Abstract
Description
陽性T細胞から上記のT細胞抗原受容体をコードする核酸配列をクローニングするステップ1)と、
初代T細胞を単離、培養するステップ2)と、
ステップ1)で得られた核酸配列をステップ2)の前記初代T細胞に送達し、上記CDR、上記α鎖ポリペプチド、上記β鎖ポリペプチド又はT細胞抗原受容体を発現させる組換えT細胞を得るステップ3)と、を含む組換えT細胞の製造方法を提供する。
陽性T細胞から上記抗体又はその抗原結合断片、又は上記のT細胞抗原受容体をコードする核酸配列をクローニングして得るステップ(1)と、
ステップ(1)で得られた核酸配列をベクター骨格に連結し、発現ベクターを得るステップ(2)と、
ステップ(2)で得られた発現ベクターを宿主細胞に形質転換し、次に、その発現を誘導するステップ(3)と、
抗体又はその抗原結合断片又はT細胞抗原受容体を得るステップ(4)と、を含む抗体又はその抗原結合断片又はT細胞抗原受容体の製造方法を提供する。
(1)好ましくは配列番号5~26のいずれか1つに示されるか、又は配列番号5~26のいずれか1つに示されるアミノ酸配列とは少なくとも80%の相同性を有するT細胞抗原受容体と、

C末端にavi-tag配列が連結されたMHC分子α鎖細胞外領域及びβ2m鎖を発現して精製するステップI)と、
抗原ペプチド、ステップI)で得られたC末端にavi-tag配列が連結されたMHC分子α鎖細胞外領域及びβ2m鎖をリフォールディングし、単量体を製造するステップII)と、

一、EBV抗原エピトープ4量体の構築
1)配列が最適化されたHLA-A*0201(そのアミノ酸配列は配列番号1、ヌクレオチド配列は配列番号118に示される)、HLA-A*242(そのアミノ酸配列は配列番号2、ヌクレオチド配列は配列番号119に示される)、HLA-A*1101(そのアミノ酸配列は配列番号3、ヌクレオチド配列は配列番号120に示される)のα鎖及びβ2m鎖(そのアミノ酸配列は配列番号4、ヌクレオチド配列は配列番号121に示される)を発現させた。ここで、α鎖の構造は対応するHLA型α鎖の細胞外領域配列にAvi-tag配列が連結され、BamHI酵素切断部位で隔てられ、それによりビオチン化部位を提供するものである。β2m鎖はシグナルペプチド配列を取り除き、成熟ペプチド配列の前に2つのアミノ酸(MとA)を付加した。発現ベクターはPET28a+であり、発現菌はtransetta又はBL21である。IPTG濃度は0.5mMで、誘導発現は4時間であった。α鎖及びβ2m鎖タンパク質の封入体を抽出した。
2)EBV抗原エピトープ(抗原ペプチド)の選択:HLA-A*0201型は抗原エピトープFLYALALLL(配列番号29)、HLA-A*2402型は抗原エピトープPYLFWLAAI(配列番号30)、TYGPVFMSL(配列番号31)、TYGPVFMCL(配列番号32)、HLA-A*1101型は抗原エピトープSSCSSCPLSK(配列番号33)、SSCSSCPLTK(配列番号34)に対応する。
3)pMHC I単量体のフォールディング及び精製:ステップ2)の前記抗原ペプチド、対応するステップ1)の前記β2m鎖リフォールディングタンパク質及びα鎖タンパク質をモル比40:2:1の割合で還元系に順に加え、72時間フォールディング反応させた。得られた産物をSuperdex75 10/300GLカラムで精製した。精製産物を収集してavidityキットでビオチン化し、再度精製してビオチン化単量体を得、ゲル電気泳動により単量体純度を検出した。
4)ステップ3)の前記ビオチン化単量体とAPC標識ストレプトアビジンとの結合反応により、対応する4量体を得、それぞれA0201-FLYALALLL-4量体、A2402-PYLFWLAAI-4量体、A2402-TYGPVFMSL-4量体、A2402-TYGPVFMCL-4量体、A1101-SSCSSCPLSK-4量体及びA1101-SSCSSCPLTK-4量体と命名した。
二.EBV抗原エピトープ4量体の効果検出
1.ヒト末梢血単球細胞(PBMC)を単離し、細胞密度1×106細胞/mLの細胞懸濁液を製造した。
2.細胞を3000rpmで5分遠心分離した。上清を除去し、1%血清を含むPBS 50μlで重懸濁させた。
3.4量体2μlを加えて室温で30分インキュベートした。
4.CD8抗体2μlを加え、氷上で20分インキュベートした。
5.PBS 1mlを加えて、3000rpmで5分処理した。
6.上清を除去し、PBS 1mlを加えて、3000rpmで5分処理した。
7.上清を除去し、4%パラホルムアルデヒド500μlで細胞を再懸濁させ、細胞懸濁液をろ過膜でろ過した。
8.フローサイトメータで陽性細胞を検出した。
三、実験結果
単量体のSDS PAGE検出結果を図1に示す。図に示すように、リフォールディングとHPLC精製により得られた単量体は重鎖(C末端にAvi-tag配列が連結されたα鎖の細胞外領域)と軽鎖(シグナルペプチド領域を除去したβ2m鎖)に対応する大きさのタンパク質を明確に示し、純度が高い。
構築した4量体をそれぞれ対応するHLA型TCRに感染した細胞と共インキュベートした。一例として、構築されたA0201-FLYALALLL-4量体、A2402-TYGPVFMSL-4量体、A2402-TYGPVFMCL-4量体、A2402-PYLFWLAAI-4量体、A1101-SSCSSCPLTK-4量体、A1101-SSCSSCPLSK-4量体は、それぞれ対応するHLA型TCRに感染した細胞と共インキュベートした(LLL4量体はTCR E23、AAI4量体はTCR E29、MSL/MCL4量体はTCR E44、LSK/LTK 4量体はTCR E141に対応する)、図2Aに示すように、MBL社が市販した4量体と比較して、A0201-FLYALALLL-4量体で検出された陽性細胞の割合は70.5%で、市販したFLYALALLL-4量体(59.7%)を大きく上回り、A1101-SSCSSCPLSK-4量体で検出された陽性細胞の割合は20.3%で、市販したSSCSSCPLSK-4量体(18.1%)を明らかに上回っている。本発明の4量体が、細胞陽性率の検出において高い特異性と染色効果を示すことが十分に示されている。更なる体積勾配検出実験において、独自に開発したA2402-TYGPVFMSL4量体の特異的結合効果が市販4量体よりはるかに高いことを発見した(図2B参照)。
本発明により構築された4量体で抽出されたTCRは、野生型抗原エピトープと突然変異型抗原エピトープとを同時に認識し、免疫逃避を防止することができる。図3-Aに示すように、A2402 HLA型野生型エピトープ(MCL)と突然変異型エピトープ(MSL)4量体はいずれもMSL4量体で抽出されたTCR E44で認識され得る。図3-Bに示すように、A1101 HLA型野生型エピトープ(LSK)と突然変異型エピトープ(LTK)4量体は、いずれもLSK4量体で抽出されたTCR E141で認識され得る。同一HLA型の異なる抗原エピトープ4量体について対応する特異的TCRを抽出すると、図3-Cに示すように、AAI4量体では、対応するTCR E29陽性細胞が検出され得。
一、EBV LMP2エピトープ特異的TCRのβとα遺伝子断片の取得
1)実施例1で製造されたA0201-FLYALALLL-4量体、A2402-PYLFWLAAI-4量体、A2402-TYGPVFMSL-4量体、A2402-TYGPVFMCL-4量体、A1101-SSCSSCPLSK-4量体及A1101-SSCSSCPLTK-4量体を末梢血とともに染色し、4量体染色陽性のT細胞をフローサイトメトリーにより選別し、逆転写してcDNA(SuperScript登録商標IV Reverse Transcriptase、Invitrogen)を得た。多重PCR(Multiplex PCR)原理により、2回のPCR(KOD-Plus-Neo、TOYOBO)によりTCRβ遺伝子の可変領域断片を増幅した。

具体的には、PCRポリメラーゼKOD-Plus-Neoの製品取扱書によると、1回目のPCR系は20μL、アニーリング温度は60℃、反応は30サイクルであった。1回目のPCR反応の産物1μLを取り、2回目のPCRのテンプレートとし、2回目のPCR系は30μL、アニーリング温度は60℃、反応は30サイクルであった。次に、2回目のPCR産物をアガロースゲル電気泳動にかけ、対応する大きさのバンドについてゲルをカットして回収し(QIAGEN社のゲル回収キット)、シーケンシングに送り、シーケンシングプライマーは下流プライマー2である。TCRβ遺伝子配列を得、具体的には、E23、E240、E29、E180-1、E44、E141、E149、E168、E170、E244、E245、E254、E301、E304、E305、E307、E314、E315、E316、E317、E318、E320のTCRβ遺伝子配列はそれぞれ配列番号122~143の「二重下線」で示したヌクレオチド配列である。
2)以上のように、4量体染色陽性T細胞を逆転写してcDNA(SuperScript(登録商標)IV Reverse Transcriptase、Invitrogen)を得た。取扱説明書に基づき、TCRα遺伝子断片を、2回PCR(KOD-Plus-Neo、TOYOBO)により増幅した。

二、pHAGE-TCRベクターの構築
TCRβ、fp2A、TCRαを長いプライマー(fp2A配列を含む)overlap-PCR(KOD-Plus-Neo、TOYOBO)により増幅し、TCRβ-fp2A-TCRα断片を得、それぞれE23、E240、E29、E180-1、E44、E141,E149、E168、E170、E244、E245、E254、E301、E304、E305、E307、E314、E315、E316、E317、E318、E320のpHAGE-TCRプラスミドと命名した。
増幅用プライマー:
上流プライマー5を表1に示す。

具体的には、プライマー5とプライマー6でTCRβとTCRαをそれぞれ増幅し、PCR系は50μL、アニーリング温度は60℃、反応は30サイクルであった。PCR産物をアガロースゲル電気泳動にかけて、ゲルを回収し(QIAGEN社のゲル回収キット)、産物1μLをそれぞれ回収してテンプレートとし、上流プライマー5と下流プライマー6を用いてoverlap PCRを行い、PCR系は50μL、アニーリング温度は60℃、反応は30サイクルであった。アガロースゲル電気泳動を行って、約1800bpのバンドを得、ゲルをカットして回収した。
レンチウイルスベクターpHAGE-IRES-RFPをNotIとNheIで二重酵素切断した。酵素切断系は40μLであり、NotIとNheIをそれぞれ1.5μL、プラスミド2~3μgを含み、37℃で酵素切断6時間後、系にアルカリホスファターゼ(NEB)1μLを加え、1時間処理してプラスミドの自己結合を減少させ、酵素切断後のプラスミドをアガロースゲル電気泳動にかけて回収し、nanodropで濃度を測定し、骨格としてプラスミド構築に用いた。
Clone Express II One Step Cloning kitの製品取扱書によると、overlapで酵素切断後線形化したpHAGE-IRES-RFPベクターにTCRを連結し(図4参照)、Stbl3菌株に形質転換し、アンピシリンを含むLBプレート上で12~16時間培養し、単クローン菌を選んでシークエンシングを行い、シークエンシングプライマーはpHAGEベクター上のプライマーseq-pHAGE-Fとseq-pHAGE-R、及び下流プライマー4を用いた。それぞれE23、E240、E29、E180-1、E44、E141,E149、E168、E170、E244、E245、E254、E301、E304、E305、E307、E314、E315、E316、E317、E318、E320と略称される対応するTCRを得た。
1、内因性TCRノックアウトJurkatT細胞株の構築
Jurkat細胞TCRの配列特徴に基づき、α鎖とβ鎖の定常領域にguide配列(TRA_oligo1-CACCGTCTCTCAGCTGGTACACGGC(配列番号362)、TRA_oligo2-AAACGCCGTGTACCAGCTGAGAGAC(配列番号363)、TRB_oligo1-CACCGGGCTCAAACACAGCGACCTC(配列番号364)、TRB_oligo2-AAACGAGGTCGCTGTGTTTGAGCCC)(配列番号365)を設計した。
合成したα鎖とβ鎖のguide配列をそれぞれsgRNA-LentiCRISPR-puroとsgRNA-LentiCRISPR-BSDレンチウイルスベクターに構築し、パッケージングプラスミドpsPAX2、pMD2.G及びPEIトランスフェクション試薬と一定の割合で293T細胞に共トランスフェクションし、48時間と72時間の細胞培養上清を採取し、濃縮した2つのウイルスを同時にヒトJurkatT細胞株に感染した。感染48時間後、適切な濃度のピューロマイシンとブラストサイジンで殺滅し、2つの薬物のそれぞれの対照群の細胞を全て死亡させた。生存している細胞について単細胞をフローサイトメトリーにより選別し、96ウェルプレートに入れて培養した。得られたモノクローナル細胞株に対して、TCRα鎖とβ鎖の抗体を用いて発現をそれぞれ同定し、両鎖が欠損した細胞株は内因性TCRノックアウトJurkat T細胞とし、JC5と命名した。
2、EBV TCRを安定に組み込んだJC5細胞株の構築
実施例2で構築した上記のE23、E240などのpHAGE-TCRプラスミドをそれぞれパッケージングプラスミドpsPAX2、pMD2.G及びトランスフェクション試薬PEIと一定の割合で混合し、293T細胞にトランスフェクションした。48時間と72時間の細胞培養上清を採取し、濃縮した後に対数成長期のJC5細胞(MOI=0.3)に感染した。感染3日後、抗ヒトCD3と抗ヒトTCRαβフロー抗体を用いて細胞を染色し、同じTCR発現レベルの細胞を選別して培養したものをJC5-TCR細胞株とした。
3、TCRの細胞膜への形成及び親和性の検出
1×106JC5-TCR細胞を取り、Brilliant Violet 421商標anti-humanTCRαβ(Biolegend)及び対応するEBV LMP2 pMHC4量体-APC(Tetramer-APC)で染色した後、フローサイトメトリー分析を行った。
図5、6より、製造されたEBV LMP2 HLA-A*A0201 FLYALALLL特異的E23-TCR、E240-TCR、EBV LMP2HLA-A*A2402 PYLFWLAAI特異的E29-TCR、E180-1-TCR及びEBV LMP2HLA-A*A2402 TYGPVFMSL/TYGPVFMCL特異的E44-TCRはいずれも正しく発現し、細胞膜外に表示され、対応する4量体プローブとの一定の親和性があることが分かった。
図7より、製造したEBV LMP2 HLA-A*A1101 SSCSSCPLSK/SSCSSCPLTK特異的TCRのうち、E244-TCRとE307-TCRを除いて、他の構築したTCRはいずれもSSCSSCPLSK/SSCSSCPLTKエピトープによく結合していることが分かった。この結果は、自作4量体が特異的に結合するT細胞の同定への適用に成功しただけでなく、得られたTCRが良好な親和性を有することを示している。
実施例3のpMHC4量体についてはTCRの構造親和性を検出し、4量体はJC5表面のTCRに4価結合していることを考慮して、TCRの活性をさらに同定するために、T2細胞にHLA-A*1101分子を安定的に組み込み、TCRの半最大効果抗原濃度(EC50)定量に用いるT2-HLA-A*1101細胞株を構築し、TCRの機能活性を比較した。
1.HLA-A*1101を安定に組み込んだT2細胞株の構築
HLA-A*1101分子とβ2m分子(ヒト由来)をクローニングし、fp2Aで連結した後、pHAGE-BSDベクターに構築し、パッケージングプラスミドpsPAX2、pMD 2.G及びPEIトランスフェクション試薬と一定の割合で293T細胞に共トランスフェクションし、ウイルスをパッケージングし、T2細胞系の感染に用いた。感染48時間後のT2細胞を、対照組の細胞が全て死亡するまで、適切な濃度のブラストサイジンで殺滅し、T2-HLA-A*1101細胞株を得た。
2、TCR機能活性及びEC50の測定
合成したLMP2抗原エピトープをDMSO溶媒で4mg/mLのストック濃度に希釈した。その後、完全培地を用いてエピトープペプチドセグメントを連続的に勾配希釈し、2×10-8~2×10-4MのLSKとLTKのペプチドセグメント溶液を得、1:100の体積で1×106細胞/mLのT2-HLA-A*1101細胞懸濁液に添加し、均一に混合した後、細胞を96ウェルプレートに1孔あたり100μL敷き、また、濃度1×106細胞/mLのJC5-TCR細胞100μLを加え、均一に混合すると、ペプチドセグメント濃度が1×10-10~1×10-6MのT2インキュベーション系となった。24時間共培養した後、培地の上清を収集し、ELISAキットでIL2生産量を検出した。実験を3回繰り返した。図8及び図10は、それぞれ異なる濃度のLSK及びLTKペプチドセグメント刺激下でのJC5-TCR細胞のIL2産生量を示している。対応するEC50値はprism計算により得られ、3回繰り返したEC50値は具体的には図9と図11に示される。図8~11より、合成したLSK抗原エピトープに対しては、TCR E149、E304、E170、E315が優れた機能活性を示し、LTKエピトープに対しては、E149、E254、E170、E316、E317、E318がいずれも良好な機能活性を示したことが分かった。E244とE307の結果は4量体染色と一致し、いずれもSSCSSCPLSK/SSCSSCPLTKエピトープに対する認識能力が弱かった。
1、ヒト初代T細胞の単離、培養及びレンチウイルス感染
スクリーニングされたTCRのEBV LMP2抗原に対する認識と殺傷機能をさらに検証するために、リンパ球単離液Ficollを用いてボランティアの末梢血から単核球(PBMC)を単離して得た後、EasySep Human T cell isolation kit(stem cell technologies)の製品取扱書に従って、PBMCから陰性選別法によりT細胞を取得し、100U/mL IL2を含む1640完全培地で細胞を1×106細胞/mLに再懸濁させ、抗CD3/CD28抗体で被覆された培養皿で培養して活性化した。活性化48時間後、TCRをロードしたウイルス顆粒(実施例3で製造)をレンチウイルス系でT細胞に感染させ、感染方法は32℃、1500rpmで2時間遠心分離し、取り出した後37℃細胞インキュベータで10時間培養し、その後、培地の交換により感染を停止し、37℃細胞インキュベータで培養を続けることである。感染3日後、フローサイトメータでTCR陽性細胞を選別し、TCRT細胞(上記E23、E240、E29、E180-1、E44、E141,E149、E168、E170、E244、E245、E254、E301、E304、E305、E307、E314、E315、E316、E317、E318、E320を含む)を得た。
2、標的細胞の構築
LMP2-RFP、HLA-A*0201-BSD/HLA-A*2402-BSD/HLA-A*1101-BSD及びLuciferase-GFPをそれぞれロードしたウイルス顆粒をレンチウイルス系で対数成長期のRaji細胞に感染させた。薬物スクリーニング及びフローサイトメトリー選別により、LMP2、HLA-A分子及びLuciferas-GFPを同時に安定に発現させたRaji細胞を得、Raji- HLA-A*A0201/2402/1101-LMP2-luciferaseと命名した。また、対数成長期のEBV-LCL細胞にHLA-A*0201-BSD/HLA-A*2402-BSD/HLA-A*1101-BSDのウイルス粒子を感染させた。薬物スクリーニングにより、HLA-A分子を安定に発現させたEBV-LCL細胞を得、それぞれEBV-LCL-HLA-A*0201、EBV-LCL-HLA-A*2402、EBV-LCL-HLA-A*1101細胞と命名した。
3、ヒト初代T細胞におけるTCRのインビトロ機能検証
1)内在性レベルの抗原エピトープに対するTCR認識能力の検証
EBV-LCLは、即ち、EBウイルスに感染された後不死化したヒトB細胞であり、生体内腫瘍細胞中の抗原レベルをリアルにシミュレーションした。そこで、SSCSSCPLSK/SSCSSCPLTKエピトープを認識するTCR T細胞とEBV-LCL-HLA-A*1101細胞をエフェクター/ターゲット比8:1、4:1、2:1、1:1、0.5:1、0.25:1で共培養し、ここで、標的細胞は1×105細胞に一定させた。24時間インキュベートした後、上清を収集して、分泌したサイトカインIL2(図12)とIFN-γ(図13)を検出することに用いた。サイトカイン放出レベルから見ると、TCR:E141、E170、E254及びE315が内因性レベルのLSKに結合すると、T細胞を顕著に活性化できた。特に、E315 TCR T細胞は、それぞれのエフェクター/ターゲット比の濃度で最高のIFN-γレベルを示しただけでなく、対応するIL2値も高く、その結果から、本発明により製造されたTCRが腫瘍内因性抗原の認識を効果的に媒介できることを示した。
2)TCR T細胞の腫瘍細胞に対する長期殺傷能力の検証
SSCSSCPLSK/SSCSSCPLTKエピトープを認識するTCR T細胞とRaji-HLA-A*1101-LMP2-luciferase細胞をエフェクター/ターゲット比1:3から開始して共培養し、0日目とし、その後、それぞれ1日目、3日目、5日目に細胞を採取してフローサイトメトリー分析を行った。ここで、使用した培地はIL2を含まない1640完全培地であり、最初のTCR T細胞は1×105細胞であり、各時点のサンプルは独立してインキュベートし、残りの共インキュベートサンプルはそれぞれ2日目、4日目に、液体を半分交換し、標的細胞を補充した。フローサイトメトリーに用いた細胞をまず抗ヒトCD3抗体で染色し、投入時に指定体積の細胞を収集して記録し、換算して系内のT細胞の数を把握した(図14参照)。絶対T細胞数の増殖曲線から、E315-TCR T細胞は2つの抗原エピトープを認識した後に最も活性化し増殖したことがわかった。さらに、さらに系内のエフェクター/ターゲット比を分析すると(図15参照)、増殖結果と同様に、E315-TCR T細胞は最も強い腫瘍除去能力を示した。
3)ヒト初代T細胞におけるTCRのインビトロ機能検証
Raji-HLA-A*0201-LMP2、LMP2をトランスフェクションしていないRaji細胞、E23-TCRT、E240-TCRT細胞及び1G4 T細胞を1:0.5、1:1、1:2の数量の比で共培養し、共培養24時間後、細胞と上清をそれぞれ収集し、E23-TCRTとE240-TCRT細胞の活性化と標的細胞の死亡を予備的に検出した。細胞外サイトカインTNFα、IL2及びIFNγ(図16参照)の放出レベルから見ると、E240-TCRTとE23-TCRTは標的細胞との共培養後、対照群1G4 Tと比べ、T細胞の活性化を顕著に引き起こすことができる。そして、標的細胞の分解後に放出されたルシフェラーゼの量は標的細胞の死亡状況を反映することができる(図16参照)。実験の結果、本発明の実施例により構築されたE23-TCRT及びE240-TCRT細胞が、EBV LMP2抗原ペプチド提示細胞によって特異的に活性化され、標的細胞を著しく殺傷できることが証明された。
Raji-HLA-A*2402-抗原ペプチド、抗原ペプチドをトランスフェクションしていないRaji細胞、抗原ペプチドを添加しているT2-HLA2402細胞、及び抗原ペプチドを添加していないT2-HLA2402細胞をそれぞれE29-TCRT、E180-1-TCRT細胞、1G4 T細胞と1:3の数量の比で共培養し、共培養24時間後、細胞と上清をそれぞれ収集し、E29-TCR T及び180-1-TCRT細胞の活性化を予備的に検出した。細胞外サイトカインIL2及びIFNγ(図17参照)の放出レベルから見ると、E29-TCRT及び180-1-TCRT細胞は標的細胞との共培養後、対照群1G4 Tと比べ、T細胞の活性化を顕著に引き起こすことができる。そして、細胞外サイトカイン(図18参照)の放出レベルから見ると、E44-TCRT細胞は標的細胞との共培養後、対照群RFP Tと比べ、同じくT細胞の活性化を顕著に引き起こすことができる。実験の結果、本発明の実施例により構築されたE29-TCRT、E180-1-TCRT及びE44-TCRT細胞が、EBV LMP2抗原ペプチド提示細胞により特異的に活性化され、標的細胞を著しく殺傷できることが証明された。
SSCSSCPLSK/SSCSSCPLTKエピトープを認識するE141、E149、E254、E301、E304及びE314はTCRα鎖とβ鎖のCDR3高度可変領域で全て非常に保存的であると同定したため、配列は1つのアミノ酸が異なるほど高度に類似しているが、機能的には大きく異なっている(図9及び図11)。なぜなら、これらの保存的CDR3モチーフを有するTCRはpublic TCRと定義され、これらの保存配列を有しないTCRはprivate TCRと定義されるからである。この実施例では、public TCRの構造的親和性及び機能が分析された。
1、BFP法によるpublic TCRの構造親和性の測定
赤血球を片側の微量吸液管に固定し、pMHC分子が特異的に埋め込まれたビーズを赤血球の表面に吸着させ、極高感受性のバイオフィルム力学プローブ(BFP)を形成し、それとともに、JCR-TCR細胞を反対側の微量吸液管に固定し、圧電式センサにより両者の接触を制御し、1サイクル当たりの接触時に印加圧力10pN、接触時間0.1sとし、次に、1000pN/sの速度で分離し、次のサイクルを開始した。過程全体にわたって顕微鏡で赤血球-ビーズ表面のひずみを記録し、結合形成の有無と持続時間を判定した。図19の結合形成持続時間から分かるように、public TCR間では、LSKエピトープへの構造的親和性にばらつきがあるが、その中で、E304は良好な機能を有し、T2の結果と一致している(図9)。
2.Alanine Scanning法による保存的CDR3モチーフの機能の分析
TCR E141 α鎖とβ鎖のCDR3領域の先頭よりも後のアミノ酸を順次アラニンに突然変異し、a2-a9及びb2-b11と命名し、pHAGEレンチウイルスベクターに構築し、ウイルスをパッケージングした後、ヒト初代T細胞(MOI=10)に感染し、感染3日後、フローサイトメトリーでTCR陽性細胞を選別できた。選別したTCR T細胞とRaji-LMP2-luciferase標的細胞を1:1のエフェクター/ターゲット比で共培養し、ここで、T細胞は1×105細胞であった。24時間共インキュベート後、細胞と上清をそれぞれ収集し、細胞は沈殿分解後、生存している標的細胞から放出されたルシフェラーゼ量の測定に用い(図20)、上清はTCR T細胞から分泌されたサイトカインIL2の測定に用いた(図21)。図20及び図21より、HLA unmatched-Raji細胞、又はTCR-T細胞無添加LA matched-Raji及びTCR未トランスフェクションpan-T細胞添加HLA matched-Rajiの対照群のバックグラウンド値と比較して、突然変異のないE141はHLA matched-Rajiに遭遇すると標的細胞を効果的に除去し、多くのサイトカインIL2を特異的に産生していることが分かった。しかし、a3、a5、a6及びb6、b7、b8の部位では、個々のアミノ酸の突然変異がTCR T細胞を完全に不活性化させるのに十分であった。これらの実験結果から、これらのpublic TCRが同じ保存的モチーフを共有しているにもかかわらず、1つのアミノ酸の違いによって相互の機能が大きく異なっている可能性があることを共に示唆している。
EBウイルスは主に鼻咽頭上皮細胞とB細胞に感染し、鼻咽頭がんと多種のB細胞リンパ腫の発生、発展と密接に関連している。本実施例では、同定されたTCRの生体内機能を検証するために、B細胞リンパ腫マウスモデル及び鼻咽頭がん固形腫瘍モデルをそれぞれ構築した。
1.リンパ腫モデル及びTCRTの生体内機能の検出
3×105 Raji-HLA-A*1101/0201/2402-LMP2-luciferase腫瘍細胞を5~6週齢のNOD/Scid IL-2Rγ null (NCG)雌マウスに尾静脈を通して接種し、リンパ腫モデルを構築し(図22、23、24参照)、これを1日目とし、5日目に、マウスを、A:PBS注射群(等体積PBS注射)、B:対照TCRT細胞注射群(TCR-1G4 T細胞)、C:EBV TCRT注射群(E141-TCRT細胞)の3群に分け、B/C群マウスは5×106 TCR T細胞を尾静脈注射し、A群は等体積200μLのPBSを注射した。8日目に、二次注射を行い、具体的な操作は5日目と一致した。他のTCR T細胞の具体的な再輸注量と再輸注時間を図22、23に示す。その後の数週間で、マウス体内の腫瘍細胞の成長状況、T細胞の生体内増殖状況、及びマウスの生存状況をモニタリングした。図24に示すように、本発明の実施例で構築されたEBV特異的E141-TCRT細胞は、対照群と比較して、マウスの体内腫瘍細胞を顕著に殺傷し、マウスの生存率を向上させることができる。さらに、図22、23に示すように、本発明の実施例で構築されたEBV特異的E23-TCRT、E240-TCRT、E29-TCRT、E44-TCRT細胞は、対照群と比較して、同じくマウスの体内腫瘍細胞を顕著に殺傷し、マウスの生存率を向上させることができる。
2、固形腫瘍モデル
1×106C666-1-HLA-A*1101-LMP2-luciferase腫瘍細胞を5~6週齢のNCG雌マウスに皮下接種し、鼻咽頭がん固形腫瘍モデルを構築し(図25参照)、これを0日目とし、7日後、マウスを、A:PBS注射群(等体積PBS注射)、B:対照TCR-T細胞注射群(TCR-1G4 T細胞)、C:EBV TCR-T注射群(E141-TCRT細胞)の3群に分け、B/C群マウスは3×106T細胞を尾静脈注射し、A群は等体積200μLのPBSを注射した。その後の数週間で、マウスの体内腫瘍細胞の成長状況、T細胞の体内増殖状況、及びマウスの生存状況をモニタリングした。図25、図26、及び図27に示すように、本発明の実施例で構築されたEBV特異的E141-TCRT細胞は、対照群と比較して、マウスの腫瘍細胞を顕著に殺傷でき(図25、図26)、再輸注されたTCR T細胞はインビボで特異的に増殖する(図27)。
