Abstract
Description
本発明の一態様に係るナノ構造体は、親水性ブロックと疎水性ブロックとを含む第1の両親媒性分子が複数個集合して構成される第1の領域と、第1の両親媒性分子とは異なる第2の両親媒性分子が複数個集合して構成される第2の領域とを含む壁部で囲まれている中空体であって、上記壁部の第2の領域が固相から液相に転移する相転移温度において上記第1の領域は固相である。本発明の一態様に係るナノ構造体は、構造安定性および温度応答性といった機能を有する。
第1の両親媒性分子としては、両親媒性ペプチド鎖等が挙げられる。なお、以下、第1の両親媒性分子が有する親水性ブロックを「第1の親水性ブロック」と称し、第1の両親媒性分子が有する疎水性ブロックを「第1の疎水性ブロック」と称する場合がある。
本明細書において「ペプチド」とは、2個以上のアミノ酸がペプチド結合によって結合した化合物を指す。本明細書において「アミノ酸」は、天然アミノ酸、非天然アミノ酸、およびそれらの修飾および/または化学的変更による誘導体を包含する概念であり、また、α−アミノ酸、β−アミノ酸、γ−アミノ酸等を包含する。好ましくは、α−アミノ酸である。本発明において「両親媒性ペプチド鎖」は、ペプチドをベースとする両親媒性の分子であり、一部にペプチド以外の構成要素が存在していてもよい。そのような構成要素として、例えば、N末端またはC末端の修飾、ブロック間の非ペプチドリンカー等が挙げられる。
第2の両親媒性分子は、第1の両親媒性分子とは異なる両親媒性分子を指し、例えば、脂質等が挙げられる。以下、第2の両親媒性分子が有する親水性ブロックを「第2の親水性ブロック」と称し、第2の両親媒性分子が有する疎水性ブロックを「第2の疎水性ブロック」と称する場合がある。
本明細書において「脂質」とは、水系媒体中において、第2の親水性ブロックが外側(水相)に向かって配向し、第2の疎水性ブロックが内側に向かって配向して層(典型的な一例では脂質二重層)を形成し得る任意の脂肪酸誘導体を指す。「脂質」には、天然の脂質だけでなく、人工脂質も包含される。
本発明の一実施形態のナノ構造体は、第1の両親媒性分子(例えば、両親媒性ペプチド鎖)が複数個集合して構成される第1の壁部(壁部全体における第1の領域)と、第2の両親媒性分子(例えば、脂質)が複数個集合して構成される第2の壁部(壁部全体における第2の領域)とを含む壁部で囲まれた中空体である。第1の両親媒性分子(例えば、両親媒性ペプチド鎖)と第2の両親媒性分子(例えば、脂質)との集合の仕方は特に限定されないが、一例において、自己集合的な配向会合による。より具体的な一例では、疎水性相互作用による集合である。
あるいは、予め加熱した状態の水系媒体に、第1の両親媒性分子(例えば、両親媒性ペプチド鎖)と、第2の両親媒性分子(例えば、脂質)とを分散して、そのまま所定時間の加熱を継続してもよい。
本発明の一実施形態のナノ構造体は、剤を保持(好ましくは内包)している。本明細書において「剤を内包している」とは、剤が中空体に共有結合されることなく、中空体の内部に存在していることを指す。典型的には、剤は液体に溶解または懸濁された状態で内包されている。当該液体は、親水性の液体であり得、上述の水系媒体であり得る。
すなわち、本実施形態のナノ構造体の製造方法は、一例において、剤と、第1の両親媒性分子(例えば、両親媒性ペプチド鎖)と、第2の両親媒性分子(例えば、脂質)とを含む水系媒体を加熱することによって、中空体の作製と剤の保持とを同時進行で行う工程を含む。この方法では、効率的且つ容易に剤を保持(好ましくは内包)させることができる。
本実施形態では、ナノ構造体を含む医薬組成物も提供される。当該医薬組成物は、剤として医薬を含んでいる。医薬としては、対象疾患に適したものを特に限定することなく用いることができるが、具体的には、抗癌剤、抗菌剤、抗ウィルス剤、抗炎症剤、免疫抑制剤、ステロイド剤、ホルモン剤、および血管新生阻害剤等が挙げられる。
(試料調製)
ポリ(サルコシン)26−b−(L−Leu−Aib)7の両親媒性ポリペプチド(以下、「S26L14」とも称する)およびポリ(サルコシン)26−b−(L−Leu−Aib)8(S26L16)を既報の参考文献1、2のように合成した。S26L14の合成は、1HNMR分光法およびMALDI−TOF MS分光測定法によって確認した。
TEM画像は、JEM−1230(JEOL社製)を用いて、80kVの加速電圧で取得した。観察するために、分散液のドロップを炭素コートCu格子(Okenshoji Co., Ltd., Japan)上に載せ、2%酢酸サマリウムで逆染色し、過剰な液体を濾紙で除去した。
生理食塩水中の分子集合体の流体力学的直径(hydrodynamic diameter)をHe−Neレーザーを用いるELSZ−2PL(大塚電子社製)によって分析した。測定は25℃で行った。
PLHV1−1分散液を透析によって洗浄し、フリーのS26L14およびDMPCを除去した。その後、PLHV1−1におけるS26L14およびDMPCの濃度を、リン脂質定量測定キット(PHOSPHOLIPID C AUTOKIT 2-10 DEGC 120TST, Wako, Japan)を用いた220nmおよび600nmにおけるUV吸光度から推定した。
TEM画像を図3のA〜DおよびF〜I、図4のA〜F、図5、並びに図6のA、Bに示す。図3のA〜DおよびF〜Iのスケールバー、並びに図4のA〜Cは、200nmを示す。cryo−TEM画像を図3のEに示す。図3のEのスケールバーは、100nmを示す。DLSの結果を図3のJ、図6のC、および表1に示す。リン脂質定量測定の結果を表2に示す。
(円偏光二色性(CD))
光学距離が1cmのセルを用いてJASCO J−720(JEOL社製)によってCD測定を行った。データは25℃で記録した。0.35mMの分散液をCDデータ収集前に5倍に希釈し、測定試料とした。
CD測定の結果を図7および8に示す。
(蛍光分光計)
JASCO FP−6500蛍光分光計(JEOL社製)を用いて、25℃、透過型セルで、集合させた分散液の蛍光スペクトルを得た。
PLHVの均一性/不均一性を調べるために、N−(7−ニトロ−2−1,3−ベンゾキサジアゾール−4−イル)−1,2−ジパルミトイル−sn−グリセロ−3−ホスホエタノールアミン(NBD−PE)およびN−(lissamineローダミン B スルホニル)−1,2−ジパルミトイル−sn−グリセロ−3−ホスホエタノールアミン(Rhod−PE)を用いて、蛍光共鳴エネルギー転移(FRET)分析を行った。FRET測定で、NBD−PEおよびRhod−PEは、それぞれドナーおよびアクセプターとして振る舞う。
さらに、アクセプターの非存在下および存在下におけるFRET効率Eを下記式(1)に従って計算するために、各ドナー(NBD−PEのみ)ラベルされたベシクルも調製した。
ここで、IDは、ドナーのみでラベルされたベシクルを含むサンプルのドナー強度であり、IDAは、ドナーおよびアクセプターの両方でラベルされたベシクルのドナー強度である。
PLHV、S26L14ナノチューブ、およびDMPCリポソームの膜の流動性をN,N−ジメチル−6−ドデカノイル−2−ナフチルアミン(Laurdan)で測定した。Laurdanは、周囲の環境の極性に感受性の高い蛍光色素である。全体の分極値GP340は、下記式(2)によって算出される。
ここで、I440およびI490は、340nmで励起したLaurdanの440nmおよび490nmでの発光強度である。循環水槽によって、温度範囲は10〜42℃に制御した。
DSCはリポソームおよび他の分子の相転移を検出するのに広く用いられている。所望の濃度の20μLの自己集合したハイブリッドベシクル、DMPCリポソーム、またはペプチドナノチューブを蓋で密閉したアルミニウムパン上に置いた。生理食塩水のみを置いた参照試料パンも用いた。走査試料の温度範囲は1℃/分の加熱率で10〜60℃とした。
FRET解析の結果を図9に示し、Laurdanテストの結果を図10に示す。
340nmの励起光を用いることによって、440nmおよび490nmにおけるLaurdanの発光強度から計算された、全体の分極GP340値は、膜表面の水和の度合いの有用な指標として示されている(式(2))(参考文献11、12)。温度の関数としてのGP340値を調べて、相転移温度を評価した。10〜42℃のS26L14ナノチューブのGP340値は一定であり、当該温度範囲に亘って相転移温度(Tm)は示されなかった(図11のA)。一方、DMPCのGP340値は〜25℃において劇的な変化が見られ、DMPCの既知のTmに対応していた。PLHV1−1およびPLHV4−1のGP340値は、〜35℃において減少が見られた。膜の相転移を確認するために、示差走査熱量(DSC)測定も行った。PLHV1−1およびDMPCリポソームのサーモグラムは、PLHV1−1が38度の相転移温度を有し、DMPCリポソームが〜23℃の値を有していることが示された(図11のB)。さらに、DSC分析の結果も、S26L14ナノチューブは14〜46℃に相転移温度を有していないことを示した(図11のB)。上述したように、様々な温度におけるPLHVのCDスペクトル(図8)は、42℃は、PLHVにおけるペプチドの二次構造およびペプチド間の束形成に影響を与えないことを示した。これらの結果による考察によると、脂質ドメインおよびS26L14に相当するハイブリッドベシクルの相転移は、スペクトルに影響を与えなかった。
(FITC−PEG2KおよびFITC−PEG5Kの合成)
FITC−PEG2KおよびFITC−PEG5Kの合成方法、および得られたFITC−PEG2KおよびFITC−PEG5KのNMRおよびMSスペクトルを以下に示す。

自己消光特性を示す濃度である3mMのFITC−PEGを用いて、PLHVの内包特性および放出特性を調べた。所望のモル比のハイブリッドベシクルおよびDMPCリポソームストック溶液をFITC−PEGを含む生理食塩水に添加(インジェクション)し、2時間撹拌し、次いで、90℃で1時間加熱した。次いで、試料を遠心分離速度2000rpm、4℃で、Millipore社製の遠心分離フィルターユニット(30kDa分子量カットオフ)に通して、フリーのFITC−PEGを除去した。FITC−PEGの内包効率を下記式(3)のように算出した。
ここで、Fafは、遠心分離フィルターに通した後の蛍光強度であり、Fbfは、遠心分離フィルターに通す前の蛍光強度である。蛍光発光アッセイを用いて、ハイブリッドベシクルおよびDMPCリポソームからのFITC−PEG放出を測定した。要するに、100μLの、FITC−PEGを内包したハイブリッドベシクルまたはDMPCリポソームを20mLの生理食塩水に添加(インジェクション)し、均一に混合した。0.5、1、2、および4時間の間隔で、1mLの試料を取り出し、氷上で冷却して、更なる薬剤放出を停止した。FITC−PEGの放出率は、以下の式(4)を用いて算出した。
ここで、Fは、特定の点での試料の蛍光強度であり、F0は、サンプルのバックグラウンドの蛍光であり、F100は、溶解緩衝液によって溶解したキャリアからの最大蛍光放出であり、3つの試料の平均である。溶解緩衝液(コード番号:CK12、ロット番号:KU749、Dojindo Molecular Technologies, INC., Japan)(5μL)を、FITC−PEGを内包したハイブリッドベシクル分散液(1mL)に添加し、分散液を60℃で10分加熱し、F100値を得た。490nmの励起波長、および520nmの発光波長で、JASCO FP−6500蛍光分光計(JEOL社製)を用いた。本放出実験で用いられた温度は、4℃、37℃、および42℃であった。PEGの分子量は2および5kDaであった。
フルオロセインイソシアネートでラベルされたポリエチレングリコール(Mw:2000)(FITC−PEG2K)を、FITCの蛍光を消光する十分高いFITC−PEG2K濃度で、PLHVに被包した。DMPCリポソーム、PLHV1−1、およびPLHV4−1におけるFITC−PEG2Kの放出効果は、それぞれ3.9%、7.1%、および5.8%であった。FITC−PEG2K放出実験は、4℃、37℃、および42℃で行われた。図12に示すように、FITC−PEG2Kは、一定温度下で時間の関数として放出された。FITC−PEG2Kの被包は、上述したすべての集合体について、4℃で4時間維持した(図12のA〜C)。DMPCリポソームについて、70%を超えるFITC−PEG2Kが、37℃および42℃において迅速に放出された。これは、DMPCリポソームが、37℃および42℃において無秩序な相であることによるものである(図11)。一方、PLHV1−1は、42℃でのみFITC−PEG2Kを放出し(図12のB)、37℃はTmよりも低いため、FITC−PEG2Kは37℃では被包されたままであった。従って、DMPCドメインは、温度応答性のゲートとして振る舞っていた。同様の方法におけるPLHV4−1について、FITC−PEG2Kは37℃で放出されなかった。しかしながら、42℃において、ゆっくりとした放出が観測された(図6のC)。おそらく、DMPCドメインのサイズがより小さいと、PLHV4−1からのFITC−PEG2Kの放出率が減少した。さらに、他の小さな疎水色素sulforhodamine B(SRB)も、37℃でナノ粒子の内側にトラップされたままであり、42℃でPLHV1−1およびPLHV4−1から放出された(図13)。42℃で4時間加熱した後、SRBでは、FITC−PEG2Kよりもより多くの放出が観測された。これらの結果も、脂質ドメインのサイズが、トラップされる物質のサイズおよび放出率を潜在的に定めることができることを支持している。それゆえ、この結果は、ハイブリッドベシクルが薬剤を閉じ込め、かつ放出することができることを示している。より大きな分子量を有する、フルオロセインイソシアネートでラベルされたポリエチレングリコール(Mw:5000)(FITC−PEG5K)の放出動態を評価した。PLHV1−1からのFITC−PEG5Kの放出は、42℃で抑制された(図12のD)。一方、DMPCリポソームは、37℃(破線)および42℃(実線)の両方でFITC−PEG2Kと同様にFITC−PEG5Kを放出した(図14)。

Claims (11)
- 親水性ブロックと疎水性ブロックとを含む第1の両親媒性分子が複数個集合して構成される第1の領域と、第1の両親媒性分子とは異なる第2の両親媒性分子が複数個集合して構成される第2の領域とを含む壁部で囲まれている中空体であって、
上記壁部の第2の領域が固相から液相に転移する相転移温度において上記第1の領域は固相である、ナノ構造体。 - 親水性ブロックと疎水性ブロックとを含む第1の両親媒性分子が複数個集合して構成される第1の領域と、第1の両親媒性分子とは異なる第2の両親媒性分子が複数個集合して構成される第2の領域とを含む壁部で囲まれている中空体であって、
上記第1の両親媒性分子は、親水性ペプチドブロックと疎水性ペプチドブロックとを含む両親媒性ペプチド鎖であり、
上記第2の両親媒性分子は、脂質である、ナノ構造体。 - 上記第2の両親媒性分子は、脂質である、請求項1に記載のナノ構造体。
- 上記第2の両親媒性分子は、リン脂質である、請求項3に記載のナノ構造体。
- 上記第1の両親媒性分子は、親水性ペプチドブロックと疎水性ペプチドブロックとを含む両親媒性ペプチド鎖である、請求項1、3又は4に記載のナノ構造体。
- 上記疎水性ペプチドブロックがヘリックス構造を形成している、請求項5に記載のナノ構造体。
- 25℃で動的光散乱法によって測定した、150mM NaCl、pH7.4の生理食塩水中の平均流体力学的直径が10〜500nmである、請求項1〜6の何れか1項に記載のナノ構造体。
- 上記第1の両親媒性分子と上記第2の両親媒性分子との占有表面積の比は、40:60〜95:5である、請求項1〜7の何れか1項に記載のナノ構造体。
- 剤を保持している、請求項1〜8の何れか1項に記載のナノ構造体。
- 請求項9に記載のナノ構造体を含む、医薬組成物。
- 請求項1〜10の何れか1項に記載のナノ構造体の製造方法であって、
上記第1の両親媒性分子と、上記第2の両親媒性分子とを含む水系媒体を加熱して、中空体を作製する工程を含む、ナノ構造体の製造方法。
