Abstract
Description
(1)抗原の精製
抗原として利用したサルモネラ属菌由来の分子は、以下のとおりである。
SsaKタンパク質(226アミノ酸、分子量26162、菌体外へサルモネラのタンパク質を分泌させるIII型分泌装置の一部)(配列番号:1、2)
(i)SseGタンパク質(230アミノ酸、分子量24237、III型分泌装置より菌体外へ分泌されるタンパク質)(配列番号3、4)
(ii)SseFタンパク質(261アミノ酸、分子量26595、III型分泌装置より菌体外へ分泌されるタンパク質)(配列番号5、6)
(iii)LptDタンパク質(786アミノ酸、分子量89540、菌体膜タンパク質)(配列番号7、8)
(iv)LPS(Lipopolysaccharide)
Salmonella enterica serovar Typhimurium x3306株のDNAを鋳型に、表1に記載の各種プライマーを用いてPCR行った。
5mLのLB培地(アンピシリン入り)中で、一晩、前記サルモネラ属菌を前培養(静置培養)した。次いで、100mLのLB培地にて吸光度(600nm)が0.3になるまで振盪培養し、Isopropyl-β-D-thiogalactopyranosideを加えて、さらに6時間培養した。培養後、菌体を回収し、超音波処理により菌体を破砕した。次いで、破砕した菌体をUrea lysisバッファーにて溶解し、ニッケルカラムを用いて各種タンパク質を捕捉して精製し、imidazole溶出バッファーにて溶出した。
ニワトリ(白色レグホン)に対して、生後1週間目と3週間目に、Salmonella enterica serovar Typhimuriumまたは不活化ワクチン(熱不活化した死菌)を2回接種し、2週間後に血清を採取した。
0.5μg/mlに調整した抗原(組み換えサルモネラタンパク質)を37℃、1時間反応させ抗原を固相化した後、ブロッキングワン(ナカライテスク)を用いて室温で1時間ブロッキングした。次いで、固相化した各種タンパク質に対して、サルモネラ感染群およびワクチン接種群の血清をそれぞれ、37℃で1時間反応させ、さらに、2次抗体(HRP標識ウサギ抗ニワトリIgY抗体)を、37℃で1時間反応させた。次いで、基質としてのTMB(3,3’,5,5’-テトラメチルベンジジン)を添加して、HRP標識を発色させ、吸光プレートリーダー(450nm)を用いて、ニワトリIgYを定量した。
(1)ニワトリにおける組換えSsaKタンパク質を抗原としたエライザ法
III型分泌装置タンパク質であるSsaKタンパク質を抗原として用いて、ニワトリ血清に対してエライザを行った結果、サルモネラ属菌感染群においては、ワクチン接種群と比較して、血清中の抗体価が高かった(図1A)。一方、III型分泌装置より分泌されるSseGタンパク質、SseFタンパク質、膜タンパク質であるLptDタンパク質、およびSalmonella enterica serovar Typhimurium x3306由来のLPSを、それぞれ抗原として用いた場合には、サルモネラ属菌感染群とワクチン接種群との間で血清中の抗体価に有意差は認められなかった(図1B~E)。以上の結果から、SsaKタンパク質は、ワクチン接種個体とサルモネラ属菌感染個体とを区別できる抗原であることが判明した。
