12%〜15%のグリセリン、PPG又はそれらの混合物と、
8%〜12%の液体油と、
を含む、チュアブル組成物を提供する。
有効成分部は、液体又は固体の形態の有効成分(複数の場合がある)単独であってもよい。固体形態の場合、有効成分部は有効成分(複数の場合がある)単独であってもよく、例えば、純粋な形態、又は有効成分と、少なくとも実質的にチュアブル組成物の製造においてその完全な状態を維持する賦形剤(複数の場合がある)及び粉末を形成する製剤用添加剤(複数の場合がある)との造粒としての有効成分(複数の場合がある)を含む粉末形態、又は粒子若しくは顆粒であってもよい。
有効成分(複数の場合がある)に加えて、有効成分部が固体形態である場合、固形分は、香味料、崩壊剤、増量剤、甘味料、可塑剤、結合剤、防腐剤の少なくとも1種を含む。可塑剤及び/又は結合剤と共に、少なくとも香味料及び崩壊剤が含まれることが好ましい。例えば、上に列挙される成分はいずれも有効成分(複数の場合がある)と共に含まれる。単一の成分が、上に列挙される幾つかの成分の役割を果たす場合がある。例えば、香味料は甘口香味料である及び/又は増量剤として作用する場合があり、可塑剤は結合剤としても作用し得る。
肉食動物、特にイヌ及びネコによる受容を改善するため、上記組成物に高い割合の肉香味料を含むことが有利である。
5%〜25%、例えば約15%又は少なくとも約15%の割合の崩壊剤は、チュウにおいて有用となる場合がある。崩壊剤の割合が低すぎる場合、チュウが摂取後に容易に崩壊しないというリスクがある。崩壊剤の割合が高すぎる場合、チュウを製造することが困難であり、チュウの構造が粘着力を欠くというリスクがある。
少なくとも幾つかの実施形態では、デンプンは結合剤及び/又は崩壊剤として有用である。所望の粘度に応じて、デンプンを予め糊化してもよく(pre-gelatinized:アルファ化してもよく)、例えば部分的に又は全てを予め糊化してもよい。化工デンプンは、予め糊化したデンプンに対する代替物又は補足物である。限定的な量のデンプンは、デンプン以外の崩壊剤、例えばいわゆるスーパー崩壊剤によって補足される。崩壊剤の量は、動物による摂取後にチュウの所望の分解と共に適切な粘度を得るように調整される。
幾つかの実施形態では、可塑剤及び/又は結合剤は、例えば最大15%、又は最大12%、又は更には最大10%、例えば5%〜15%、2%〜12%、5%〜12%、又は更には5%〜10%の量で存在する。
本発明の一態様では、液体であってもよい流動分は、製品の最終粘度に重要な役割を果たす。特に、流動分及び/又は液体は、例えば、製品が圧縮及び/又は押出しによって得られる場合、固形分の組み合わせにおいて最終製品を形成するように支援する。
本発明において有用な可能性のある湿潤剤として、湿潤助剤(複数の場合がある)と共に、エチレングリコール(EG)、ポリエチレングリコール(PEG)、プロピレングリコール(PG)、ポリプロピレングリコール(PPG)、及びそれらの少なくとも2種の組み合わせ、又はそれらの少なくとも1種が挙げられる。
流動性又は液体の油(複数の場合がある)は植物油であってもよく、又は鉱物油であってもよい。ダイズ油は押出しによってチュウを製造するのに有用である。ダイズ油は、典型的には、最終ブレンドに均一に添加される場合、投薬の端のきれいな押出しを可能とし、また切断及び包装の際の取り扱いを容易にする、液体滑剤として作用する。代替的な又は追加的な成分として以下が挙げられ得る:PEG、オレイン酸グリセリル、中鎖脂肪酸、トウモロコシ油、硬化ココナッツ油及び硬化パーム核油、又はMygliol(商標)(SASOL)等の化工油、植物性ショートニング、ヒマシ油、鉱油、魚油、様々なエステル、他の植物油。
上記組成物は、防腐剤又は防腐剤の組み合わせを含んでもよい。また、他の添加剤、例えば抗酸化剤、着色剤、及び/又は塩等の味感向上剤が存在してもよい。
製品は、例えば圧縮成形及び/又は押出しによって得られてもよい。
クロスカルメロースナトリウム含有率の効果を検証するため、開発バッチ(development batch)を変動する量の崩壊剤を用いて作製した。プラジクアンテル及びイベルメクチンの両方について、中型の錠剤及びブロック(10%又は15%のいずれかのクロスカルメロースナトリウムを含む)に対して崩壊の比較を行った。

この実験は、標準のソフトチュウ製剤中の或る特定の成分の重要性、並びにそれらがチュウの混合及び押出しにおいて果たす役割を検証した。検証される成分は、ポリエチレングリコール(PEG)、ダイズ油、及びグリセリンであった。PEGは、例えばPEGが流動分として添加される場合、流動分又は流動分に類するものとみなすことができ、又は押出し若しくは加熱等のチュウ製造プロセスの少なくとも1つの工程の間に流動性とされてもよい。
バッチ1(PEGなし)
バッチで使用される標準的製剤との観察される相違はなかった。
このバッチは、標準ブレンドよりもHobartミキサーに負荷を与えるように見える。ほとんどの材料がパドルにこびりつき/引っかかって、20分間のうち最後の5分間は適切に混合することができなかった。最終ブレンドは、先のバッチよりも粘性で、よりガム様であった。
この最終ブレンドは、先の2つのバッチと比較して乾燥した外観を有した。最終ブレンドを「ウェットサンド」と記載してもよく、典型的な生地の粘度を有しない。
3.3g〜3.6gのチュウ重量に対して、別段の指示がない限り、華氏85度、ジャケット温度華氏55度にて、中型のダイサイズにより押出しを行った。
このバッチは、錠剤に類似するがPEG粒子を含まないチュウを押出した。一部過剰な油があり、端がボロボロになっていた(frayed edge)。
このブレンドは押出したが、押出し物はボロボロとしており、チュウの表面は滑らかではなかった。
このブレンドは押し出さなかった。最終的にカッターによって押したわずかなチュウは乾燥していて崩れやすかった。過剰の油が押出し機から浸み出し、先述のどのソフトチュウバッチよりも多くの油が観察された。
以下の実験では、混和性液体の予備混合を除くため、液体を添加する順番を修正した。ブレンド及びチュウの肉眼的観察を行った。
本明細書に従って押出し可能なブレンドを調製した。
25℃/60%相対湿度において、玩具サイズ又は大サイズのチュウにて実施例4の組成物に対して、安定性チャンバーにおける安定性の試験に成功した。
25℃/60%相対湿度−T=0ヶ月、1ヶ月、2ヶ月、3ヶ月、6ヶ月、9ヶ月、12ヶ月、18ヶ月、24ヶ月、30ヶ月、及び36ヶ月
40℃/75%相対湿度−T=0ヶ月、1ヶ月、2ヶ月、3ヶ月、及び6ヶ月
30℃/65%相対湿度の試料を保存したが、40℃/75%相対湿度の試料により不合格がない場合は試験しなかった。
目的は、様々な比較のデンプン系チュウ(典型的には25%〜30%超)の崩壊を本発明によるソフトチュウ試料と比較することであった。

- 前記流動性油が液体油である、請求項1〜4のいずれか一項に記載のチュアブル組成物。
- 前記湿潤剤がグリセリンであり、前記流動性油がダイズ油である、請求項1〜4のいずれか一項に記載のチュアブル組成物。
