【発明の詳細な説明】

本発明は、好ましくは微晶質セルロース(MCC)のような錠剤用賦形剤(t
ablet filler)と好ましくはβ−シクロデキストリンのような結合
剤とをベースとする直接錠剤形成助剤(direct tabletting
auxiliary)に関する。この助剤は湿式混合法によって製造したもので
ある。

製薬業では現在、直接錠剤形成、即ち錠剤形成助剤と活性物質との乾式混合及
び圧縮に、多数のキャリヤー物質、例えばセルロース粉末、リン酸ジカルシウム
、ソルビトール、MCC、デキストロース、ラクトース又はラクトース/セルロ
ースを使用している。
【0003】
この種の直接錠剤形成助剤に要求される主な条件は、流動性が高く、低圧下で
十分な圧縮性を示し、且つ充填容量(loading capacity)が大
きいことである。

この種の助剤を用いて製造した錠剤は、十分な硬度と、小さい摩損度と、良好
な崩壊分散特性及び溶解性とを示す筈である。

市販の製品はこれらの要件を部分的にしか満たしていない。

直接錠剤形成法は、活性物質にストレスをかけずに処理でき且つ錠剤の処理及
び製造コストが低いという理由から、製薬業にとって特に有利な方法である。

直接錠剤形成では助剤として微晶質セルロースが長年にわたり世界中で使用さ
れてきた(例えば、Avicel(登録商標)PH101及びPH102)。こ
れについては特に、USP XXII/NF XVII、1915ページを参照
されたい。

本発明の目的は、前述の要件を満たし、更に充填容量を増大させ、即ち活性物
質含量70〜75%の錠剤を形成できるようにし、且つ通常は崩壊剤の添加が不
要な直接錠剤形成助剤を提供することにある。

助剤。Claims: 1. A tableting aid, selected from the following main components: A) microcrystalline cellulose or cellulose powder;
A tablet excipient comprising from 0 to 98% by weight; and B) a binder selected from α-, β- or γ-cyclodextrin, which directly comprises from 2 to 40% by weight of the tableting aid, of a homogeneous mixture. A direct tableting aid, comprising in form, the intimate mixture of A and B being prepared using a wet mixing method and subsequent drying in the presence of water or a water / alcohol mixture. 2. A direct tableting aid according to claim 1, wherein the content of component A) is from 80 to 98% by weight and the content of component B) is from 2 to 20% by weight. 3. The direct tableting aid according to claim 1, wherein the binder is β-cyclodextrin. 4. The direct tablet-forming auxiliary according to claim 1, which is prepared by wet granulation.
