本明細書において使用される場合、用語「治療」又は「治療すること」は、(1)対象の状態若しくは疾患を改善させる若しくは安定させること、又は(2)対象の状態若しくは疾患に関連する症状の発症若しくは悪化を予防する若しくは軽減すること、を意味する。
本明細書で考察されている通り、ミトコンドリアは、細胞エネルギー代謝及び細胞死を調節する必須の細胞小器官である。従って、機能不全のミトコンドリア及びマイトファジーを介するそれらの除去における欠陥は、多くの病態生理学的障害及び疾患と関係があるとされてきた。例えば、β−アミロイドフラグメントは、ミトコンドリアを標的とし、アルツハイマー病においてミトコンドリア機能不全を引き起こすことが実証されており、ミトコンドリア機能及び生理機能の崩壊は、ハンチントン病の原因となる単一遺伝子への突然変異によってもたらされる。従って、マイトファジー障害は、パーキンソン病及びアルツハイマー病等の種々の神経変性疾患に関与するとされてきた。
単一遺伝子パーキンソン病(PD)に関連する遺伝子の中でも、parkin及びPINK1における突然変異は、常染色体劣性の若年性PD(EOPD)の最も一般的な遺伝的原因として同定されている。ParkinはE3ユビキチンリガーゼをコードし、PINK1はミトコンドリアのセリン/トレオニンキナーゼをコードし、これらはいずれも、健全なミトコンドリア機能及び形態の維持に関与している。PDにおけるミトコンドリア機能不全の役割は、1−メチル−4−フェニル−1,2,5,6−テトラヒドロピリジン(MPTP)及びロテノン誘導パーキンソニズム等のミトコンドリア毒素への暴露を実証するデータから理解されてきた。最近の研究により、Parkinが、カルボニルシアニド3−クロロフェニルヒドラゾン(CCCP)によるミトコンドリア膜電位の散逸時に、PINK1依存的方式でミトコンドリアに動員され、それにより、ユビキチン・プロテアソーム系を介してミトコンドリア融合タンパク質マイトフュージン(Mfh)1及び2等のミトコンドリア外膜タンパク質のユビキチン化及び分解を促進することが実証された。このプロセスは、機能不全のミトコンドリアと健全なミトコンドリアのプールとの融合を防止し、本明細書においてはマイトファジーと称されるプロセス、すなわち、オートファジー・リソソーム系を介する機能不全のミトコンドリアのクリアランスを促進する。一貫して、parkin又はPINK1のいずれかにおける突然変異は、患者由来の細胞又はparkin若しくはPINK1における突然変異が導入された細胞のいずれかを使用したPDの細胞モデルにおいて、マイトファジーを損ない、機能不全のミトコンドリアの蓄積につながることが実証されてきた。ミトコンドリアの品質管理に対するその有害な影響により、マイトファジー障害は、PDにおける神経変性及び疾患進行に関与する。
本発明は、Nix及び/又はGABARAP−L1ポリペプチド又はフラグメント又はバリアント又はアナログ、並びにNix及び/又はGABARAP−L1ポリペプチド又はフラグメント又はバリアント又はアナログをコードする発現ベクターの使用を提供する。一実施形態において、本発明は、配列における変異(alteration)を生成することによって、Nix及び/又はGABARAP−L1アミノ酸配列又は核酸配列を最適化する方法を提供する。このような変異は、ある特定の突然変異、欠失、挿入又は翻訳後修飾を含んでよい。他の実施形態において、本発明は、任意の自然発生Nix及び/又はGABARAP−L1ポリペプチドのバリアント又はアナログを更に含む。バリアントは、アミノ酸配列差異によって、翻訳後修飾によって、又はその両方によって、本発明の自然発生ポリペプチドと異なることができる。Nix及び/又はGABARAP−L1ポリペプチドのバリアントは、概して、本明細書において記述される自然発生アミノ酸配列の全て又は一部と、少なくとも85%、より好ましくは90%、最も好ましくは95%、又は更には99%の同一性を呈することになる。配列比較の長さは、少なくとも5、10、15又は20アミノ酸残基、好ましくは少なくとも25、50又は75アミノ酸残基、より好ましくは100超のアミノ酸残基である。
本明細書で考察されている通り、parkin又はPINK1への突然変異の結果としてのParkin関連マイトファジーの機能障害は、Nix介在性マイトファジーの増大によって補償され得る。ミトコンドリアの欠陥を有する対象が、健全なミトコンドリア及び欠陥があるミトコンドリアの混合集団を有することを考慮すると、欠陥があるミトコンドリアの数を選択的に低減させる作用物質は、神経変性障害の治療に有用である。所望の場合、Nix及び/又はGABARAP−L1の発現又は生物活性を増大させる作用物質を、細胞(例えば、mtDNAに遺伝的欠陥を含む細胞、parkin若しくはPINK1に遺伝子突然変異を含む細胞、黒質の細胞又はドーパミン作動性神経細胞)における、欠陥があるミトコンドリアの選択的排除を強化する効能について試験する。このような方法は、個別の薬剤適用のために、例えば、神経変性障害を有する対象に有益である可能性が高い作用物質を同定するのに、特に有用である。一例において、候補化合物は、ミトコンドリアの脱共役剤又はマイトファジーを誘導する他の作用物質の添加前に、添加と同時に、又は添加後に、細胞の培養培地(例えば、神経培養)に添加される。次いで、細胞におけるミトコンドリア機能及びマイトファジーの程度を、本明細書において記述されているものを含む当業者に公知の標準的な方法を使用して測定する。候補作用物質の存在下でのミトコンドリア機能及び/又はマイトファジーの程度を、候補作用物質を受けなかった対応する対照培養物において測定されたレベルと比較する。
一実施形態において、本明細書において記述されているスクリーニングは、parkin又はPINK1に突然変異を有する細胞中で行われる。
概して、マイトファジーを変調することができる作用物質は、天然生成物若しくは合成(若しくは半合成)抽出物両方の大きなライブラリー若しくは化学ライブラリーから、又はポリペプチド若しくは核酸ライブラリーから、当技術分野において公知の方法に従って、同定される。創薬及び開発の分野の当業者であれば、被験抽出物又は作用物質の正確な供給源が、本発明のスクリーニング手順に決定的ではないことを理解するであろう。スクリーニングにおいて使用される作用物質は、公知の作用物質(例えば、他の疾患又は障害に使用される公知の治療薬)を含み得る。代替として、本明細書において記述されている方法を使用して、事実上いくつもの未知の化学抽出物又は作用物質をスクリーニングすることができる。このような抽出物又は作用物質の例としては、植物、菌、原核生物又は動物ベースの抽出物、発酵ブロス、及び合成作用物質、並びに現存する作用物質の修正品が挙げられるがこれらに限定されない。
本発明は、神経変性障害の治療のためのNix及び/又はGABARAP−L1の発現又は活性を増大させる作用物質(上記スクリーニングにおいて同定される作用物質を含む)を提供する。一実施形態において、本発明は、Nix及び/又はGABARAP−L1ポリペプチドをコードする発現ベクターを含む医薬組成物を提供する。別の実施形態において、本明細書に記述されている方法を使用して薬理価値を有することが発見された化学的実体は、薬物として、又は、例えば合理的な薬物設計による、現存する作用物質の構造的修飾のための情報として有用である。治療的使用のために、本明細書に開示されている方法を使用して同定される組成物又は作用物質を、全身的に投与(例えば、薬学的に許容される担体中で製剤化)してよい。好ましい投与経路は、例えば、患者において連続的な持続レベルの薬物を提供する皮下、静脈内、腹腔内、筋肉内若しくは皮内注射、鼻腔内(例えば、鼻腔用スプレー)又は経皮(例えば、局所パッチ)投与を含む。ヒト患者又はその他の動物の治療は、生理学的に許容される担体中の、治療有効量の神経変性障害治療薬を使用して、行われることになる。好適な担体及びそれらの製剤化は、例えば、Remington’s Pharmaceutical Sciences by E.W.Martinに記述されている。投与される治療剤の量は、投与方式、患者の年齢及び体重、並びに神経変性障害の臨床症状に応じて変動する。概して、量は、ミトコンドリア病の治療において使用される他の作用物質について使用されるものの範囲内となるが、ある特定の場合において、化合物の特異性増大により、必要となる量はより低くなる。化合物は、当業者に公知の診断方法によって、又は神経変性障害に関連する若しくはNix介在性マイトファジーに関連する(例えば、Nix)遺伝子の転写調節を測定する任意のアッセイを使用して決定される通り、神経変性障害の臨床症状又は生理的症状を制御する投薬量で投与される。
神経変性障害の治療のための本発明の作用物質又はそのアナログの投与は、その他の成分と組み合わせて、神経変性障害又はその症状を、和らげる、低減させる又は安定させるのに有効な治療薬の濃度をもたらす任意の好適な手段によるものであってよい。一実施形態において、作用物質の投与は、細胞における機能不全の若しくは欠陥があるミトコンドリアのパーセンテージを低減させ、且つ/又は健全なミトコンドリアのパーセンテージを増大させる。
医薬組成物は、注射、注入又は移植(皮下、静脈内、筋肉内、腹腔内等)によって、剤形、製剤で、又は従来の非毒性の薬学的に許容される担体及びアジュバントを含有する好適な送達デバイス若しくは移植片を介して、非経口的に投与されてよい。このような組成物の製剤及び調製は、医薬製剤分野の当業者に周知である。
制御放出非経口組成物は、懸濁液、ミクロスフェア、マイクロカプセル、磁性ミクロスフェア、油溶液、油懸濁液又はエマルションの形態であってよい。代替として、活性薬物は、生体適合性担体、リポソーム、ナノ粒子、移植片又は注入デバイスに組み込まれていてよい。ミクロスフェア及び/又はマイクロカプセルの調製において使用される材料は、例えば、ポリガラクティア(polygalactia)ポリ−(シアノアクリル酸イソブチル)、ポリ(2−ヒドロキシエチル−L−グルタム−ニン)及びポリ(乳酸)等の生分解性生体内分解性ポリマーである。制御放出非経口製剤を製剤化する場合に使用してよい生体適合性担体は、炭水化物(例えば、デキストラン)、タンパク質(例えば、アルブミン)、リポタンパク質又は抗体である。移植片において使用される材料は、非生分解性(例えば、ポリジメチルシロキサン)又は生分解性(例えば、ポリ(カプロラクトン)、ポリ(乳酸)、ポリ(グリコール酸)若しくはポリ(オルトエステル)又はそれらの組み合わせ)であり得る。
経口使用のための製剤は、活性成分を非毒性の薬学的に許容される添加剤との混合物として含有する錠剤を含む。このような製剤は、当業者に公知である。添加剤は、例えば、不活性賦形剤又は充填剤(例えば、スクロース、ソルビトール、砂糖、マンニトール、微結晶性セルロース、バレイショデンプン等のデンプン、炭酸カルシウム、塩化ナトリウム、ラクトース、リン酸カルシウム、硫酸カルシウム又はリン酸ナトリウム);造粒及び崩壊剤(例えば、微結晶性セルロース等のセルロース誘導体、バレイショデンプン等のデンプン、クロスカルメロースナトリウム、アルギン酸塩又はアルギン酸);結合剤(例えば、スクロース、グルコース、ソルビトール、アカシア、アルギン酸、アルギン酸ナトリウム、ゼラチン、デンプン、アルファ化デンプン、微結晶性セルロース、ケイ酸アルミニウムマグネシウム、カルボキシメチルセルロースナトリウム、メチルセルロース、ヒドロキシプロピルメチルセルロース、エチルセルロース、ポリビニルピロリドン又はポリエチレングリコール);並びに平滑剤、流動促進剤及び付着防止剤(例えば、ステアリン酸マグネシウム、ステアリン酸亜鉛、ステアリン酸、シリカ、水素化植物油又はタルク)であってよい。その他の薬学的に許容される添加剤は、着色剤、香味剤、可塑剤、湿潤剤、緩衝剤等であり得る。
経口使用のための制御放出組成物は、活性物質の溶解及び/又は拡散を制御することによって活性神経変性障害治療薬を放出するように構築されていてよい。溶解又は拡散制御放出は、作用物質の錠剤、カプセル剤、ペレット剤若しくは顆粒製剤の適切なコーティングによって、又は化合物を適切なマトリックスに組み込むことによって、実現することができる。制御放出コーティングは、上記で言及したコーティング物質の一つ又は複数(例えば、セラック、ビーズワックス、グリコワックス、キャスターワックス、カルナウバワックス、ステアリルアルコール、モノステアリン酸グリセリル、ジステアリン酸グリセリル、パルミトステアリン酸グリセロール、エチルセルロース、アクリル樹脂、dl−ポリ乳酸、酢酸酪酸セルロース、ポリ塩化ビニル、ポリ酢酸ビニル、ビニルピロリドン、ポリエチレン、ポリメタクリレート、メタクリル酸メチル、2−ヒドロキシメタクリレート、メタクリレートヒドロゲル、1,3ブチレングリコール、メタクリル酸エチレングリコール及び/又はポリエチレングリコール)を含んでよい。制御放出マトリックス製剤において、マトリックス材料は、例えば、水和メチルセルロース、カルナウバワックス及びステアリルアルコール、Carbopol 934、シリコーン、トリステアリン酸グリセリル、アクリル酸メチル−メタクリル酸メチル、ポリ塩化ビニル、ポリエチレン並びに/又はハロゲン化フルオロカーボンを含んでもよい。
本発明の作用物質の半固体局所投与のための剤形としては、軟膏剤、ペースト剤、クリーム剤、ローション剤及びゲル剤が挙げられる。剤形は、皮膚への塗布領域における活性成分の持続放出のために、粘膜付着性ポリマーと共に製剤化されてよい。活性化合物を、滅菌条件下で薬学的に許容される担体と、必要とされ得る任意の保存剤、緩衝剤又は噴射剤と、混合してよい。このような局所用調製物は、所望の化合物を、局所用液体、クリーム及びゲル製剤において一般に使用される従来の医薬賦形剤及び担体と組み合わせることによって調製することができる。
当業者は、動物モデルと比較してヒトの投薬量を修正することが当技術分野において日常的であると認識しているため、ヒト投薬量は、マウスにおいて使用した化合物の量を外挿することによって最初に決定することができる。或る実施形態において、投薬量は、体重1Kg当たり化合物約1mg〜約5000mg(約1mg/Kg〜約5000mg);又は体重1Kg当たり化合物約5mg〜約4000mg(約5mg/Kg〜約4000mg/Kg)又は約10mg〜約3000mg(約10mg/Kg〜約3000mg/Kg);又は体重1Kg当たり化合物約50mg〜約2000mg(約50mg/Kg〜約2000mg/Kg);又は体重1Kg当たり化合物約100mg〜約1000mg(約100mg/Kg〜約1000mg/Kg);又は体重1Kg当たり化合物約150mg〜約500mg(約150mg/Kg〜約500mg/Kg)の間で変えてよいことが想定される。他の実施形態において、この用量は、約1、5、10、25、50、75、100、150、200、250、300、350、400、450、500、550、600、650、700、750、800、850、900、950、1000、1050、1100、1150、1200、1250、1300、1350、1400、1450、1500、1600、1700、1800、1900、2000、2500、3000、3500、4000、4500、5000mg/Kg体重であってよい。他の実施形態において、より高い用量を使用してよく、そのような用量は、体重1Kg当たり約5mg〜約20mg(約5mg/Kg〜約20mg/Kg)の範囲内であってよいことが予想される。他の実施形態において、用量は、約8、10、12、14、16又は18mg/Kg(体重)であってよい。当然ながら、この投薬量は、治療プロトコールにおいて日常的に為されるように、初期臨床試験の結果及び特定の患者の必要性に応じて、上方又は下方に調整されてよい。
本発明は、Nix介在性マイトファジーを介して機能不全の又は欠陥があるミトコンドリアの排除を変調することにより、神経変性障害又はその症状を治療する方法を提供する。この方法は、細胞からの機能不全の又は欠陥があるミトコンドリアのクリアランスを変調する作用物質を治療有効量含む医薬組成物を、本明細書において記述されている方法を使用して、対象(例えば、ヒト等の哺乳動物)に投与することを含む。故に、一実施形態は、神経変性障害に罹患している対象又はかかりやすい対象を治療する方法である。この方法は、該障害を治療するのに十分な治療有効量の、本明細書において記述されている作用物質を、該障害が治療されるような条件下で対象に投与する工程を含む。
本発明は、神経変性障害の治療又は予防用キットを提供する。一実施形態において、キットは、有効量の本発明の作用物質を含有する治療組成物又は予防組成物(例えば、Nixの発現及び/又は活性を増大させる作用物質等の、細胞におけるNix介在性マイトファジーを増大させる作用物質;Nixポリペプチド又はそのフラグメント若しくはバリアント、及び/又は、GABARAP−L1ポリペプチド又はそのフラグメント若しくはバリアント、又は、これらをコードする発現ベクター)を単位剤形で含む。いくつかの実施形態において、キットは、治療化合物又は予防化合物を含有する無菌容器を含み、このような容器は、ボックス、アンプル、ボトル、バイアル、チューブ、バッグ、ポーチ、ブリスターパック、又は当技術分野において公知である他の好適な容器であり得る。このような容器は、プラスチック、ガラス、ラミネート紙、金属箔、又は医薬を保持するのに好適な他の材料製であり得る。
場合により、治療効能又は予防効能を有する作用物質を、神経変性障害を治療するための任意のその他の標準的な療法と組み合わせて投与してよい。所望の場合、本発明の作用物質は、単独で投与してもよく、又は神経変性障害の治療に有用な従来の治療薬と組み合わせて投与してもよい。例えば、パーキンソン病の治療に有用な治療薬は、デプレニール、アマンタジン又は抗コリン薬、レボドパ、カルビドパ、エンタカポン、プラミペキソール、ラサギリン、抗ヒスタミン薬、抗鬱薬、ドーパミンアゴニスト、モノアミン酸化酵素阻害剤(MAOI)等を含むが、これらに限定されない。
本発明者らは、機能的Parkinタンパク質を有さない健全なホモ接合性parkin突然変異キャリアを同定した。ミトコンドリアは、ATPの形態でエネルギーを生成する主要な細胞器官であり、parkinにおける突然変異の影響を受ける主要標的であるため、Parkin欠損突然変異キャリア(以後「キャリア」)及び患者(機能的Parkinを欠いている複合ヘテロ接合体;以後「患者」)からの繊維芽細胞におけるミトコンドリアATP合成速度を決定した。
細胞培養
ヒト繊維芽細胞及びヒト嗅覚神経球細胞株の確立及び培養のためのプロトコールは、既に記述されている(Koentjoro,et al,2012,Mov Disord 27(10),1299−303;Park,et al,2011;Hum Mutat 32(8),956−64)。細胞は、全ての実験について最高で第15継代まで継代培養した。
ATP合成速度は、既述の通りに決定した(Shepherd,et al.,2006)。簡潔に述べると、トリプシン処理後、製造業者の説明書に従い、BCAタンパク質アッセイキット(サーモサイエンティフィック、米国イリノイ州ロックフォード)を使用して総タンパク質濃度を決定することにより、繊維芽細胞を採取した。細胞を、細胞懸濁緩衝液(150mMのKCl、25mMのトリス−HCl pH7.6、2mMのEDTA pH7.4、10mMのKPO4 pH7.4、0.1mMのMgCl2及び0.1%(w/v)BSA(シグマ))中、1mg/mL総タンパク質で希釈した。ATP合成は、250μLの細胞懸濁液を、750μLの基質緩衝液(10mMのリンゴ酸塩、10mMのピルビン酸塩、1mMのADP、40μg/mLのジギトニン及び0.15mMのアデノシン五リン酸(シグマ))と共に37℃で10分間インキュベーションすることによって誘導した。このインキュベーション後、50μLに分取した反応混合物に450μLの沸騰しているクエンチ緩衝液(100mMのトリス−HCl、4mMのEDTA pH7.75(シグマ))を添加し、100℃で2分間インキュベートすることにより、反応を停止させた。得られた反応混合物を更に1:10でクエンチ緩衝液に希釈し、ATP量を、FB10照度計(ベルトールド・ディテクション・システムズ、ドイツ、プフォルツハイム)内、ATP生体発光アッセイキット(ロシュ・ダイアグノスティックス、スイス、バーゼル)を用い、製造業者の説明書に従って測定した。
細胞死は、サイトトックス96非放射性細胞毒性アッセイキット(プロメガ、米国ウィスコンシン州マディソン)を使用して、製造業者のプロトコールに従って決定した。簡略に述べると、ヒト嗅覚神経球細胞を、24ウェル培養プレートに1ウェル当たり50,000細胞で播種し、48時間にわたって培養した。次いで、細胞を、漸増用量のロテノン(シグマ;1.5μΜ、2.5μΜ及び12.5μΜ)に72時間にわたって暴露した。50μLに分取した培養培地を、基質混合物と共に30分間にわたってインキュベートした。ベンチマーク・マイクロプレート・リーダー(バイオ・ラッド、米国カリフォルニア州ハーキュリーズ)を使用して、490nmで分光光度法により乳酸デヒドロゲナーゼ(LDH)活性を測定した。
対照(37.4±1.2)と比較した場合、ATP合成速度は、患者細胞では有意に低減した(31.4±3.0、p<0.05)が、キャリア細胞では低減していなかった(36.4±3.5)(図1A)。次いで、細胞をロテノンで処理して、キャリア細胞におけるそれらの効果を調査した。ロテノンは、ミトコンドリア複合体I阻害剤であり、Parkin関連PD細胞モデル等のミトコンドリア機能不全を有する細胞において毒性を増大させることが示されている。ロテノンに暴露すると、患者細胞は毒性の増大を提示したのに対し、対照とキャリア細胞とでは同様に反応した(図1B及び図1C)。
キャリア細胞において観察された正常なミトコンドリア機能は、Parkinの損失を補償するミトコンドリアの品質管理の正常機能を示唆するものであった。CCCPを使用してキャリア細胞中でマイトファジーを誘導し、以下の三つの異なる方法の組み合わせを使用して調査した:クエン酸シンターゼ活性(ミトコンドリアのマトリックス酵素)によるミトコンドリアの質量の測定;定量的リアルタイムPCRによるmtDNA含有量の測定;並びに共焦点顕微鏡法によるミトコンドリア及びオートファゴソームの共局在化。共焦点顕微鏡法を使用してマイトファジーを視覚的にモニターするために、オートファゴソームマーカーについてはGFP−LC3を、ミトコンドリアマーカーについてはRFP−Mitoを発現している繊維芽細胞を生成した。
レンチウイルス産生及び細胞株の確立
緑色蛍光タンパク質(GFP)タグ付きLC3ベクターは、Ernst Wolvetang博士から寄贈されたものであった。GFP−LC3の発現のためのレンチウイルスは、レンチ−XレンチウイルスHTXパッケージングシステム(クロンテック、米国カリフォルニア州マウンテンビュー)及びリポフェクタミン2000(インビトロジェン、米国カリフォルニア州カールスバッド)を使用し、製造業者の説明書に従って生成した。レンチウイルスを含有する培地を、トランスフェクション後48時間及び72時間で収集し、続いて、レンチ−X濃縮器(クロンテック)を使用して濃縮した後、ウイルスの力価を測定した。
繊維芽細胞を、6ウェル培養プレートに1ウェル当たり200,000細胞で播種し、10μΜのDMSO又はCCCP(シグマ)のいずれかで24時間にわたって処理して、ミトコンドリア膜電位を低減させることによってマイトファジーを誘導した。
CCCPへの暴露により、対照(ビヒクル対照と比較して平均51.8%低減、p<0.001)及びキャリア細胞(ビヒクル対照と比較して38.7%低減、p<0.001)ではミトコンドリア質量が有意に低減したが、患者細胞(p=0.16)においては低減しなかった。同様に、mtDNA含有量についても、対照(35.4%、p<0.01)及びキャリア細胞(27.6%、p<0.01)ではCCCP処理によって有意に減少したが、患者細胞(p=0.84)では減少しなかった。共焦点顕微鏡法において、GFP−LC3とRFP−Mitoとの間の最小の共局在化は、CCCP処理前に観察された(データは示されていない)。CCCPによるマイトファジーの誘導時に、GFP−LC3とRFP−Mitoとの間の共局在化が著しく増大することが対照及びキャリア細胞において観察され、マイトファジーの上昇を示唆したのに対し、患者細胞においてはこのような変化は観察されなかった。共局在の定量化は、患者細胞(8.6±9.5%、p<0.01)において、対照(100±28.3%)と比較して有意に低い共局在化度を示し、欠陥があるマイトファジーを示唆したのに対し、キャリア細胞において観察された共局在化(101.9±36.5%)は、対照細胞と同等であった。
キャリア細胞のマイトファジーにおけるParkinの機能に対する補償の欠如を確認するために、CCCP処理時の、Parkinのミトコンドリアの動員及びマイトフュージン2(Mfn2)のユビキチン化を評価した。
ミトコンドリア単離
標準的なダウンス型ホモジナイザー及びマンニトール−スクロース−EDTA(MSE)緩衝液(25mMのマンニトール、75mMのスクロース、100mMのEDTA(シグマ))を用いるプロトコールを使用して、繊維芽細胞からミトコンドリアを単離した。簡潔に述べると、細胞をトリプシン処理によって収集し、3mLの冷MSE緩衝液に再懸濁した。電動式ダウンス型ホモジナイザー(キカラボテクニック(Kika Labortechnik)、ドイツ、シュタウフェン)を使用して、細胞を溶解させた。追加で3mlのMSE緩衝液をホモジネートに添加し、続いて、600×gで4℃にて10分間にわたって遠心分離した。次いで、ミトコンドリアを含有する上清を、12,000×gで4℃にて15分間にわたって遠心分離して、ミトコンドリアを収集した。遠心分離後、上清(「細胞画分」)を保存し、9kDa分子量カットオフの遠心式タンパク質濃縮器(サーモサイエンティフィック)を介し、製造業者の説明書に従って濃縮した。一方で、ミトコンドリアを含有するペレット(「ミトコンドリア画分」)を1mlのMSE緩衝液で二回洗浄し、最後に60μLの細胞溶解緩衝液(プロテアーゼ阻害剤カクテルを補充したセルリティックM細胞溶解試薬(シグマ))に再懸濁した。
タンパク質発現は、エクセルシュアロックミニセル電気泳動システム及びエクセルIIブロットモジュール(インビトロジェン)を使用するウエスタンブロッティングによって決定した。簡潔に述べると、20〜30μgの全細胞溶解物又はミトコンドリア/細胞画分のいずれかを、NuPAGEノベックス4%〜12%ビス−トリスSDS/ポリアクリルアミドゲル(インビトロジェン)を使用して分割し、ポリフッ化ビニリデン(PVDF)膜(サーモサイエンティフィック)に移した。次いで、膜にブロットされたタンパク質を、タンパク質特異的一次抗体及び西洋ワサビペルオキシダーゼ抱合二次抗体の順次適用でプローブした。アッセイにおいて使用した抗体を以下の表2において詳述する。スーパーシグナル・ウエスト・ピコ又はフェムト化学発光基質(サーモサイエンティフィック)を使用して化学発光を発生させ、LAS4000(富士フィルム株式会社、日本、東京)を使用して検出した。
繊維芽細胞からの全RNAをRNイージーミニキット(キアゲン)を使用し、製造業者の説明書に従って調製し、次いで、スーパースクリプトIIIファーストストランド合成システム(インビトロジェン)を用い、製造業者の説明書に準拠して、cDNAに逆転写した。得られたcDNAを使用して、Rotor Gene 6000(キアゲン)でクオンティテクトSYBRグリーンPCRキット(キアゲン)を使用する定量的リアルタイムRT−PCR(qRT−PCR)において、製造業者の説明書に従って遺伝子発現を決定した。反応において使用したプライマーを、以下の表3に収載する。
CCCPへの暴露時に、Parkinは対照細胞のミトコンドリア画分において高度に蓄積していたのに対し、細胞画分におけるタンパク質は減少しており、Parkinのミトコンドリアの動員を示していた。加えて、対照細胞は、CCCP後、ユビキチン化Mfn2の増大と共に非ユビキチン化形態の量の減少を示し、Parkin介在性ユビキチン化及びMfn2の分解を示していた。しかしながら、これらのParkin関連事象は、CCCP処理時にキャリア細胞においては観察されず、マイトファジーのプロセスにおけるParkin機能の欠如を裏付けた。加えて、PINK1転写の発現レベルは、対照と比較して、キャリアにおけるCCCP処理前後には上昇せず、キャリア細胞のParkin/PINK1介在性マイトファジーにおける代償性活性化の欠如を裏付けた。加えて、高活性オートファジーがミトコンドリアの非特異的分解を媒介している可能性がある。そこで、この可能性について、オートファジー機能のインジケーターとして、CCCP処理時のLC3−IからLC3−IIへの変換をモニターすることによって試験した。更に、CCCPは、HeLa細胞、HCT116細胞及びMEFにおけるオートファジーを、ラパマイシンと同等の活性化の大きさで誘導することが他の場所で実証された。調査した全ての細胞株において、CCCP処理前後にLC3−II/β−アクチン比の同様の増大が検出され、基本及び/又はマイトファジー誘導条件下でオートファジー機構の正常機能を示した。まとめると、これらの結果は、キャリア細胞において観察されたCCCP誘導マイトファジーが、PINK1/Parkin経路によって又はオートファジーの異常活性化によってのいずれでも媒介されていないことを示し、Parkin非依存性マイトファジー経路の関与を示唆している。
キャリア細胞においてCCCPによって誘導されるマイトファジーの増大をNix介在性マイトファジーが司るか否かを評価するために、基本条件及びCCCP処理条件下での、Nix、GABARAP−L1及びGABARAP−L2の発現レベルを、qRT−PCRを使用して評価した。
繊維芽細胞を、基本条件下で培養するか、又は10μΜのCCCPで6時間にわたって処理した後、全RNA及びcDNA合成を抽出した。Nix、GABARAP−L1及びGABARAP−L2の発現を、qRT−PCRによって決定した。
Nixの発現は、基本条件下での対照とキャリア細胞との間では同等であったが、CCCPによるマイトファジーの導入時には、キャリア細胞において有意に増大(p<0.01)した。キャリア細胞は、両方の条件下の対照と比較した場合、GABARAP−L1のレベル上昇を示したが、GABARAP−L2の発現低減を示した。その一方で、これらの遺伝子の発現は、CCCP処理後であっても、対照細胞と比較した場合、患者細胞において有意に低いままであることが分かった。まとめると、これらの結果は、キャリア細胞におけるCCCP処理及び高発現レベルのその結合パートナーGABARAP−L1によるNixの誘導を示し、代替的なマイトファジーにおけるそれらの関与を示唆している。
CCCP誘導マイトファジーにおけるその関与を裏付けるために、本発明者らは、siRNAを使用してNixを発現抑制し、CCCP誘導マイトファジーにおける変化を評価した。
siRNA介在性Nixノックダウン
ダーマコンON−TARGETプラスSMARTプールヒトBNIP3L(Nix siRNAと称する;サーモサイエンティフィック、L−011815−00−0005番)及びダーマFECT1 siRNAトランスフェクション試薬(サーモサイエンティフィック、T−2001−01番)を使用し、製造業者の説明書に準拠して、繊維芽細胞におけるNixをノックダウンした。ON−TARGETプラス非標的化siRNA1番(スクランブルsiRNAと称する;サーモサイエンティフィック、D−001810−01−05番)を陰性対照として使用した。
siRNAのトランスフェクション48時間後のNixの発現レベルは、mRNAレベル(>95%)及びタンパク質レベルにおいて劇的に低減し、ノックダウン成功を示した。CCCPへの暴露後、スクランブルsiRNAをトランスフェクトした細胞は、クエン酸シンターゼ活性によって測定されたミトコンドリア質量の有意な低減を示し(それぞれのビヒクル対照と比較して、対照細胞において63.0%低減、p<0.001及びキャリア細胞において30.1%、p<0.05)、正常なマイトファジーを示した。しかし、Nix siRNAのトランスフェクションは、キャリアにおいてミトコンドリア質量の低減を抑止したが、対照細胞においてはしなかった(対照細胞において47.6%低減、p<0.01及びキャリア細胞において8.5%、p=0.15)。スクランブルsiRNA(対照細胞において20.7%低減、p<0.001及びキャリアにおいて33.0%、p<0.05)及びNix siRNA(対照細胞において36.0%低減、p<0.01及びキャリア細胞において8.1%、p=0.39)をトランスフェクトした細胞におけるmtDNA含有量の評価からも、同様の結果が得られた。加えて、Nix siRNAをトランスフェクトしたキャリア細胞は、CCCPによるマイトファジーの誘導時に、スクランブルsiRNAをトランスフェクトした細胞と比較して、GFP−LC3及びRFP−mitoの共局在化の著しい低減を示したのに対し、スクランブル及びNix siRNAをトランスフェクトしたキャリア細胞の間では、同様の低い程度の共局在化しか基本条件下では観察されなかった(データは示されていない)。共局在化の定量化は、それぞれのスクランブルsiRNA細胞と比較した場合、Nix siRNAトランスフェクトキャリア細胞における有意な低減(63.0%低減、p<0.001)を示し、CCCP誘導マイトファジーの機能障害を実証した。まとめると、これらの結果は、Nixが、キャリア細胞におけるCCCP誘導マイトファジーを、Parkin機能喪失によって容易にすることを示す。
Nix発現とCCCP誘導マイトファジーとの関係を確認するために、本発明者らは、Nix発現欠損を有する細胞におけるNix発現を、対照及びキャリア細胞に対して増大させ、CCCP誘導マイトファジーにおける変化を評価した。
Nixの発現増大
Nix発現に対するホルボールミリスチン酸アセテート(PMA)の効果を評価するために、対照細胞及び患者細胞(「発端者(proband)」)をPMA(10nM又は20nM)に24時間にわたって暴露した。細胞を24時間後に採取し、Nix及びGABARAP−L1タンパク質の発現を、上記で概説した通りのウエスタンブロッティングによって決定した。
図6は、対照及び患者細胞におけるNix(図6A〜図6B)及びGABARAP−L1(図6C〜図6D)の発現がウエスタンブロッティングによって決定され、バンドが密度測定を使用して定量化されたことを示す。β−アクチン(42kDa)を負荷対照として使用した。Νix/β−アクチン比のレベルは、PMA処理時に、ビヒクル対照(図6A〜図6B)と比較して増大した。PMA(図6C〜図6D)への暴露時に、GABARAP−L1発現の増大はなかった。
Nixの発現を誘導する作用物質で処理した患者細胞におけるマイトファジーの観察された修復の特異性を評価するために、parkinに複合ヘテロ接合変異を有する個体から単離された細胞及びPINK1にホモ接合変異を有する個体から単離された細胞において、マイトファジーを評価した。
NixのsiRNA介在性ノックダウン
マイトファジーのホルボールミリスチン酸アセテート(PMA)誘導修復の特異性を評価するために、対照細胞、parkinに複合ヘテロ接合変異を有する個体から単離された細胞(「患者」)、及びPINK1にホモ接合変異を有する個体から単離された細胞(「PINK1」)を、上記の実施例5で概説した通りのNixのsiRNA介在性ノックダウンに供した。簡潔に述べると、細胞を、非標的化siRNA(スクランブルsiRNA)又はsiRNA標的化Nix(Nix siRNA)のいずれか、続いてCCCP及びPMAによる24時間にわたる共処理に供した。
図8Aは、スクランブルsiRNA又はsiRNA標的化Nixによる細胞の処理後のNixの発現を示す。Nixのノックダウンが成功した。図8Bは、各Nix siRNA−ビヒクル処理細胞と比較した場合、Nix siRNAで処理した患者及びPINK1細胞が、PMA及びCCCP共処理後に、mtDNAの有意な減少を示さなかったことも示す。一元ANOVAに続くポストホックテューキーのHSD多重比較検定おいて、NS;有意ではない及び*;p<0.05。
Nix発現とCCCP誘導マイトファジーとの関係を確認するために、本発明者らは、機能的parkinを欠いている(且つ対照及び「キャリア」細胞に対してNix発現欠損を有する)患者細胞においてNixを過剰発現させ、CCCP誘導マイトファジーにおける変化を評価した。
Nixの過剰発現
pCMV6−Nix(Origene;RC203315番)中の野生型Nix cDNA(NM_004331)を、FLAGタグを含有するpER4レンチウイルスベクター中にサブクローン化した。Nix−FLAGの発現のためのレンチウイルスは、レンチ−X HTXレンチウイルスパッケージングシステム(クロンテック、米国カリフォルニア州マウンテンビュー)及びリポフェクタミン2000(インビトロジェン、米国カリフォルニア州カールスバッド)を使用し、製造業者の説明書に従って生成した。レンチウイルスを含有する培地をトランスフェクション後、48時間及び72時間で収集し、続いて、レンチ−X濃縮器(クロンテック)を使用する濃縮ステップの後、ウイルスの力価を測定した。繊維芽細胞に、レンチウイルス空ベクター(pEmpty)又はレンチウイルスNix−FLAGベクター(pNix−FLAG)のいずれかを、4μg/mLのポリブレンの存在下、一細胞当たり10感染単位のレンチウイルスの比率で24時間にわたって形質導入し、その後の実験に使用した。
図9は、Nixの過剰発現が、機能的parkinを欠いている細胞(患者細胞を含む;「Parkin mut.」)及びPINK1にホモ接合変異を有する個体から単離された細胞(「PINK1 mut.」)におけるCCCP誘導マイトファジーを修復することを示す。繊維芽細胞に、空ベクター(pEmpty)又はNix−FLAGベクター(pNix−FLAG)を含有するレンチウイルスのいずれかを形質導入した。DNAをビヒクル処理細胞及びCCCP処理細胞から単離した後、定量的リアルタイムPCRを使用してミトコンドリアDNA定量化を行った。図9Aでは、Nix−FLAGを発現しているParkin及びPINK1突然変異体において、核DNA(nDNA)に対するミトコンドリアDNA(mtDNA)の相対量は、ビヒクル処理細胞と比較した場合、CCCP処理後に有意に減少した。一元ANOVAに続くポストホックテューキーのHSD多重比較検定おいて、NS;有意ではない**;p<0.01。図9Bでは、レンチウイルスを形質導入したGFP−LC3(緑色)及びRFP−Mito(赤色)を発現している患者細胞を、20μΜのCCCPで4時間にわたって処理した。オートファゴソーム及びミトコンドリアの共局在化(右パネルにおける黄色点)が、Nix−FLAGを発現している患者細胞において観察され、マイトファジーの活性化を示したが、空ベクターを発現している患者細胞においては観察されなかった。尺度:10μm。共局在化速度を、50個の個々の細胞画像から、ライカアプリケーションスイートアドバンスト蛍光(LAS AF)ソフトウェアを使用して算出した(図9C)。CCCP処理後、Nix−FLAGを発現している患者細胞は、空ベクター形質導入細胞と比較した場合、有意に高い共局在化速度を提示した。スチューデントの両側t検定において、***p<0.001。

- 前記作用物質が、Nixポリペプチド若しくはそのフラグメント若しくはバリアント若しくはアナログ、及び/又はGABARAP−L1ポリペプチド若しくはそのフラグメント若しくはバリアント若しくはアナログ、の細胞における生物活性又は発現を増大させる、請求項22に記載の使用又は請求項23に記載の作用物質。
- 前記作用物質が、Nixポリペプチド若しくはそのフラグメント若しくはバリアント、及び/又はGABARAP−L1ポリペプチド若しくはそのフラグメント若しくはバリアントを含む、請求項22に記載の使用又は請求項23に記載の作用物質。
- 前記作用物質が、Nixポリペプチド若しくはそのフラグメント若しくはバリアントをコードする発現ベクターを含む、請求項22に記載の使用又は請求項23に記載の作用物質。
- 前記神経変性障害がミトコンドリア機能不全を伴う、請求項22若しくは請求項24〜請求項26のいずれか一項に記載の使用、又は請求項23〜請求項26のいずれか一項に記載の作用物質。
- 前記神経変性障害がマイトファジー障害を含む、請求項22若しくは請求項24〜請求項27のいずれか一項に記載の使用、又は請求項23〜請求項27のいずれか一項に記載の作用物質。
- 前記神経変性障害が、パーキンソン病、アルツハイマー病、レヴィー小体認知症、クロイツフェルト・ヤコブ病、ハンチントン病、多発性硬化症、又は筋萎縮性側索硬化症を含む群から選択される、請求項22若しくは請求項24〜請求項28のいずれか一項に記載の使用、又は請求項23〜請求項28のいずれか一項に記載の作用物質。
- 前記神経変性障害がパーキンソン病である、請求項29に記載の使用又は請求項29に記載の作用物質。
- 前記神経変性障害が、parkin及び/又はPINK1における突然変異を伴う、請求項22若しくは請求項24〜請求項30のいずれか一項に記載の使用、又は請求項23〜請求項30のいずれか一項に記載の作用物質。
