Abstract
Description
機械的感受性薬物送達という新たな分野において、健康な組織と疾患組織との間の物理的特性の違いを薬物ターゲティングの誘発要因として利用することが求められている。剪断力は、血管中における誘発要因である。マレイの法則によれば、体内血液運搬に必要な仕事量を最小限とするために、血管系全体にわたって剪断力が1.5Pa未満で平衡に保たれている。狭窄部位においては剪断力が少なくとも10倍高くなるため、このような力の変化を用いて、ナノ粒子凝集体またはレンズ型ベシクルからの薬物放出を活性化することができる。
本明細書の文脈において、用語「ファセット形態」は、レンズ型(レンズ豆形状)または非球状、多くの場合は多面体の形状を有するベシクルに関し、かつ、非球状のベシクル形状を特徴とし得る。リポソームがレンズ型形状を有することによって、その表面領域が、障害および/または欠陥に関連した異なる湾曲を有することになる。そのため、湾曲が大きく規則性が小さい領域では、すなわち中心線に沿った領域では、破壊点となり得る箇所が生じる。こうしたレンズ型形態は、球状ベシクルと平坦表面との間のエネルギーが最小値をとる準安定状態であるとみなされる。冷却速度などのカイネティック因子およびベシクル調製法の選択が、ファセット形態の形成に影響を及ぼす。

・拡張障害処置、具体的には、ベータ遮断薬、ACE阻害剤、またはARBから選択される医薬品、

本発明の別の一態様は、ベシクルの調製方法であって、
i. 1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートを、有機溶媒中、具体的には揮発性有機溶媒(solgent)中、より具体的にはメタノールまたはエタノールまたはエタノールアセテート、またはジクロロメタンまたはクロロホルム中に溶解させた状態で提供する工程と、
ii. 不活性雰囲気下(真空下または窒素気流下)において有機溶媒を除去することによって、脂質シートを得る工程と、
iii. 生理的pHを有する第1の水性緩衝溶液(具体的には10mmol/L HEPESを含む)中に脂質シートを懸濁する工程であって、第1の緩衝溶液が、
・クロリド塩を含む、具体的にはNaClを約5mmol/L〜約15mmol/Lの範囲、具体的には約10mmol/Lの濃度で含む、および/または
・200mOsm/L〜400mOsm/Lの範囲、具体的には280mOsm/L〜320mOsm/Lの範囲の容量オスモル濃度を有する、より具体的には308mOsm/Lの容量オスモル濃度を有する、工程と、
iv. 少なくとも1回の凍結工程と少なくとも1回の解凍工程とを適用する工程であって、
具体的には、凍結工程が、液体窒素などの低温貯蔵用の剤によって行なわれ、解凍工程が、試料の温度を急速に55℃〜70℃の範囲、具体的には65℃にして、懸濁液にすること(ベシクル断片化)によって行なわれる、工程と、
v. 押出工程において、懸濁液を、具体的には、孔径が50nm〜150nmの範囲、より具体的には80nm〜120nmの範囲、さらにより具体的には孔径が100nmであるフィルターを通してろ過することによって、押し出し、
任意に、この押出工程を、具体的には5〜15回繰り返すことによって、
押出物を得る工程と、
vi. 生理的pHおよび重量オスモル濃度を有する第2の緩衝溶液中に押出物を透析して、低分子量成分を除去する工程とを含む、方法に関する。
本発明のさらに別の一態様は、
・血管疾患、または
・皮膚科疾患、または
・関節症の処置における、上述される化合物の使用であって、
当該化合物は本発明に係るベシクルの形態で投与されることを特徴とする、使用に関する。
実施例1:1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートの合成
1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートを、スキーム1に示される合成経路に従って合成した。オキシ三塩化リンを1,3−ジクロロプロパノールで置換することにより、ホスホロクロリデートを得た。さらにBoc保護エタノールアミンで置換することにより、適度に安定なホスホロアミデート(5)を得た。この中間体をジアジドに変換し、続いて還元してジアミンとした。ヘプタデカン酸と塩化チオニルとから調製したヘプタデカン酸クロリド(6)を用いてヘプタデカノイル鎖を得て、これをジアミンと反応させて、頭部が保護されたホスホロアミデート(7)を得た。酸性の頭部を脱保護し、硫酸ジメチルで4級化して、最終産物であるジアミドリン脂質1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェート(4)を得た。頭部保護ジクロロプロピルホスホロアミデート(5)から開始して5つの工程の後に得られた全収量は、37%であった。次いで、この化学的に純粋なジアミドリン脂質1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートを、空気−水界面において単層として(2Dモデル)、およびバルク系として(3Dモデル)、分子レベルで特徴づけた。
表面圧/1分子あたり面積の等温線の測定を、ウィルヘルミーの紙秤(精度0.2mN/m)を備えたラングミュア−ポッケルストラフ上で、空気−水界面にて行なった。部分相の温度を5℃〜25℃の間で変えて、等温線を測定した(図2)。
GIXD実験により、凝縮単層の格子構造についてのオングストロームスケールでの洞察が得られる。異なる測定値について、散乱ベクトルの面内要素(QXY)および面外要素(QZ)の各最大値が表1中に示され、ヒートマップの例が図4中に示される。1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートについては、測定したすべての温度/表面圧の組み合わせに対して、2つのブラッグピークが観察された。高いQZにおけるシグナルは、2つの等しいシグナルからなる縮重シグナルである。1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェート(4)の単層は、歪んだ矩形の単位胞で描かれる斜方格子構造を有し、NN(最近接)方向に歪んでおり、脂質尾部の同NN方向への傾斜角tが相当大きい(27°〜38°)。20℃において、鎖の断面積(A0)は約19.3Å2であり、したがって、これより低温における観察値(10℃にて19.0Å2、および15℃にて19.1Å2)よりもわずかに大きいだけである。これは、ヘリンボーン型(herringbone)充填様式の鎖に典型的な値である。この充填様式は、歪みとsin2(t)の外挿により得られる歪み値によって裏付けられる。d0値は−0.0775に達し、格子の歪みの原因が傾斜のみである場合に予期され得るゼロ値とは明らかに異なる。ここに提示される場合において、ヘリンボーン型充填は、さらなる原因であり、頭部と頭部との間の水素結合ネットワークによって生じている可能性がある。
本発明者らは、以前に記載されたプロトコール(Ishikawaら,J.Mol.Biol.2007,368(5),1249−1258)に従って、cryo−TEM画像を測定した。Rad−PC−Radのみからなる大きな単層ベシクル(LUV)について記録したcryo−TEM画像は、膜の膜主相転移温度よりも低温において、相当数の強ファセット状の形状を示す(Tm≒45℃)(図6および図7)。図6から計算した、ファセット状ベシクルと球状ベシクルとの比率は、21:4である。依然として球状の形状が生じているが、これは、調製した脂質膜中の小さな不純物によるものと考えられる、というのは、少量の界面活性化合物が膜の形状および挙動に対して既に相当な影響を及ぼしている可能性があるためである。
膜の小葉の指組みは、膜の主相転移温度にも影響を及ぼす。加熱および冷却の速度を0.5℃/分として行なった、1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートを含有するLUVからの示差走査熱量測定実験の結果、主相転移温度は44.7℃で、エンタルピー変化は24.16kJ/molであることが分かる。主相転移温度は、マルガリン酸鎖を有する天然1,2−ジエステルリン脂質の48.6℃よりも低い。また、二重層の主相転移温度の値は、指組みが起こり得ない単層実験から計算した臨界温度よりも、11.5K高い。
以前に、本発明者らは、1,3−パルミトイルアミド−1,3−デオキシ−sn−グリセロ−2−ホスファチジルコリン(2)を含有するベシクルからの5(6)−カルボキシフルオレセインの放出について報告している(Holmeら,Nat.Nanotechnol.2012,7,536−543)。1,3−パルミトイルアミド−1,3−デオキシ−sn−グリセロ−2−ホスファチジルコリン(2)が、その主相転移温度(Tm=37℃)よりもわずかに高いだけの温度において強い自発性放出を示すことから、この脂質を人間の平均体温である37℃で使用することは理想的ではない。Tmを上げることにより放出特性が改善されることが見込まれる。1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートは、1,3−パルミトイルアミド−1,3−デオキシ−sn−グリセロ−2−ホスファチジルコリンよりも長い鎖を有するため、主相転移温度も44.7℃と高い。このことにより、22℃(室温)および37℃(人間の平均体温)におけるベシクルからの漏れは小さくなる(図8)。アシル鎖を、人工C18リン脂質1,3−ステアロイルアミド−1,3−デオキシ−sn−グリセロ−2−ホスファチジルコリンまで伸ばすと、Tmが52℃に上がるが、このリン脂質はリポソームをほとんど形成しない。
実施例7:ベシクルの調製
1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェート(4)の、直径100nm(LUVET100)の大きな単層ベシクルを、押し出しプロトコールに従って調製した。25mlの丸底フラスコ中において、この脂質10mgをCH2Cl2中に溶解した。有機溶媒を蒸発させた後、得られたフィルムを高真空(40mbar)にて一晩、さらに乾燥させた。次いで、内部緩衝液(1ml、純水に50mM 5(6)−カルボキシフルオレセイン、10mM HEPES緩衝液(AppliChem)、10mM NaClを溶解したもの、pH7.4)を用いて、30分間かけてこのフィルムを含水させた。そして、少なくとも5回の凍結−解凍サイクル(液体N2から65℃)を行なった後、Mini Extruder(Avanti Polar Lipids、米国)を使用し、メッシュサイズ100nmのトラックエッチド(track−edged)フィルター(Whatman、米国)を使用して、懸濁液を11回押し出した。次いで、外部緩衝液(107mM NaClを超純水に溶解したもの、10mM HEPES、pH7.4)を用いたサイズ排除カラム(1.5×20cm セファデックスG−50カラム)を使用して、外部緩衝液の交換を行なった。
Pad−PC−Padリポソームの最終濃度は(19.6+/−1.3)mg/mLであり、DPPC/DSPE−PEGの最終濃度は(20.2+/−6.5)mg/mLであった。
調べたq範囲0:02nm−1〜1:65nm−1は、実空間距離dにして314〜3nmに対応し、したがって、リポソームの特徴づけに関連するナノメートル域のほぼ全体をカバーする。図11のダイアグラムは、好適なガラスキャピラリ中に得たDPPC−DSPE/PEG(上パネル)リポソームおよびPad−PC−Pad(下パネル)リポソームの散乱シグナルの半径方向の積分である。
ガラスキャピラリ中での測定により、静的条件においてPad−PC−Padのリポソーム構造のフィンガープリントが得られた。剪断応力勾配などの外部摂動下におけるPad−PC−Padリポソームの挙動を調べるために、マイクロ流体デバイス中に作った流れ条件下でのPad−PC−Padリポソームの2DラスターSAXSスキャンを記録した。
人工リン脂質のライブラリを合成した。表5に、温度依存性SANS測定の前および後における、流体力学的なサイズおよび多分散に関連した、リポソーム調製物の特徴づけの概要を示す。DPPCリポソームおよびPad−PC−Padリポソームのサイズは(120±2)nmであり、多分散指数は0.01〜0.20である。Rad−PC−Rad、Pes−PC−Pes、およびSad−PC−Sadのリポソーム調製物は、平均サイズ150〜170nm、多分散指数0.57および0.23を示した。Pad−Pad−PCおよびSur−PC−Surのリポソームは、サイズが400〜1,500nm、多分散指数が0.57であった。SANS実験において、リポソームを42℃に加熱した後に室温としたところ、流体力学的なサイズおよび多分散指数が実質的に変化した。概して、リポソームはサイズの増大を示した。唯一の例外がPad−PC−Padリポソームについてみられ、2分の1に減少した。このDLS測定結果を、cryo−TEMを用いて確認した。
SANS実験は、q範囲0.01〜3nm−1が実空間での周期性2〜600nmに対応するため、ナノメートル域の全体をカバーする。したがって、リポソームのサイズだけでなく、その脂質二重層の厚さについても求めることができた。半径および偏心量は、Pad−PC−Padリポソームについてのみ計算した。
リポソームの調製
2種のリン脂質、すなわち、市販のモル濃度5%のDSPE−PEG2000を含む天然1,2−ジパルミトイル−sn−グリセロ−3−ホスファチジルコリン(DPPC)(Lipoid、Zug、スイス)と、近年報告されたプロトコール(Fedotenkoら,Tetrahedron Letters 2010,51,5382−5384)に従って合成した1,3−パルミトイル−アミド−1,3−デオキシ−sn−グリセロ−2−ホスファチジルコリン(Pad−PC−Pad)とを使用して、リポソームを調製した。簡潔には、標準的な薄膜法(Waldeら,Encycl.Nanosci.Nanotechnol.2004,9,43−79;Olsonら,Biochimica et Biophysica Acta (BBA)−Biomembranes 1979,557,9−23)によってリポソームを調製して、超純水を用いて含水させた。得られた各懸濁液は、脂質含有量が20mg/mLであり、これは、Pad−PC−Padを使用して達成できる最高濃度に対応する。液体窒素冷却および水浴加熱(60℃)からなる12工程を連続して行なうことによって、懸濁液を凍結/解凍した。直径約100nmでサイズ分布の狭いリポソームを得るために、Liposofast LF−50(Avestin社、カナダ)を使用し、トラックエッチド(track−edged)ポリカーボネートフィルター膜(Whatman Nucleopore、Sigma−Aldrich、Buchs、スイス)を通して、複数回のバレル押し出しを行なった。
2種の調製物の脂質濃度を、リン酸塩試験によって求めた(Stalder and Zumbuehl.CHIMIA International Journal for Chemistry 2013,67,819−821)。リポソームのサイズおよびサイズ分布を、Delsa Nano C(Beckman Coulter、米国)を使用する動的光散乱(DLS)によって定量した。DLS測定は、温度25℃において、波長658nmで作動する2つのレーザダイオードを使用して行なった。散乱角度は165度に設定した。リポソーム懸濁液をグロー放電孔あき炭素グリッド上に載せ、Cryoplunge CP3システム(Gatan、米国)を用いて迅速に凍結させ、Gatan626クライオホルダーを用いてJEM2200FS透過電子顕微鏡(JEOL、日本)に移すことによって、Pad−PC−Padリポソーム懸濁液のCryo−TEM画像を得た。Cryo−TEM顕微鏡写真は、加速電圧200kV、倍率20,000、4〜8μm不足焦点、および線量として電子10個=Å2において、F416 CMOS検出器(TVIPS、ドイツ)を使用して記録した。
X線に対応したマイクロ流体デバイスを、以前に報告されている通りに準備した(Lutz−Buenoら,Lab on a Chip 2016,16,4028−403)。簡潔には、Siウェハ上のSU−8ネガティブレジスト(Nano SU−8 100、MicroChem社、MA、米国)のフォトリソグラフィを使用してマスターを作製した。厚さ250μmのフォトレジストの層を、マイクロ流体設計を有するフォトマスクを通して紫外光に曝露した後、現像した。このマイクロ流体デバイス設計は、1000μm幅の水平な流路と、中程にある長さ2000μmのくびれとからなるものであった。くびれ幅が125μmのものと250μmのものとの、2通りの設計を作製した。ポリジメチルシロキサン(PDMS、Sylgard 184、Dow Corning社、Midland、米国)と架橋剤とを10:1の比率で混合して、可撓性のレプリカスタンプを形成した。このPDMS混合液をフォトリソグラフィマスター上に注ぎ、温度80℃において一晩硬化させた。その後、PDMSスタンプをマスターから剥がした。Norland Optical Adhesive 81(Norland Products社、Cranbury、米国)を、厚さ25μmのポリイミドフィルム(Goodfellow社、Cambridge、英国)上に注いだ。NOA 81で被覆したポリイミドフィルム上にPDMSスタンプを載せ、NOA 81被覆中にマイクロ流体構造の跡をつけた。λ=366nmの紫外光に1分間曝露してNOA 81を架橋させた後、可撓性のPDMSスタンプを剥がした。直径0:75mmの穴あけ器を使用して、入口および出口の孔をあけた。次いで、マイクロ流体NOA 81/ポリイミドフィルムを、厚さ25μmの第2のポリイミドフィルムで封止した。
空間分解SAXS測定を、スイスの放射光施設Swiss Light Source(PSI、Villigen、スイス)のcSAXSビームラインで行なった。試料の位置においてX線ビームの焦点を25μm×50μm(垂直×水平)のスポットサイズに設定し、光子エネルギーを11:2keV(λ=1:1Å)に設定し、かつ、試料と検出器との間の距離を7:102mに設定し、ベヘン酸銀試料の第1散乱次数(first scattering order)によって求めた。Pilatus 2M検出器(Kraftら,J.Synchrotron Radiat.2009,16,368−375)(ピクセルサイズ:172μm×172μm)を使用して、散乱シグナルを記録した。アルミニウム/ポリエーテルエーテルケトン/アルミニウム試料ホルダー上に載せたマイクロ流体デバイスを使用して、リポソーム懸濁液の動的空間分解SAXS測定を行なった。デバイスを絶えず水平方向に動かしながら検出器でバーストモードでデータを記録することによって、1列毎に2次元(2D)スキャンを実現した。マイクロ流体デバイスの予め選択した領域において、1ポイントあたりの曝露時間を1:5秒として、ステップサイズ(step size)25μm(v)×50μm(h)にて48(v)×96(h)ポイントを得た。ビームラインから入手できるMatlabパッケージを使用して(https://www.psi.ch/sls/csaxs/software)、データ処理を行なった。実験中、ポリテトラフルオロエチレン(PTFE)チューブを用いてマイクロ流路に接続したシリンジポンプシステム(Nemesys、Cetoni GmbH、Korbussen、ドイツ)を使用して、流速を調整した。ポイントに関するバックグラウンド補正のために、リポソーム懸濁液のSAXSシグナルを取得するよりも前に、測定したすべての流速について、マイクロ流体デバイス中における超純水の散乱シグナルを記録した。矩形の流路断面を有するマイクロ流体デバイスについては、下記等式により、体積流量vから壁剪断速度を計算した:(Molloyら,Journal of Thrombosis and Haemostasis 2016,15,972−982)

以前に報告されたプロトコール(Holmeら,Nature Nanotechnology 2012,7(8),536−543;Neuhausら,Langmuir 2018,34(10),3215−3220;Neuhausら,Soft matter 2018,14(19),3978−3986;Neuhausら,Angewandte Chemie 2017,129(23),6615−6618)に従って脂質を配合し、標準的な押し出しプロトコール(Holmeら,Nature Nanotechnology 2012,7(8),536−543)に従ってリポソームを調製した。要約すると、脂質10mgをCH2Cl2中に溶解した。有機溶媒を蒸発させた後、得られた薄いフィルムをさらに真空条件下(40mbar)において一晩乾燥させた。このフィルムを、D2Oを用いて、30分間かけて含水させた。凍結−解凍サイクル(液体N2から65℃水浴へ)を5回行ない、続いて、Mini Extruder(Avanti Polar Lipids、米国)および孔サイズ100nmのトラックエッチド(track−edged)フィルター(Whatman、米国)を使用して押し出しサイクルを11回行なった。得られたリポソーム調製物の脂質濃度は約10mg/mLであった。
スイスのスパレーション中性子源施設(Swiss Spallation Neutron Source)SINQ(Paul Scherrer Institute、Villigen、スイス)のSANS−Iビームラインにて、SANS測定を行なった。経路長2mmのホウ素非含有石英ガラスセル中に試料を入れ、温度制御ホルダー中に入れた。このシステムは、試料の温度を1Kより良い精度で制御できるものである。3〜4Kステップにおいて22〜42℃にて、リポソーム懸濁液の測定を行なった。続いて、この懸濁液を室温(22℃)に冷却し、再度測定した。1.6、6.0、および18.0mの3通りの検出器距離を用い、散乱ベクトル範囲0.01nm−1<q<10nm−1、中性子波長(λ)0.45および1.2Åにて、データを採取した。128×128ピクセルのアレイを有する2次元3He検出器を用いてデータを採取し、1次元のI(q)散乱曲線にして、その結果、2次元画像を半径方向に平均化した。標準的な手順に従い、BerSANSソフトウェアパッケージを使用して、透過、バックグラウンド散乱、および検出器効率についてデータを補正した(Keiderling,Applied Physics A 2002,74(1),s1455−s1457;Strunzら,Journal of Applied Crystallography 2000,33(3 Part 1),829−833)。
このリポソーム懸濁液を、D2Oで1:2の比率で希釈して、最終濃度5mg/mLとした。次に、各懸濁液の4μLを孔あき炭素被覆グリッド(Lacey、Tedpella、米国)に吸着させ、ワットマン1ろ紙を用いてブロッティングして、Leica GPプランジャ(Leica、オーストリア)を使用して、液体エタン中において温度−178℃でガラス化した。凍結させたグリッドを、Gatan 626クライオホルダーを使用して、Talos電子顕微鏡(Thermo Fisher、米国)上に移した。低線量系(20e−/Å2)を使用し、試料を液体窒素の温度に保ちながら、加速電圧200kVおよび公称倍率45,000倍にて、電子顕微鏡写真を記録した。顕微鏡写真の記録はCETAカメラによって行なった。試料レベルでのピクセルサイズは(3.26Å)2であった。cryo−TEM顕微鏡写真のいくつかが、ゆっくりと変動する明るさのばらつきを呈したため、この不均質性を、fuzzy C−meansアルゴリズムの変形(Schulzら,Sci Rep 2012,2,826;Ahmedら,IEEE Trans Med Imaging 2002,21(3),193−9)を使って補填した。
DLS測定を、DelsaMax PRO(Beckman Coulter、米国)を用いて、室温で、D2O中の脂質濃度0.3mg/mLにて行なった。加熱サイクルよりも前に行なった5回の独立した測定で得たデータと、加熱サイクルを適用して8か月後に行なった3回の測定で得たデータとを平均した。データ処理は、キュムラント分析法によって行なった。
SASfitソフトウェア(Bresslerら,Journal of Applied Crystallography 2015,48(5),1587−1598)および対応するモデルを使用して、SANSデータを分析した。Pad−PC−Pad試料を、測定した全q範囲にわたってフィッティングしたが、他の試料についてのフィッティングを、脂質二重層の厚さに対応するq範囲に限定した。Pad−PC−Padについて、本発明者らは、厚さと半径との両方の観点において均質な断面(Guinier,Annales de Physique,1939;pp161−237)およびサイズ分布を有する楕円シェルモデルを使用した。また、本発明者らは、粒子間相互作用を説明するために、sticky hard sphereモデル(Baxterら,The Journal of Chemical Physics 1968,49(6),2770−2774;Mays,Langmuir 1989,5,422−428)を含めた。残りの調製物について、本発明者らは、一般化されたGuinier近似(Hjelmら,The Journal of Physical Chemistry B 2000,104(2),197−211;Fratzl,Journal of Statistical Physics 1994,77(1−2),125−143)を使用して、二重層の厚さを調べ、さらに、本発明者らは、適宜、多重層の寄与を追加した(準晶質理論)(Schwartzら,Biophysical journal 1975,15(12),1201−1233;FruE`hwirthら,Journal of Applied Crystallography 2004,37(5),703−710)。
Pad−PC−Padリポソームは、Tm未満において、脂質二重層の指組みに関連するファセット状の形態を呈し得る。SAXSを使用して、Pad−PC−Padリポソームの層間隔を測定したところ、Tmよりも高温においてリン脂質鎖の指組みが外れていることが示された。SAXSデータから、Pad−PC−Pad二重層内の脂質分子の頭部間距離が(3.3±0.8)nmに対応することが明らかになった。指組みの喪失は、加熱サイクル後の二重層の厚さにおける21%の増加を説明する。明らかに、冷却しても元の状態にはならず、リポソームが崩壊して、加熱サイクル前に測定した元のサイズの半分のナノ構造になった。生理学的に妥当な温度においてPad−PC−Padリポソームが不安定であるということから、これを人体内での標的薬物送達のためのキャリアとして使用することは示唆されない。

- 膜と前記膜により境界が画定される容積とを含む、または本質的にこれらからなるベシクルであって、前記膜は、本質的に1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートからなる、ベシクル。
- 前記膜は二重層構造である、請求項1に記載のベシクル。
- 前記膜は単層構造である、請求項1に記載のベシクル。

- 前記ベシクルは、医薬剤もしくは医薬品または診断剤を含む、請求項1〜4のいずれか1項に記載のベシクル。
- 前記医薬品または診断剤は、
・線維素溶解剤、
・抗凝固剤、
・抗凝集剤、
・アテローム硬化性プラーク安定化剤、具体的にはスタチン、
・血管拡張剤、具体的には、直接または間接作用型の血管拡張薬から選択される血管拡張剤、より具体的には、酸化窒素遊離剤、アルファアドレナリン受容体拮抗薬、アンジオテンシン変換酵素(ACE)阻害剤、アンジオテンシン受容体遮断薬(ARB)、直接型レニン阻害剤、カルシウムチャネル遮断薬(CCB)、エンドセリン受容体拮抗薬、ホスホジエステラーゼ阻害剤、カリウムチャネル開口薬から選択される血管拡張剤、
・抗不整脈薬、具体的には、ナトリウムチャネル遮断薬、ベータ遮断薬、カリウムチャネル遮断薬、カルシウムチャネル遮断薬から選択される抗不整脈薬、
・イノトロープ陽性薬(inotrope positive medication)、具体的には、カテコールアミンまたは非カテコールアミンから選択される医薬品、
・心筋リモデリング薬、具体的には、ACE阻害剤またはARBから選択される医薬品、
・拡張障害処置のための薬物、具体的には、ベータ遮断薬、ACE阻害剤、またはARBから選択される医薬品、
・鬱血解消のための薬物、具体的には、脳性ナトリウム利尿ペプチドまたは窒素放出薬(nitro−release drugs)から選択される医薬品、
・放射線造影マーカー、具体的には、コンピュータ断層撮影、磁気共鳴画像法、陽電子放射断層撮影、シンチグラフィー、または経皮的冠動脈形成術において使用するための放射線造影マーカー、
・化学療法剤、
・凝固因子剤、および
・抗炎症剤から選択されるいずれか1つである、請求項5に記載のベシクル。 - 前記製薬学的活性剤、医薬品、または診断剤は、アルテプラーゼ(alteplasum)、ヘパリン、アセチルサリチル酸、クロピドグレル(clopidogrelum)、糖タンパク質IIb/IIIa阻害剤、ロスバスタチン(rosuvastatinum)、酸化窒素遊離剤、ニトロプルシド、モルシドミン、フェントラミン、エナラプリル、カンデサルタン、ジルチアゼム、ボセンタン、ミルリノン、レボシメンダン、ミノキシジル(minoxidilum)、アリスキレン(aliskirenum)、キニジン、メトプロロール、アミオダロン、ベラパミル、エピネフリン、ノルエピネフリン、ドーパミン、ドブタミン、イソプレナリン(isoprenalin)、レボシメンダン、バソプレシン、グリプレシン(glypressin)、脳性ナトリウム利尿ペプチド、ネシリチド(nesiritidum)、ニトログリセリン、アルテプラーゼ(alteplasum)、エプタコグアルファ(eptacogum alfa)、遺伝子組換え第VII因子(ノボセブン)、ヨウ素またはヨウ素含有造影剤、ガドリニウム含有造影剤、心筋細胞、および幹細胞から選択される、請求項5〜6のいずれか1項に記載のベシクル。
- ベシクルの調製方法であって、
i. 1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェートを、有機溶媒中に溶解させた状態で提供し、
不活性雰囲気下において前記有機溶媒を除去することによって、脂質シートを得る工程と、
ii. 生理的pHを有する第1の水性緩衝溶液中に前記脂質シートを溶解する工程と、
iii. 少なくとも1回の凍結工程と少なくとも1回の解凍工程とを適用する工程と、
iv. 前記懸濁液を、具体的には、孔径が50nm〜150nmの範囲、より具体的には80nm〜120nmの範囲、さらにより具体的には孔径が100nmであるフィルターを通して、押し出すことによって、
押出物を得る工程と、
v. 生理的pHを有する第2の緩衝溶液中に前記押出物を透析して、低分子量成分を除去する工程とを含む、方法。 - 請求項8に記載のプロセスによって得られるベシクル。
- ・心血管疾患、
・皮膚科疾患の処置において使用するための医薬品であって、
前記医薬品は、請求項1〜7のいずれか1項に記載のベシクル、もしくは請求項8に記載の方法によって提供されるベシクルの形態にて、または請求項9に記載のベシクルとして投与されることを特徴とする、請求項7に記載の医薬品。 - 血管疾患の処置において使用するための医薬品であって、前記血管疾患は、急性冠症候群(ACS)、心筋梗塞、急性心不全、慢性心不全、脳血管発作(CVA)、脳卒中、アテローム性動脈硬化、血管攣縮、腫瘍処置、喀血、肺塞栓症、肺動脈性肺高血圧症、腸管虚血、腸管出血、腎梗塞、腎出血、高血圧処置のための腎自己調節、自己免疫性糸球体腎炎、間質性腎炎、胎児疾患、胎盤梗塞、胎盤出血、網膜虚血、網膜出血、および網膜血管新生から選択される、請求項10に記載の医薬品。
- 請求項10に記載の皮膚科疾患の処置において使用するための化合物であって、前記皮膚科疾患は、
・ざ瘡、
・ナプキン皮膚炎、
・アトピー性皮膚炎、
・脂漏性皮膚炎、
・乾癬、
・疣贅、
・足白癬、
・脂漏性角化症、
・蕁麻疹、
・酒さ、
・皮膚科性ウイルス感染、および
・皮膚科性細菌感染から選択される、化合物。 - モニタリングまたは診断方法において使用するための、請求項1〜7のいずれか1項に記載のベシクル、または請求項8に記載の方法によって提供されるベシクル、または請求項9に記載のベシクル。
- 1,3−ジヘプタデカンアミドプロパン−2−イル(2−(トリメチルアンモニオ)エチル)ホスフェート(4)。
