Abstract
Description
(a)第1項~第4項のいずれか一項の組成物を甲状腺髄様がんの発症が疑われる個体に投与する段階、
(b)投与した組成物の診断用放射性核種の信号を検出する段階、及び
(c)検出された診断用放射性核種の信号を正常個体の信号と比較する段階。
(a)第1項~第4項のいずれか一項の組成物を甲状腺髄様がんが発症した個体に投与する段階と、
(b)投与した組成物の診断用放射性核種の信号を検出する段階、及び
(c)検出された診断用放射性核種の信号が甲状腺以外の部位で表示されるかどうかを確認する段階。
1-1.甲状腺組織及び甲状腺がん細胞株染色
甲状腺で嗅覚受容体(olfactory receptor)が発現するか確認するため、嗅覚マーカータンパク質(olfactory marker protein;以下、OMPという)の発現を確認した。正常甲状腺(thyroid gland)組織と甲状腺髄様がん(medullary thyroid cancer、MTC)細胞をOMP抗体(Rabbit anti-OMP;Santa Cruz、CA、USA)で染色した結果、甲状腺でOMPが発現することを確認でき、甲状腺を構成する細胞の中で濾胞傍C-細胞(parafollicular C-cell)のみでOMPが発現することが分かった(図1)。甲状腺髄様がんは、濾胞傍C-細胞に由来する悪性甲状腺腫瘍で、カルシトニンは、濾胞傍C-細胞のマーカータンパク質である。
甲状腺髄様がん細胞株であるMZ-CRC1及びTT、未分化甲状腺癌(anaplastic thyroid cancer)細胞株であるFRO及びSW1736、甲状腺乳頭がん(papillary thyroid cancer)細胞株であるTPC-1、甲状腺濾胞癌(follicular thyroid cancer)細胞株であるFTC-133をOMP抗体で染色した。
甲状腺がん細胞株で発現する嗅覚受容体の種類を確認するためにRNAを分離した後、cDNAを合成して当業界に知られている方法によってリアルタイム-ポリメラーゼ連鎖反応(real-time polymerase chain reaction、RT-PCR)を行った。
2-1.濾胞傍C-細胞に嗅覚受容体リガンド処理
甲状腺髄様がん細胞株でOR51E1及びOR51E2が多く発現するので、濾胞傍C-細胞株であるMZ-CRC1及びTT細胞株に嗅覚受容体リガンド(ligand)であるアセテート(acetate)、ノナン酸(nonanoic acid)、プロピオネート(propionate)及びアゼライン酸(azelaic acid)をそれぞれ100μMずつ処理してその影響を確認した。
8週齢の雌C57BL/6マウスから甲状腺を分離して組織切片を作製し、OR51E2、カルシトニン及びOMPをそれぞれの抗体で染色した。その結果、OR51E2、OMP及びカルシトニンが甲状腺組織で同じ部位に位置(co-localizaiton)することが分かった(図6)。
OR51E2遺伝子の位置に緑色蛍光タンパク質(GFP)が挿入されたC57/BL6マウスを購入(Jackson Laboratory、USA)して無菌条件で飼育した。一週間の純化期間を経た後、99%アセテート100μlを注射し、30分後にマウスを犠牲にして甲状腺及び血清を分離した。甲状腺は、カルシトニン及びGFP抗体でそれぞれ染色し、分離した血清では、カルシトニンの濃度を測定した。
3-1.11C-アセテート処理
起源が異なる甲状腺がん細胞株に放射性同位元素で標識された11C-アセテートを処理(20μCi)し、陽電子放出(positron emission)の程度を確認した。対照群としては、最もよく用いる放射性医薬品であるF-18フルデオキシグルコース(F-18-fluorodeoxyglucose)を処理した。
甲状腺髄様がん細胞株であるMZ-CRC-1及びTT細胞株にコールドアセテート(cold acetate;放射性同位元素で標識されないアセテート)100μMを処理した後、11C-アセテートを2、5、10、20、40及び60μCi濃度で処理してCPMを測定した。また、MZ-CRC-1及びTT細胞株に11C-アセテートを処理(20μCi)した後、コールドアセテートを0、20、40、80、150及び300μMの濃度で処理してCPMを測定した。
アセテートとOR51E2の結合が甲状腺がんの診断に活用できるのか確認するため、MZ-CRC-1細胞株(濾胞傍C-細胞)にアセテート100μMを処理し、1分、5分、及び30分後に細胞を染色してOR51E2を確認した。その結果、アセテート処理によってOR51E2が細胞内に移動することが確認できた(図10)。
Claims (10)
- 診断用放射性核種で標識された嗅覚受容体OR51E2のリガンドを含む、甲状腺髄様がん標的用組成物。
- 前記甲状腺髄様がんは、濾胞傍C-細胞(parafollicular C-cell)に由来するものである、請求項1に記載の甲状腺髄様がん標的用組成物。
- 前記診断用放射性核種は、炭素-11(C-11)、窒素-13(N-13)、酸素-15(O-15)、フッ素-18(F-18)、リン-32(P-32)、銅-64(Cu-64)、ガリウム-67(Ga-67)、ガリウム-68(Ga-68)、ルビジウム-82(Rb-82)、ジルコニウム-89(Zr-89)、テクネチウム-99m(Tc-99m)、インジウム-111(In-111)、ヨウ素-123(I-123)、ヨウ素-131(I-131)、キセノン-133(Xe-133)、タリウム-201(TI-201)及びトリウム-229(Th-229)からなる群から選ばれるものである、請求項1に記載の甲状腺髄様がん標的用組成物。
- 前記嗅覚受容体OR51E2のリガンドは、アセテート(acetate)、プロピオネート(propionate)、ノナン酸(nonanoic acid)、ベータ-イオノン(β-ionone)、アゼライン酸(azelaic acid)、エストリオール(estriol)、エピテストステロン(epitestosterone)、19-OH AD(19-hydroxyandrost-4-ene-3,17-dione)、パルミチン酸(palmitic acid)、アンドロステンジオン(androstenedione)、D-アラニル-d-アラニン(D-alanyl-d-alanine)、グリシルグリシン(glycylglycine)、コージビオース(kojibiose)、尿素(urea)、AFMK(N-acetyl-N-formyl-5-metoxykynurenamine)、ペラルゴニジン(pelargonidin)、ヒドロキシピルビン酸(hydroxypyruvic acid)、ブラジキニン(bradykinin)、8-ヒドロキシグアノシン(8-hydroxyguanine)、イミダゾロン(imidazolone)、2-ピロリドン(2-pyrrolidinone)、2-ケトグルタル酸(2-ketoglutaric acid)、L-グリセル酸(L-glyceric acid)、グリシン(glycine)または13-シスレチノイン酸(13-cis retinoic acid)であるものである、請求項1に記載の甲状腺髄様がん標的用組成物。
- (a)第1項~第4項のいずれか一項の組成物を甲状腺髄様がんの発症が疑われる個体に投与する段階と、
(b)投与した組成物の診断用放射性核種の信号を検出する段階と、
(c)検出された診断用放射性核種の信号を正常個体の信号と比較する段階と、を含む、甲状腺髄様がんの診断方法。 - 前記甲状腺髄様がんの診断方法は、(c)段階で前記検出された信号が正常個体の信号よりも高い場合、個体に甲状腺髄様がんが発症したと判断する段階をさらに含むものである、請求項5に記載の甲状腺髄様がんの診断方法。
- (a)第1項~第4項のいずれか一項の組成物を甲状腺髄様がんが発症した個体に投与する段階と、
(b)投与した組成物の診断用放射性核種の信号を検出する段階と、
(c)検出された診断用放射性核種の信号が甲状腺以外の部位で表示されるかどうかを確認する段階と、を含む、甲状腺髄様がんの転移有無を確認する方法。 - アセテート及びこれに結合された治療用放射性核種を有効成分として含む、甲状腺髄様がん治療用薬学的組成物。
- 前記治療用放射性核種は、炭素-11(C-11)、窒素-13(N-13)、酸素-15(O-15)、フッ素-18(F-18)、リン-32(P-32)、銅-64(Cu-64)、ガリウム-67(Ga-67)、ガリウム-68(Ga-68)、ルビジウム-82(Rb-82)、ジルコニウム-89(Zr-89)、テクネチウム-99m(Tc-99m)、インジウム-111(In-111)、ヨウ素-123(I-123)、ヨウ素-131(I-131)、キセノン-133(Xe-133)、タリウム-201(TI-201)、トリウム-229(Th-229)、イットリウム-90(Y-90)、ホルミウム-166(Ho-166)及びレニウム-188(Re-188)からなる群から選ばれるものである、請求項8に記載の甲状腺髄様がん治療用薬学的組成物。
- 請求項8または請求項9の甲状腺髄様がん治療用薬学的組成物を治療が必要な個体に投与する段階を含む、甲状腺髄様がんの治療方法。
