Abstract
Description
1.(i)成熟ヒト細胞を直接再プログラミングするインビトロの方法であって、その成熟ヒト細胞を転写因子の混合物の存在下で培養する工程を含み、その混合物は、因子BRN2、SOX2、KLF4、ZIC3を含んでおり、その培養を、GSK-3阻害剤(特にChir99021);Alk5阻害剤(特にAlk5阻害剤II);ヘッジホッグ/スムーズンドアゴニスト(特にプルモルファミン)の存在下で実施する方法、または(ii)多能性ヒト幹細胞(特に胚性幹(ES)細胞または人工多能性幹(iPS)細胞)を直接分化させるインビトロの方法であって、その多能性ヒト幹細胞を、GSK-3阻害剤(特にChir99021);Alk5阻害剤(特にAlk5阻害剤II);ヘッジホッグ/スムーズンドアゴニスト(特にプルモルファミン)の存在下で培養することを含む方法。
2.前記培養を、モノアミン-オキシダーゼの阻害剤(特にトラニルシプロミン)を追加して存在させて実施する、項1に記載のインビトロの方法。
3.モノアミン-オキシダーゼの前記阻害剤(特にトラニルシプロミン)が誘導相の間だけ存在し、特に前記培養の最初の12〜21日目の間にだけ存在し、特にADFに関しては最初の12〜21日目の間、pHSCに関しては最初の12〜16日目の間、PBMCに関しては最初の17〜21日目の間にだけ存在する、項2に記載のインビトロの方法。
4.(i)誘導神経ボーダー幹細胞を生成させるインビトロの方法であって、成熟ヒト細胞を転写因子の混合物の存在下で培養する工程を含み、その混合物は、因子BRN2、SOX2、KLF4、ZIC3を含んでおり、その培養を、GSK-3阻害剤(特にChir99021);Alk5阻害剤(特にAlk5阻害剤II);ヘッジホッグ/スムーズンドアゴニスト(特にプルモルファミン)の存在下で実施する方法、または(ii)神経ボーダー幹細胞を生成させるインビトロの方法であって、多能性ヒト幹細胞(特に胚性幹(ES)細胞または人工多能性幹(iPS)細胞)を、GSK-3阻害剤(特にChir99021);Alk5阻害剤(特にAlk5阻害剤II);ヘッジホッグ/スムーズンドアゴニスト(特にプルモルファミン)の存在下で培養する工程を含む方法。
5.前記培養を、モノアミン-オキシダーゼの阻害剤(特にトラニルシプロミン)を追加して存在させて実施する、項4に記載のインビトロの方法。

7.前記成熟ヒト細胞が体細胞である、項1(i)〜6のいずれか1項に記載のインビトロの方法、または前記多能性幹細胞がiPS細胞である、項1(ii)〜6のいずれか1項に記載のインビトロの方法。
8.前記体細胞の選択が、成人線維芽細胞;膵臓由来間葉系間質細胞;末梢血細胞(特に末梢血単核細胞)からなされる、項7に記載のインビトロの方法。
9.前記培養工程を、支持フィーダー細胞の上、特にマウス線維芽細胞の上で実施する、項1〜8のいずれか1項に記載のインビトロの方法。
10.前記成熟ヒト細胞を含む培養物に前記因子BRN2、SOX2、KLF4、ZIC3を形質導入する、項1(i)〜9のいずれか1項に記載のインビトロの方法。
11.前記因子BRN2、SOX2、KLF4、ZIC3がベクターに含まれる、項10に記載のインビトロの方法。
12.前記ベクターがポリシストロン性ベクターである、項11に記載のインビトロの方法。
13.前記ベクターがドキシサイクリン誘導ベクターであり、特に配列ID番号1に従うベクターpHAGE2-TetOminiCV-BRN22AKlf4-IRES-Sox2E2AZic3-Wである、項10または11に記載のインビトロの方法。
14.前記培養を形質導入後の少なくとも12日間にわたって、特に12日間、または13日間、または14日間、または15日間、または16日間にわたって、ドキシサイクリンの存在下で実施する、項13に記載のインビトロの方法。
15.単一コロニーをクローンで増やす工程をさらに含む、項13または14に記載のインビトロの方法。
16.形質導入後19日目〜24日目から選択された日まで前記コロニーを増やす、項15に記載のインビトロの方法。
17.前記ベクターが、前記因子BRN2、SOX2、KLF4、ZIC3をコードする核酸配列に隣接するloxP部位をさらに含んでいて、特にこのベクターが、配列ID番号2に従うベクターpHAGE2-TetOminiCV-BRN22AKlf4-IRES-Sox2E2AZic3-W-loxpである、項11〜16のいずれか1項に記載のインビトロの方法。

神経系の形成は、原腸胚形成から短時間経過した後の神経板段階から始まる。シグナル伝達経路(WNT、BMP、SHHなど)が協調し、神経になる予定の細胞を多様化させる。この細胞がもとになり、さまざまな脳領域、脊髄のほか、神経堤を発生させる(13、14、15)。われわれは、段階特異的な転写因子の過剰発現に、増殖培地による十分なシグナル伝達のきっかけの提供が組み合わされると、成人の体細胞の直接的な再プログラミングによって幹細胞の特徴(自己複製性と多能性が含まれる)を持った初期胚性神経前駆細胞にすることができる可能性があると仮定した。
初期原腸胚形成後神経発生の間、シグナル伝達分子(Wnt、BMP、FGF、SHHなど)が神経板の発生とパターニングを細かく調節することによって多様化をガイドして中枢神経系と神経堤にする。そこでわれわれは、iNBSCがCNS系列とNC系列に分かれる前に1つの発生段階を示すかどうかを調べた。その目的でiNBSCクローンを、(1)CNSへの分化を誘導するためChir99021、SB431542(ALK 4, 5, 7阻害剤)、プルモルファミン(CPS)の後にbFGFを追加して存在させて培養するか、(2)NC運命への分化を促進するためChir99021、Alk-5阻害剤、BMP4を含有する培地の存在下で培養した(図2a)。
iNBSCが成熟タイプの細胞へと分化する潜在力を調べるため、iNBSCに分化への特殊なきっかけを与えた。CNSアイデンティティを有するニューロンを誘導するため、最初に細胞の培養を、プルモルファミンの存在下にて、FGF8(ドーパミン作動性ニューロン)、全トランスレチノイン酸(ATRA)(運動ニューロン)を補足するか、追加のきっかけ(GABA作動性/グルタミン酸作動性分化)なしで、7日間実施した。その後、培地を成熟培地に切り換え、5週間後に分析した(図3a)。
iNBSCの存在と、インビトロで多能性細胞からiNBSCを安定化させる能力は、通常の胚発生の間に「一次神経ボーダー幹細胞(pNBSC)」も出現するどうかという疑問を生む。その目的で、E7.5〜E10.5のマウス胚を単離した。視組織と非神経組織を物理的操作で取り出して酵素で消化させた後、得られた単一細胞懸濁液を支持用マウス胚性線維芽細胞の層の上に播種し、CAPの存在下で培養した(図4a、拡張データ図4a)。播種してから2〜3日後、単独のコロニーを採取することによってクローン株を確立した。注目すべきことに、すべての胚段階が多彩な神経前駆細胞を生じさせたにもかかわらず、E8.5段階の胚からだけ安定に増殖する株を確立することができたため、これは、発生過程の短い期間の間だけ、その細胞が胚の中に存在することを示している(拡張データ図4b)。神経マーカーの発現は、上記のヒトiNBSCの発現と同様であった。したがってpNBSCの中に、Sox1、Sox2、Pax6、Zfp521のほか、神経ボーダーマーカーMsx1 、Pax3が検出された(図4b、図4c、拡張データ図4c、図4d)。さらに、pNBSCは自己複製能力を維持しており、初期マーカーの発現が消えることなく40継代を超えて(4ヶ月超)培養し続けることができた(拡張データ図4e)。さらに、pNBSCは、単一細胞ソーティングをすると、E13.5段階の胚から単離して培養した一次放射状グリア幹細胞と比べて10倍も大きいクローン生成能力を示した(拡張データ図4f)(1)。興味深いことに、pNBSCは、フィーダーの不在下でヒトiNBSCよりも迅速に分化するにもかかわらず、pNBSCの培養もフィーダーに依存していた。フィーダーの不在下での迅速な分化は、NotchリガンドであるDll4とJagged1の存在下でpNBSCをMG上で培養したときに部分的に停止させることができた(拡張データ図4g)。これは、フィーダーを媒介としたNBSCニッチの維持の少なくとも一部がNotchシグナル伝達によって媒介されることを示している。
ヒトiNBSCとマウスpNBSCは多くの特徴を共有しており、そのような特徴に含まれるのは、長期の自己複製能力、特定のマーカーの発現、発生潜在力である。比較をさらに分子発現風景へと拡張するため、マウスpNBSCとヒトiNBSCの網羅的遺伝子発現プロファイルを分析した。
iNBSCは自己複製潜在力と大きな分化潜在力を有するため、遺伝子に基づいてヒト神経症候群をモデル化するための強力なツールである。そのことを直接証明するため、CRISPR/Cas9を媒介とした遺伝子編集を利用して、SCN9a遺伝子によってコードされる侵害受容体である電位依存性ナトリウムチャネルNav1.7を変異させた。このチャネルは末梢神経系で発現しており、SCN9aにおける機能獲得変異によって原発性肢端紅痛症と発作性疼痛障害になる一方で、機能喪失変異によって疼痛に対して先天的に非感受性になる。感覚ニューロンに由来するヒトiNBSCでの機能喪失状況をモデル化するため、特殊なガイドRNAでSCN9aを標的としてエキソン22/27を変異させると、この遺伝子の切断バージョンが得られた(図6aと拡張データ図5a)。欠失した対立遺伝子を有するSCN9-/- iNBSCサブクローンを増やした後、感覚ニューロンへと分化させた。ウエスタンブロット分析から、SCN9a-/- iNBSCに由来するニューロンにはSCN9aが不在であることが確認されたが、対照はそうでなかった(図6b、拡張データ図5b)。感覚ニューロンマーカーBRN3aとペリフェリンに関する3週齢超のニューロン培養物の染色から、SCN9aにおける変異が感覚ニューロンの分化を妨げることが除外された(図6c)。ペリフェリン/BRN3a二重陽性ニューロンは、対照とSCN9a-/-由来の培養物の間で有意差を示さなかった(拡張データ図5c)。次に、感覚ニューロンで発現していて、活性化することで炎症性疼痛を媒介するP2RX3受容体の選択的アゴニストであるα,β-メチレン-ATP(29)を用いて刺激したときのニューロンの活性を、カルシウム流測定によって評価した。予想通り、α,β-メチレン-ATPの添加によって大半の細胞で活性が同期カルシウム流に伴って大きく増加した(図6d)。それとは対照的に、SCN9-/-培養物が示す同期カルシウム流の活性と欠乏はより少なかった(図6d、図6e)。α,β-メチレン-ATPがP2RX3受容体によって媒介されることを確認するため、α,β-メチレン-ATPによって誘導されるカルシウム流の測定を選択的P2RX3アンタゴニストA-317491の存在下で実施したところ、予想通り、ニューロンの活性は対照と比べて有意に低下した(図6d、図6e)。まとめると、データは、疼痛が媒介するシグナル伝達にSCN9Aが必要であることを示しており、より重要なこととして、ヒトiNBSCには、ヒトで遺伝子によって起こる神経疾患の文脈において、将来のモデル化または遺伝子救済のほか、関連する機能的スクリーニングで使用される大きな潜在力があることを示している。
本明細書では、ヒトの皮膚線維芽細胞と血液細胞から多能性クローンiNBSCへの直接変換を証明する。したがってこのiNBSCはこれまで同定されていない神経前駆細胞であり、自己複製能力とCNS系列およびNC系列への分化能力が保持されている。われわれは、ヒトiNBSCがE8.5段階の杯性神経褶に由来するタイプのマウス神経前駆細胞を模倣していることの証拠を提供する。さまざまなヒト細胞とマウス細胞の両方が、似た発現プロファイルのほか、分化と増殖の潜在力を共有している。したがってNBSCは新規な真正の胚性多能性自己複製性前駆細胞であり、それは、成人の体細胞の直接的再プログラミングによって誘導すること、または一次胚性脳組織から単離して取得することができる(図6f)。
実施例1:ウイルス産生と、誘導神経ボーダー幹細胞への再プログラミング
レンチウイルス粒子を産生させるため、別の箇所に記載されている(40)ようにして、pHAGE2-TetO-BKSZ-floxまたはm2RTTA(39、Addgeneプラスミド#20342)をコードするプラスミドをヘルパープラスミドpsPAX2(Addgeneプラスミド#12260)およびpMD2.G(Addgeneプラスミド#12259)とともに293FT細胞株(Life technologies社)にトランスフェクトした。
導入遺伝子カセットを切除するため、以前に報告されている(42)ようにして5×105個のiNBSCを新鮮なフィーダーの上に播種し、Cherry-Creをコードするプラスミドをトランスフェクトした。48時間後、細胞を回収し、Cherry蛍光を探すソーティングを実施した。フィーダーで覆った10 cmの皿の上に5000個のCherry陽性細胞を播種し、コロニーが見えるようになるまでインキュベートした。単独のコロニーを採取し、ゲノムDNAに関する導入遺伝子特異的PCRによって導入遺伝子の除去をチェックした。
PBMCは健康な男性ドナー(年齢24〜30歳)に由来するものであり、Ficoll勾配法を利用して単離した。PBMCは、直接使用するか、以前に報告されているように90%血清代替物/10%DMSOの中で凍結させて使用した(詳細は41を参照されたい)。
ヒト一次膵臓線維芽細胞をVitro Biopharma社から取得し、10%FCSを補足したMSC-Gro(商標)培地(Vitro Biopharma社)の中で培養した。細胞は、4継代にわたって増やした後に使用した。細胞は、5%CO2と20%O2の中で37℃にて増殖させた。
ヒトiPSCを、フィーダー細胞の上にあるDMEM/F12、64μg/mlのLAAP、1×NEAA、15%血清代替物、20 ng/mlのbFGFの中で常法に従って増殖させた。ヒトiPSCは、NBSCへと分化させる前に、マトリゲル(商標)で覆ったプレートの上にあって20 ngmlのbFGFと1 ng/mlのTGFβ1を補足した基本培地の中で1継代培養した。
実施例5.1:cNPCへの分化
マトリゲル(商標)で覆ったプレート(1:36、増殖因子が減らされている、BD Biosciences社)の上にNBSCを播種することによってcNPCへの分化を開始させ、NBSC維持培地から、4μMのChir99028と、3μMのSB431542(Sigma社)と、0.5μMのプルモルファミンを含む基本培地へと切り換えた。5日後、10 ng/mlのbFGF(Peprotech社)と10 ng/mlのLIFを培地(Peprotech社)(CSPFL)に添加した。
マトリゲル(商標)で覆ったプレートの上にNBSCを播種し、1μMのプルモルファミンと10 ng/mlのFGF8を補足した基本培地の中で1週間培養した後、1μMのプルモルファミンを含む基本培地の中でさらに追加して1日間培養した。その後、培養物を、10 mg/mlのBDNFと10 mg/mlのGDNFを含有する基本培地の中でさらに7週間増殖させた。その後、細胞を、基本培地と、20 ng/mlのbFGFと、20 ng/mlのEGFと、10 ng/mlのLIFからなる放射状グリア培地の中で培養した。RG様細胞は増殖すると見えるようになったため、培養物をAccutaseで処理し、マトリゲルで覆ったプレートの上にある放射状グリア培地の中で増やした。最後に、CD133/2、SSEA1、GLASTに関する細胞ソーティングによって培養物のRG様細胞を富化した。すべての培養物を37℃、5%CO2、20%O2にて増殖させた。
堤への分化を開始させるため、マトリゲルで覆ったプレートの上に1×105個/6ウエルのiNBSCを播種し、4μMのChir99028と、5μMのAlk5阻害剤IIと、10 ng/mlのBMP4(CAB)(Peprotech社)を補足した基本培地の中で3日間増殖させた。その後、培地を、4μMのChir99028と、10 ng/mlのFGF8(Peprotech社)と、10 ng/mlのIGF1(Peprotech社)と、1μMのDAPT(Sigma社)を補足した基本培地に切り換えた。5日後、NCSC様細胞をSSEA-1negCD133negP75+HNK1+に関する細胞ソーティングによって精製した。すべての培養物を37℃、5%CO2、20%O2にて増殖させた。
間葉系堤細胞を誘導するため、マトリゲルで覆ったプレートの上にiNBSCを最初に播種し、4μMのChir99028と、10 ng/mlのBMP4と、10μMのDAPTを補足した基本培地の中で7日間増殖させた。その後、細胞を、10 ng/mlのbFGFと10 ng/mlのIGF-1を含有する基本培地の中で5継代を超えて培養した。その後、培地を間葉系幹細胞培地(Gibco社)に切り換えることによってMSC様細胞を安定化させた。MSC様細胞を2継代超にわたって培養した後に分析した。
ニューロンへの分化を誘導するため、マトリゲルで覆ったプレートの上にiNBSCを播種し、iNBSC維持培地を、1μMのプルモルファミン(不定方向分化)、1μMのプルモルファミン、および10 ng/mlのFGF8(ドーパミン作動性ニューロンへの分化)を含有する神経誘導培地、または1μMのプルモルファミンと1μMの全トランスレチノイン酸(Sigma社)(運動神経への分化)を含有する神経誘導培地に1週間切り換えた。iNBSCを、基本培地、3μMのChir99028、3μMのSB431542、1μMのプルモルファミンの中で1週間培養した後、3μMのChir99028、3μMのSB431542、1μMのプルモルファミン、10 ng/mlのFGF4の中でさらに1週間培養し、最後に1μMのプルモルファミンに2日間切り換えることによってセロトニン作動性ニューロンへの分化を開始させた。
iNBSCとcNPCを三次元培養物として分化させるため、2×104個の単一細胞懸濁液を10μlの基本培地の中に再懸濁させ、氷の上で150μlのマトリゲルと混合した。次に、この混合物の30μlの液滴をカバーガラスの上に分配し、37℃で10分間インキュベートした。ゲル化の後、包埋された細胞に分化培地を適用した。
マウス:
本研究を通じて6〜12週齢のマウスを使用した。異なる妊娠段階の胚はトマト-マウスに由来するものであった(Gt(ROSA)26Sortm4(ACTB-tdTomato,-EGFP)Luo)。雌のNOD.Prkdcscid.Il2rgnull(NSG)マウスへの移植を実施した。すべてのマウスを、DKFZ(ドイツがん研究センター)において、特定の病原体がない(SPF)条件のもとで個別換気ケージ(IVC)の中に維持した。サンプルサイズを推定するのに統計学の方法を利用せず、マウス実験は無作為化も盲検化もしなかった。
移植の前に、MGで覆ったプレート上にあって1μMのプルモルファミンと10 ng/mlのFGF8を補足した基本培地の中で8日間培養することにより、iNBSCの分化を開始させた。移植の当日、プライミングした培養物を解離させて単一細胞懸濁液にし、培地の中に1μl当たり細胞5×104個の密度で再懸濁させた。6週齢の雌のNSGマウスをイソフランで麻酔し、定位装置の中に取り付け、手術中はイソフラン麻酔下に維持した。3μlの神経細胞懸濁液をガラス製マイクロピペットで線条体の中に両側から輸液した。移植では以下の座標を使用した:ブレグマと脳表面から前/後:0 mm;中央/側面±2.5;背/腹-2.5 mm。
雄と雌のトマトマウスをつがいにし、翌日に膣栓を調べた。栓試験の陽性を性交後0.5日と見なした。胚を性交後8.5日目に回収し、視組織と非神経組織を物理的操作で取り出した。神経組織をAccutaseで消化させ、得られた単一細胞懸濁液をマウス胚性線維芽細胞の層の上に播種し、NBSC維持培地の中で5%CO2、5%O2、37℃にて培養した。播種してから2〜3日後、単独のコロニーを物理的操作で採取し、クローンpNBSC株を確立した。pNBSC株をAccutaseで処理することによって新鮮なフィーダー細胞上で3日ごとに常法に従って分割すると、40継代を超えて(4ヶ月超)培地の中に維持することができた。
pNBSCをCNSへと分化させるため、マトリゲルで覆ったプレートの上にpNBSCを播種し、1μMのプルモルファミンを補足した基本培地の中で3日間培養した。分化の開始後2〜4日目にロゼット様SC構造を観察することができた。ロゼット様細胞の培地を、20 ng/mlのbFGFと20 ng/mlのEGFを補足した基本培地に切り換えることにより、RG様SCを安定化させた。細胞を3継代超にわたって増やした後、Ssea1+Glast+に関するFACSソーティングによってRG様SCをさらに富化した。1μMのプルモルファミンを補足した基本培地の中でpNBSCを3日間培養した後、10 ng/mlのBDNFと10 ng/mlのGDNFを含有する基本培地に3週間超にわたって切り換えることにより、成熟した子孫細胞(ニューロン、アストロサイト、オリゴデンドロサイトなど)を誘導した。
雄と雌のトマト-マウスをつがいにし、翌日に膣栓を調べた。栓試験の陽性を性交後0.5日と見なした。胚を性交後13.5日目に回収し、内側と外側の基底核原基を物理的操作で単離した。個々の胚からの神経組織をAccutaseで処理することによって消化させた。得られた単一細胞懸濁液をペトリ皿に移し、20 ng/mlのbFGF と20 ng/mlのEGFを補足した基本培地の中で5日間培養した。得られた球を、フィブロネクチンで覆った細胞培養皿の上に載せ、少なくとも3継代にわたって増やした後、Glast+Ssea1+細胞を探すFACSソーティングを実施して下流分析に使用した。
2×104個のpNBSCからなる単一細胞懸濁液を10μlの基本培地の中に再懸濁させ、氷の上で150μlのマトリゲルと混合した。次に、混合物の30μlの液滴をカバーガラスの上に分配し、37℃で10分間インキュベートした。ゲル化の後、包埋された細胞に分化培地を適用した。
スライスの調製
電気生理学的記録を6〜12週齢の雌マウスで実施した。線条体を含有する急性冠状スライス(300μm)から記録した。
浴の温度を30〜32℃にして全細胞パッチ-クランプ記録を実施した。直立顕微鏡(Olympus社BW-X51)に取り付けた水没記録チェンバーの中に個々のスライスまたはカバーガラスを配置し、酸素化細胞外溶液を灌流させた。記録ピペットをホウケイ酸ガラスキャピラリーから引っ張ると、先端の抵抗は5〜8MΩであった。液間電位差は補正しなかった。直列抵抗をできるだけ補償し、記録中は連続してモニタした。「ギガシール」が最初に得られなかった場合、または直列抵抗が20%を超えて変化するか40MΩよりも大きかった場合には、細胞を廃棄した。
MECの中のビオシチン充填細胞を含む急性スライスを4%パラホルムアルデヒドの中で一晩固定した後、PBSで徹底的に洗浄した。
細胞培養物を、細胞透過性蛍光Ca2+インジケータFluo-3 AM(2μM;F1242、Thermo Scientific(商標))とCell Tracker Red CMTPX(1μM;C34552、Thermo Scientific(商標))を含む溶液の中でインキュベートし、細胞膜を明確にした。染料ローディングは、インキュベータの中で37℃、5%CO2にて30分間実施した。
細胞をAccutaseで処理することによって回収し、補足なしのDMEM/F12で2回洗浄した。その後細胞を培地の中に再懸濁させ、抗体を用いて4℃で30分間染色した。使用した抗体は以下の通りである:抗SSEA1 (MC480)-V450抗体(Becton Dickinson社)、抗CD133/2 (293C3)-FITC抗体(Miltenyi Biotec社)、抗CD271 (ME20.4-1.H4)-PE抗体(Miltenyi Biotec社)、抗CD271 (ME20.4-1.H4)-APC抗体(Miltenyi Biotec社)、抗GLAST (ACSA-1)-APC抗体(Miltenyi Biotec社)、抗HNK1 (TB01)-PeCy7抗体(eBioscience社)、マウス抗HNK1抗体(クローンVC1.1、Sigma社)、ロバ抗マウスAlexa Fluor 488抗体(Abcam, ab150105社)、抗CXCR4 (12G5)-PeCy5抗体(BioLegend社)、抗Cxcr4(L276F12)-BV421抗体(BioLegend社)、抗PSA-NCAM(クローン:2-2B)-PE抗体(Miltenyi Biotec社)、抗CD44(G44-26)-PE抗体(BD Pharmingen社)、抗CD105 (43A3)-FITC抗体(BioLegend社)、抗CD90 (5E10)-BV421抗体(BioLegend社)、抗CD146-Alexa647抗体(Biolegend社)、抗CD13 (WM15)-APCCy7抗体(BioLegend社)。FACS分析をLSRIIまたはLSR Fortessaフローサイトメータ(Becton Dickinson社、サンノゼ、カリフォルニア州)で実施した。データはFlowJoソフトウエア(Tree Star社、アッシュランド、オレゴン州)を利用して分析した。細胞ソーティング実験はBD FACSAria(商標)IIIソーター(Becton Dickinson社、サンノゼ、カリフォルニア州)で実施した。分別パラメータとして、100μmのノズル;約2000事象/秒を使用した。
4%パラホルムアルデヒドを含むPBS(Electron Microscopy Sciences社、19208)の中で細胞を15分間固定した。次に、固定した細胞をPBSで3回洗浄し、0.1%トリトンX-100と1%BSAを含有するPBSの中で1時間ブロックした。一次抗体を4℃の0.1%トリトンX-100と1%BSAに適用した。本研究で使用した一次抗体と希釈液のリストに関しては、拡張データ表1を参照されたい。細胞をPBSで3回洗浄した後、二次抗体とともに室温で2時間インキュベートした。
IHC分析のため、移植されたマウスの脳を4%パラホルムアルデヒドの中で一晩固定し、20%スクロースを含むPBSを用いて連続的に脱水した。その後、脳を3%アガロースの中に包埋し、Leica VT1000S滑走式ミクロトームを用いて100μmの冠状切片を調製した。脳のスライスを、3%ヤギ血清と0.25%トリトン-Xを含むTBSの中で4℃にて1時間ブロックした。一次抗体(拡張データ表1)を、3%ヤギ血清と0.25%トリトン-Xを含むTBSに4℃で72時間適用した。スライスをTBSの中で3回洗浄した後、二次抗体を、3%ヤギ血清と0.25%トリトン-Xを含むTBSの中で室温にて2時間インキュベートした。脳のスライスをDAPIで染色した後、LSM 710 ConfoCor 3共焦点顕微鏡(Zeiss社)にマウントして分析した。
脂肪細胞分化物を室温にて4%パラホルムアルデヒドの中で15分間固定した。細胞をddH2Oで2回洗浄した後、60%イソプロパノールとともに5分間インキュベートし、あとで完全に乾燥させた。3.5 mg/mlのRed O(Sigma社)を含む60%イソプロパノールからなる新鮮なオイルレッド染色溶液を室温で細胞に10分間適用した。細胞をddH2Oで4回洗浄した後、顕微鏡(Nikon社、Eclipse Ti-E)の画像を取得した。
軟骨細胞分化物を4%パラホルムアルデヒドの中で15分間固定した後、PBSで3回すすいだ。10 mg/mlのアルシアンブルーGX(Sigma社)を含む3%酢酸からなるアルシアンブルー溶液を1時間適用し、0.1 MのHClで2回すすいだ。細胞をPBSで2回洗浄した後、顕微鏡(Nikon社、Eclipse Ti-E)の画像を取得した。
野生型iNBSCとSCN9a-/- iNBSCを少なくとも3週間分化させて感覚ニューロンにした後、RIPAバッファー(Cell Signaling Technology社)と、1 mMのPMSF(Sigma社)と、1 mMのEDTAと、Halt Protease-Phosphates Inhibitor Cocktail(Pierce社)を用いて全細胞ライセートを調製した。5%SDSの中で95℃にてライセートを変性させた後、タンパク質サンプルを4〜12%TGXゲル(Criterion、Bio-Rad社)の上でTGS(トリス-グリシン-SDS)ランニングバッファー(Bio-Rad社)を用いて解像し、PVDF膜(Trans-Blot TURBO、Bio-Rad社)の上にブロットした。0.3%(vol/vol)のトゥイーン-20と5%(wt/vol)のBSA粉末を含有するTBSを用いて膜を1時間ブロックした。一次抗体(拡張データ表1)をシェイカー上で4℃にて一晩インキュベートした。徹底的に洗浄した後、0.3%のトゥイーン-20を含有するTBSの中で二次HRP結合抗体を室温で1時間インキュベートした。膜を洗浄し、ECLキット(Amersham International社)を用いて免疫複合体を可視化した。
ガイドRNAをDNAオリゴ(Sigma社)として注文し、別の文献(46)に記載されているようにしてpSpCas9(BB)-2A-Puroベクター(PX459)(Feng Zhang氏からAddgeneを通じて寄贈)でクローニングした。マウス神経幹細胞のためのNucleofector(商標)キット(Lonza社)を用いて0.5〜2×106個のiNBSCを0.5〜2 ngのプラスミドDNAとともにプログラムA-033でヌクレオフェクト(nucleofect)し、新鮮なフィーダー細胞の上に播種した。トランスフェクトされた培養物を48時間後に回収し、GFP蛍光に関してソーティングし、フィーダーの上に載せた。5〜7日後に個々のコロニーを手作業で単離した。QIAamp DNAミニキット(QIAGEN社)を用いてゲノムDNAを抽出し、ガイドRNA標的部位を増幅し、サンガーシークエンシング(GATC)によって分析した。CRISP-ID(47)を用いて二遺伝子座配列を復元した。
オン-カラムDNA消化(Qiagen社、79254)を含むARCTURUS PicoPure RNA単離キット(Life Technologies社、Invitrogene社)を利用して全RNAを単離した。大容量cDNA合成キット(Applied Bioystems社)を利用して逆転写を実施した。ViiA7機械上のABI Power SYBR Green Mastermix(Applied Bioystems社)でリアルタイム定量PCRを実施した。結果は、ViiA7-ソフトウエアV1.2.4を利用して分析した。発現は、ハウスキーピング遺伝子GAPDHに規格化した。すべてのPCR反応を技術的トリプリケートとして実施した。25回超の増幅サイクルでRNAの品質が低かったサンプル、および/またはハウスキーピング遺伝子が検出されたサンプルは、研究から除外した。本研究で使用したプライマーについては拡張データ表2を参照されたい。

Claims (15)
- 成人線維芽細胞(AFC);膵臓由来間葉系間質細胞(pMSC);および末梢血単核細胞(PBMC)から選択された成熟ヒト細胞を直接再プログラミングするインビトロの方法であって、その成熟ヒト細胞を転写因子の混合物の存在下で培養する工程を含み、その混合物は、因子BRN2、SOX2、KLF4、およびZIC3を含んでおり、そしてその培養を、GSK-3阻害剤Chir99021;Alk5阻害剤II;およびプルモルファミンの存在下で実施する方法。
- (i)初期神経マーカーであるPAX6、ASCL1、BRN2、およびSOX1と;(ii)幹細胞マーカーであるネスチンおよびSOX2の両方に対して陽性であることを特徴とする、単離された誘導神経ボーダー幹細胞株。
- 請求項2に記載の単離された誘導神経ボーダー幹細胞株を増殖させるインビトロの方法であって、その単離された誘導神経ボーダー幹細胞株からの細胞を培養する工程を含み、特にその培養を、増殖支援サイトカインであるDLL4およびJAGGED-1の存在下で実施する方法。
- 誘導神経ボーダー幹細胞、特に請求項2に記載の単離された誘導神経ボーダー幹細胞株の細胞、または請求項3に記載のインビトロの方法によって得られる細胞を分化させるインビトロの方法であって、その誘導神経ボーダー幹細胞を分化因子の存在下で培養する工程を含む方法。
- (i)妊娠8〜12週の胚から取得可能な一次神経前駆細胞と同じ発生状態であり、(ii)前駆細胞マーカーLONRF2およびZNF217を特徴とするとともに、MSX1、PAX3、およびTFAP2に関して陰性であることを特徴とし、そして(iii)成熟ヒト細胞にエピジェネティックに対応することを特徴とする、中枢神経系系列の単離された中枢神経系プライムド神経前駆細胞株。
- CNS前駆細胞を生成させるインビトロの方法であって、誘導神経ボーダー幹細胞を、GSK-3阻害剤Chir99021;ALK4, 5, 7阻害剤IIプルモルファミン;bFGF;およびLIFを含む培地の中で培養する工程を含む方法。
- 成熟ヒト細胞にエピジェネティックに対応することを特徴とする、単離された中枢神経系前駆細胞株。

- 神経堤前駆細胞を生成させるインビトロの方法であって、誘導神経ボーダー幹細胞を、GSK-3阻害剤Chir99021;Alk5阻害剤II;およびBMP4の存在下で3日間培養した後、GSK-3阻害剤Chir99021;FGF8;IGF1;およびDAPTの存在下で培養する工程を含む方法。
- 成熟ヒト細胞にエピジェネティックに対応することを特徴とする、単離された神経堤前駆細胞株。
- 請求項13に記載の神経堤前駆細胞株を分化させるインビトロの方法であって、その神経堤前駆細胞株からの細胞を分化因子の存在下で培養する工程を含む方法。
