試料を混合した気体をカメムシの口先約1から2cmのと
ころに噴出せしめて、カメムシが口吻を伸長するかどう
かにより摂食刺激活性の有無を判断した。 2.活性成分物質の単離 ハスモンヨトウの幼虫をインゲン豆粉末を主成分とする
人工試料を餌として飼育し、6齢幼虫20頭を用いてヘ
キサンとアセトンの1:2混合溶媒で30分間抽出し、
粗抽出物を得た。粗抽出物には明確な摂食刺激活性が認
められた。この粗抽出物をシリカゲルを充填したカラム
クロマトグラフィーにより分画した。溶出溶媒としてヘ
キサン、5%エーテルを含んだヘキサンおよび15%エ
ーテルを含んだヘキサン溶媒を用いて順次溶出した。得
られた各画分について摂食刺激活性を評価したところ、
15%エーテル/ヘキサン画分にのみ明確な摂食刺激活
性が認められたが、他の画分には活性がほとんど認めら
れなかった。さらに、この15%エーテル/ヘキサン画
分を溶媒抽出法を用いて酸性、中性、塩基性画分に分画
したところ、中性画分に強い摂食刺激活性が認められ
た。 3.活性成分物質の同定 上記摂食刺激活性が認められた中性画分をガスクロマト
グラフ直結型質量分析計(GC−MS)を用いて分析し
たところ、炭素数20個のアルコール化合物を主成分と
していることが明らかにされた。この成分のマススペク
トルを後掲図1に示す。この成分のマススペクトルが別
途化学合成した(E)−3,7,11,15−テトラメチ
ル−2−ヘキサデセン−1−オール(E−フィトール)
のマススペクトルと全く一致した。さらにガスクロマト
グラフ分析においてこの成分の保持時間が別途化学合成
したE−フィトールの保持時間と一致することを確認し
た。この結果から、上記の15%エーテル/ヘキサン画
分中に含まれる活性成分物質は、E−フィトールである
と同定された。なお、活性成分物質のハスモンヨトウの
6齢幼虫1頭当たりからの抽出量は約480ngであっ
た。 4.E−フィトール標品 別途化学合成により得られるフィトールは、E−及びZ
−異性体の混合物であるので、これをアセチル化してフ
ィトール アセテートとした後、硝酸銀を含有させたシ
リカゲルカラムクロマトグラフィーを用いて両異性体の
分離・精製を行い、それぞれ加水分解してE−及びZ−
フィトールとしたものを精製して、各標品とした。 5.摂食刺激活性の確認試験 ハリクチブトカメムシに対する合成E−及びZ−フィト
ールと上記15%エーテル/ヘキサン画分の摂食刺激活
性を上記1.記載の確認法により試験した。この摂食刺
激活性の試験の結果を表1に示す。
