本発明は、血液溶解物中の共有結合した化合物によるリアルタイムインビボ受容体占有率を直接測定するための方法に関する。より具体的には、本発明は、臨床薬の開発のための薬力学的バイオマーカーとしてのリアルタイムインビボBTK受容体占有率の測定に関する。
創薬と開発は、長期かつ高リスクのプロセスである。成功した新しい薬を開発するための平均コストは、早期の発見と開発プロセス間の数えきれない薬物候補の失敗について260億ドルとも推定され、臨床試験に入る薬物候補の全生存率は、12%未満と推定されている(PhRMA Report, "Biopharmaceutical Research & Development: The Process Behind New Medicines", 2015)。これらの薬物候補の失敗を生じる主な問題は、後期開発段階、特に、概念実証(第II期)臨床試験間における臨床的有効性が不十分であるか、または欠如することに関するものであった(Kola, I., et. al., J. 本発明は、本発明の好ましい実施態様の下記の詳細な説明および本明細書に含まれる実施例を参照することによってより容易に理解されうる。
本発明の1の実施態様において、前記目的の化合物は、前記受容体の少なくとも1つのアミノ酸に不可逆的に結合する。目的の化合物の一例は、BTKに不可逆的に結合する図1Aで示される化合物である。
受容体占有(RO)アッセイ開発の選択および感度に影響を与える多くの試行が行われた。特に、血液試料中の遊離の薬物および遊離の受容体の存在は、溶解工程の間に生体外で薬物が結合した受容体を生成しうるため、ROの過大評価を生じうる。溶解工程の間に受容体特異的なクエンチャーを加えることは、遊離の受容体を、クエンチャーが結合した(QB)受容体に急速に変換し、それにより生体外において薬物が結合した受容体の形成を妨げることが知見された。
免疫捕捉は、標的タンパク質および捕捉薬剤の特有の免疫親和力を利用する高度に選択的な試料浄化方法である(Stevenson, L., et. al., Bioanalysis 2013, 5, 2903-2918)。内在性標的のために高い免疫捕捉効率を有する適切な捕捉抗体を選択することは、そのアッセイを成功させるために重要である。市販品として入手できない特異的な抗体の場合、それらの標的タンパク質に特異的な抗体を調製する方法は、当業者に公知である。
ROアッセイで使用するために調製した受容体サロゲートペプチドは、目的の化合物の結合部位を含んでいる必要がある。また、前記サロゲートペプチドは、LC−MS検出に感度を有する必要がある。
本発明の1の実施態様において、受容体占有率は、クエンチャーが結合し、かつ薬物が結合したサロゲートペプチドを、1回のLC−MS/MSランで同時に測定することによって決定することができる。
DB−QRPおよびQB−QRPのための定量化の下限値(LLOQ)は、標準曲線における両方の分析物について最も低い濃度である0.250nMおよび0.125nMであった。図8bおよび8eに示されるように、LLOQ濃度レベルにおけるDB−QRPまたはQB−QRPのシグナル対ノイズ(S/N)は、少なくとも3またはそれ以上高かった。
血液溶解物中のBTK ROを定量化するための方法の検証について、全ての標準曲線を、DB−BTKについて0.250〜12.5nM、およびQB−BTKについて0.125〜12.5nMの範囲の標準曲線を用いて1/x加重二次(weighted quadratic)回帰モデルに適合させた。各ランにおいて、較正標準の少なくとも3分の2については、それらの設定値(nominal values)から逆算された濃度の偏差が、±20.0%以内(LLOQレベルにおける±25.0%)であった。表2〜3に示されるように、それらの設定値から逆算された濃度の偏差は、3回の正確および精度の高いランについて、DB−BTKでは、±11.1%以内であり、QB−BTKでは、±19.8%であった。前記正確性および精度は、品質管理サンプルを用いて評価した。

サル血液溶解物におけるDB−BTKおよびQB−BTKについて示された安定性を表6にまとめる。この結果により、DB−BTKおよびQB−BTKの安定性は、DB−BTKまたはQB−BTKの絶対濃度が、−80℃での2回の凍結融解サイクル後、RTで24時間保存後、または−80℃で224日間後に減少するにも関わらず、測定BTK RO%と理論BTK RO%との差が、全てのQCについて±5%であったため、BTK RO%において影響がないか、最小限の影響であることが示された。

RO測定は、前臨床動物モデルおよび臨床試験においてメカニズムに関連する有効性のために不可欠な測定である。実用上、ROは、最初のヒト(FIH)試験において用量漸増を決定する際に特に有用である。
薬物が結合したBTK(DB−BTK)およびクエンチャーが結合したBTK(QB−BTK)標準物および品質管理サンプルの調製
試薬と材料
図1Aおよび1Bで示される薬物およびクエンチャー化合物は、各々、Bristol−Myers Squibb(ニュージャージー州、プリンストン)における研究開発により合成した。ビオチン化ウサギ抗BTKモノクローナル抗体(ロット番号:D3H5,カタログ番号:12624)および10x緩衝液(カタログ番号:9803)は、Cell Signaling Technology(マサチューセッツ州、ダンバーズ)から入手した。Dynabeads MyoneストレプトアビジンT1ビーズ(カタログ番号:65602)は、ThermoFisher Scientific(カリフォルニア州、カールスバッド)から入手した。組み換えBTKタンパク質(カタログ番号:PV3587)は、Life Technologies(カリフォルニア州、カールスバッド)から入手した。前記プロテアーゼ阻害剤(カタログ番号:539134)は、Millipore Sigma(マサチューセッツ州、ビレリカ)から入手した。アッセイで用いた内部標準(IS)の[13C9,15N]−QB−QRP(N末端およびC末端の両方に隣接アミノ酸を含む)(表8)は、Bristol−Myers Squibb(BMS)(米国ニュージャージー州、プリンストン)における化学合成部門で合成した。サルACD−A全血は、Bioreclamation Inc.(ニューヨーク州、ウェストベリー)から入手した。全ての他の試薬は、分析級のものであった。MagnaBot 96マグネット分離デバイス(カタログ番号:V8151)は、Promega Corporation(ウィスコンシン州、マディソン)から入手した。
組み換えBTK(5.9μM,200μL)を400μLのPBSと合わせ、次いでアセトニトリル中の1.0mg/mLの薬物溶液(図1Aで示される化合物)の15μL(2701μMの薬物,15μL)を添加した。次に、該溶液をRTで1時間インキュベートし、1919nMのDB−BTK濃度とした。DB−BTK濃度は、遊離BTKのDB−BTKへの変換が過剰量の薬物の存在下で〜100%であったため、反応で用いられる合計遊離BTK量を基準にした。
組み換えBTK(5.9μM,200μL)を400μLのPBSと合わせ、次いでアセトニトリル中の1.0mg/mLのクエンチャー溶液(図1Bで示される化合物)の15μL(2806μMのクエンチャー,15μL)を添加した。該溶液をRTで1時間インキュベートし、1919nMのQB−BTK濃度とした。QB−BTK濃度は、遊離BTKのQB−BTKへの変換が過剰量の薬物の存在下で〜100%であったため、反応で用いられる合計遊離BTK量を基準にした。
15mLの10x溶解緩衝液を含む1つのバイアルを、DMSO中の1.0mLのプロテアーゼ阻害剤および80μLの0.5mg/mLのクエンチャー溶液(図1Bで示される化合物)と混合した。次いで、該溶液を、75mLの合計体積とするために59mLの脱イオン化水で希釈した。
15mLの10x溶解緩衝液を含む1つのバイアルを、DMSO中の1.0mLのプロテアーゼ阻害剤および83μLの0.5mg/mLの薬物溶液(図1Aで示される化合物)と混合した。次いで、該溶液を、75mLの合計体積とするために59mLの脱イオン化水で希釈した。
サルACD−A全血(3.5mL)を、1.5μMのクエンチャー(図1Bで示される化合物)を含む7mLの溶解緩衝液と合わせた。該試料を室温で1時間振盪した。

BTK RO QCは、異なる割合の溶解物+433および溶解物+195を混合することによって調製する。25%、50%、および90%のBTK RO%である低QC(LQC)、中間QC(MQC)、および高QC(HQC)は、表7で示されるように、それぞれ、2250/750、1500/1500、および300/2700(v/v)の溶解物+433/溶解物+195を混合することによって調製した。

b溶解物+195は、サル溶解物を薬物溶液で前処理することによって得た(図1A)。
DB−BTKの0.250、0.500、1.00、2.00、5.00、10.0、および12.5nMにおける較正標準(STD)を、DB−BTK保存溶液(緩衝液中で1919nM)の各希釈によって0%のDB−BTKを含むサル血液溶解物中で調製した。調製中、DB−BTKの200および20nMの中間希釈物を、1919nMのDB−BTK保存溶液を血液溶解物+433で希釈することによって調製し、次いで最終STD試料にさらに希釈した。設定濃度は血液体積に基づき、血液溶解物中の実際の濃度は、3の希釈因子により各々の1/3であった。
QB−BTKの0.125、0.250、0.500、1.00、2.00、5.00、10.0、および12.5nMにおける較正標準(STD)を、QB−BTK保存溶液(緩衝液中で1919nM)の各希釈によって0%のQB−BTKを含むサル血液溶解物中で調製した。調製中、QB−BTKの200および20nMの中間希釈物を、1919nMのQB−BTK保存溶液を血液溶解物+195で希釈することによって調製し、次いで最終STD試料にさらに希釈した。設定濃度は血液体積に基づき、血液溶解物中の実際の濃度は、3の希釈因子により各々の1/3であった。
免疫沈澱のためのDynabeads MyoneストレプトアビジンT1ビーズの濃度は、10mg/mLであった。ビオチン化mAbの結合能は、20μg mAb/mgビーズであった。合計25mLのストレプトアビジンT1ビーズを、各2.5mLの10個のチューブに分注した。該試料を3mLのPBST溶液で洗浄した。DynaMag(登録商標)−5マグネットを用いて該マグネットビーズを液体試料マトリックスから分離した。最後の洗浄液を捨て、該ビーズを各チューブ中の2.5mLのPBSTで再懸濁し、次いで0.500mLのビオチン化抗BTK抗体(1mg/mL,ロット番号:D3H5)を加えた。10個のチューブ中の試料(0.500mL)をRTで1時間インキュベートした。該ビーズを該溶液から分離し、各チューブ中のビーズを3mLのPBSTで洗浄した。該チューブ中のビーズを2.5mLのPBSTで再懸濁した。最終抗BTK抗体濃度は、0.2μg/μLのビーズであった。該試料を次の使用のために4℃で保存した。
コントロールサル血液溶解物、較正標準、品質管理サンプル、またはサル試験試料についての3000μLの血液溶解物試料の一定分量を室温で遠心分離した。ビーズ上の抗BTK捕捉抗体(0.2μgmAb/μLビーズで60μL)を、遠心分離した試料の上澄み物に加え、該試料をRTで120分間インキュベートし、続いて遠心分離した。該上澄み物を除去し、PBSTを該ビーズ試料に加えた。該試料をTECAN液体ハンドラー上のLowBind 96ウェルプレートに移し、MagnaBot 96マグネット分離デバイスの上に置いた。該ビーズを800μLのPBSTで2回洗浄し、続いて500μLの25mM NH4OAc(0.05% Tween−20を含む)で洗浄した。洗浄緩衝液を除去し、LowBind 96ウェルプレート中のビーズを下記に記載されるようなトリプシン消化に用いた。
(100μLの25mM NH4OAc(0.05% Tween−20を含む)を加えた)ブランク溶解物を除き、IS(25mM NH4OAc(0.05% Tween−20を含む)中の100μLの0.5μg/mL SIL−QB−QRP)を、96ウェルプレート中のビーズに加え、25μLの100mM NH4HCO3を各試料に加えた。該試料を、25μLの新たに調製した100mM NH4HCO3中で160ng/μLのトリプシン溶液を加えることによって消化し、50℃で30分間インキュベートし、次いで20μLの50%/50%のMeOH/ギ酸(v/v)でクエンチした。該試料を短くボルテックスし、次いでMagnaBot 96デバイスの上に置き、該上澄み物を新たな96ウェルプレートに移し、続いて4000rpmで2分間遠心分離した。該試料をLC−MS/MSにより分析した。
アッセイ適格性および試料分析のために、Sciex(カリフォルニア州、フォスターシティ)からのAB Sciex Triple Quad 5500質量分光計をLC−MS/MSデータ取得のために用いた。

bF*の記号は、[13C9,15N]−フェニルアラニンを表す。下線のペプチド部分は、トリプシンペプチドであり、LC−MS/MSアッセイでモニターした。
正確性および精度によるアッセイ適格性を、さらなる安定性試験(BTK RO QCの凍結融解、RTおよび長期安定性を含む)とともに行う。方法の直線性は、全ての適格性の実行において7つのDB−BTK(0.250nM〜12.5nM)または8つのQB−BTK(0.125nM〜12.5nM)転正曲線点を用いて評価した。1/x加重による二次回帰モデルを定量化のために用いた。受容基準は下記のとおりであった:実施それぞれにおいて、較正標準の少なくとも3分の2について、それらの設定チから逆算した濃度の偏差が±20.0%以内であった(LLOQレベルの±25.0%)。BTK RO QCのためには、各レベルのQCの少なくとも50%について、測定BTK RO%と理論BTK RO%との差異が±5%以内であるべきである。例えば、BTK RO QC_25%について、測定BTK RO%は、25.0±5.0%以内であるべきである。前記アッセイ選択性は、ブランク血液溶解物のみであるQC0(ISのみを含むブランク溶解物)を用いて評価した。
サルPD試験への適用
確率されたELISAに基づくアッセイと比較したLC−MS/MSによるBTK ROアッセイの有用性およびロバスト性を評価するために、図1Aで示される化合物の単回漸増用量試験を行った。カニクイザルに、薬物化合物1Aを0.1、0.2、および0.5mg/kgのPOで投与した。血液試料は、実施例3に記載のLC−MS/MSおよびリガンド結合アッセイ(LBA)によるBTK ROアッセイを用いるPK評価のために1日目に1、3、5、7、および24時間で採取し、PD評価のために投与前、1、3、5、7、24、48、72、144、および168時間に採取した。
PD試料の採取前に、2x溶解緩衝液を0.0133xプロテアーゼ阻害剤および1.5μMのクエンチャー(図1Bで示される化合物)溶液とともに含む下記の溶解緩衝混合液を調製した:15mLの10x溶解緩衝液を含む1つのバイアルに、1.0mLのプロテアーゼ阻害剤および80μLのDMSO中の0.5mg/mLのクエンチャー化合物1Bを混合した。次いで、該溶液を75mLの合計体積とするために59mLの脱イオン水で希釈した。
LBAによるBTK ROアッセイ
サルACD−Aで処理した全血を、投薬後の設定時点で採取し、96ウェルのV底の2mLプレート(Costar,カタログ#3960)に加え、次いでプロテアーゼ阻害剤(Calbiochem,カタログ#539134)およびビオチン化プローブ(BMT−105186)を含む2x溶解緩衝液(Cell Signaling,カタログ#9803)で溶解した。該溶解物を、ストレプトアビジンでコーティングしたプレート(ThermoFisher Scientific,neutrAvidin(登録商標)カタログ#15128)に移し、室温で1時間振盪しながらインキュベートした。洗浄後、抗BTK抗体(Cell Signaling,カタログ#8547,PBS+0.05% Tween20+0.5% BSA中で1:1000希釈)を加え、室温で1時間振盪しながらインキュベートした。該プレートを洗浄し、続いてヤギ抗ウサギ西洋ワサビペルオキシダーゼ二次抗体(インビトロジェン,G21234)をPBS+0.05% Tween20+0.5% BSA中において1:2,500希釈で加えた。該ELISAを、テトラメチルベンジジン(TMB)(Sigma,カタログ#T0440)を加えて発色させ、2.0N 硫酸を加えて停止させた。450nmにおける吸光度を読み、試験試料のBTK不活性の相対%を、溶解前の完全なBTK不活性を考慮に入れるために、2μM BMT−126867で前処理した血液に由来するクエンチされた溶解物と混合した通常のサル血液に由来する異なる比率の溶解物を含む試料の標準曲線から算出した。
0.5mLのサル血液を、PK評価のために設定時点で採取した。各血液試料を遠心分離して血漿を分離した。10μLの125μM クエンチャー溶液(図1Bで示される化合物)を200μLの血漿にすぐに加えた。混合後、該試料を−20℃で保存し、LC−MS/MS分析に使用した。
Claims (7)
- 受容体占有率を測定するためのアッセイ方法であって、
a.受容体に不可逆的に結合する目的の化合物で治療した対象から血液試料を採取し、
b.クエンチャーを含む溶解溶液を前記血液試料に加え、
c.目的の受容体を単離し、
d.前記単離された受容体を消化させてサロゲートペプチドを調製し、
e.前記サロゲートペプチドの量を測定し、次いで薬物が結合したサロゲートペプチドの量を、薬物が結合し、かつクエンチャーが結合したサロゲートペプチドの総量と比較することによって薬物の受容体占有率を決定する
工程を含む、アッセイ方法。 - 目的の受容体に共有結合する目的の薬物を投与した対象における薬物の受容体占有率を決定するためのアッセイ方法であって、
a.前記目的の薬物で治療した対象から血液試料を採取し、
b.前記目的の薬物と同一の受容体部位に不可逆的に結合するクエンチャーを含む溶解溶液を、前記血液試料に加え、
c.薬物が結合し、かつクエンチャーが結合した受容体を免疫捕捉により単離し、
d.前記単離された薬物が結合し、かつクエンチャーが結合した受容体を消化させて、薬物が結合し、かつクエンチャーが結合したサロゲートペプチドを調製し、
e.前記サロゲートペプチドの量を、LC−MS/MSを用いて測定し、次いで薬物が結合したサロゲートペプチドの量を、薬物が結合し、かつクエンチャーが結合したサロゲートペプチドの総量と比較することによって薬物に結合した受容体の量を決定する
工程を含む、アッセイ方法。 - 前記目的の薬物が、図1Aで示されている、請求項2に記載のアッセイ方法。
- 前記工程(b)のクエンチャーが、図1Bで示されている、請求項2に記載のアッセイ方法。
- 前記消化工程(d)が、トリプシン消化である、請求項1または2に記載のアッセイ方法。
- 前記サロゲートペプチドの量が、LC−MS/MSによって測定される、請求項1に記載のアッセイ方法。
- 図1Aで示される薬物を投与した対象におけるBTK受容体占有率を測定するための方法であって、
a.前記薬物を投与した対象から血液試料を採取し、
b.図1Bで示されるクエンチャーを含む溶解溶液を前記血液試料に加え、
c.薬物が結合し、かつクエンチャーが結合したBTKを、ストレプトアビジンT1ビーズに結合させた抗BTKビオチン化抗体を用いて単離し、
d.前記単離された薬物が結合し、かつクエンチャーが結合したBTK抗体複合体をトリプシンで消化させて、サロゲートペプチドを調製し、
e.前記サロゲートペプチドの量を、LC−MS/MSクロマトグラフィーを用いて測定し、次いで薬物が結合したサロゲートペプチドの量を、薬物が結合し、かつクエンチャーが結合したサロゲートペプチドの総量と比較することによって薬物に結合したBTKの量を決定する
工程を含む、方法。
