動的光散乱試験(DLS):
DLSは、タンパク質、ポリマー、ミセル、炭水化物、およびナノ粒子を含む、様々な粒子のサイズを特徴づけるために使用し、その測定は、媒体中の粒子コアのサイズ、表面構造のサイズ、粒子濃度、およびイオンの種類に依存する。ナノコンテナの刺激誘発分解は、DLSを用いて試験し、ここで、6mMのドデシル硫酸ナトリウム溶液を評価し、これは、2〜20nmの範囲、特に6nmである(図3)。しかしながら、酸化導電性ポリマー、すなわち、ポリアニリンの接近後、SDS集合体サイズにおける変化は、0.1〜5nmの範囲、特に1nmであると決定され、基本的に溶液中のナノスケール集合体の不存在を示す(図4)。
UV−vis分光法を用いて、導電性ポリマーと相互作用後のSDSミセルの分解から放出されるゲスト分子の量を定量する。分解および隔離ゲスト分子の同時放出は、時間の関数として評価し、これは、多重発光ピーク、および372nmでのピーク強度(I372)対393nmでのピーク強度(I393)の比を示す。SDS集合体中のゲスト分子は、疎水性環境に対応し、これは、集合体へのOxCPの添加後に、封入材に親水性環境を与えることが認められる。この環境における変化は、SDSミセルの分解および隔離分子または封入材の水中への同時放出による。
カチオン性酸化導電性ポリマー(OxCP)、ポリアニリン(PANI)と相互作用させるイオン性ミセルとして、ドデシルスルホン酸ナトリウム(SDS)を選択した。表面誘発不均一分解過程は、動的光散乱(DLS)を用いて試験した。6mMのSDS溶液の凝集体サイズは、6nmであることがわかった(図3)。水中5mlのSDS溶液に、100mgの乾燥OxCP(PANI)粉末を添加し、これらの分量を今度の実験のすべてで維持した。酸化ポリアニリンの接近後、SDS集合体サイズの変化は、1nmであると決定され、基本的に溶液中のナノスケール集合体の不存在を示した(図4)。
この例は、分解が、OxCPの正電荷とSDSの負電荷との間の引力相互作用によるかどうかを決定するために作成した。そうであるならば、集合体は、中性導電性ポリマー(NeuCP)の存在下で壊離(break apart)しないはずである。NeuCPをSDSミセル溶液に添加し、集合体サイズをDLSにより決定した。集合体のサイズは、6nmであることがわかり、モニタリングの期間にわたって一定のままであった。図3参照。
ピレンを用いた分子の送達
分解および隔離ゲスト分子の同時放出を、ピレンを用いて時間の関数として調査し、これは、多重発光ピーク、および372nmでのピーク強度(I372)対393nmでのピーク強度(I393)の比を示す。水中ピレンの1マイクロモルの溶液を、SDS集合体中に取り込み、I372/I393が0.87であると決定されたが、これは、疎水性環境におけるピレンの存在に対応する。ピレン隔離SDS集合体へのOxCPの添加後、I372/I393は、0.87から増加を開始し、60分間で1.09の値に達し、これは、ピレンに直面する新たな環境が親水性であることを示した。このピレンの環境における変化は、SDSミセルの分解および隔離ピレンの水中への同時放出のためである。NeuCPを用いて同じ実験を行い、ピレンのI372/I393の変化は認められなかったが、これは、集合体が、NeuCPの存在下で無傷であることを裏付ける。
UV−vis分光法を用いて、OxCP原料粉末との相互作用後のSDSミセルの分解から放出されたピレンの量を定量した。ナノスケールコンテナの分解後にピレンの約96%が放出され、放出プロファイルは、図7に示す。ピレンの無変化UV−visスペクトルの特徴は、SDSミセルが、NeuCPの存在下で分解せず、ゲスト分子を放出しなかったことを示す。
OxCP表面形態に応じた放出反応速度の変動
OxCPナノ繊維は、界面重合によって合成し、直径約50nmを有した(図6)。次いで、100mgのナノ繊維をSDSミセル溶液に添加し、集合体の寸法を決定した。6nmの集合体を1nmに分解し、これは、おおよそモノマーのサイズである。放出の速度を調査するために、これらのナノサイズ繊維を、ピレン封入SDSミセルに添加し、ピレンのI372/I393を時間の関数としてモニターした。本発明者らは、分解および同時放出が15分で開始し、これは、バルクOxCP放出と比較して約2倍だけ速いことに気付いた。対照実験を、NeuCPナノ繊維を用いて行い、これは、微量の放出を示した。本発明者らはまた、CTABミセルと、OxCPおよびNeuCPナノサイズ繊維との相互作用を試験し、これは、分解も、その後の封入ゲスト分子の放出もまったく示さなかった。図7参照。
分解および隔離ゲスト分子の同時放出を、ナイルレッドを用いて時間の関数として調査した。水中SDSの6mM溶液に、水中ナイルレッドの1マイクロモル溶液を添加し、続いて、100mgのポリ(3,4−エチレンジオキシチオフェン)の原料粉末を添加した。ナイルレッドの放出を、670nmでの強度をモニターすることによって、ポリマーの添加後に時間の関数として試験し、結果を図9に示す。時間の関数としての発光強度の低下は、ゲスト分子の放出を示す。

Claims (10)
- 部位特異的送達に有用なナノコンテナの刺激誘発分解のための組成物であって、不溶性カチオン性酸化導電性ポリマー、ミセル集合体およびゲスト分子を含み、ミセル集合体とポリマーの非局在電荷との、前記分解を引き起こす多価相互作用によって、ゲスト分子の部位特異的送達が達成される組成物。
- 請求項1に記載の組成物であって、前記カチオン性酸化導電性ポリマーが、組成物の1〜50重量%の範囲内のポリチオフェン(PTs)、ポリピロール(PPy)、およびポリ(3,4−エチレンジオキシチオフェン)ポリ(スチレンスルホネート)(PEDOT/PSS)、ポリアニリン(PANI)、ポリスチレン(PS)から選択される組成物。
- 請求項1に記載の組成物であって、前記カチオン性酸化導電性ポリマーが、バルク、ナノ、フィルム、パッチまたはデバイスの形態である組成物。
- 請求項2に記載の組成物であって、前記カチオン性酸化導電性ポリマーが、ポリアニリン(PANI)である組成物。
- 請求項2に記載の組成物であって、前記カチオン性酸化導電性ポリマーが、ポリ(3,4−エチレンジオキシチオフェン)である組成物。
- 請求項1に記載の組成物であって、前記ミセル集合体が、そのCMCでイオン性ミセルを含む組成物。
- 請求項5に記載の組成物であって、前記ゲスト分子が、組成物の1重量%以下の疎水性分子である組成物。
- 請求項1に記載の組成物であって、前記ゲスト分子が、薬剤、色素、小分子、生体分子、タンパク質およびそれらと同様のものから選択される組成物。
- 請求項1に記載の組成物であって、前記組成物が再生利用可能である組成物。
- ナノコンテナの刺激誘発分解の準備のための方法であって、
a)イオン性ミセルをそのCMCで溶媒に溶解させて、ミセル集合体を形成すること、
b)ステップ(a)の溶液にゲスト分子を添加すること、および
c)酸化導電性ポリマーの添加により、ミセルを分解し、ゲスト分子を同時放出させること
を含む方法。
