Abstract
Description
本出願は、2018年8月10日に特許出願された、米国仮出願第62/717,656号の利益を主張し、本明細書で全体として参照により組み込まれる。
本発明は、アメリカ国立衛生研究所(NIH)によって授与されたR21EB022652を受けて政府の支援で実行された。政府は本発明に一定の権利を有する。
インビボでは、腫瘍微小環境は、腫瘍細胞、内皮細胞等の血管細胞、および繊維芽細胞等の間質細胞を含む多数の細胞類を包含する複雑な環境である。さらに、インビボでは、これらの細胞は血流および様々な生物学的な送達条件にさらされる。インビボでは、腫瘍内の微小血管細胞は、血流の影響を受け、物理的、および拡散性要因によって、腫瘍細胞および非腫瘍細胞と連絡する。さらに、腫瘍血管系は異常であり、無秩序な分岐、低流量、および漏洩毛細血管を特徴とするため、腫瘍細胞を標的にする抗癌治療に対して主な送達障壁として機能する。腫瘍細胞と内皮細胞と間質細胞との相互作用は、各細胞の種類に影響を及ぼし、血管形成と腫瘍細胞増殖の増大させる。このクロストークは、抗癌剤に対する腫瘍細胞の反応性を求めるのに重要な要因になり得る。
ペプチドCRV(CRVLRSGSC)は、末端システイン残基間のジスルフィド結合を有する環状マクロファージ標的化ペプチドである。CRVは、選択的に腫瘍へ移動定着され、前記腫瘍内のTAMと結合する。CRVは、腫瘍内のTAMの表面上でRXRBを認識してそれに結合する。CRVは、TAMのみを認識し、アテローム動脈硬化性プラーク内のマクロファージを認識しない。
RAW(腫瘍由来のマウスのマクロファージ細胞株RAW264.7)、J774(腫瘍由来のマウスマクロファージ細胞株J774A.1)、THP-1分化マクロファージ(ヒト単球細胞株THP-1から分化されたヒトマクロファージ)および4T1(マウスの乳癌細胞株)の各細胞は、10%FCSを添加したDMEMにおいて培養された。1×106の細胞は、エッペンドルフチューブの中の完全増殖培地の300μLにおいてFAM-CRVまたは対照ペプチドFAM-GGS(10μM)でインキュベートされた。4°Cで1時間インキュベション後に、ペプチド含有培地は遠心分離によって取り除かれて、細胞はPBSによって2回洗浄された。その後、100μL のPFA(4%緩衝液)が固定のために細胞に加えられ、フローサイトメトリーデータはFACSCanto(BD Biosciences、サンホセ)にて得られた。実験は、別の日に3回繰り返された。
CRVがマクロファージにインビボで結合することができるかを調査するために、FAM-CRVは、担癌のマウスに静脈注射された。この目的のために、4T1およびMCF10CA1aのヒト乳癌細胞、およびKRASインクのマウスPDAC細胞は、10%ウシ胎仔血清、ペニシリン100U/mLおよびストレプトマイシン100μg/mLを添加したダルベッコ変法イーグル培地において培養された。Py8119細胞は、5%FCS、アンフォテリシンB2.5μg/mL、ゲンタマイシン50μg/mLおよびMITO+を含むHam’s F12K培地において培養された。ヒト細胞株はSanford Burnham Prebys Medical Discovery Institute(ラ・ホーヤ、カリフォルニア州)のDNA Analysis Core Facilityによって認証され、KRASインク細胞株はDDC Medical(フェアフィールド、オハイオ州)によって認証された。試験した全ての細胞では、マイコプラズマ汚染は陰性であった。4T1腫瘍を生成するために、1×106の腫瘍細胞(100μLのPBS中に懸濁された)が、正常なBALB/cマウスの乳房脂肪体に同所的に注入された。MCF10CA1a腫瘍を生成するために、2×106の腫瘍細胞(100μLのPB中に懸濁された)が、メスのBALB/c無胸腺ヌードマウスの乳房脂肪体に同所的に注入された。Py8119腫瘍を生成するために、1×106の腫瘍細胞(100μLのPBS中に懸濁された)が、C57BL6マウスの乳房脂肪体に同所的に注入された。KRASインクPDAC腫瘍を生成するために、1×106の細胞が、メスのBALB/cマウスに注入された。H1975腫瘍を生成するために、1×106の細胞(100μLのPBS中に懸濁された)が、注入された。(マウスの種類は? 注入した場所は?)動物実験の全ては、Sanford Burnham Prebys Medical Discovery InstituteのAnimal Research Committeeから承認を得た。

マクロファージがアテローム動脈硬化性プラーク等の他の疾患の部位に存在するため、CRVがマクロファージを含む他の病理組織を認識するかどうかを知ることが興味深い。したがって、FAM-CRVのインビボの移動定着は、大動脈内にアテローム動脈硬化性プラークを有するApoE-/-マウスにおいて試験された。p32受容体を介して腫瘍とプラークの両方に移動定着し、マクロファージに結合するLyP1と異なり、FAM-CRVは全身投与後に大動脈のプラークに移動定着せず(図4A)、このことはCRVがインビボでアテローム動脈硬化性プラーク内のマクロファージとTAMを識別することができると示唆した。大動脈の免疫蛍光染色では、さらにプラーク内のFAM-CRV蓄積が観察されなかった。単個細胞浮遊液もアテローム動脈硬化性プラークを有するマウス大動脈から得られ、インビトロの結合の研究は、FAM-CRVが、FAM-GGSと比較して、プラーク内のマクロファージに特異的に結合しないことを確認した(図4B)。この特徴は、CRVを疾患特異的な用途のためにマクロファージを標的とするために望ましいペプチドにする。
CRVとLyP-1の間の相違は、CRVがTAM認識のためにおそらく異なる受容体に結合することを示唆する。したがって、マクロファージ上のCRV受容体が同定された。複雑さを低減させるため、RAW細胞の細胞膜画分は単離され、実行されたアフィニティークロマトグラフィーは推定CRV受容体を単離するために実行された。この目的のために、CRVはカラム上で固定された。4T1腫瘍組織の溶解物およびRAW細胞の膜タンパク質(ThermoFisherからのMemPERPlusキットで単離された)は、2つの推定受容体の独立源として利用された。緩衝液および対照ペプチドGGSでの洗浄後に、推定受容体は過剰のフリーCRVペプチドで溶出された。その後、質量分析はレチノイドX受容体ベータ(RXRB)を受容体候補として同定した。

マクロファージとのCRV結合とRXRB表面発現との間の相関性をさらに調査するために、4T1腫瘍モデルから分離された腫瘍細胞に対するCRVおよびRXRB抗体のエクスビボの結合が実行された。約8mmの腫物サイズを備えたマウスは、深い麻酔下(アベルチン、つま先ピンチに対して無反応)で頸椎脱臼によって安楽死させられた。標的(腫瘍)および対照(例えば肝臓、脾臓)組織は収集され、さらに単細胞に分離された。腫瘍細胞は、MACS腫瘍分離キットを使用して分離された。その後、細胞は、蛍光標識CRVまたは他の細胞マーカー(CD11b、CD68、F4/80)で4°Cで1時間インキュベートされた。陽性細胞は、BD LSRFORTESSAにて定量化され、データはFCS Express Version 3(De Novo Software)を使用して分析された。
RXRBとCRVの間のよい相関性は、インビボのCRV体内分布を説明する可能性がある、異なる組織内のRXRB表面発現を調査する関心の高まりにつながる。RXRB抗体(または対照としてのウサギIgG)およびFAM-CRVの両方は、同じマウスに静脈注射され(RXRB抗体:4時間の循環; FAM-CRV:1時間の循環)、RXRBの提示は、経心腔的かん流後に評価された。組織切片は、0.1%トリトンX-100を備えた1%ウシ血清アルブミンで1時間ブロックされ、適切な一次抗体および二次抗体とインキュベートされた。血管は、CD-31に対するモノクローナル抗体によって組織切片の染色により視覚化された。一次抗体は、ネズミ抗マウスCD31CD11b(BD Biosciences)、ネズミ抗マウスCD11b(BD Biosciences)、ネズミ抗マウスF4/80モノクローナル(BD Biosciences)およびウサギ抗フルオレスセイン/オレゴン・グリーン(Invitrogen)ポリクローナル抗体であった。二次抗体、アレクサ・フルーアー488ヤギ抗ウサギIgGおよび594ロバ抗ネズミIgGは、Invitrogenからであった。PBSで洗浄された後、切片はDAPI含の封入剤(Vector Laboratories、バーリンゲーム、カリフォルニア州)に埋め込まれ、ツァイスLSM 710 NLO共焦点顕微鏡で検査された。
50μgのウサギ抗マウスRXRB抗体(GeneTex、PBSで希釈され、全容積は100μLであった)は、各々のマウス(n=3)に静脈注射された。マウス当たり50μgのウサギIgGは、対照群(n=3)に注入された。3時間後に、FAM-CRV(100μL内の100μg)は全ての動物に注入された。FAM-CRVの1時間の循環後、動物は経心臓的かん流によって殺された。組織は、上記の通りに収集され、固定されてスライスされた。
CRVを標的とするアプローチの翻訳の可能性を調査するために、ナノ粒子のCRVを介した腫瘍への移動定着は評価された。様々な種類の薬物の制御された負荷および放出を備えた治療キャリアーとして多孔質シリコンナノ粒子(pSiNP)を示す広範囲な研究があった(24、25)。それらの薬物負荷の高い効率および時間ゲート画像化の特性は、それらを薬物キャリアーとして特に望ましくする。
