本発明に係るナノ粒子は、高分子ナノシステムを形成している。高分子ナノシステムは、ヒアルロン酸(HA)およびポリL-アルギニンから調製される。高分子ナノシステムにより、白金系薬物(特に、ジアクア(1,2-ジアミノシクロヘキサン)白金(II)などの白金(II)系薬物)が封入される(または会合する)。
上記のように、本発明に好適な白金系薬物としては、白金(II)系薬物、白金(IV)系薬物、およびこれらの混合物が挙げられる。
カルボプラチンでは、シスプラチンに存在していた2個の塩素原子が、酸素を含んでいるリガンドに置換されている。このリガンドは、C3ジカルボン酸に由来しており、C3ジカルボン酸の中央の炭素原子は、3個の炭素原子とともにシクロブチル環を形成している。
従来、ポリL-アルギニンは、塩酸塩型(すなわち、ポリL-アルギニン塩酸塩(PArg-Cl))が使用されている。多くの場合、ポリL-アルギニン塩酸塩は、Alamanda (R) Polymersが販売しているものである(Mwは5kDa~100kDa、好ましくは5kDa~15kDa、または好ましくは50kDa~100kDa、または好ましくは20kDa~50kDa)。
ヒアルロン酸(ヒアルロナンまたはヒアルロナートとも称する)は、N-アセチル-D-グルコサミンおよびD-グルクロン酸を含んでいる二糖ユニットから構成されるアニオン性多糖である。人体(生物流体および生体組織)に天然に存在し、薬学において広汎に使用されている。

上述した通り、本発明は、本発明に係るナノ粒子の製造方法に関する。この製造方法は、少なくとも、以下の工程を含む。
(i)塩素原子を含んでいないアクア錯体形態の白金系薬物を提供する工程
(ii)塩化物イオンを含んでいないポリL-アルギニンの水溶液を提供する工程
(iii)工程(i)のアクア錯体形態の白金系薬物と、工程(ii)の水溶液とを混合する工程
(iv)ナノ粒子の形成に適した条件下にて、工程(iii)で得られた混合物に、ヒアルロン酸を加える工程

工程(i)では、塩素原子を含んでいないアクア錯体形態の白金系薬物を提供する。
工程(ii)は、塩化物イオンを含んでいないポリL-アルギニンの水溶液の提供からなる。
工程(iii)は、工程(i)の塩素原子を含んでいない白金系薬物のアクア錯体と、工程(ii)の塩化物イオンを含んでいないポリL-アルギニンの水溶液と、の混合に相当する。
工程(iv)では、ヒアルロン酸(例えばLifecore Biomedicalが販売しているHA(LMWHA(Mw≒20kDa))を、工程(iii)で得られた混合物に添加し、所望のナノ粒子を得る。
工程(v)は任意工程であり、工程(iv)で得られるナノ粒子の単離からなる。
実施例に例示されるように、本発明で定義されるナノ粒子は、癌を予防および/または治療できる。癌の例としては、膵臓癌(特に、膵管腺癌(PDAC))、大腸癌、肺癌、小細胞肺癌および非小細胞肺癌、卵巣癌、精巣癌、乳癌、脳腫瘍、肉腫、リンパ腫、頭頸部癌、転移性大腸癌、胃癌、卵巣癌、食道癌、膀胱癌、子宮頸癌、白血病(慢性骨髄性白血病など)、前立腺癌、肝臓癌、結腸癌、腎臓癌、皮膚癌、骨肉種、子宮癌、リンパ腺癌、胃癌、腸癌が挙げられる。
本発明のもう一つの態様によれば、本発明は、本発明に係るナノ粒子を有効成分として含んでいる医薬組成物に関する。より詳細には、これらの医薬組成物は、本発明に係るナノ粒子の効果的な用量と、薬学的に許容可能な添加剤の1種類以上とを含有している。
例えば、被験体の循環系に組成物を注射するなどして、医薬組成物を静脈内投与する場合が挙げられる。

高用量または低用量が適切である特定の症例はありうる。このような用量であっても、本発明の範囲を外れるものではない。通常の慣行においては、各患者にとって適切な用量は、投与方法ならびに患者の体重および反応に応じて医師が決定する。
癌を予防および/または治療するために、本発明に係るナノ粒子を単独療法に使用することができる。それを必要とする患者または被験体が、他の特定の抗癌剤に対する耐性を有しているか否かにかかわらず、単独療法は好ましく推奨される。
他の態様によれば、本発明は、上述した病理に対する処置方法を提供する。この方法は、本発明に係るナノ粒子の効果的な用量を患者に投与する工程を含む。任意構成で、上記に定義された他の抗癌剤および/または他の有効成分と組合せて投与してもよい。
[A-1.化学物質]
ジクロロ(1,2-ジアミノシクロヘキサン)白金(II)(DACHPtCl2、Mw=380.17Da)、硝酸銀AgNO3(Mw≒169.97Da)、マンニトール(Mw=182Da)およびN1培地サプリメント(100×)は、Sigma Aldrich(Saint Quentin Fallavier, France)から購入した。N1培地サプリメント(100×)には、0.5mg/mLの組換えヒトインスリン、0.5mg/mLのヒトトランスフェリン(一部鉄飽和)、0.5μg/mLの亜セレン酸ナトリウム、1.6mg/mLのプトレシン、および0.73μg/mLのプロゲステロンが含まれている。
(A-2-1.ナノ粒子の物理化学的キャラクタライゼーション)
実験に使うナノ粒子の粒径分析およびゼータ電位の測定には、Malvern Zetasizer (R) apparatus DTS 1060(Nano Series ZS, Malvern Instruments S.A., Worcestershire, UK)を用いた。脱イオン水で1/60に稀釈したナノ粒子分散液を、25℃にて3回測定した。
封入効率(EE)は、間接法で特定した。DACHPtを担持したサンプルを、Amicon Ultra-filter遠心分離チューブ(カットオフ:30kDa、Merck Millipore, Cork, Ireland)に入れて遠心分離した(7000g、30分間、20℃)。下層の溶液の濃度を、ICP-OES(誘導結合プラズマ発光分光法)(ICP-OES ICAP 7200 duo Thermo Scientific)により測定した。
ナノシステムを凍結乾燥するために、HAおよびPArgを10%w/vマンニトール溶液に溶解させて、ブランクナノ粒子およびDACHPtを担持したナノ粒子をそれぞれ調製した。マンニトール10%(w/w)を凍結防止剤として選択したのは、ナノ粒子の物理化学的特性を最もよく維持できたためである(データ省略)。凍結乾燥工程は、RZ6 Vacubrand pomp(Fisher Scientific, Illkirch, France)を備えたALPHA 1-4 LSC(CHRIST)凍結乾燥機により実行した。凍結乾燥させた粉末を水中に再懸濁した後、粒径、多分散度、ゼータ電位および封入効率を評価した。
fSPT(Fluorescence Single particle tracking)により、緩衝液中および血清中におけるナノ粒子の拡散をキャラクタライズした。
ブランクナノ粒子およびDACHPtを担持したナノ粒子を、ヒアルロン酸(HA)およびポリアルギニン(PArg)から調製した。調製に際しては、以下に説明するイオン向性ゲル化法を用いた。
DACHPtCl2(5mM、9.504mg)を蒸留水で懸濁し、硝酸銀(AgNO3)(4.24mg)と混合して([AgNO3]/[DACHPt]=1)、DACHPtのアクア錯体を形成させた。25℃にて暗所に24時間、磁気攪拌しながら溶液を保持した。反応後、塩化銀(AgCl)の沈澱が見られた。混合物を2000gにて20分間遠心分離し、AgClの沈澱を除去した。その後、0.22μmフィルタによる濾過で上清を精製して、無菌性を確保した。
PArg-Clをイオン交換樹脂(Amberlite (R) IRA 900 Cl)で処理して、PArgを調製した。簡潔に説明すると、1mLの湿潤状態の樹脂が充填されているカラムに、3mLのNaOH(1M)を通した。30分後、溶液のpHが中性になるまで、精製水でカラムをリンスした。次に、1mLのPArg-Cl溶液(50mg/mL)をカラムの頂端から投入し、PArg-OHを末端から回収した。その後、溶液が目的の濃度(12.5mg/mL)に達するまで、2~3mLの精製水でリンスした。体積を測定して、12mg/mLになるまでの稀釈度を確認した。
ブランクナノ粒子(白金系薬物を担持していないナノ粒子)は、Oyarzun-Ampuero et al (Eur J Pharm Biopharm Off J Arbeitsgemeinschaft Fuer Pharm Verfahrenstechnik eV. 2011;79(1):54-57)に開示されている方法を改変して得た。
DACHPtを担持したナノ粒子を得るために、実施例2で得たPArg-OH溶液に、水に代えて実施例1で得たDACHPt溶液500μLを加え、何も担持していないナノ粒子について記載した実施例3と同様の工程を施した。詳細には、実施例1で得られたDACHPt溶液を、実施例2で得られたPArg-OH溶液と混合し、攪拌しながら10分間放置した。DACHPt-PArg-OH溶液にHAを加えることにより、DACHPtを担持したナノ粒子を形成させた。
PArg-HAナノ製剤を標識する手段として、Alexa Fluor 647カルボン酸を使用した。
HA溶液(濃度:9~11.25mg/mL)とDACHPt溶液とを混合して、PArgの存在がナノ粒子の形成に必要か否かを調べた。
凍結乾燥させた種々の製剤(異なるHA/PArgの比率で調製されている)の、DACHPt封入効率を測定した。
粒径およびゼータ電位に関するナノ粒子の安定性は、凍結乾燥後に測定した。測定方法は、上述の通りである。
生物流体中のタンパク質の存在によりDLS測定中に光が散乱され、その結果、ナノ粒子の特性評価が妨げられる場合があることが実証されている。生物流体中におけるナノ粒子の物理化学的性質を調査する際にはDLSは採用し難い技術であることを鑑みて、稀釈されていない生物流体(ヒト血清、血液、腹水など)中におけるナノシステムの粒径を測定する有力な方法として、SPTが提案された。
[G.1.細胞培養]
(細胞株)
ヒト肺胞癌A549細胞(Prof. L. Migliore, University of Pisaよりご提供いただいた)を、10%ウシ胎仔血清(FBS)、1mMのGlnおよび抗生物質を添加したHam's F12培地中で増殖させた。ヒト大腸腺癌HT-29細胞(Prof. P. Petronini, University of Parmaよりご提供いただいた)を、10%FBS、2mMのGlnおよび抗生物質を添加したDMEM高グルコース(4.5g/L)中で増殖させた。これらの細胞を、37℃、5%CO2存在下にてインキュベートした。全ての細胞は、解凍後10継代を経ないうちに使用した。
(B6KPC3細胞、A549細胞およびHT29細胞の生存性)
3.5×103個のB6KPC3細胞およびA549細胞、ならびに7.5×103個のHT-29細胞を96ウェルプレートに播種した。24時間後、1%NEAA(Non Essential Amino Acids 100x, Gibco, Monza, Italy)、1mMのピルビン酸ナトリウム(Sigma-Aldrich, Milan, Italy)、1%N1培地サプリメント(0.5mg/mLの組換えヒトインスリン、0.5mg/mLの一部鉄飽和ヒトトランスフェリン、0.5μg/mLの亜セレン酸ナトリウム、1.6mg/mLのプトレシンおよび0.73μg/mLプロゲステロンを含有している;Sigma-Aldrich)を添加した、FBSを含んでいない新しい培養培地に交換した。このとき、市販のオキサリプラチン(Oxaliplatin Hospira (R)の5mg/mL溶液)、DACHPt、ブランクHAPArg-NPおよびDACHPtを担持したHAPArg-NPの濃度を、種々に変化させて(0~200μM)培地に加えた。24時間後、MTS Cell Proliferation Colorimetric Assay Kit(Novus Biologicals, Abingdon, UK)を用いて細胞生存性を評価した。簡潔に説明すると、培地を除去し、無血清培地の中で1:10に稀釈したMTS溶液を100μLずつウェルに注入した。予備実験において、各細胞型の代謝に応じたMTSへの曝露時間を決定しておいた(HT-29細胞:1時間、A549細胞:3時間)。マイクロプレートリーダ(EnSpire (R) Multimode Plate Reader, Perkin Elmer, Boston, MA, US)を用いて、492nmの吸光度を測定した。
CV(%)=(吸光度(処理ウェル))/(吸光度(対照ウェル))×100
薬物を含まない培地中でインキュベートした未処理細胞の吸光度を、吸光度(対照ウェル)とした。
B6KPC3細胞(図2A)HT-29細胞(図2B)およびA549細胞(図2C)を48時間インキュベートした。この際、オキサリプラチン(Oxaliplatin Hospira (R) 5mg/ml)、DACHPt、ブランクナノ粒子およびDACHPtを担持したNPの濃度を、種々に変化させた(0~200μM)。したがって、オキサリプラチン、DACHPtおよびブランクナノ粒子は、対照として使用されている。次に、MTSアッセイにより生存性を評価した(材料および方法の欄を参照)。対照(未処理細胞)に対する百分率(%)でデータを表す。3者の測定のいずれも、データは、2回の実験の平均値±標準偏差である(各実験で3回の測定を行った)。
(A.材料および方法)
健康なマウス(8週齢の雌、C57BL/6)に、200μLのDACHPtを担持したナノ粒子およびオキサリプラチン溶液を、マウス1匹あたりに投与する白金当量が35.9μgとなるようにIV注入した。
・1日目、4日目、8日目、11日目、15日目、18日目、22日目および25日目にIV点滴(2時間)
・1週間あたり合計2用量を4週間投与
いくつかの薬物について、静脈内ボーラス注入後の血漿濃度-時間の曲線は、明らかな二相性パターンを示した。この二相性パターンは、白金誘導体が様々な組織に広範囲に分布した後、最終的に体内から排出された結果である。得られたプロファイル(図3Aおよび3B)に基づいて、2コンパートメントモデルを用いてデータを分析した。DACHPtを担持したナノ粒子に由来するPt濃度およびDACHPt濃度を評価・比較したところ、その結果は同程度であった。最後に、オキサリプラチン溶液に由来するPt濃度、およびDACHPtを担持したナノ粒子に由来するDACHPt濃度を、時間の関数としてプロットした。
(PArgOHおよびPArgCl(混合系)-HAナノ粒子を得るためのプロトコル)
PArgClに由来するClイオンの影響を調べるために、PArgOHおよびPArgCLを異なる質量比(または重量比)で含んでいるナノ粒子を作製した。
(シスプラチンを担持したHA-PArgナノ粒子を得るためのプロトコル)
シスプラチンを担持したHa-PArgナノ粒子を得るために、上述したDACHPtナノ粒子の作製と同様のプロトコルを施した。DACHPtと同じ濃度のシスプラチンをParg-OH溶液と混合した後、上記と同じプロトコルを施した。ナノシステムを10%w/vマンニトールの存在下にて調製し、凍結乾燥させた。再懸濁した後、カプセル化効率を上記のプロトコルと同様に評価した。
(3D細胞モデル:MCTSの形成および処理)
多細胞腫瘍スフェロイド(MCTS)を、既報に従って形成させた(Virgone-Carlotta A, Lemasson M, Mertani HC, et al. In-depth phenotypic characterization of multicellular tumor spheroids: Effects of 5-Fluorouracil. PLOS ONE. 2017;12(11):e0188100. doi: 10.1371/journal.pone.0188100)。簡潔に説明すると、細胞と基板との接着を避けるためにUltra Low Attachment (ULA) 96 wells Round-Bottom plate(Greiner bio-one)中にて、HTC-116細胞株を用いてMCTSを形成させた。細胞をトリプシン処理し、1mLあたり2400個の細胞となるように、Malassez gridを用いて計数した。この細胞濃度(体積200μLのウェルあたり480個の細胞)は、実験の終了時において、1ウェルあたり1個の直径500μm未満のスフェロイドが存在するように選択した。1200g、室温にて、5分間プレートを遠心分離し、スフェロイドの形成を開始させた。実験の全過程にわたり、37℃、5%CO2存在下のインキュベータにプレートを置き、振盪し続けた。播種後第1日目の終わりに100μLの培養培地を加え、3次元的に適切に成長させた。播種から2日後、オキサリプラチン水溶液またはDACHPtを担持したナノ粒子(NP)を用いてMCTSを処理した。薬物の濃度は、5μM、25μMまたは50μMである。3日間にわたり処理を更新した。結果を図4~6に示す。
倒立顕微鏡(Leica DMIRB)で、96ウェルプレート内のMCTSの位相差写真を撮影した。撮影時期は、オキサリプラチンに曝露してから0日目、1日目、2日目、3日目、4日目、7日目の時点である。Image J softwareを利用して、Soble閾値により各スフェロイドをエッジ検出した。ImageJ "Analyse Particles" pluginを利用して、得られた二値画像を、長軸(LM)および短軸(Lm)を有する楕円に近似した。これにより、平均直径Dを計算した(D=(LM+Lm)/2)。次に、スフェロイドを球体であると仮定し、体積Vを求めた(V=πD3/6)。結果を図7に示す。
より生体に類似したin vitro培養系におけるナノ粒子の腫瘍生存に対する影響を検討した。そのために、オキサリプラチン溶液およびDACHPtを担持したNPの用量に対する反応を比較した。3日間にわたり処理を施し、HCT-116細胞株由来の多細胞腫瘍スフェロイド(MCTS)に対する影響を調査した。位相差顕微鏡像に基づいてMCTSの体積を評価し、これを細胞毒性効果の出力とした。図4~6のデータは、種々の処理に関して、対照の体積(未処理のMCTS)で正規化したMCTSの体積を示す。3種類の異なる薬物濃度(5μM、25μMおよび50μM)を検討した。
Claims (15)
- 薬物送達システムとして有用なナノ粒子であって、少なくとも下記(a)~(c)から形成されており、
(a)白金系薬物;
(b)ポリL-アルギニン;
(c)ヒアルロン酸;
上記白金系薬物は、白金錯体(II)、白金錯体(IV)、およびそれらの混合物から選択される白金錯体である、ナノ粒子。 - 上記(b)ポリL-アルギニンは、ポリL-アルギニン水酸化物、ポリL-アルギニン塩酸塩、およびポリL-アルギニン水酸化物とポリL-アルギニン塩酸塩との混合物から選択される、請求項1に記載のナノ粒子。
- 上記白金系薬物は、ジアクア(1,2-ジアミノシクロヘキサン)白金(II)、オキサリプラチン、カルボプラチン、ネダプラチン、ロバプラチン、ヘプタプラチン、cis-ジアンミンジアクア白金(II)、ジアクア(1,2-ジアミノメチルシクロブタン)白金(II)、ジアクア(4,5-ジアミノメチル-2-イソプロピル-1,3-ジオキソラン)白金(II)、およびそれらの混合物から選択される白金(II)系薬物であり、
好ましくは、ジアクア(1,2-ジアミノシクロヘキサン)白金(II)である、
請求項1または2に記載のナノ粒子。 - 上記(a)白金系薬物は、塩素原子を含んでいない形態で存在し、
上記(b)ポリL-アルギニンは、ポリL-アルギニン水酸化物であり、
好ましくは、上記(a)白金系薬物は、ジアクア(1,2-ジアミノシクロヘキサン)白金(II)である、
請求項1~3のいずれか1項に記載のナノ粒子。 - 上記(a)白金系薬物は、塩素原子を含んでおり、
上記(b)ポリL-アルギニンは、ポリL-アルギニン塩酸塩である、
請求項1に記載のナノ粒子。 - [HA]/[Parg]の重量比が、0.5/2.5超であり、
好ましくは(0.6/2.5)~(15/2.5)であり、
より好ましくは(3/2.5)~(12/2.5)であり(例えば3/2.5、4/2.5、7/2.5、9/2.5、10/2.5、11.25/2.5および12/2.5)、
さらに好ましくは11.25/2.5である、
請求項1~5のいずれか1項に記載のナノ粒子。 - [白金系薬物]/([HA]+[PArg])の重量比が、0.01~1.00であり、
好ましくは0.03~0.50であり、
より好ましくは0.04~0.10である、
請求項1~6のいずれか1項に記載のナノ粒子。 - 請求項1~7のいずれか1項に記載のナノ粒子の製造方法であって、少なくとも下記(i)~(v)の工程を含む方法:
(i)塩素原子を含んでいないアクア錯体形態の白金系薬物を提供する工程;
(ii)塩化物イオンを含んでいないポリL-アルギニンの水溶液を提供する工程;
(iii)工程(i)の上記アクア錯体形態の白金系薬物と、工程(ii)の上記水溶液とを混合する工程;
(iv)ナノ粒子の形成に適した条件下にて、工程(iii)で得られた混合物に、ヒアルロン酸を加える工程;
(v)任意構成で、工程(iv)で得られたナノ粒子を回収する工程。 - 上記アクア錯体形態は、ジアミンジアクア白金(II)、ジアクア(1,2-ジアミノシクロヘキサン)白金(II)(DACHPt)、ジアクア(1,2-ジアミノメチルシクロブタン)白金(II)、ジアクア(4,5-ジアミノメチル-2-イソプロピル-1,3-ジオキソラン)白金(II)、およびそれらの混合物から選択され、
好ましくは、ジアクア(1,2-ジアミノシクロヘキサン)白金(II)である、
請求項8に記載の方法。 - 工程(ii)の上記塩化物イオンを含んでいないポリL-アルギニンは、ポリL-アルギニン塩酸塩から得られる、請求項8に記載の方法。

- 請求項8~11のいずれか1項に記載の方法によって得られるナノ粒子。
- 請求項1~7および12のいずれか1項に記載のナノ粒子の1種類以上と、
薬学的に許容可能な添加剤の1種類以上と、
を含んでいる医薬組成物。 - 癌の予防および/または治療に用いる、請求項1~7および12のいずれか1項に記載のナノ粒子。
- 上記癌は、膵臓癌(特に、膵管腺癌(PDAC))、大腸癌、肺癌、小細胞肺癌および非小細胞肺癌、卵巣癌、精巣癌、乳癌、脳腫瘍、肉腫、リンパ腫、頭頸部癌、転移性大腸癌、胃癌(gastric cancer)、卵巣癌、食道癌、膀胱癌、子宮頸癌、白血病(慢性骨髄性白血病など)、前立腺癌、肝臓癌、結腸癌、腎臓癌、皮膚癌、骨肉種、子宮癌、リンパ腺癌、胃癌(stomach cancer)および腸癌から選択される、請求項14に記載のナノ粒子。
