Abstract
Description

アデノウイルスベクターは、ゲノムからのウイルス遺伝子発現を担う初期(E1AおよびE1B)遺伝子を欠失させることによって複製不全にされ得、染色体外形態で宿主細胞中に安定に維持される。これらのベクターは、複製細胞および非複製細胞の両方にトランスフェクトする能力を有する。アデノウイルスベクターは、インビボで治療遺伝子の一過性発現をもたらすことが示されており、7日でピークに達し、約4週間持続する。加えて、アデノウイルスベクターは、非常に高い力価で産生され得、少量のウイルスを用いた効率的な遺伝子治療を可能にする。
組換えアデノ随伴ウイルス(rAAV)は、非病原性パルボウイルスに由来し、細胞免疫応答を本質的に誘起せず、大部分のシステムにおいて数ヶ月持続する導入遺伝子発現をもたらす。さらに、アデノウイルスと同様に、アデノ随伴ウイルスベクターもまた、複製細胞および非複製細胞に感染する能力を有する。
プラスミドDNAは、より入念なパッケージングシステムが存在しないために、しばしば「裸のDNA」と称される。インビボでの心筋細胞へのプラスミドDNAの直接注射が達成されている。プラスミドベースのベクターは、相対的に非免疫原性であり、非病原性であり、細胞ゲノム中に安定的に組み込まれる潜在力を有し、インビボでの有糸分裂後細胞における長期遺伝子発現をもたらす。さらに、プラスミドDNAは、血流中で迅速に分解され、したがって、遠位の器官系における導入遺伝子発現の見込みは無視できる。プラスミドDNAは、巨大分子複合体、例えば、リポソームまたはDNA-タンパク質複合体の一部として細胞に送達され得、送達は、エレクトロポレーションを含む技術を使用して増強され得る。
