Abstract
Description
(1)口腔バイオフィルムの内部に存在する病原性細菌の細胞外電子伝達機構を制御することにより、前記病原性細菌の活性を低減させる方法。
(2)前記病原性細菌の細胞内から細胞外への電子移動を抑制することにより、前記病原性細菌の細胞外電子伝達機構を制御する、(1)に記載の方法。
(3)口腔バイオフィルムの内部から外部への電子移動を抑制することにより、前記病原性細菌の細胞内から細胞外への電子移動を抑制する、(2)に記載の方法。

S. mutans(UA159株)は、口腔バイオフィルム内に存在する代表的な病原性細菌であり、Rexタンパク質を有していることが知られている。Rexタンパク質は、レドックスセンサーであり、エネルギー代謝の初期調節メカニズムに関与する遺伝子に結合することによって、DNA転写抑制因子として作用する。図1に示すように、Rexタンパク質は、細胞質のNADH/NAD+の比率が低い場合には、ピルビン酸デヒドロゲナーゼ(pdhAB)の転写を抑制して乳酸発酵経路を促進する。一方、NADH/NAD+の比率が高い場合には、標的遺伝子に対するRexタンパク質の結合が阻害されるため、エタノール生成経路が促進される。そのため、NAD+に対してNADHが過剰であるかどうかの指標として、グルコース酸化によって生成されるエタノールの量を利用することによって、実験系の電気化学システムにおける電子移動を定量的に評価することができる。
ATCC(American Type Culture Collection)から入手したS. mutans(UA159株)を、ブチルゴム栓をしたバイアル内で、20mLのブレインハートインフュージョン(BHI)培地中で、37℃で前培養した。バイアル内のヘッドスペースは、対数増殖期の後期に達するまでCO2/N2(20:80 v/v)を充填して無酸素状態とした。このときの波長600nmにおける吸光度(OD600)は1.0であり、培地のpHは4.6±0.2まで低下した。得られた培養物を、7800rpm、37℃で10分間遠心分離し、得られた細胞ペレットを合成培地(DM)で2回洗浄した。なお、合成培地は、一般に知られている手法を一部改良して調製した。合成培地の成分組成(1リットル当たり)は、以下の通りである:NaHCO3 2.5g;CaCl2・2H2O 0.09g;NH4Cl 1.0g;MgCl2・6H2O 0.2g;NaCl 10g;HEPES 7.2g;酵母抽出物 0.5g。
作用極、対極、参照極で構成される3電極システムを備えた測定装置(シングルチャンバー)を用いて、電気化学測定を行った。作用極のスズドープ酸化インジウム(ITO)は、スプレー熱分解法による薄膜形成装置(SPD Laboratory社製)を用いてガラス基板上に成膜し、測定装置の底部に設置した(抵抗値 8Ω/□;厚さ 1.1mm;表面積 3.14cm2)。対極および参照極としては、それぞれ、白金ワイヤおよびAg/AgCl(KClsat)を用いた。
この電気化学セルに、電解質(電子供与体(electron donor))として10mMのグルコースを添加した合成培地(4.8mL)、またはグルコースを添加した合成培地に電子受容体(electron acceptor)として10μMのリボフラビン(RF)を追加した培地(4.8mL)を注入し、セル内の溶液をN2ガスで少なくとも15分間パージして、溶存酸素を除去した。次いで、合成培地で前洗浄したS. mutansを用いて新たに調製した細胞懸濁液0.2mL(600nmでの光学濃度(OD):2.5)を注入し、標準水素電極(SHE)を基準としてITO電極の電位差を+0.4Vとする定電位条件で、最終のOD600を0.1とした。電気化学測定中、測定装置の温度は37℃に維持し、装置の振とうや溶液の撹拌は行わなかった。
また、比較試験として、電極が電子を受け取らない状態とする開路電圧(OCV;Open Circuit Voltage)条件で、同様の電気化学分析を行った。
測定装置から8時間おきに約200μLのアリコートを回収し、孔径0.22μmのフィルターを通して細胞を取り除いたろ液を−20℃で保管し、分析に供した。グルコース濃度は、グルコースアッセイキット(GAGO−20、Sigma−Aldrich製)を用いて測定した。代謝産物の測定では、上記のろ液を蒸留水で100倍に希釈し、pHを8.0に調整したものを試料として用いた。乳酸、酢酸、ギ酸の濃度は、イオンクロマトグラフィーシステム(HIC−20Asuper、Shimadzu社製)を用いて測定した。試料の注入量は50μLとし、ノンサプレッサ方式で陰イオン測定を行った。分析カラムおよびガードカラムは、それぞれ、Shim−pack IC−A3、およびShim−pack IC−GA3(いずれもShimadzu社製)を用いた。移動相には8mM p−ヒドロキシ安息香酸、3.2mM Bis−Trisおよび50mM ボロン酸を含有させ、流速は1.2mL/minとした。カラム温度は40℃に維持し、検出器(CDD−10ASP)のパラメーターは当該システムの取扱説明書に記載の通りに設定した。ピーク面積の分析は、Shimadzu社より提供された分析用ワークステーションソフトウェア(LabSolutions)を用いて行った。酢酸、乳酸およびギ酸の溶離時間は、それぞれ、約2.4分、約3.25分、約3.4分であり、検量線は十分に高い直線性(R2=0.999)を示した。なお、イオンクロマトグラフィー分析では、乳酸、酢酸およびギ酸以外の代謝産物は検出されなかった。
重クロム酸の酸性溶液を用いる標準的な滴定法を一部変更して、試料中のエタノール濃度を測定した。滴定に用いる溶液は以下のようにして調製した。
(1)重クロム酸溶液(5.0mol/L 硫酸中、0.01mol/L):125mLの水を500mLのコニカルフラスコに入れ、一定の速度で旋回させながら70mLの濃硫酸をゆっくりと滴下する。フラスコを冷却した後、0.75gの重クロム酸カリウム(K2Cr2O7)を加え、この混合物を蒸留水で250mLに希釈する。
(2)デンプン溶液(指示薬溶液;1.0%):100mLの沸騰水に1.0gの可溶性デンプンを添加し、溶解するまで撹拌する。
(3)チオ硫酸ナトリウム溶液(Na2S2O3・5H2O;0.03mol/L):7.44gのNa2S2O3・5H2Oを1Lのメスフラスコに入れ、蒸留水に溶解させて1L溶液とする。
(4)ヨウ化カリウム溶液(KI;1.2mol/L):25mLの水に5gのKIを溶解させる。
対照の無菌培地を用いた場合には、リボフラビンの存在下(Sterile medium w/ RF)および非存在下(Sterile medium)のいずれも、S. mutansの添加による電流密度の変化は見られなかった。一方、電子供与体として10mMのグルコースを添加した合成培地を用いた場合(EET)には、リボフラビンの非存在下で、約30nA/cm2の電流密度を示した。また、リボフラビンの存在下(RF EET)では、リボフラビンの非存在下よりも早いタイミングで電流密度の上昇が見られ、その最大値は約40nA/cm2であった。
また、図3(b)〜図3(d)に示すように、グルコースを添加した合成培地を用いた場合(EET)、グルコースおよびリボフラビンを添加した合成培地を用いた場合(RF EET)、OCV条件でグルコースを添加した合成培地を用いた場合(OCV)、OCV条件でグルコースおよびリボフラビンを添加した合成培地を用いた場合(RF OCV)のいずれも、電流の発生に伴い、グルコース濃度は低下し(図3(b))、有意な量の乳酸の産生が確認された(図3(c))が、エタノールの生成は確認されなかった(図3(d))。
なお、図3(b)、3(c)においては、各条件とグラフとの対応関係を、(1)〜(4)の符号を用いて示している。後述する図4(b)、図4(c)、図8についても同様である。
すなわち、本発明によって活性が低減される病原性細菌は、一般に電流発生菌として知られているシュワネラ属(Shewanella)細菌、ジオバクター属(Geobacter)細菌等と比べて、細胞外電子伝達機構によって細胞内から細胞外へ移動する電子の量が少ない場合があることに留意されたい。
しかしながら、このような少量の電子移動に起因する微小な電流の発生が、S. mutansによる乳酸発酵の促進とエタノール生成の抑制に有意に関係していることに加え、S. mutansの細胞内の還元エネルギーの緩和に有意に寄与しているという事実は、本発明者等による新たな知見であり、本発明の根幹をなす技術的思想である。
以下、上記の実験系による電気化学分析の結果について、より詳細に説明する。
電気化学測定の開始から5時間を超えると、EET条件での電流密度の値は飽和に達し(図3(a))、その後電流密度が低下するにつれて、グルコースの消費速度および乳酸の産生速度も低下した(図3(b)、図3(c))。
一方、電気化学測定の開始から24時間が経過しても、EET条件ではエタノールの生成はほとんど見られず(図3(d))、酢酸およびギ酸の生成量はごくわずかであった(図4(b)、図4(c))。
これらの結果から、S. mutansの細胞外電子伝達機構が、乳酸発酵およびグルコース酸化と相関関係を有することが示唆された。
乳酸産生と比較して、エタノールの生成によってより多くのNAD+が再生されることを考慮すると、OCV条件では、Rex遺伝子が、NADH量が多いことを検知し、エタノールの生成を促進することでNAD+の生成を促していると考えられる。
このことは、本実験系で確認された電流の発生は、発酵で生じた水素の酸化によって支配されているのではなく、電子授受のプロセス(electron shuttling process)と関連していることを示している。
10μMのリボフラビンの530nmにおける蛍光強度を基準として(A)、OD600=0.1となるようにS. mutansを添加し(B)、その後、10mMのグルコースを添加するにつれて(C)、リボフラビンの530nmの蛍光強度は有意に減少した。
この結果は、リボフラビンが酸化されていること(酸化型のリボフラビンが生成していること)を示しており、このリボフラビンの酸化が、S. mutansによるものであること、また、S. mutansによるグルコース酸化と関連付けられることを示している。
なお、図5中、「DM」は、合成培地の発光スペクトルである。
すなわち、発酵性細菌(嫌気性細菌)について、細胞内電位のバランスを保つことを補助する電子伝達物質(電子シャトル)が存在し、実際に細菌がそのような電子シャトルを利用して酸化還元バランスを保っていることが、本発明者等によって初めて明らかにされた。また、口腔バイオフィルムを構成する微生物の中に、病原性細菌の細胞内から細胞外への電子の移動を補助する電子伝達物質を生成する細菌が存在し得ることが示された。
次に、細胞に特異的な安定同位体を用いて、二次イオン質量分析(NanoSIMS分析)を行うことによる、S. mutansの代謝活性の定量的評価手法およびその結果について説明する。
15Nによる同位体効果の影響と思われる変化は見られたものの、非標識のHN4Clを用いた場合(図示せず)と同様の電流の生成が確認された。また、リボフラビンの存在下(W/ riboflavin)では、リボフラビンの非存在下(W/o riboflavin)の場合よりも、15時間の電気化学測定で得られた電流値の最大値が22%高かった。
電気化学測定を行った後、S. mutansを含むバイオフィルムが付着したITO電極を反応装置から回収し、2.5%のグルタルアルデヒドを含む0.1Mのリン酸緩衝液中で10分間固定し、電極を新たな0.1Mのリン酸緩衝液に浸漬させて洗浄した。洗浄した電極試料を、リン酸緩衝液中のエタノールのグラジエント(25%、50%、75%、90%)および100%エタノールで脱水し、t−ブタノールによる溶媒交換、真空乾燥の後、Quick Coater SC−701(サンユー電子社製)を用いて白金でコーティングした。CAMECA NanoSIMS 50 L system(CAMECA社製)を用いて、試料の表面にCs+ビームを照射し、二次イオン(12C14N−および12C15N−)の量を測定した。一細胞毎に対象領域(ROI:Regions of Interest)をマーキングし、各ROIにおけるイオン強度の合計値を算出し、全窒素(Ntotal)に対する15Nの割合(15N/Ntotal)から、一細胞毎のNの同位体比(%)を計算した。各試料について、200以上の細胞を対象として計測を行った。
リボフラビン非存在下のOCV条件(図7(a);OCV)では、比較的低い活性(15N/Ntotal < 10%)を示した細胞が全体(n=231)の42%であった。一方、リボフラビン存在下のOCV条件(図7(c);RF OCV)、およびリボフラビン存在下のEET条件(図7(b);RF EET)では、15N/Ntotalの値が10%未満の細胞の数は、それぞれ14%および9%であり、図7(a)に示すリボフラビン非存在下のOCV条件と比較して、有意に低かった。なお、リボフラビン非存在下のEET条件(図7(d);EET)における、15N/Ntotalの値が10%未満の細胞の数は、4%であった。
各条件で得られた結果をまとめた図8からも理解されるように、これらの結果から、リボフラビンがS. mutansの細胞内から細胞外への電子移動を媒介すること、また、リボフラビンが存在することによって、活性の低い細胞の代謝活性が高められることが示唆された。
・小さい電流値として確認されるS. mutansの細胞内から細胞外への電子の移動は、細胞内のNADH/NAD+の比を制御するのに大きな役割を果たしていること。
・リボフラビンが、口腔バイオフィルムにおいて、S. mutansの細胞内外、バイオフィルムの内外の電子移動を媒介すること。
・細胞内の還元的な環境から細胞外の酸化的な環境へ電子を排出することによってNAD+の再生が起こり、S. mutansによる乳酸発酵が促進されること。
次に、上述したような口腔バイオフィルム内の病原性細菌による直接的な電子伝達能を評価するための実験手法およびその結果について説明する。
その結果、培地を交換した後も、交換前と同程度のレベルの電流の生成が継続的に確認された。すなわち、培地の変更によって、S. mutansによる電流生成への影響は確認されず、また、培地にプランクトン細胞が含まれない場合でもS. mutansによる電流生成が確認された。
図12(b)は、図10(b)に示したDPボルタモグラムにおける、標準水素電極(SHE)に対する電位が約20mVでのレドックスピーク電流値(μA)に対して、S. mutansによる電流生成値(μA)をプロットしたグラフである。
図12(b)中の左上に挿入したグラフに示すように、電気化学測定の開始2時間後の結果を除いて、20mVでのピーク電流値とS. mutansによる電流生成値の間に、相関係数の二乗(R2)=0.9489で線形関係が得られ、さらに、図12(b)に示した回帰直線が原点を通過していることから、レドックス分子が優先的に電流生成を媒介しており、水素や、酵母抽出物中に含まれる少量のリボフラビンは、このプロセスにわずかに関与しているのみであることが示された。
20mLの細胞培養物を7800rpmで10分間遠心分離することによって、S. mutans細胞を回収し、回収したS. mutans細胞を、2%パラホルムアルデヒドおよび2.5%グルタルアルデヒドを含む溶液中で氷冷して固定した。固定後のすべての操作は、2mLのエッペンドルフチューブ中で行った。1.5mLの洗浄液で穏やかに懸濁させながら5回洗浄を行い、50mM Na+−HEPES(pH 7.4、35g/L NaCl)を用いて5000×gで4分間遠心分離した。次いで、3,3’−ジアミノベンジン(DAB)と過酸化水素(H2O2)による処理および四酸化オスミウム(OsO4)溶液によるポスト固定を行い、細胞膜およびペプチドグリカン層を染色した後、樹脂包埋を行った。得られた樹脂ブロックから80nmの切片を切り出し、銅製のマイクログリッド上に載せ、JEM−1400電子顕微鏡(日本電子製)を用いて80kVの加速電圧条件で観察した。S. mutansの細胞膜による電子線吸収の定量分析は、DAB陽性の染色細胞のLINEプロファイルから取得されたLRホワイトレジン、細胞質、および細胞膜の領域の電子線透過(ET)量を比較することによって行った(Deng, X., Dohmae, N., Nealson, K. H., Hashimoto, K. & Okamoto, A. Multi-heme cytochromes provide a pathway for survival in energy-limited environments. Sci. Adv. 4, eaao5682 (2018)を参照されたい。)。
過酸化水素の存在下でDAB染色を行うポジティブコントロール条件(DAB positive)では、図10(c)および図13(a)に示すように、細胞膜およびペプチドグリカン層の染色が確認されたのに対して、ネガティブコントロール条件として過酸化水素を用いなかった場合(DAB negative)には、図10(d)および図13(b)に示すように、細胞膜やペプチドグリカン層の染色はほとんどもしくは全く確認されなかった。図10(c)では、白色矢印で示すように、細胞表面またはペプチドグリカン層にレドックス活性剤が存在する様子が確認できるが、図10(d)では、白色矢印で示した箇所にそのような物質が存在することは確認できない。なお、図10(c)、図10(d)のスケールバーは、100nmである。
図14Aは、ポジティブコントロール条件(DAB positive)でS. mutans細胞をDAB染色した結果であり、図14Bは、ネガティブコントロール条件(DAB negatiev)でS. mutans細胞をDAB染色した結果である。
各条件で得られたTEM画像から、図14Aおよび図14Bの左上に示すように1つの細胞を任意に選定し、分析対象として3箇所を選択し、電子線透過量をLINEプロファイル分析した。1〜3の符号を付したLINEプロファイル分析結果について、Y軸は電子線透過量であり、X軸は分析に用いたフレームの長さ(nm)である。
それぞれのプロファイリング結果は、大まかに3つの領域に分けることができる。電子線透過量の値が最も低い領域は、黒色矢印で示すように細胞膜(membrane)であり、点線で囲んだ部分の両側の領域は、LRホワイトレジン(resin)または細胞の内側(cytoplasm)である。図14Aにおける白色矢印は、細胞表面もしくはペプチドグリカン層に局在化したDAB活性剤を示している。図14Aに示した3つのプロファイリング結果から得られた正規化した電子線透過量の平均値は、1.23と見積もられ、自然界に存在する細菌について報告された値(1.88)と近い値が得られた。
Claims (12)
- 口腔バイオフィルムの内部に存在する病原性細菌の細胞外電子伝達機構を制御することにより、前記病原性細菌の活性を低減させる方法。
- 前記病原性細菌の細胞内から細胞外への電子移動を抑制することにより、前記病原性細菌の細胞外電子伝達機構を制御する、請求項1に記載の方法。
- 口腔バイオフィルムの内部から外部への電子移動を抑制することにより、前記病原性細菌の細胞内から細胞外への電子移動を抑制する、請求項2に記載の方法。
- 口腔バイオフィルムの内部に存在する、電子伝達物質生成能を有する細菌による電子伝達物質の生成を阻害することにより、前記病原性細菌の細胞内から細胞外への電子移動を抑制する、請求項2または3に記載の方法。
- 前記病原性細菌の細胞外膜に存在する電子伝達酵素の活性を阻害することにより、前記病原性細菌の細胞内から細胞外への電子移動を抑制する、請求項2に記載の方法。
- 前記電子伝達物質がリボフラビンである、請求項4に記載の方法。
- 前記病原性細菌が口腔疾患の原因となる細菌である、請求項1から6のいずれか一項に記載の方法。
- 前記病原性細菌が、う蝕原性細菌または歯周病原性細菌である、請求項7に記載の方法。
- 前記病原性細菌が、Streptococcus mutans、Streptococcus sanguinis、Porphyromonas gingivalis、またはAggregatibacter actinomycetemcomitansである、請求項8に記載の方法。
- 口腔バイオフィルムの内部に存在する、電子伝達物質生成能を有する細菌によって生成される電子伝達物質の生成阻害剤を含有する口腔用組成物。
- 前記電子伝達物質がリボフラビンである、請求項10に記載の口腔用組成物。
- 口腔バイオフィルムの内部に存在する病原性細菌の細胞外膜に存在する電子伝達酵素に対する活性阻害剤を含有する口腔用組成物。
