポリマー;セルロースアセテート、セルロースプロピオネート、セルロースブチレート、セルロースアセテートブチレート、ポリスチレン、ならびに、スチレンと、メチルメタクリレート、ビニルアセテートおよびアクリロニトリルとのコポリマーである。
光学的に透明な重合体、即ち、平面レンズおよび眼用レンズ、窓、ならびに自動車の透明物のような光学的用途に適した物質、を製造するために使用される光学的有機樹脂モノマーと共に、本発明のフォトクロミックナフトピランを使用することが特に考えられる。そのような光学的に透明な重合体は、約1.48〜約1.75、例えば、約1.495〜約1.66の範囲の屈折率を有する。特に考えられるのは、PPG Industries,Inc.によって、商品名CR−307およびCR−407として販売されている光学樹脂である。
下記実施例において本発明をより詳しく説明するが、それらの実施例において多くの改質および変更が当業者に明らかであるので、それらの実施例は説明することのみを意図するものである。
実施例1
段階1
カリウムt−ブトキシド(75g、0.67モル)を、トルエン200ミリリットル(mL)を含有する反応フラスコに加えた。反応フラスコに、オーバーヘッド攪拌器、滴下漏斗、および窒素入口を備えた冷却器を取り付けた。反応フラスコの内容物を還流温度に加熱し、ベンゾフェノン(91g、0.5モル)、琥珀酸ジメチル(90g、0.62モル)、およびトルエン(100g)の混合物を半時間かけて加えた。得られるぺ−スト状の混合物をさらに2時間還流し、冷却し、約400mLの水を加え、充分に混合した。水性層を分離し、稀塩酸で酸性にし、トルエン200mLで抽出した。溶媒、トルエンおよび残留t−ブタノールを回転蒸発器で除去して、近似定量収量(a near quantitative yield)の未精製半エステル、4,4−ジフェニル−3−メトキシカルボニル−3−ブテン酸を生成した。この物質をさらに精製することなく、次の段階に直接使用した。
段階2
段階1からの未精製半エステルを、トルエン200mLを含有する反応フラスコに加えた。無水酢酸(100g)および無水酢酸ナトリウム(15g)を加え、混合物を17時間還流した。混合物を冷却し、溶媒、トルエンを回転蒸発器で除去した。得られる残留物を塩化メチレン200mLに溶解し、攪拌した。水(200mL)を加え、続いて、二酸化炭素の発生が止むまで固体炭酸ナトリウムをゆっくりと加えた。塩化メチレン属を分離し、水洗した。溶媒、塩化メチレンを回転蒸発器で除去して、主に1−フェニル−2−メトキシカルボニル−4−アセトキシ−ナフタレンを含有する粘稠油を得た。この物質をさらに精製することなく、次の段階に直接使用した。
段階3
段階2からの1−フェニル−2−メトキシカルボニル−4−アセトキシ−ナフタレンを含有する油を、メタノ−ル400mLを含有する反応フラスコに加えた。濃塩酸2mLを加え、混合物を加熱還流した。約4時間後、混合物の容量を回転蒸発器で半分に減少させた。混合物を冷却すると、生成物が結晶化を開始した。得られる結晶を吸引濾過し、新たなメタノールで洗浄し、乾燥した。回収生成物100gは、174〜176℃の融点を有し、核磁気共鳴スペクトル(NMR)は、その生成物が4−フェニル−3−メトキシカルボニル−1−ナフトールと一致する構造を有することを示した。
段階4
段階3からの4−フェニル−3−メトキシカルポニル−1−ナフトール100gを、10重量%水酸化ナトリウム水溶液350mLおよびメタノール50mLを含有する反応フラスコに加えた。混合物を1時間還流し、冷却し、次に、約1リットルの冷却(約4℃)稀塩酸を含有するビーカーにゆっくりと注いだ。210℃〜212℃の融点を有する得られる結晶性生成物、1−フェニル−4−ヒドロキシ−2−ナフトエ酸、約90gを、真空濾過によって集めた。
段階5
段階4からの1−フェニル−4−ヒドロキシ−2−ナフトエ酸35gを、85重量%燐酸溶液35gを含有する反応フラスコに加えた。得られる混合物を190℃〜200℃に加熱し、この温度で1時間維持した。この間に、深い赤色の固体生成物が形成された。混合物を冷却し、水200mLを加えた。固形物をスパチュラで砕き、濾過し、水、5重量%重炭酸ナトリウム水溶液、および水で、連続的に洗浄した。赤色生成物、5−ヒドロキシ−7H−ベンゾ[C]−フルオレン−7−オン、18gを、真空濾過によって回収した。
段階6
段階5からの5−ヒドロキシ−7H−ベンゾ[C]−フルオレン−7−オン(6g)を、1,1−ジ(4−メトキシフェニル)−2−プロピン−1−オール(6g)およびトルエン100mLを含有する反応フラスコに加えた。得られる混合物を攪拌し、50℃に加熱し、ドデシルベンゼンスルホン酸を3滴加え、反応混合物を50℃で5時間維持した。反応混合物を室温に冷まし、濾過し、集めた濾液を5重量%水酸化ナトリウム水溶液で3回洗浄した。溶媒、トルエンを、回転蒸発器で除去し、残留物にアセトンを添加したときに、所望生成物が結晶化した。固形物を真空濾過し、新たなアセトンで洗浄し、乾燥して、190℃〜191℃の融点を有する赤色−オレンジ色の生成物6.2gが得られた。NMRは、その生成物が3,3−ジ(4−メトキシフェニル)−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
段階7
段階6からの3,3−ジ(4−メトキシフェニル)−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピラン3gを、無水ジエチルエーテル50mLを含有する反応フラスコに加えた。赤色−オレンジ色が完全に消えるまで、水素化リチウムアルミニウムを少量ずつ攪拌混合物に加えた。反応混合物をさらに10分間攪拌し、少量のエタノールで急冷し、5重量%塩酸200mLに注いだ。有機層を分離し、水洗し、濾過し、溶媒、ジエチルエーテルを、回転蒸発器で除去した。ヘキサン:酢酸エチルの2:1混合物約10ミリリットルを残留物に加えることによって、所望生成物の結晶化が生じた。回収した結晶を、ヘキサン:酢酸エチルの2:1混合物の少量で洗浄し、乾燥して、融点127℃〜129℃を有する生成物2.6gを得た。NMRスペクトルは、その生成物が、3,3−ジ−(4−メトキシフェニル)−13−ヒドロキシ−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例2
段階7において、3,3−ジ(4−メトキシフェニル)−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピラン3gを、無水テトラヒドロフラン50mLを含有する反応フラスコに加えたことを除いては、実施例1の工程に従った。過剰量のメチルグリニャール試薬を攪拌しながら反応に加える間、混合物を氷浴で冷却し、窒素パッドによって湿気から保護した。さらに10分間攪拌した後、5重量%塩酸200mLを加え、有機層を分離し、水洗した。溶媒、テトラヒドロフランを回転蒸発器で除去した。ヘキサン:酢酸エチルの2:1混合物約10ミリリットルを、残留物に添加することによって、非フォトクロミック物質の結晶化が生じた。この物質を濾過によって分離した。ヘキサン:酢酸エチルの3:1混合物を溶離剤として用いて、濾液をシリカ上でカラムクロマトグラフィーに掛けた。エーテルから結晶化した所望生成物を、濾過し、乾燥して、融点125℃〜126℃を有する生成物0.7gを得た。NMRスペクトルは、この生成物が、3,3−ジ(4−メトキシフェニル)−13−ヒドロキシ−13−メチル−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例3
段階6において、1−(2,3−ジヒドロベンゾフル−5−イル)−1−(4−メトキシフェニル)−2−プロピン−1−オールが、1,1−ジ(4−メトキシフェニル)−2−プロピン−1−オールの代わりに使用されたことを除いては、実施例1の工程に従った。得られる生成物、3−(4−メトキシフェニル)−3−(2,3−ジヒドロベンゾフル−5−イル)−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピランを、実施例2に記載のように、段階7における3,3−ジ(4−メトキシフェニル)−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピランの代わりに使用した。得られるジアステレオマー混合物をエーテルヘキサン混合物から結晶化し、濾過し、乾燥した。回収した結晶3.7gは、121℃〜135℃の温度範囲で融解した。NMRスペクトルは、生成物が、3−(4−メトキシフェニル)−3−(2,3−ジヒドロベンゾフル−5−イル)−13−ヒドロキシ−13−メチル−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例4
段階1において、ベンゾフェノンに代わって使用された4,4′−ジメチルベンゾフェノン(105g、0.5モル)を可溶化するために、トルエン100gの代わりに200gを使用して、4,4−ジ(4−メチルフェニル)−3−メトキシカルボニル−3−ブテン酸を生成したことを除いては、実施例1の工程に従った。このストッブ半エステルを段階2に使用して、融点144℃〜146℃を有する1−(4−メチルフェニル)−2−メトキシカルボニル−4−アセトキシ−6−メチルナフタレン100gを生成した。実施例1の工程の段階3を省略した。段階4において、4−フェニル−3−メトキシカルボニル−1−ナフトールに代わって1−(4−メチルフェニル)−2−メトキシカルボニル−4−アセトキシ−6−メチルナフタレンを使用して、融点210℃〜213℃を有する1−(4−メチルフェニル)−4−ヒドロキシ−6−メチル−2−ナフトエ酸を生成した。段階5において、この生成物100gを、1−フェニル−4−ヒドロキシ−2−ナフトエ酸の代わりに使用し、キシレン(250g)および85重量%燐酸溶液250gと混合した。攪拌混合物を、ジーン−スタークトラップ(Dean−Stark trap)を備えた1リットルフラスコ中で、20時間還流して、3,9−ジメチル−5−ヒドロキシ−7H−ベンゾ[C]−フルオレン−7−オン90gを生成し、その生成物2.0gを、1−(2,3−ジヒドロベンゾフラ−5−イル)−1−(4−メトキシフェニル)−2−プロピン−1−オール3.0gと共に、段階6に使用した。得られる生成物、3−(4−メトキシフェニル)−3−(2,3−ジヒドロベンゾフラ−5−イル)−6,11−ジメチル−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピランを段階7において、エーテルヘキサン混合物を生成物を結晶化するために使用したことを除いては実施例2に記載のように使用した。回収生成物1.2gは、198℃〜200℃の融点を有した。NMRスペクトルは、この生成物が、3−(4−メトキシフェニル)−3−(2,3−ジヒドロベンゾフラ−5−イル)−13−ヒドロキシ−6,11,13−トリメチル−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例5
段階6において、1,1−ジ(4−メトキシフェニル)−2−プロピン−1−オール10gを、3,9−ジメチル−5−ヒドロキシ−7H−ベンゾ[C]−フルオレン−7−オン10gと反応させて、融点227℃〜229℃を有する生成物16gを生成したこと以外は、実施例4の工程に従った。NMRスペクトルは、この生成物が、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。段階7において、段階6からのピラン生成物10gを、過剰量のメチルグリニャールと反応させて、所望生成物を、エーテルヘキサン混合物の代わりにメタノールから結晶化した。回収生成物8gは、233℃〜235℃の融点を有した。NMRスペクトルは、この生成物が、3,3−ジ(4−メトキシフェニル)−6,11,13−トリメチル−13−ヒドロキシ−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例6
メチルグリニャール試薬の代わりに、エチルグリニャール試薬の過剰量を、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピラン3gと反応させて、融点153℃〜155℃を有する結晶生成物1.4gを生成したことを除いては、実施例5の工程に従った。NMRスペクトルは、この生成物が、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−エチル−13−ヒドロキシ−インデノ[2,1−f]ナフト[1.2−b]ピランと一致する構造を有することを示した。
実施例7
段階7において、メチルグリニャールの代わりに、イソプロピルグリニャール試薬の過剰量を、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピラン3gと反応させて、融点209℃〜210℃を有する結晶性生成物1.7gを生成したことを除いては、実施例5の工程に従った。NMRスペクトルは、この生成物が、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−(1−メチルエチル)−13−ヒドロキシ−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例8
段階7において、メチルグリニャールの代わりに、t−ブチルグリニャール試薬の過剰量を、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピラン3gと反応させて、240℃よりも高い融点を有する結晶性生成物1.0gを生成したことを除いては、実施例5の工程に従った。NMRスペクトルは、この生成物が、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−(1,1−ジメチルエチル)−13−ヒドロキシ−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例9
段階7において、メチルグリニャールの代わりに、n−ブチルリチウム試薬の過剰量を、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−オキソ−インデノ[2,1−f]ナフト[1,2−b]ピラン3gと反応させて、148℃〜150℃の融点を有する結晶性生成物1.0gを生成したことを除いては、実施例5の工程に従った。NMRスペクトルは、この生成物が、3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−ブチル−13−ヒドロキシ−インデノ[2,1−f]ナフト[1,2−b]ピランと一致する構造を有することを示した。
比較例1
実施例1の段階3からの4−フェニル−3−メトキシカルボニル−1−ナフトール(2g)および1,1−ジ(4−メトキシフェニル)−2−プロピン−1−オ−ル(2g)を、トルエン100ミリリットル(mL)を含有する反応フラスコに加えた。得られる混合物を攪拌し、40℃に加熱し、ドデシルベンゼンスルホン酸を2滴加え、この反応混合物を40℃で3時間維持した。反応混合物を室温に冷ました後、同容量の水に加えた。有機層を分離し、溶媒、トルエンを、回転蒸発器で除去した。得られる残留物を、ヘキサン:酢酸エチルの2:1混合物を溶離剤として用いて、シリカ上でクロマトグラフィーに掛けた。フォトクロミック画分を集め、溶媒を留去し、所望生成物をヘキサン/ジエチルエーテル混合物から結晶化させた。回収した結晶を乾燥し、濾過して、融点168℃〜169℃を有する生成物2gを得た。NMRスペクトルは、この生成物が2,2−ジ(4−メトキシフェニル),5−メトキシカルボニル,6−フェニル−[2H]−ナフト[1,2−b]ピランと一致する構造を有することを示した。
実施例10
パートA

(d)3−(4−メトキシフェニル)−3−(2,3−ジヒドロベンゾフル−5−イル)−13−アセトキシ−6,11−ジメトキシ−13−メチル−インデノ[2,1−f]ナフト[1,2−b]ピラン;
(e)3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−メトキシ−インデノ[2,1−f]ナフト[1,2−b]ピラン;
(f)3,3−ジ(4−メトキシフェニル)−6−メチル−11−フルオロ−13,13−ジエトキシ−インデノ[2,1−f]ナフト[1,2−b]ピラン;
(g)3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−(1−メチルエチル)−13−ヒドロキシ−インデノ[2,1−f]ナフト[1,2−b]ピラン;
(h)3−(4−メトキシフェニル)−3−(3,4−ジメトキシフェニル)−6,11−ジメチル−13,13−ジプロピル−インデノ[2,1−f]ナフト[1,2−b]ピラン;および
(i)3,3−ジ(4−メトキシフェニル)−6,11−ジメチル−13−ブチル−13−ヒドロキシ−インデノ[2,1−f]ナフト[1,2−b]ピラン;

(B) 3,3-di (4-methoxyphenyl) -13-hydroxy-13-methyl-indeno [2,1-f] naphtho [1,2-b] pyran;
(C) 3- (4-methoxyphenyl) -3- (2,3-dihydrobenzofur-5-yl) -13-hydroxy-13-methyl-indeno [2,1-f] naphtho [1,2- b] pyran;
(D) 3- (4-methoxyphenyl) -3- (2,3-dihydrobenzofur-5-yl) -13-acetoxy-6,11-dimethoxy-13-methyl-indeno [2,1-f] Naphtho [1,2-b] pyran;
(E) 3,3-di (4-methoxyphenyl) -6,11-dimethyl-13-methoxy-indeno [2,1-f] naphtho [1,2-b] pyran;
(F) 3,3-di (4-methoxyphenyl) -6-methyl-11-fluoro-13,13-diethoxy-indeno [2,1-f] naphtho [1,2-b] pyran;
(G) 3,3-di (4-methoxyphenyl) -6,11-dimethyl-13- (1-methylethyl) -13-hydroxy-indeno [2,1-f] naphtho [1,2-b] Piran;
(H) 3- (4-methoxyphenyl) -3- (3,4-dimethoxyphenyl) -6,11-dimethyl-13,13-dipropyl-indeno [2,1-f] naphtho [1,2-b Pyran; and (i) 3,3-di (4-methoxyphenyl) -6,11-dimethyl-13-butyl-13-hydroxy-indeno [2,1-f] naphtho [1,2-b] pyran ;
