産業上の利用分野
本発明は、イブプロフェンの投与による消化管障害が
生じることが可及的に防止されたイブプロフェン製剤に
関する。
従来の技術及び発明が解決しようとする課題
イブプロフェンは、解熱鎮痛剤等として効果の高いこ
とが従来より知られているが、イブプロフェンを経口投
与した場合、胃部不快感、胃痛、嘔気などが生じること
があり、副作用として消化管障害を起こさせることが指
摘されている。
このために従来、イブプロフェンの経口投与による胃
腸障害を軽減させることを目的としてイブプロフェンを
アニリン誘導体系解熱剤と併用したり(特開昭56−9722
4号、同56−154416号公報)、アセトアミノフェンと併
用したり(特開昭59−104315号公報)、サリチル酸系薬
物と併用したり(特開昭61−134315号公報)することが
提案されている。しかしながら、これらは2種の解熱鎮
痛剤を併用するものであり、有効成分としてイブプロフ
ェンのみを用いた場合にもその副作用を軽減させること
が望まれる。
更に、イブプロフェンのみを用いる場合の胃障害軽減
法としては、固体脂肪と混合溶融し、得られた液を噴霧
凝固する方法(特開昭57−145807号公報)があるが、操
作性に難があり、かつ表面が油性となるため、内服した
場合の有効性を低下させることが予想され、問題があ
る。
本発明は上記事情に鑑みなされたもので、イブプロフ
ェンを投与した場合に生じる副作用、特に消化管障害が
軽減されたイブプロフェン製剤を提供することを目的と
する。
課題を解決するための手段及び作用
本発明者らは上記目的を達成するため鋭意検討を進め
た結果、炭酸マグネシウムをイブプロフェンと併用する
こと、より好ましくは更にアルミニウムグリシネートを
配合することにより、イブプロフェンの有する解熱鎮痛
作用等の薬効を阻害することなく、胃粘膜障害を著しく
改善し、消化管障害を軽減することができ、このためイ
ブプロフェンを有効成分とする安全性の高い製剤が得ら
れることを知見し、本発明をなすに至った。
従って、本発明はイブプロフェンに炭酸マグネシウム
を併用したイブプロフェン製剤を提供するものである。
以下、本発明につき更に詳しく説明する。
本発明に係るイブプロフェン製剤は、上述したように
イブプロフェンに炭酸マグネシウム、より好ましくは炭
酸マグネシウムとアルミニウムグリシネート(ジヒドロ
キシアルミニウムアミノアセテート)を併用するもので
ある。なお、これらに加え、更に炭酸水素ナトリウム、
水酸化マグネシウム、水酸化アルミナマグネシウム、乳
酸アルミニウム、ケイ酸マグネシウム、ケイ酸アルミン
酸二マグネシウム、ケイ酸アルミン酸マグネシウムビス
マス、メタケイ酸アルミン酸マグネシウム、乾燥水酸化
アルミニウムゲル、合成ケイ酸アルミニウム、ショ糖硫
酸エステルアルミニウム塩(アルサルミン)、合成ヒド
ロタルサイド、酸化マグネシウム、ロートエキス、乳酸
カルシウム、アミノ酢酸、水酸化アルミニウム・炭酸マ
グネシウム混合乾燥ゲル、水酸化アルミニウム・炭酸水
素ナトリウムの共沈生成物、水酸化アルミニウム・炭酸
カルシウム・炭酸マグネシウムの共沈生成物、水酸化マ
グネシウム・硫酸アルミニウムカリウムの共沈生成物な
どの1種又は2種以上を併用するようにしてもよい。
この炭酸マグネシウム又はこれとアルミニウムグリシ
ネートとの合計の配合量は、イブプロフェン100重量部
に対し5〜500重量部の割合とすることが好ましい。
本発明のイブプロフェン製剤において、イブプロフェ
ンの投与量は常用量(成人の場合1日600mg程度)とす
ることができるが、この場合有効成分としてはイブプロ
フェンを単独で使用してもよく、他の解熱鎮痛剤、その
他の有効成分と併用することもできる。ここで、他の解
熱鎮痛剤や有効成分を併用する場合は必要によりイブプ
ロフェンの投与量を少なくすることができる。
本発明のイブプロフェン製剤は、種々の形態、例えば
錠剤、顆粒剤、細粒剤、散剤、硬カプセル剤、軟カプセ
ル剤等として調製することができ、その形態に応じた通
常の用法に従って投与することができる。この場合、こ
れらの製剤は常法に従って調製し得、この際上述した成
分に加えてその製剤に応じた通常の配合成分を配合して
製剤化することができる。
発明の効果
本発明のイブプロフェン製剤は、イブプロフェンに対
し炭酸マグネシウムを併用したことにより、イブプロフ
ェンの副作用、特に消化管障害が軽減され、使用上の安
全性が高いものである。
次に、実験例を示し、本発明の効果を具体的に示す。
〔実験例〕
ジャーン(Jahn)らの方法によりラットの胃粘膜障害
を試験した。
即ち、体重130g前後の雄性ラットを18時間絶食させた
後、1群7〜8匹を使用し、第1表に示す薬物を10%ア
ラビアゴム水溶液に懸濁したものを投与した。3.5時間
後に動物を殺して胃を摘出し、大湾に沿って縦切し、肉
眼的に胃粘膜障害の有無並びに程度を判定した。この場
合、潰瘍の程度はアダミ(Adami)らの方法〔Arch.int.
Pharmacodyn.147,113(1964)〕により潰瘍係数(ulcer
index)として表わした。
結果を第1表に示す。
第1表の結果に見られる通り、イブプロフェン(200m
g/kg)の経口投与により潰瘍係数3.2程度の胃粘膜障害
が認められるが、炭酸マグネシウム又はこれとアルミニ
ウムグリシネートを併用投与することによって胃粘膜障
害が著しく改善されることが認められた。
以下、実施例(製剤例)を示し、本発明を具体的に示
すが、本発明は下記の例に制限されるものではない。
〔実施例1〕錠剤
イブプロフェン 75mg/l錠
炭酸マグネシウム 100
アルミニウムグリシネート 50
トウモロコシデンプン 50
結晶セルロース 50
ステアリン酸マグネシウム 2
上記成分を常法により打錠する。

The present invention has been made in view of the above circumstances, and an object of the present invention is to provide an ibuprofen formulation in which side effects caused by administration of ibuprofen, particularly, gastrointestinal disorders are reduced. Means and Action for Solving the Problems The present inventors have conducted intensive studies to achieve the above object, and as a result, it has been found that magnesium carbonate is used in combination with ibuprofen, and more preferably, aluminum glycinate is further blended to obtain ibuprofen. It is possible to significantly improve gastric mucosal damage and reduce gastrointestinal tract damage without inhibiting the drug efficacy such as antipyretic analgesic effect of having a highly safe formulation containing ibuprofen as an active ingredient.
