Description
本発明は、鉄バクテリアが産生した鞘状酸化物に由来するリン酸イオンの吸着技術に関する。
従来、レプトスリックス属やガリオネラ属といった鉄バクテリアが知られている。鉄バクテリアは、鉄イオンを用いて水酸化鉄系の殻をつくり、赤褐色の沈殿物を形成する。浄水場ではこの鉄イオンの除去能に着目し、鉄バクテリアが用いられている。
一方で、自然界では、この沈殿物は、時に配管をつまらせたり、赤水被害をもたらせたりするなど敬遠され、廃棄物として処理される。

本発明は、自然物由来の新たなリン酸イオン除去技術を提供することを目的とする。
また、除去されたリンを含む肥料を提供することを目的とする。

請求項1に記載の発明は、鉄バクテリアが産生した鞘状酸化物を主成分とすることを特徴とするリン酸イオンの選択的吸着剤である。
鉄バクテリアの例としては、レプトスリックス属(Leptothrix)、ガリオネラ属(Gallionella)、スフェロチルス属(Sphaerotilus)、クロノスリックス属(Clonothrix)、トクソスリックス属(Toxothrix)、シデロモナス属(Sideromonas)、シデロカプサ属(Siderocapsa)又はシデロコッカス属(Siderococcus)を挙げることができる。
レプトスリックス属においては、レプトスリックス・オクラセア(Leptothrix ochracea)や、レプトスリックス・コロディニ(Leptothrix cholodnii)を挙げることができる。
なお、鞘状酸化物の形態としては、直線的な筒状のほか、らせん状、中空ひも状、カプセル状、ひも状と球状の凝集体も含まれるものとする。
レプトスリックス属においては、レプトスリックス・オクラセア(Leptothrix ochracea)や、レプトスリックス・コロディニ(Leptothrix cholodnii)を挙げることができる。
なお、鞘状酸化物の形態としては、直線的な筒状のほか、らせん状、中空ひも状、カプセル状、ひも状と球状の凝集体も含まれるものとする。
請求項2に記載の発明は、鉄バクテリアが、レプトスリックス・オクラセアであることを特徴とする請求項1に記載の吸着剤である。
請求項3に記載の発明は、リン酸イオンを含む処理対象液に対して、請求項1または2に記載の吸着剤を用いることによりリン酸イオンを選択的に吸着除去することを特徴とするリン酸イオン除去方法である。
請求項4に記載の発明は、リン酸イオン濃度が100ppm以下になるまで希釈した処理対象液を用いることを特徴とする請求項3に記載のリン酸イオン除去方法である。
請求項5に記載の発明は、請求項3または4に記載の方法によりリン酸イオンが吸着された吸着剤を含有することを特徴とする肥料である。
本発明によれば、自然物由来の新たなリン酸イオン吸着剤(請求項1および請求項2)、並びに、リン酸イオン除去方法(請求項3および請求項4)を提供することができる。
また、除去されたリンを含む肥料を提供することができる(請求項5)。

以下、本発明の実施の形態を図面を参照しながら詳細に説明する。
まず、鉄バクテリアとして、レプトスリックス・オクラセアを用い、鉄イオンが含まれる溶液にて鞘状酸化物を産生させた。

レプトスリックス・オクラセア(Leptothrix ochracea)は、鞘状酸化鉄の生成能を有する。試験にて用いた鞘状酸化鉄は浄水場から精製したものである。
なお、レプトスリックス・オクラセアは、浄水場の自然ろ過施設に存在し、堆積物中に存在するため、適宜これを用いて分離培養可能である。堆積物自体は以下に示すように遠心分離、減圧乾燥等を施して精製することができる。

<精製・分析>
京都府城陽市の浄水場にて20Lのタンクに採取した酸化鉄(レプトスリックス・オクラセアが生産した鞘状のセラミックス)を含んだ混濁液を静置し、酸化鉄を沈殿させた。上澄み液10Lをデカンテーションにより取り除き、イオン交換水を10L加えて混濁させた。これを1日静置し、酸化鉄を沈殿させた。これらの沈殿操作を5回繰り返した。残った混濁液を遠心管(800mLサイズ)に取り、5分間超音波を当てた後、遠心分離(9000rpm、10分間)により酸化鉄を沈殿させ、上澄み液を除いた。この操作を繰り返しタンク内の混濁液がなくなったら、イオン交換水で洗いながら1つの遠心管にバイオジナス酸化鉄をまとめた(この時、遠心管の底にたまった砂を混ぜないように注意した)。
遠心分離後、上澄み液を捨て、遠心管内の酸化鉄にエタノール(約2L)を加えてナスフラスコに移し、1時間撹拌した。エバポレーターでエタノールを留去し真空乾燥し、赤褐色粉末として75gの酸化鉄が得られた。
なお、元素分析の結果、C:1.87%,N:0.00%であった。また、酸化鉄の元素比をEDXにより決定すると、Fe:Si:P=73:22:5であった。酸化鉄のその他のキャラクタリゼーションの方法は、前記非特許文献1に記載の通りである。
鞘状構造物の大きさは、通常、直径約1μm程度、長さ約200μm程度である。得られた鞘状酸化物のバルキング状態、および、その光学顕微鏡写真とSEM写真とを、図1に示す。

水中に産生された鞘状酸化物に対して洗浄と遠心分離をおこない、乾燥した鞘状酸化物を得、これを用いて吸着試験をおこなった。

<吸着試験1:吸着能>
試験液として、リン酸二水素カリウム、塩化カリウム、臭化カリウム、硫酸カリウム、硝酸カリウムを水にとかし、PO4 3−、Cl−、Br−、SO4 3−,NO3 −の各20ppmの水溶液、各100ppmの水溶液、各1000ppmの水溶液を準備した。
試験液として、リン酸二水素カリウム、塩化カリウム、臭化カリウム、硫酸カリウム、硝酸カリウムを水にとかし、PO4 3−、Cl−、Br−、SO4 3−,NO3 −の各20ppmの水溶液、各100ppmの水溶液、各1000ppmの水溶液を準備した。
20ppmの試験液50mlに、鞘状酸化物を0.1g添加し、20時間経過後の吸着量(mg/g)をイオンクロマトグラフィーによって測定した。結果を図2に示す。図示したように、鞘状酸化物には、PO4 3−に対して特異的に高い吸着性能があることが確認できた。
また別途、2−line ferrihydrite 0.1gを、20ppmの試験液50mlに添加し、20時間経過後の吸着量を測定した。鞘状酸化物との対比を図3に示す。図示したように、2−line ferrihydriteはPO4 3−に対して高い吸着能を示すが、SO4 3−も同程度吸着する。換言すると、鞘状酸化物にはPO4 3−への選択的な吸着能があるといえる。
<吸着試験2:吸着速度>
次に、100ppmに調整した試験液50mlを用い、鞘状酸化物を0.1g添加し、10時間経過後の吸着量を測定し、20時間経過後のものと比較した。結果を図4に示す。図示したように、20時間経過後の方が吸着量が若干多くなるものの、吸着反応は比較的速やかに進行することが推察された。そこで、別途吸着速度を測定したところ、5分〜10分で濃度が半減し、添加後およそ40分で反応が鈍くなることを確認した(反応の鈍化は、吸着の進行に伴いPO4 3−濃度が小さくなり、鞘状酸化物との接触機会が低減することに由来するものと思われる)。この試験によれば、鞘状酸化物は、PO4 3−を速やかに吸着する性質があることがわかった。

次に、PO4 3−の1000ppm水溶液50mlに、鞘状酸化物0.1gを添加し、20時間経過後の吸着量を測定し、100ppm水溶液のものと比較した。結果を図5に示す。図示したように、吸着量自体は1000ppm水溶液の方が大きいが、吸着率は小さい。したがって、鞘状酸化物の効率的な吸着能の発揮を考えると、図2bの結果も加味すれば、PO4 3−濃度は100ppm以下とするのが好ましいといえる。
次に、PO4 3−の1000ppm水溶液50mlに、鞘状酸化物0.1gを添加し、20時間経過後の吸着量を測定し、100ppm水溶液のものと比較した。結果を図5に示す。図示したように、吸着量自体は1000ppm水溶液の方が大きいが、吸着率は小さい。したがって、鞘状酸化物の効率的な吸着能の発揮を考えると、図2bの結果も加味すれば、PO4 3−濃度は100ppm以下とするのが好ましいといえる。
以上の実験から、本技術を用いて、吸着剤を提供することができる。

なお、処理対象液に、リン酸イオンでなく、リン酸二水素イオン(H2PO4 −)、リン酸水素イオン(HPO4 2−)が含まれている場合は、適宜解離処理を前処理としておこない、リン酸イオンリッチな処理対象液化する工程を含ませてもよい。

なお、処理対象液に、リン酸イオンでなく、リン酸二水素イオン(H2PO4 −)、リン酸水素イオン(HPO4 2−)が含まれている場合は、適宜解離処理を前処理としておこない、リン酸イオンリッチな処理対象液化する工程を含ませてもよい。
また、本技術を用いて、肥料を得ることもできる。

特に、肥料については、図3に示した様に、硫酸イオンが混在する処理対象液を処理しても、硫酸イオンは事実上吸着しないため、効率的にリンを施肥できることとなる。
本発明によれば、従来廃棄されてきた鉄バクテリア由来の鞘状酸化物に新たな用途を見つけ、資源として活用することができる。

- 鉄バクテリアが産生した鞘状酸化物を主成分とすることを特徴とするリン酸イオンの選択的吸着剤。
- 鉄バクテリアが、レプトスリックス・オクラセアであることを特徴とする請求項1に記載の吸着剤。
- リン酸イオンを含む処理対象液に対して、請求項1または2に記載の吸着剤を用いることによりリン酸イオンを選択的に吸着除去することを特徴とするリン酸イオン除去方法。
- リン酸イオン濃度が100ppm以下になるまで希釈した処理対象液を用いることを特徴とする請求項3に記載のリン酸イオン除去方法。
- 請求項3または4に記載の方法によりリン酸イオンが吸着された吸着剤を含有することを特徴とする肥料。
