Description
項1.
被検物質(化合物も組成物も包含する)が適用された脂肪細胞(脂肪前駆細胞も含む)における5−リポキシゲナーゼ(5−LO)遺伝子の発現量(好ましくはmRNA又はタンパク質の発現量)を測定する工程を含む、抗インスリン抵抗性物質(NASH予防及び/
又は治療薬を含む)をスクリーニングする方法。(当該方法においては、被験物質の添加により5−LO遺伝子の発現量が減少した場合に、その被験物質を抗インスリン抵抗性物質として好ましく選択することができる。)
項2.
脂肪細胞が、単離された脂肪細胞である、項1に記載の方法。
項3.
被検物質が適用された単離脂肪細胞(脂肪前駆細胞も含む)の培養液中のロイコトリエンB4量を測定する工程を含む、抗インスリン抵抗性物質(NASH予防及び/又は治療薬候補物質を含む)をスクリーニングする方法。(当該方法においては、被験物質の添加により培養液中のロイコトリエンB4量が減少した場合に、その被験物質を抗インスリン抵抗性物質として好ましく選択することができる。)
項4.
脂肪細胞が、初代培養細胞である、項2又は3に記載の方法。
項5.
脂肪細胞が、5−LO遺伝子が過剰発現した脂肪細胞(インスリン抵抗性患者やNASH患者から単離した脂肪細胞でもよいし、モデル非ヒト哺乳動物〔遺伝子組み換え動物も含む〕由来脂肪細胞でもよいし、単離後に5−LO過剰発現遺伝子操作された細胞でもよい)である、項1〜4のいずれかに記載の方法。
項6.
5−LO遺伝子が脂肪細胞において(好ましくは脂肪細胞特異的に)過剰発現した(好ましくはインスリン抵抗性又はNASHのモデルである)非ヒト哺乳動物。

抗インスリン抵抗性物質スクリーニング用キット。
、初代培養細胞以外の細胞(例えば株化された脂肪細胞)を用いた場合には、5−LO遺伝子の発現量変化が検出しづらいおそれがあるためである。
コトリエンB4量が減少した場合に、その被験物質を抗インスリン抵抗性物質として好ましく選択することができる。
イマーセットはリアルタイムPCR用のプライマーセットであることが好ましい。また、上記の通り、抗体は5−LOタンパク質を特異的に認識するものが好ましい。また、ポリクローナル抗体でもモノクローナル抗体でもよく、モノクローナル抗体が好ましい。好ましい抗体の一例として、Anti−5 Lipoxygenase 抗体(abcam社)が挙げられる。なお、本発明において「抗体」は5−LOタンパク質認識能を有する抗体の一部をも包含する意味で用いられる。また、アプタマーは核酸アプタマー及びペプチドアプタマーを包含する。
細胞におけるインスリンによる信号伝達カスケードとして、次のようなことが知られている。すなわち、インスリン受容体にインスリンが結合すると、内在するチロシンキナーゼが活性化され、自己リン酸化される。そのリン酸化チロシンにインスリンレセプター基質1(IRS1)が結合し、このIRS1がリン酸化される。リン酸化されたIRS1はホスファチジルイノシトール−3−キナーゼ(PI3K)に結合し活性化をおこす。活性化されたPI3KはさらにプロテインキナーゼB(PKB、Aktとも呼ばれる)を細胞膜に引き寄せを活性化し、PKBはPDK1(phosphatidylinositol−dependent protein kinase 1)によりリン酸化され、活性型PKBとなる。この活性型PKBが種々のタンパク質をリン酸化することにより代謝調節が行われる。そこで、このカスケードを更に詳細に調べるため、PDK1を脂肪細胞特異的に欠損したマウスを作製し解析した。
2006, 3 : 267−75.)と脂肪細胞特異的CreマウスであるAdiponectin−Creマウス(Eguchi J et al, Cell Metab. 2011, 13 : 249−59.)を交配することにより脂肪細胞特異的PDK1欠損マウス(A−PDK1KOマウス)を作製した。当該マウスに通常食を摂取させ飼育した。また、A−PDK1KOマウスとともに産出する同腹仔PDK1−floxマウスを、以下の検討においてコントロールマウスとして用いた。なお、前記交配においては、目的とするマウスを得るためには何回か交配する必要がある。よって、目的マウスを得る最終の交配としては、次に示す交配となる。

ン固定後パラフィン包埋し、切片を作製した。当該切片をHE(ヘマトキシリン・エオジン)又はシリウスレッドにて染色した。結果を図2に示す。
Foxo1経路の恒常的な活性化がインスリン抵抗性やNASH発症に関与していないかを検討するため、脂肪細胞特異的PDK1/Foxo1ダブル欠損マウスの解析を行った。
ASHを発症したと考えられた一方、A−PDK1/Foxo1DKOマウスでは正常マウスと同様の染色像が観察され、NASHは発症していないと考えられた。
PDK1/Foxo1の下流でどのような分子がインスリン抵抗性(特にNASH)の発症に寄与するのかを調べるため、A−PDK1/Foxo1DKOマウス、A−PDK1KOマウス、及びコントロールマウスの間でオミックス解析を行い、振る舞いが変動する分子を探索した。
al, Am J Physiol Cell Physiol. 2014, 307 : C39−54.)により脂肪組織および血漿中脂質メディエーター濃度を測定した。濃度に変動が見られた物質の解析結果を図6に示す。なお、図6中の*はP<0.05を示す。
DK1KOマウスではコントロールマウスに比べて有意に脂肪組織中のロイコトリエンB4(LTB4)の濃度が高くなっているところ、A−PDK1/Foxo1DKOマウスではA−PDK1KOマウスに比べて有意に脂肪組織中のLTB4濃度が低くなった。また、同様に、A−PDK1KOマウスではコントロールマウスに比べて有意に血漿中のLTB4濃度が高くなっているところ、A−PDK1/Foxo1DKOマウスではA−PDK1KOマウスに比べて有意に血漿中のLTB4濃度が低くなった。
以上の結果から、5−LO遺伝子の発現抑制を指標として、抗インスリン抵抗性物質(インスリン抵抗性治療薬候補物質)をスクリーニングすることができると考えられた。そこで、(i)インスリン処理した脂肪細胞、(ii)活性型Aktを強制発現させた脂肪細胞、及び(iii)Foxo1阻害剤(Calbiochem, AS1842856)で処理した脂肪細胞、のそれぞれについて、5−LOのmRNA発現量を測定した。なお、AS1842856の化学構造式は次のとおりである。
(i)インスリン処理:上記で調整した細胞を、16時間血清を含まないDMEM−high glucose培地で培養後、10−7Mのインスリンで24時間処理した。
(ii)アデノウイルスを用いた活性型Aktの導入:活性型Aktを発現するアデノウイルス(Kitamura T et al, Mol Cell Biol. 1999 : 6286−96.)又はLacZを発現するアデノウイルスを上記細胞に5.5MOI(multiplicity of infection)の量で感染させ、48時間後に細胞を回収した。
(iii)Foxo1阻害剤処理:10μMのFoxo1阻害剤(AS1842856)で24時間処理した後、細胞を回収した。

Claims (10)
- 被検物質が適用された脂肪細胞における5−リポキシゲナーゼ(5−LO)遺伝子の発現量を測定する工程を含む、抗インスリン抵抗性物質をスクリーニングする方法。
- 脂肪細胞が、単離された脂肪細胞である、請求項1に記載の方法。
- 被検物質が適用された単離脂肪細胞の培養液中のロイコトリエンB4量を測定する工程を含む、抗インスリン抵抗性物質をスクリーニングする方法。
- 脂肪細胞が、初代培養細胞である、請求項2又は3に記載の方法。
- 脂肪細胞が、5−LO遺伝子が過剰発現した脂肪細胞である、請求項1〜4のいずれかに記載の方法。
- 5−LO遺伝子が脂肪細胞において過剰発現した非ヒト哺乳動物。
- 非ヒト哺乳動物の脂肪細胞において5−LO遺伝子を過剰発現させる工程を含む、インスリン抵抗性非ヒト哺乳動物を製造する方法。
- 被験単離脂肪細胞における5−LO遺伝子の発現量を測定する工程を含む、当該被験単離脂肪細胞がインスリン抵抗性の対象から単離されたかを判定する方法。
- 被験単離脂肪細胞における5−LO遺伝子の発現量が、正常対象から得た単離脂肪細胞における5−LO遺伝子の発現量より高い場合に、当該被験単離脂肪細胞がインスリン抵抗性の対象から単離されたと判定する、請求項8に記載の方法。
- 5−LOのmRNAを増幅するプライマーセット、及び/又は、5−LOタンパク質を認識する抗体若しくはアプタマー、を備える、
