Description
本出願は、米国特許法35U.S.C.§119(e)に基づいて2010年6月16日に出願の米国特許仮出願第61/355,364号の優先権を主張する2011年6月15日に出願の米国特許出願第13/161,251号の一部継続出願である、2015年5月22日に出願された米国特許出願第14/719,921号の優先権を主張する。これらの特許文献のそれぞれは、参照によりその全体が本明細書に組み込まれる。

本発明は、放射性ナノ粒子ならびに放射性ナノ粒子の製造およびブラキセラピー用を含む使用方法に関する。
一態様では、中空ナノ粒子が本明細書で記載され、該ナノ粒子は、いくつかの実施形態では、以前のナノ粒子に比べて、1つまたは複数の利点を提供し得る。いくつかの実施形態では、例えば、本明細書で記載の中空ナノ粒子は、約600nm〜約900nmの調整可能なSPRピークを示し、それにより、種々の画像化、治療、セラノスティックおよびセンシング用途に有用な特性を与える。いくつかの実施形態では、本明細書で記載の中空ナノ粒子は、望ましい磁気および/または光熱特性を示す。いくつかの実施形態では、本明細書で記載の中空ナノ粒子は、磁気共鳴画像法(MRI)および陽電子放出断層撮影(PET)にとって有用である。いくつかの実施形態では、本明細書で記載の中空ナノ粒子は、望ましい触媒特性を示す。いくつかの実施形態では、本明細書で記載の中空ナノ粒子は、非毒性である。いくつかの実施形態では、本明細書で記載の中空ナノ粒子は、光熱療法用として機能させることが可能である。

別の態様では、表面上に中空ナノ粒子を選択的に沈着させる方法が本明細書で記載され、該方法は、いくつかの実施形態では、以前の方法に比べて、1つまたは複数の利点を提供し得る。例えば、いくつかの実施形態では、本明細書で記載の表面上に中空ナノ粒子を選択的に沈着させる方法は、簡単で効率的である。いくつかの実施形態では、表面上に中空ナノ粒子を選択的に沈着させる方法は、異なる疎水性を有する複数の領域を有する基板を用意すること、複数の気泡を形成すること、および複数の気泡の内の少なくとも1個の表面上にシェルを形成し、中空ナノ粒子を形成することを含む。いくつかの実施形態では、中空ナノ粒子は、第1の疎水性を有する1個または複数の領域上に選択的に沈着される。いくつかの実施形態では、基板はパターン形成基板を含む。
別の態様では、放射性ナノ粒子が本明細書で記載される。いくつかの実施形態では、本明細書で記載の放射性ナノ粒子は、金属ナノ粒子コアおよび金属ナノ粒子コアのまわりに配置された外側金属シェルを含む。さらに少なくとも1種の金属放射性同位体が金属ナノ粒子コアおよび/または外側金属シェル内に配置される。さらに、いくつかの事例では、本明細書で記載の放射性ナノ粒子は、金属ナノ粒子コアと外側金属シェルとの間に配置された内側金属シェルをさらに含む。
別の態様では、放射性ナノ粒子の製造方法が本明細書で記載される。いくつかの実施形態では、本明細書で記載の放射性ナノ粒子の製造方法は、金属ナノ粒子コアを用意すること、および金属ナノ粒子コアのまわりに内側金属シェルを形成することを含み、内側金属シェルは第1の金属または金属の組み合わせを含む。さらに、方法は、金属ナノ粒子コアのまわりに外側金属シェルを形成することをさらに含み、外側金属シェルは、少なくとも1種の金属放射性同位体を含む第2の金属または金属の組み合わせを含む。いくつかの事例では、第2の金属または金属の組み合わせは、非放射性金属同位体をさらに含む。
さらに別の態様では、ブラキセラピーを実施する方法が本明細書で記載される。いくつかの実施形態では、ブラキセラピーの実施方法は、本明細書で記載の組成物を生体コンパートメント内に配置することを含む。例えば、いくつかの事例では、組成物は複数の分散した放射性ナノ粒子を含み、「分散した」放射性ナノ粒子は、非凝集化または実質的に非凝集化ナノ粒子であり、本明細書セクションVIIIで前述の平均寸法を有するナノ粒子が含まれる。例えば、いくつかの実施形態では、組成物は、複数の放射性ナノ粒子のコロイド状分散体である。当業者ならわかるように、このようなコロイド状分散体は、ナノ粒子を分散させる溶媒または分散媒をさらに含むことができる。本開示の目的と矛盾しないいずれの溶媒または分散媒も使用し得る。例えば、いくつかの事例では、コロイド状分散体は水性分散液である。さらに、いくつかの実施形態では、放射性ナノ粒子は血清または緩衝液中に分散される。本明細書でさらに前述したように、このような放射性ナノ粒子のコロイド状分散体は、ブラキセラピーを実施する以前の方法で使用される比較的大きなカプセル化放射線療法剤よりも多くの利点を提供できる。
積層膜を使って形成した中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。図1に示すように、参照電極としての3MのNaCl溶液中のAg/AgCl電極および対極としての白金メッシュを備えた三電極電着セルを使ってナノ粒子を形成した。Princeton Applied Research 273A Potentiostat/Galvanostatを使って、作用電極に電位を印加した。2〜5枚の市販のアルミナ膜の積層体(Whatman Corp.)が複数の核生成基板を提供した。それぞれの膜は約60μmの厚さで、全体厚さを貫通するチャネルが設けられた。それぞれの膜の片側には、小さい分岐部が認められた。チャネル直径は、約300nmであり、分岐部の直径は、約20nm〜約200nmに変動した。チャネル密度は、約109/cm2であった。膜の上面図および断面図を図2に示す。それぞれの膜の分岐側が、たとえ存在しても上の膜ではなく、下の膜に最近接となるように、膜を積層した(すなわち、分岐側を膜積層体の各層の上端ではなく下端になるように配向させた)。500nmのCu層を積層体の下端の膜の底側上にスパッタリング蒸着し、作用電極として機能させた。1cmの直径のオーリングを備えたテフロン(登録商標)セルを膜積層体の最上部に配置した。
パターン形成基板を使って形成した中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。図8に示すように、参照電極としての3MのNaCl溶液中のAg/AgCl電極および対極としての白金メッシュを備えた三電極セルを使って中空Auナノ粒子を形成した。ガラス基板(光学顕微鏡スライド)上のAgストライプからなるリソグラフィーによりパターン形成した電極を作用電極として使用した。光学顕微鏡ガラススライドを脱イオン水で濯いだ後、使用の前にプラズマ処理で清浄化した。フォトリソグラフィを使って、ガラス基板にパターン形成した。フォトマスク設計を図9Aに示す。ストライプの幅は50μmとし、ストライプを100μm毎に繰り返した。Agストライプパターン形成基板を図9Bに示す。基板の非コーティングガラス領域は、中空Auナノ粒子の核生成および成長のための表面を提供した。Princeton Applied Research 273A Potentiostat/Galvanostatを使って、作用電極に電位を印加した。
TEMグリッドを使った中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。TEMグリッドを作用電極および核生成基板として使用したこと以外は、実施例2で記載のものに類似の方法を使って中空Auナノ粒子を形成した。TEMグリッドは、炭素膜でコーティングされた銅メッシュから構成された。図11に示すように、中空Auナノ粒子が炭素膜上で観察された。スケールバーは1μmである。電着後に、何らそれ以上の処理をすることなく、高解像度TEM(HR−TEM)による特性評価を実施した。
多孔質シェルを有する中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。実施例1で記載のものに類似の方法を使ってナノ粒子を形成した。5枚の陽極酸化アルミナ濾過膜の代わりに2枚の積層体を使い、700nm層のCuをスパッタリング沈着して、底部膜の細孔を塞ぎ、作用電極として機能させたことを除いて、実施例1に記載のように電着セルを使った。市販のAu亜硫酸塩溶液(Techni−Gold 25 ES RTU(Technic,Inc.製))を電解質として使用した。溶液の初期pHは約7.0であった。溶液に0.4Mのスルファミン酸Ni溶液を加えて、pHを約6.0にした。Princeton Applied Research 273A Potentiostat/Galvanostatを使って、−0.80V(対Ag/AgCl参照電極)の電位を作用電極に印加した。この電位で水素放出が発生した。多孔質シェルを有する中空Auナノ粒子を得るために、反応時間を10分未満に保持した。実施例1に記載のようにして、多孔質シェルを有する中空Auナノ粒子を精製し、単離した。
中空Auナノ粒子を含む中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。印加電位をパルス出力したことを以外は、実施例1で記載のものに類似の方法を使って中空Auナノ粒子を形成した。ナノ粒子の製作を、600秒間の−0.8V(対Ag/AgCl)の印加電位で開始し、続けて、300秒間の開回路とした。2回の追加のサイクルにわたりパルス出力電位を繰り返した。図13に示すように、二重シェルナノ粒子が得られた。内側の空洞の平均直径は約50nmで、全体寸法は約300nmであった。スケールバーは200nmである。その他の二重シェルナノ粒子を図14に示す。図14A〜Cのスケールバーは100nmである。図14Dのスケールバーは20nmである。
Fe3O4ナノ粒子を含む中空Auナノ粒子
本明細書のいくつかの実施形態と一致するFe3O4ナノ粒子を含む中空Auナノ粒子を以下のように形成した。多孔質シェルを有する中空Auナノ粒子を実施例4に記載されているものに類似の方法で調製した。0.2Mの亜硫酸ナトリウムを使って電解質のpH値を約6.5に調節し、反応時間を400〜600秒とした。得られた中空Auナノ粒子は、直径約50nmの空洞および約25nm未満の厚さのシェルを有していた。シェルは多孔質で、HR−TEMで測定して、約2〜3nmの細孔寸法を示した。
ドープFe3O4ナノ粒子を含む中空Auナノ粒子
本明細書のいくつかの実施形態と一致するドープFe3O4ナノ粒子を含む中空Auナノ粒子が以下のように形成される。Fe2+およびFe3+イオンと共にドーパントイオン源も供給されること以外は、Fe3O4ナノ粒子を含む中空Auナノ粒子を実施例6に記載されているものに類似の方法で調製した。ドーパントイオンには、陽電子放出断層撮影(PET)画像化に有用な64Cu2+または89Zr4+などの核種が含まれる。実施例6に記載されているものに類似の方法によって、Au/ドープFe3O4ナノ複合材料が調製および精製されると、表面が以下のように官能化される。アルミナ膜の溶解の前に、リポ酸またはジヒドロリポ酸(DHLA)の溶液が膜に添加され、この配位子と中空Auナノ粒子表面との結合が生じる。その後、膜は脱イオン水でさらに濯がれる。その後、リポ酸/DHLA配位子のカルボン酸基は、N−(3−ジメチルアミノプロピル)−N”−エチルカルボジイミド塩酸塩(EDC)およびN−ヒドロキシスルホスクシンイミド(スルフォNHS)によるカルボジイミドカップリングを用いてNH2GR11にカップリングされる。NH2GR11は、例えば、Gao et al.,Amino Acids,2010 March 11:20221650に記載されている前立腺癌特異的ポリアルギニンペプチドである。この文献は参照によりその全体が本明細書に組み込まれる。ペプチド標的化薬剤を含むナノ複合粒子は、その後、サイズ排除高速液体クロマトグラフィー(HPLC)または3サイクルの約30kDaの分画分子量のセントリコンフィルターを使った遠心分離濾過により精製される。このプロセスを図19に示す。
治療薬を含む中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子が以下のように形成される。多孔質シェルを有する中空Auナノ粒子が実施例4または実施例6に記載されているものに類似の方法で調製される。その後、多孔質中空Auナノ粒子を充填したアルミナ膜が薬物などの治療薬の濃縮溶液中に浸漬される。膜を溶液中に十分な時間にわたり保持し(例えば、10〜600分)、治療薬をAuナノ粒子空洞中に拡散させる。その後、Na3Au(SO3)2などの金属含有前駆物質を膜中の溶液に添加し、多孔質Auシェルを成長させて、少なくとも一部の細孔を密封する。その後、得られた治療薬を含む中空Auナノ粒子は医学的処置に使用できる。封入された治療薬は、Auシェルの破壊により放出される。シェルのSPR周波数またはそれに近い波長を有する光をシェルに照射することによりシェルは破壊される。
ラマン活性種を含む中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。中空金ナノ粒子を実施例4に記載のように合成した。中空金ナノ粒子は、約50nmの空洞直径および約100nmの外径で、730nmを中心とする吸収ピークを有する。約1.1x1011個のナノ粒子/mLを含む陽極酸化アルミニウム膜を新たに作製した20mLの、5μMのジエチルチアトリカルボシアニン(DTTC)ラマンレポーター染料の溶液中に浸漬し、室温で3時間保持した。その後、アルミナ膜を脱イオン水で数回濯いだ。その後、ナノ粒子充填膜を、20μMのSH−mPEG(MW 5kDa、mPEGはメトキシポリエチレングリコールを意味する)の溶液中に4℃で一晩浸漬した。次に、1MのNaOH溶液を使って、アルミナ膜を溶解した。ナノ粒子を脱イオン水中への数回の分散とそれに続く遠心分離により精製した。このプロセスは、図20に図示されている。
標的化化学種を含む中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。ラマンレポーター染料およびポリエチレングリコールの添加を次のように実施したこと以外は、実施例9に記載のようにしてラマンナノタグを調製した。新たに調製したラマンレポーター染料の溶液を、多孔質中空Auナノ粒子を充填したアルミナ膜を通して流し、続けて、SH−mPEG(10μM、MW 5kDa)およびSH−PEG−COOH(1μM、MW 2kDa)の体積比2:7の溶液の混合物を膜を通過させた。精製ラマンナノタグの1mLの溶液に、EDCおよびスルフォNHSを添加し、30分間インキュベートした。その後、複合粒子を、3回の遠心分離とそれに続くPBS中の再分散により精製した。次に、抗PSMA(「PSMA」は、前立腺癌中で過剰発現したII型膜貫通型糖タンパク質を意味する)を活性化エステルナノタグの溶液に加え、混合物を4℃で一晩反応させた。得られた抗体−ナノタグコンジュゲートをサイズ排除カラム分離またはセントリコン遠心分離により精製した。
粗面化表面を有する中空Auナノ粒子
本明細書のいくつかの実施形態と一致する中空Auナノ粒子を以下のように形成した。電解質のpHを下記のように変更した以外は、粗面化表面を有する中空Auナノ粒子を実施例1に記載されているものに類似の方法で調製した。約6.0のpH有する電解質を使って調製した中空Auナノ粒子は、比較的平滑なシェルを示した。表面粗さを高めるために、2MのNa2SO3(pH約9.0)の添加により、電解質のpHを約6.5または7.0に高めた。理論に束縛されることを意図するものではないが、粗度のpH依存は、pHによるNa3Au(SO3)2の自己触媒反応の速度の増加(およびそれに伴う、結晶粒成長および最終結晶粒径の増加)に起因し得る。図24は、異なるpH値の電解質を使って合成した中空金ナノ粒子の表面形態を示す。図24Aは、約6.0の電解質pHで形成された平滑なシェルを示す。図24Bは、約5nmの表面粗さを有し、約6.5の電解質pHで形成されたシェルを示す。図24Cは、約8nmの表面粗さを有し、約7.0の電解質pHで形成されたシェルを示す。図25は、中空Auナノ粒子の水性懸濁液の対応する吸収スペクトルを示す。図25の破線は、シミュレートした吸収プロファイルに対応する。プラズモンピークは、電解質pHの増加と共により長い波長にシフトした。pHは、約6.0〜6.5〜7.0に変化する場合、SPRピークは、約600nm〜630nm〜730nmにシフトした。
半球形および管状Auナノ粒子
本明細書のいくつかの実施形態と一致する半球形および管状Auナノ粒子を以下のように形成した。実施例1に記載されているものに類似の電気化学的沈着セルを使用した。電解質を電着セル中に配置した。最初に、約3%の硫酸、約3%のエチレンジアミン(EDA)、約10%の亜硫酸金ナトリウム(Na3Au(SO3)2、および約7%の亜硫酸ナトリウム(Na2SO3)から構成される水溶液を調製することにより電解質を調製した。溶液は、約7.0のpHであった。次に、溶液に0.01Mの硫酸(H2SO4)を加えて、pHを約4.0に下げた。電解質を電着セル中に配置した後、−0.6V(対Ag/AgCl)より負の電位を約10〜60分間印加した。約10分の沈着時間では、半球形金ナノ粒子が細孔壁上に観察された(図26A)。約60分の沈着時間では、管状金ナノ粒子が観察された(図26B)。図26のスケールバーは200nmである。理論に束縛されることを意図するものではないが、ナノ粒子の形態は、H2気泡の細孔壁面に対する接触角により影響を受け、次に、この接触角は細孔壁面の疎水性に影響されると考えられている。
中空Auナノ粒子の光熱特性
近赤外(NIR)レーザー照射下での組織様ファントム中の中空Auナノ粒子の光熱特性を調査した。1%イントラリピッド、ゼラチン粉末、およびパラホルムアルデヒドを使って、ゲル状ファントムを調製した。簡単に説明すると、2400mgの高度精製ゼラチン粉末を228mLの脱イオン水と混合した。次に、混合物を、間欠的に混合を加えながら、ゼラチンが完全に溶解し、溶液が透明で無色に見えるまで、マイクロ波で2〜4分間(約900℃に)加熱した。室温で連続的な混合を行って、ゼラチン溶液を600℃まで冷却させ、この時点で、12mLの1%イントラリピッド(20%脂肪乳剤、Sigma−Aldrich)および140mgのパラホルムアルデヒド(95%、Sigma−Aldrich)を加え、これにより、その溶液を白色で不透明にした。ゲル状ファントムの形成後、ファントム中に細長いポケットを形成した。750nmを中心とする波長のSPRピークを有する中空Auナノ粒子の懸濁液(50μL、3.0x109個のナノ粒子/mL)をピペットを使ってポケット中に配置した。ダイオードレーザーファイバー(平均波長810nm)をナノ粒子懸濁液に接触させてポケットの中央に配置した。レーザーファイバーを使って、中空Auナノ粒子を照射した。ファントム中の温度変化を温度計により距離と時間の関数として記録した。図27は実験構成を示す。
放射性ナノ粒子
本明細書で記載のいくつかの実施形態による放射性ナノ粒子を以下のように形成した。
ブラキセラピーの実施方法
実施例14の放射性ナノ粒子のブラキセラピーのための使用を動物モデルで以下のように評価した。
実施例14の放射性ナノ粒子または「ナノシード」の、それらの腫瘍部位での保持、毒性、および治療効力を含むインビボ評価のために、ヒト前立腺癌腫瘍担持SCIDマウスを使用した。Matsuno et al.,“Induction of lasting complete regression of preformed distinct solid tumors by targeting the tumor vasculature using two new anti−endoglin monoclonal antibodies,” Clinical Cancer Research,5,371− 382(1999);およびVanWeelden et al.,“Apoptotic regression of MCF−7 xenografts in nude mice treated with the vitamin D3 analog,EB1089,” Endocrinology,139,2102−2110(1998)に従い、わずかな修正を加えて、腫瘍誘導を実施した。さらに詳細は下記に提供される。3x106個のPC3細胞を含む細胞懸濁液をSCIDマウスの両肩部の皮下に移植した。腫瘍を4週間増殖させ、約181.67±62.14mm3の触知可能寸法に達した。0日目に動物を3つの群(n=6)、すなわち、(a)PBS溶液、(b)「コールド」Pd@Auナノ粒子PBS懸濁液(このような「コールド」ナノ粒子は、放射性のPd−103を含まないこと以外は、実施例14ノナのシードと同じであった)、または(c)「ホット」ナノシードPBS懸濁液、に無作為化し、SCIDマウス担持腫瘍に注入した。群(a)および(b)は、対照としての役割をする。6〜9箇所の無作為に選択した部位に注入を実施し、全腫瘍性腫瘤中に投与量を均一に分配した。実験試料(すなわち、ホットナノシード試料)として、Pd−103を含む1.5mCiのホットナノシードを腫瘍中に注入した。懸濁液の体積は、2.03x1010個のナノ粒子/mLのナノ粒子濃度で40μLであった。同じ量のPBS溶液およびコールドナノシード組成物を2つの対照群の腫瘍中に注入した。
実施例14のホットナノシードで処置した群では、腫瘍内注入後、時間経過に従って(注入後0、1、2、4日目(d.p.i)および注入後、1、2、4および5週目(w.p.i.))、SPECT/CTイメージングを実施し、小動物用SPECT/CTスキャナーによるPd−103の低エネルギーX線放射の取得により、ナノシードの保持を非侵襲的にモニターした。結果を図39と図40に示す。1d.p.iでの定量的SPECT分析(図40)は、注入投与量は投与部位で留まり(101.50±23.72%ID/g)、無視できる量の放射能が肝臓(0.11±0.06%ID/g)でおよび脾臓(0.14±0.01%ID/g)で観察されたことを示した。調査が進むに伴い、定量的SPECT分析で測定された腫瘍中の取り込みレベルは、5w.p.i.で、徐々に274.48±77.62%ID/gまで増加した。この理由は、ナノシードの放射線療法効果に起因して、腫瘍体積が収縮するためである。
実施例14の放射性ナノ粒子の毒性を評価し、上述の対照群および「コールド」ナノシードと比較した。5週の処置期間にわたり、全血球数(CBC)、アラニンアミノ基転移酵素(ALT)、アスパラギン酸アミノ基転移酵素(AST)、血中尿素窒素(BUN)、およびクレアチニンレベルを10および30d.p.i.にモニターした。赤血球(RBC)数およびRBC当たりの平均ヘモグロビン量(MCH)は、調査の全期間にわたり影響を受けず、治療が溶血作用を誘発しなかったことを示す。10d.p.iで、放射線療法でよく認められる、放射性ナノ粒子による初期の白血球(WBC)数の低下があったことがさらに注目される。しかし、30d.p.i後にWBC数の正常値への復帰が認められることから、この効果は可逆的であることが明らかになった。類似の効果は、血小板数およびその他のパラメータの場合にも観察された。BUN、ALT、ASTおよびクレアチニン試験は、3つの群のマウスの間で顕著な変化を示さず、放射性ナノ粒子に伴う腎臓および肝臓関連毒性がないことを示した。
3つの群の担腫瘍マウスの腫瘍体積(図41)を、ノギスを使って1日おきに二重盲検方式で測定した。15d.p.i後、明確に異なる腫瘍成長傾向が認められた(p<0.0001)。放射性ナノ粒子で処置した対象では、腫瘍体積の顕著な減少が認められたが、PBSおよびコールドナノシードの両群では、腫瘍体積の徐々に進行する増大が観察された。放射性ナノ粒子試験群における2匹のマウスの2つの腫瘍の体積が、35d.p.i後には認められなくなるほどに収縮したことは注目に値する。PBSおよびコールドナノシード群の平均腫瘍体積は、67.08±30.96mm3および58.75±35.29mm3から、それぞれ、187±80.11mm3および122.14±4.082mm3まで増大した。一方、放射性ナノ粒子群では、35日の処置後、82.75±46.25mm3から19.83±20.12mm3(p<0.001)への有意な腫瘍体積の低減が観察された。
Pd−103の低エネルギー放射を使ったSPECTイメージング法
NanoSPECT/CT Plus System(Bioscan,Washington,DC,USA)によるPd−103を使ったSPECTイメージング法を開発した。エネルギーピークおよび幅を、それぞれ、18keVおよび60%に設定することにより、Pd−103同位体をNanoSPECT/CT Plus同位体ライブラリーに加えた。その後、3mLのシリンジおよび1.2mCiのPd−103を使って、定量化較正を実施した。
放射性ナノ粒子投与量(約1.5mCi)をPBS中で調製し(pH7.4)、PC3−腫瘍担持SCIDマウスの腫瘍内に注入した。6〜9箇所の無作為に選択した部位に腫瘍内注入を注意深く実施した。注入後、NanoSPECT/CT Plusシステムを使って、小動物画像化を実施した。それぞれの投与量の腫瘍内注入後、SPECTおよびCT画像を、0、1、2、4、7、14、21および35d.p.i.に取得した。SPECT/CTの視野(FOV)の中心をマウスの肩部に置いた。55kVp、1000ms露出、および1:1のビニング数の条件で1回転当たり360投影によりCTイメージングを実施した。0.73mmの再構築後の解像度をもたらすマルチピンホール開口でコリメートした4検出器アレイでSPECTデータを収集した。SPECT像再構築を、35%平滑化、100%分解能、および3x3繰り返し(標準モード)で、HiSPECT NG(Scivis wissenschaftliche Bildverarbeitung GmbH,Germany)を使って実施した。InVivoScope2.0ソフトウェアパッケージ(Bioscan,Washington,DC,USA)を使って腫瘍活性の定量化を実施した。CTおよびSPECT画像の同時位置合わせ後、臓器を含む全ての面の腫瘍および肝臓を包含する円筒形関心領域(ROI)を描出した。
視野(FOV)の中心で有効空間分解能が1.4mmのSiemens Inveon PET/CTマルチモダリティーシステム(Siemens Medical Solutions,Knoxville,TN)を使って、マウスPET/CTイメージングを実施した。PETイメージングの前12時間にわたり全動物を絶食させた。各マウスは、150μLの生理食塩水中の150μCiのFDGを尾静脈注射により静脈内に投与された。画像収集の前およびその間、マウスをヒートパッド上に置いた。注射後(P.I.)1時間目に、動物に2.5%イソフルランを投与して、PET画像を15分間取得した。PET画像を3D Ordered Subsets Expectation Maximization(OSEM3D/MAP)アルゴリズムを使ってシングルフレームに再構築した。PET直後に、マウスの肩部を中心とするFOVで、CT画像を取得した。80kVpの電力、500μAの電流、145msの露出時間、4のビンニング、および102μmの有効ピクセルサイズの条件でCT投影像(360ステップ/回転)を取得した。ダウンサンプル因子2を用いたCT像再構築プロトコルを設定して、双一次的に補間し、Shepp−Loganフィルターを使用した。分析を行うために、Inveon Acquisition Workplace(Siemens Medical Solutions,Knoxville,TN)によりPETおよびCT画像を同時位置合わせした。組織を含む全ての面の腫瘍を包含する関心領域(ROI)を手作業で描出した。グラムあたりのパーセンテージ注入投与量として標的活性を計算した。
放射性ナノ粒子の注入後、1日目、1週目、2週目、3週目、および5週目に、3匹のマウスを屠殺し、血液、心臓、肺、筋肉、骨、脂肪、肝臓、脾臓、腎臓、胃、小腸、大腸、脳、尾、および腫瘍を含む目的の臓器を採取して秤量し、20mLのバイアルに移した。それぞれの臓器の放射能を測定するために、それぞれのバイアルの放射能をγカウンター(Perkin Elmer 2480 Wizard)で測定し、1分当たりのカウント数として記録した。その後、バイアルに王水を加えて一晩放置し、臓器を消化した。24時間後、150℃で王水を煮沸除去した。煮沸後、10mLの1%HCl溶液をバイアルに加えた後、30分間超音波処理した。次に、誘導結合プラズマ質量分析計(ICP−MS、Agilent 7700x)でAuおよびPd濃度を測定した。各試料について、この測定を少なくとも3回繰り返した。
定量データは、平均±平均の標準誤差(SEM)で表した。1元配置分散分析により平均値間の比較および有意性評価を実施した。0.05未満のP値を統計的に有意と見なした。異なる時点の、異なる群およびそれぞれ個々の群由来のデータを比較し、それらが統計的に区別可能かどうかを判定した。全データ解析は、SPSSバージョン16.0ソフトウェア(IBM SPSS Statistics)を使って実施した。
放射性ナノ粒子およびブラキセラピーの実施方法
以下のように、本明細書で記載の一実施形態による放射性ナノ粒子が調製され、ブラキセラピーを実施するために使用される。
放射性の磁気ナノ粒子ならびにブラキセラピーおよび画像化の実施方法
以下のように、本明細書で記載の一実施形態による放射性ナノ粒子が調製され、同時またはセラノスティックス法によるものを含む、ブラキセラピーおよびMRIを実施するために使用される。
最初に、複合磁気ナノ粒子が調製される。具体的には、Fe3O4ナノ粒子を含む中空Auナノ粒子が実施例6で上記されたように調製される。しかし、NaOHを使った膜の溶解の前に、インサイツ複合磁気ナノ粒子が本明細書の実施例14で前述の方法の金属ナノ粒子コアとして使用される。さらに、実施例14に記載の追加のステップを実施する前に、AAO膜の内側のチャネルの壁上に配置された中空Auナノ粒子(これはAuナノ粒子の空洞内にFe3O4ナノ粒子を含む)が完全に洗浄されるが、AAO膜内にはまだ残っている。このようにして、合成手順中に形成された小さい(<20nm)Fe3O4ナノ粒子が洗い流される。
前述の構造の1つを有する磁気ナノシードを、ブラキセラピーを、ブラキセラピー剤のリアルタイムイメージングと組み合わされる用途を含む、画像化および/またはセラノスティックス用途に使用することができる。例えば、いくつかの事例では、上記の磁気ナノシードは、インビボMRIの造影剤として使用される。当業者ならわかるように、超常磁性粒子の磁気モーメントは通常、常磁性材料より104倍高く、局所磁界の実質的外乱を生成し、プロトンの急速ディフェージング(T2緩和)に繋がり得る。T2*強調MRイメージング法は、他の超常磁性ナノ粒子による画像化に対し最も一般的に使用されている。しかし、他の超常磁性剤を使ったT2*強調画像は、多くの場合、低い信号対雑音比である。T2*強調MR像はまた、いくつかの薬剤の存在により生成された磁化率に対し極めて感受性が高く、蓄積した薬剤の実寸法より大きいシグナルボイドを生成する。さらに、このような薬剤由来の画像欠損は、その他の組織欠損または組織界面で発生し得る極端な磁気異方性が原因の欠損様信号と鑑別することが困難な場合がある。これらの制限を克服するために、オフ共鳴飽和(ORS)イメージング法を使用し得る。ORS法は、本明細書で記載の磁気ナノシードなどの超常磁性ナノ粒子系薬剤から、大幅に向上した画像化精度を備えた「ポジティブ」MRIコントラストを生成できる。本方法は通常、オフ共鳴周波数のRFパルスの適用を含む。ポジティブコントラストは、飽和のある画像と、飽和のない画像との比率または減算を取ることにより得ることができる。この技術は、造影前取得なしで、画像コントラストの可視化を可能とする。したがって、ORS−MRIを使って、組織内の磁気ナノシードを追跡することができる。
Claims (20)
- 放射性ナノ粒子であって、
金属ナノ粒子;
前記金属ナノ粒子コアのまわりに配置された外側金属シェル;および
前記金属ナノ粒子コア内または前記外側金属シェル内に配置された金属放射性同位体、
を含み、
前記放射性ナノ粒子が、3次元で30〜500nmの寸法を有する、放射性ナノ粒子。 - 3次元で80〜200nmの寸法を有する、請求項1に記載の放射性ナノ粒子。
- 前記金属ナノ粒子コアと前記外側金属シェルとの間に配置された内側金属シェルをさらに含む、請求項1に記載の放射性ナノ粒子。
- 前記内側金属シェルが、前記外側金属シェルの金属より低い還元電位を有する金属から形成される、請求項3に記載の放射性ナノ粒子。
- 前記金属ナノ粒子コアおよび前記外側金属シェルが、同じ金属または金属の組み合わせから形成される、請求項1に記載の放射性ナノ粒子。
- 前記金属ナノ粒子コアおよび前記外側金属シェルが、異なる金属または金属の組み合わせから形成される、請求項1に記載の放射性ナノ粒子。
- 前記金属ナノ粒子コアが、前記外側金属シェルの金属より高い還元電位を有する金属から形成される、請求項6に記載の放射性ナノ粒子。
- 前記金属ナノ粒子コアが、Auから形成され、前記内側金属シェルがCuから形成され、前記外側金属シェルがPd、Rh、またはAuから形成される、請求項3に記載の放射性ナノ粒子。
- 前記金属放射性同位体が、前記外側金属シェル内に配置される、請求項1に記載の放射性ナノ粒子。
