本発明の非クロマトグラフィー法で使用されるビーズ(図中の参照符号6によって示されている)は、自由に移動可能な粒子である限り、すなわち、液体を含む容器を移動させたとき、又は液体を運動させたときに液体内で浮遊することができる限り、任意の粒子状

ビーズ(直径140μm超)を本発明の方法に適用することによって確認され、実施例5を参照されたい。図6Aに示すように、小型ビーズを使用すると、バフィーコートからのCD81+細胞の効率的な単離をもたらし、一方、大型ビーズを使用すると、標的細胞のわずかな収率しかもたらさない。したがって、追加又は代替の実施形態では、ビーズは、約30~100μm、好ましくは40~60μmの直径を有することを特徴とする。本明細書で使用される場合、「小型」ビーズは40~60μmの直径を有すると定義され、「大型」ビーズは140~180μmの直径を有すると定義される。

上記のように、タグ付け剤(3)を介した生物学的実体(1)のビーズ(6)への結合は、非共有結合性タンパク質-リガンド相互作用によって間接的に媒介されてもよい。したがって、本発明の更なる実施形態又は代替の実施形態では、タグ付け剤(3)とビーズ(6)との間の結合は、連結分子(図中の参照符号10によって示される)を介して媒介される。このような間接的な結合を、図7及び10に概略的に示す。この例では、ビーズ(6)は、連結分子(10)に含まれるリガンド(5)によって認識されるリガンド結合パートナー(7)を含む。次いで、連結分子(10)は、タグ付け剤(3)によって認識される抗原(図中に参照符号2’で示す)を含む。

更なる態様では、本発明は、本発明の上記の方法のいずれかを実施するための装置に関し、装置は、本発明の生物学的実体を単離する方法を実施するように設計された構成を含む。特に、装置は、容器のためのホルダ、液体の供給及び受け取りのためのレセプタクル、容器内に存在する液体内でビーズ(6)を上下に浮遊させることを可能にするように構成された、レセプタクルから容器に液体を供給及び受け取るためのデバイス、並びに容器の一方の開口部を通して液体を浸漬し、圧送し、排出し、ビーズ(6)が適所に保持されている間に液体を排出するための手段を含む。
IBA GmbHからのアガロースビーズなどの細胞単離のために市販されているビーズは、約30μm~約180μm、最大80~120μmの様々な直径を有するビーズの混合物でである(図1)。アガロースビーズの特定の画分を得るために、市販のアガロースビーズを適切なナイロンメッシュでふるい分けした。例えば、40~60μm(「小型ビーズ」)と140~180μm(「大型ビーズ」)との間のビーズ画分を、本発明に従って行われる以下の実験に使用した。
従来から適用されている細胞単離手順の結果に対する粒径の影響を評価するために、イムノアフィニティークロマトグラフィー(IAC)を、様々なサイズのアガロースビーズ画分に適用した。アガロースビーズ(細胞グレード、ドイツゲッティンゲンのIBA GmbH)を、実施例1に記載の標準サイズ(約80~120μm)及び小型画分又は大型画分で使用した。底部にフリットを有するプラスチックカラム(ドイツゲッティンゲンのIBA GmbH)を、Strep-Tactin(登録商標)及びCD81に対するFabでコーティングした0.5mlのアガロースを充填した。緩衝液CI(PBS、1mM EDTA、0.5%BSA)で1:1に希釈し、40μmナイロンふるいで濾過した5mlのヒト血液を同時にカラムにアプライした。図2から分かるように、結合細胞がカラムを通る流れを遮断するので、小型ビーズ(左カラム)はクロマトグラフィーカラムを流れることができない。標準サイズのビーズ(中央のカラム)は、速度を遅らせてクロマトグラフィーが可能であるが、大型サイズのビーズ(右のカラム)は、遅らせることなく血液を迅速に通過させることが可能である。標準サイズのビーズを使用すると、CD81+細胞を単離することができたが、大型ビーズを使用すると、1mMビオチンを含有する緩衝液CIでカラムを溶出した後、ビーズからCD81+細胞を本質的に単離することができなかった。
CD3+細胞を、抗体コーティングアガロースビーズを含む試薬チューブ中でヒトPBMCから非クロマトグラフィーにより単離した。特に、ビーズ直径が約40~60μmのStrep-Tactin(登録商標)コーティングアガロースビーズ(ドイツゲッティンゲンのIBA GmbH)、すなわち前述の「小型ビーズ」0.5mlを、30mlチューブ(ドイツNurnbrechtのSarstedt AG&Co.KG)中で22.5μgの抗ヒトCD3 Strepタグ付きFabフラグメント(ドイツゲッティンゲンのIBA GmbH)と混合し、揺動テーブルシェーカ(Unitwist RT、ドイツプラネグのUniEquip Laborgeratebau-und Vertriebs GmbH)上で可能な限り低い速度(100回/分)で5分間インキュベートした。ビーズを重力により沈降させた後、上清をピペッティングにより除去し、廃棄した。1mlのPBMC(1.57×107細胞)をCD3-Fab/アガロースビーズに添加し、シェーカ上で可能な限り低い速度で5分間インキュベートした。ビーズを2分間沈降させた後、上清を枯渇画分として回収した。ビーズを5mlの洗浄緩衝液(CI緩衝液
:PBS、0.5%BSA、1mM EDTA)で3回洗浄し、上清を洗浄画分として回収した。細胞溶出を、CI緩衝液(ドイツゲッティンゲンのIBA GmbH)中の5mlの1mMビオチンを2回使用して行った。ビーズをビオチンインキュベーション中に振盪下で5分間インキュベートし、上清を陽性画分として回収した。それにより、「ピペットマン」及び10若しくは5mlピペット又は1mlピペットチップを使用して上清を除去し、チップの上部に30μmの孔径を有するナイロンふるいを接着して、残った未沈降ビーズを遠ざけた。CD3-PeCy7及びCD45-eFluor450抗体(いずれも米国カリフォルニア州サンディエゴのeBioscience,Inc.製)で染色した後、CyAnTM ADPフローサイトメータ(米国カリフォルニア州ブレアのBeckman Coulter)を使用してFACS分析を行った。
クロマトグラフィー法として実施された図3に概略的に可視化された手順が生物学的実体の精製に最適ではないことを示すために、実施例3に記載されるCD3+細胞を単離するための本質的に同じ構成要素を使用し、FABian(登録商標)デバイス(ドイツライプツィヒのCell.Copedia GmbH)にアプライした。特に、CD3+細胞を、「標準モード」のFABian(登録商標)デバイスを使用して、すなわち、ビーズ直径90μm(+/-50μm)のStrep-Tactin(登録商標)コーティングアガロースビーズ(ドイツゲッティンゲンのIBA GmbH)0.5mlを用いて、図5C~図5Dに提供されている使用説明書(プログラムA)に従ってクロマトグラフィー法でバフィーコートから単離した。それにより、「標準モード」FABian(登録商標)機器を以下の、位置1については4.2mlの洗浄緩衝液(ドイツゲッティンゲンのIBA GmbH)中の22.5μgの抗ヒトCD3 Strepタグ付きFabフラグメント(IBA GmbH)、位置2については4.25mlのバフィーコート(洗浄緩衝液で1:1の比で希釈)、位置3、4及び10については空のチューブ、位置5、6、7及び8についてはそれぞれ9ml、6ml、9ml及び2mlの洗浄緩衝液、位置9及び11については6mlの1mMビオチン溶液のように調製した。FABian(登録商標)プログラムAを使用した。CD3-APC及びCD45-PerCP/Cy5.5抗体(いずれも米国カリフォルニア州サンディエゴのBioLegend製)を用いて、CytoFLEXフローサイトメータ(米国カリフォルニア州ブレアのBeckman Coulter)を使用してFACS分析を行った。CD3+細胞の濃縮を、図5に、示された画分のドットプロット(A)及び棒グラフ(B)として示す。実験は、最大51%の純度及び最大63%の収率で行った。

標準的な方法(Hermanson,G.T.(2008)Bioconjucate
techniques(2nded.),Elsevier)を使用して、デキストラン(MW500.000Da)をジビニルスルホンを用いてStrep-Tactin(登録商標)(IBA GmbH)に結合させることによって、Strep-Tactin(登録商標)/デキストランポリマーを得て、これをIBA GmbHのヒトのCD4 Fab Streptamer Isolation Kit MB(カタログ番号:6-8000-206)に含まれるCD4結合Fabフラグメントと共に5分間インキュベー
トした。未結合Fabフラグメントをゲルクロマトグラフィーによって除去した。驚くべきことに、FabフラグメントでコーティングされたStrep-Tactin(登録商標)/デキストランポリマーは、生物学的実体の結合に利用可能なビーズ表面に存在する抗原結合に関与しない十分な遊離抗体を有する。カラムクロマトグラフィーは、FABian(登録商標)デバイスを使用して、Twin Strep-Tag(登録商標)として公知のストレプトアビジン結合ペプチドに融合したCD4タンパク質の細胞外ドメイン45μgとプレインキュベートした予備充填1mlアガロースカラム(IBA GmbH)を用いて行った。この融合タンパク質を大腸菌(E.coli)で組換え生産し、標準的な方法を用いて精製した。次いで、Fabフラグメントの代わりに100μg(Strep-Tactin(登録商標)含有量に関連)のStrep-Tactin(登録商標)/Fabフラグメントでコーティングされたデキストランポリマーをカラムにロードしたことを除いて、IBA FABian(登録商標)マニュアルに記載されているように、FABian(登録商標)プログラムを開始した。次の工程では、マニュアルに記載されているように、CI緩衝液で1:1に希釈したバフィーコートの形態のヒト血液6mlをカラムクロマトグラフィーにかけ、未結合細胞をFACS緩衝液で洗い流した。次いで、マニュアルに記載されているように、1mMビオチンを含有するCI緩衝液で溶出した後、CD4+細胞を得た。PE/CD4抗体染色及び抗CD235染色を用いたCytoflex FACS装置(米国のBeckton&Dickinson)を使用してFACS分析を行った。
実施例5及び6に記載される方法の更なる展開として、「移動バッチモード」におけるStrep-Tactin(登録商標)/デキストランポリマーの使用を分析したが、CD4+細胞を単離するための本質的に同じ構成要素を実施例6に記載されるように使用した。特に、ヒトPBMCからCD4+細胞を精製するために、平均直径90μm(+/-50μm)の全血標準アガロースビーズを使用した。したがって、200μgのStrep-Tactin(登録商標)/デキストランポリマー及び200μgの抗ヒトCD4
Strepタグ付きFabフラグメント(IBA GmbH)を、底部に90μmのフリットを有する15mlカラム(中国のBiocomma)中の1mlのStrep-Tactin(登録商標)コーティングアガロースビーズ(ヒトCD4抗原、すなわちStrepタグ付きCD4抗原とプレインキュベーション)に添加した。デキストランポリマーを含まない参照単離を、FabをロードしたStrep-Tactin(登録商標)ビーズを用いた標準的な方法に従って行い(重力カラムによるIBA GmbH細胞単離を参照)、45μgの抗ヒトCD4 Strepタグ付きFabフラグメントを1mlのStrep-Tactin(登録商標)コーティングアガロースビーズに添加した。その後、5mlのPBMC(2.48×107細胞)及び10mlの全血をそれぞれカラムに添加し、カラムに上部キャップ及び底部キャップを置くことによって最低速度(毎秒1回転、タンブリングローラーミキサー、RM5、A.Hartenstein)で回転させながら10分間インキュベートした。上部キャップ及びカラムの出口のキャップを除去した後、未結合細胞を重力によって排出した。ビーズを10mlの洗浄緩衝液(ドイツゲッティンゲンのIBA GmbH)で2回洗浄した。ビーズを回転(低速タンブリング)下で5分間インキュベートし、上清を廃棄した。10mlの1mMビオチン溶液(IBA GmbH)を用いて溶出を行った。CD4-FITC及びCD45-APC抗体(いずれもBioLegend製)を用いて、CytoFLEXフローサイトメータ(Beckman
Coulter)を使用してFACS分析を行った。PBMCからのCD4+標的細胞の濃縮を、開始画分、洗浄/枯渇画分及び陽性画分のドットプロット(A)及び棒グラフ(B)として図9に示し、全血からのCD4+細胞の濃縮を図9C及びDに示す。CD45ゲーティングを使用して陽性画分のFACS分析から赤血球を除外した。Strep-Tactin(登録商標)/デキストランポリマーを使用する方法の収率は、デキストランポリマーを使用しない方法の陽性画分と比較して1.5倍高かった。
Strep-Tactin(登録商標)/デキストランポリマーを移動バッチモードと組み合わせて使用して、血液試料などの体液からエキソソームを単離した。特に、10mlのヒトバフィーコートを緩衝液CIで希釈し、3500gで10分間遠心分離して細胞及び細胞残屑を除去した。上清を、それぞれCD81抗原及びCD9抗原でコーティングしたアガロースビーズ、並びにそれぞれStrep(登録商標)タグ付きCD81 Fab及びCD9 Fabを負荷したStrep-Tactin(登録商標)/デキストランポリマーを使用することによって、「hCD14ショートプログラム」の下でIBA GmbHのウェブページ「Cell Selection&Expansion」に記載されているように移動バッチモードでFABian(登録商標)デバイスを使用してイムノアフィニティー精製に使用した。

Strep-Tactin(登録商標)/デキストランポリマーを使用するための更なる展開として、実施例6及び7に記載したものと比較して異なるビーズを使用した。特に、その表面に結合したビオチン残基を有するアガロースビーズが使用されている(ドイツゲッティンゲンのIBA GmbH、カタログ番号6-0446-000)。更に、Strep-Tactin(登録商標)/デキストランポリマーを使用し、デキストランは2.05 Mio Daの平均MWを有した(デンマークのPharmacosmos A7S)。この実験中、CD3 Strepタグ付きFabフラグメント(IBA GmbH)の使用によってCD3+細胞を単離し、実施例8に記載されているように本質的に同じ細胞単離のための構成要素を使用し、実施例7に記載されているように移動バッチモードを適用した。したがって、ヒトPBMCからCD3+細胞を精製するために、90μmの平均直径(+/-50μm)を有する上述のアガロースビオチンビーズ及びStrep(登録商標)タグ付きCD3 Fabを負荷したStrep(登録商標)/デキストランポリマーを使用した。特に、FABian(登録商標)デバイスで使用する前に、1mlのビオチンコーティングビーズを、穏やかに振盪しながら200μgのStrep-Tactin(登録商標)/デキストランポリマーと共に5分間インキュベートし、次いで、実施例8に記載されているように、FABian(登録商標)デバイスでの処理のために底部に90μmのフリットを有する15mlカラム(中国のBiocomma)に充填した。約200μgの抗ヒトCD3 Strep(登録商標)タグ付きFabフラグメント(ドイツゲッティンゲンのIBA GmbH)を、Strep-Tactin(登録商標)/デキストランポリマーに結合するための機器内で使用して、アガロースビーズ上のビオチン残基に結合していないStrep-Tactin(登録商標)上の遊離結合部位を占有した。
エキソソームを、デキストランが2.05Mio Daの平均MWを有していたStrep-Tactin(登録商標)/デキストランポリマー(デンマークのPharmacosmos A7S)を使用して、ヒト血清(血液バンクからの血液から得た若年男性ドナーからプールしたもの)から単離し、移動バッチモード及びビオチンコーティングアガロースビーズと組み合わせた。特に、0.22μmのポリエチレンスルホン(PES)フィルタ(ドイツゲッティンゲンのSartorius AG)で前濾過した4.25mlの血清をPBSで1:1に希釈し、12000×gで30分間遠心分離して細胞残屑を除去した。上清を、1mlのビオチンコーティングアガロースビーズ及びStrep(登録商標)タグ付きCD81 FabをロードしたStrep-Tactin(登録商標)/デキストランポリマーを使用することによって、実施例8に記載の移動バッチモードでFABian(登録商標)デバイスを使用するイムノアフィニティー精製に使用した。特に、FABian(登録商標)デバイスで使用する前に、穏やかに振盪しながらビーズを100μgのStrep-Tactin(登録商標)/デキストランポリマーと共に5分間インキュベートし、次いでFABian(登録商標)デバイスで処理するためにカラムに充填した。CD81に対する約50μgのStrep(登録商標)タグ付きFab抗体を、Strep-Tactin(登録商標)/デキストランポリマーに結合のための機器内で使用して、アガロースビーズ上のビオチン残基に結合していないStrep-Tactin(登録商標)上の遊離結合部位を占有した。

Claims (37)
- 試料から生物学的実体を単離する方法であって、
(i)前記生物学的実体(1)を含む試料を提供することであって、前記生物学的実体が表面抗原(2)を含むこと、
(ii)前記生物学的実体上の前記表面抗原(2)に特異的に結合することができる少なくとも1つの結合ドメイン(4)を含むタグ付け剤(3)を提供すること、
(iii)自由に移動可能なビーズ(6)を提供すること、
(iv)前記試料、タグ付け剤(3)及びビーズ(6)を容器内でインキュベートし、前記ビーズが前記試料内に浮遊している間に、前記生物学的実体(1)、前記タグ付け剤(3)及び前記ビーズ(6)の間の複合体形成を可能にすることであって、前記タグ付け剤(3)を介した前記生物学的実体(1)の前記ビーズ(6)への結合が、リガンド結合パートナー(7)及びリガンド(5)を含む非共有結合性タンパク質-リガンド相互作用によって直接的又は間接的に媒介されること、
(v)上清を廃棄しながら、前記ビーズ(6)を前記容器内の適所に一時的に保持すること、及び洗浄緩衝液を添加することを含むことによって、前記生物学的実体(1)を精製すること、並びに/又は
(vi)前記ビーズ(6)及び前記タグ付け剤(3)から生物学的実体(1)をそれぞれ放出し、前記ビーズ(6)を前記容器内の所定の位置に一時的に保持しながら、前記上清から前記生物学的実体(1)を回収し、場合により前記タグ付け剤(3)及び/又は前記ビーズ(6)の回収を可能にすることによって前記生物学的実体(1)を単離すること、を含む、方法。 - (a)前記結合ドメイン(4)が、抗原結合フラグメント(Fab)中に存在し、
(b)前記リガンド結合パートナー(7)が、ストレプトアビジン又はその機能的類似体若しくは誘導体を含み、前記リガンド(5)が、ストレプトアビジン結合ペプチドを含み、及び/又は
(c)前記ビーズ(6)が、非磁性ビーズ、好ましくはアガロースビーズである、請求項1に記載の方法。 - 前記リガンド結合パートナー(7)が、Strep-Tactin(登録商標)であり、前記リガンド(5)が、Strep(登録商標)-Tagである、請求項1又は2に記載の方法。
- 前記ビーズ(6)が、約30~100μm、好ましくは約40~60μmの直径を有することを特徴とする、請求項1から3のいずれか一項に記載の方法。
- 前記試料及び場合により任意の他の液体が、同じ開口部を通して前記容器に導入され、前記容器から排出され、及び/又は前記ビーズ(6)が、フリット又はふるいによって適所に保持される、請求項1から4のいずれか一項に記載の方法。
- 前記タグ付け剤(3)が、前記リガンド(5)に連結された抗原結合フラグメント(Fab)(4)を含み、前記ビーズ(6)が、前記タグ付け剤を前記ビーズ(6)に固定化する前記リガンド結合パートナー(7)を含む、請求項1から5のいずれか一項に記載の方法。
- 前記タグ付け剤(3)が、前記リガンド(5)に連結された抗原結合フラグメント(Fab)(4)を含み、前記ビーズ(6)が、更なるリガンド(12a)を含み、前記更なるリガンド(12a)及び前記リガンド結合パートナー(7)が、更なるタンパク質-リガンド相互作用を形成し、好ましくは前記更なるリガンド(12a)が、ビオチンである
、請求項1から5のいずれか一項に記載の方法。 - 前記タグ付け剤(3)が、前記リガンド(5)に連結された抗原結合フラグメント(Fab)(4)を含み、前記ビーズ(6)が更なるリガンド(12)を含み、前記リガンド結合パートナー(7)が、更なるリガンド結合パートナー(13)を含み、前記リガンド(12)及び前記リガンド結合パートナー(13)が更なるタンパク質-リガンド相互作用を形成する、請求項1から5のいずれか一項に記載の方法。
- 前記タグ付け剤が、非共有結合性タンパク質-リガンド相互作用によって担体(11)上に固定化され、好ましくは、前記非共有結合性タンパク質相互作用が、前記タグ付け剤(3)の前記リガンド(5)と前記担体(11)上の前記リガンド結合パートナー(7)との間で媒介される、請求項7に記載の方法。
- 前記担体が、約500kDa~3,000kDa、好ましくは約1,500kDa~2,500kDaの平均分子量を有するデキストランポリマーであり、前記リガンド結合パートナー(7)の少なくとも2つの分子を含み、前記リガンド結合パートナー(7)が、好ましくは、共有結合したストレプトアビジン又はその類似体若しくは誘導体であり、前記リガンド(5)を含む前記タグ付け剤(3)及び前記ビーズ(6)上の前記更なるリガンド(12a)に結合することができ、好ましくは、単離される前記生物学的実体(1)がエキソソームである、請求項7又は9に記載の方法。
- 前記タグ付け剤(3)が、前記タグ付け剤(3)によって認識される抗原(2’)及び前記非共有結合性タンパク質-リガンド相互作用のための構成要素を含む連結分子(10)を介して前記ビーズ(6)に固定化され、好ましくは前記構成要素が前記リガンド(5)であり、前記ビーズ(6)が前記リガンド結合パートナー(7)を含む、請求項1から5のいずれか一項に記載の方法。
- 前記タグ付け剤(3)が、互いに連結された少なくとも2つの結合ドメイン(4)を含むタグ付け剤(3’)である、請求項11に記載の方法。
- 前記方法が、前記表面抗原(2)、及び前記連結分子に含まれる前記組換え抗原(2’)を認識することができる少なくとも2つのタグ付け剤(3)を含み、前記タグ付け剤(3)が、非共有結合性タンパク質-リガンド相互作用によって担体(11)上に固定化され、好ましくは、前記非共有結合性タンパク質相互作用が、前記タグ付け剤(3)の前記リガンド(5)と前記担体(11)上の前記リガンド結合パートナー(7)との間で媒介される、請求項11に記載の方法。
- 前記タグ付け剤(3)が前記担体(11)に固定化され、請求項1において特徴付けられる工程(iv)の前に、前記連結分子(10)が前記ビーズ(6)に固定化される、請求項13に記載の方法。
- 前記担体が、約500kDa~3,000kDa、好ましくは約1,500kDa~2,500kDaの平均分子量を有するデキストランポリマーであり、前記リガンド結合パートナー(7)の少なくとも2つの分子を含み、前記リガンド結合パートナー(7)が、好ましくは、共有結合したストレプトアビジン又はその類似体若しくは誘導体であり、前記リガンド(5)を含む前記タグ付け剤(3)に結合することができる、請求項13又は14に記載の方法。
- 請求項1において特徴付けられる工程(vi)において、前記生物学的実体(1)が、競合リガンドを加えることによって放出され、場合により、これはとりわけ、前記抗原(

- 前記容器が、チューブ、バイアル、シリンジ、アンプル、又はカラムである、請求項1から17のいずれか一項に記載の方法。
- 試料から生物学的実体を単離する方法であって、
(i)前記生物学的実体(1)を含む試料を提供することであって、前記生物学的実体(1)が表面抗原(2)を含むこと、
(ii)抗原(2’)及び非共有結合性タンパク質-リガンド相互作用のための構成要素を含む連結分子(10)を提供することであって、好ましくは前記構成要素がリガンド(5)であること、
(iii)前記生物学的実体上の前記抗原(2)又は前記連結分子(10)に含まれる前記抗原(2’)に特異的に結合することができる少なくとも1つの結合ドメイン(4)をそれぞれ含む少なくとも2つのタグ付け剤(3)を提供することであって、好ましくは、前記タグ付け剤が、非共有結合性タンパク質-リガンド相互作用のための構成要素を更に含み、好ましくは、前記構成要素がリガンド(5)であること、
(iv)非共有結合性タンパク質-リガンド相互作用のための少なくとも2つの構成要素を含む担体(11)を提供することであって、好ましくは前記構成要素がリガンド結合パートナー(7)であること、
(vi)タンパク質-リガンド相互作用を形成することができる構成要素を含む固定相を提供することであって、好ましくは、前記構成要素がリガンド結合パートナー(7)であること、
(vii)前記試料、連結分子(10)、タグ付け剤(3)、担体(11)及び固定相を容器内でインキュベートし、前記生物学的実体(1)、前記タグ付け剤(3)、前記連結分子(10)、前記担体(11)及び前記ビーズ(6)の間の複合体形成を可能にすることであって、前記タグ付け剤(3)を介した前記生物学的実体(1)の前記担体(11)への結合及び前記タグ付け剤(3)を介した前記連結分子(10)の前記担体(11)への結合及び前記連結分子(10)の前記固定相への結合が、非共有結合性タンパク質-リガンド相互作用によって媒介され、前記生物学的実体(1)が前記固定相に固定されること、
(viii)クロマトグラフィー手順によって前記生物学的実体(1)を精製すること、並びに/又は
(ix)前記生物学的実体(1)を、それぞれ前記担体(11)及び前記タグ付け剤(3)から放出することによって、前記生物学的実体(1)を単離すること、を含む、方法。 - (a)前記タグ付け剤(3)が、前記リガンド(5)に連結された抗原結合フラグメント(Fab)(4)を含み、及び/又は
(b)前記リガンド結合パートナー(7)が、ストレプトアビジン又はその機能的類似体若しくは誘導体を含み、前記リガンド(5)が、ストレプトアビジン結合ペプチドを含む、請求項19に記載の方法。 - 前記リガンド結合パートナー(7)が、Strep-Tactin(登録商標)であり
、前記リガンド(5)が、Strep(登録商標)-Tagである、請求項19又は20に記載の方法。 - 前記タグ付け剤(3)が前記担体(11)に固定化され、請求項19において特徴付けられる工程(vii)の前に、前記連結分子(10)が前記固定相に固定化される、請求項19から20のいずれか一項に記載の方法。
- 前記担体が3,000,000Daの平均分子量を有するデキストランポリマーである、19から22のいずれか一項に記載の方法。

(ii)抗原(2’)及び非共有結合性タンパク質-リガンド相互作用を形成することができる構成要素を含む連結分子(10)を提供することであって、好ましくは前記構成要素がリガンド(5)であること、
(iii)互いに連結された2つの結合ドメイン(4)を含み、前記生物学的実体上の前記抗原(2)及び前記連結分子(10)に含まれる前記抗原(2’)に特異的に結合することができるタグ付け剤(3’)を提供することであって、好ましくは、前記結合ドメインが抗原結合フラグメント(Fab)であること、
(vi)タンパク質-リガンド相互作用を形成することができる構成要素を含む固定相を提供することであって、好ましくは、前記構成要素がリガンド結合パートナー(7)であること、
(vii)前記試料、連結分子(10)、タグ付け剤(3’)及び前記固定相を容器内でインキュベートし、前記生物学的実体(1)、前記タグ付け剤(3’)、前記連結分子(10)及び前記固定相の間の複合体形成を可能にすることであって、前記タグ付け剤(3’)及び前記連結分子(10)を介した前記生物学的実体(1)の前記固定相への結合が非共有結合性タンパク質-リガンド相互作用によって媒介され、前記生物学的実体(1)がビーズ(6)に固定化されること、
(viii)クロマトグラフィー手順によって前記生物学的実体(1)を精製すること、並びに/又は

- 請求項9から26のいずれか一項に記載の方法における請求項36に記載のデキストランポリマーの前記担体(11)としての使用。
