解析のためのPCRプライマーは、例えば、5'側プライマーとしての5'−Rプライマー(5'−gtctaccaggcattcgcttcat−3')(SEQ ID NO:93)、および3'側プライマーとしての、TCRα鎖C領域に特異的な3−TRa−Cプライマー(5'−tcagctggaccacagccgcagcgt−3')(SEQ ID NO:94)、TCRβ鎖C1領域に特異的な3−TRb−C1プライマー(5'−tcagaaatcctttctcttgac−3')(SEQ ID NO:95)、またはTCRβ鎖C2領域に特異的な3−TRβ−C2プライマー(5'−ctagcctctggaatcctttctctt−3')(SEQ ID NO:96)であってよいが、これらに限定されない。

HLA−A*2402分子と結合するTMEM22由来の9merおよび10merペプチドを、結合予測ソフトウエア「BIMAS」(www−bimas.cit.nih.gov/molbio/hla_bind)(Parker et al.(J Immunol 1994, 152(1): 163-75)、Kuzushima et al.(Blood 2001, 98(6): 1872-81))および「NetMHC 3.0」(www.cbs.dtu.dk/services/NetMHC/)(Buus et al.(Tissue Antigens., 62:378-84, 2003)、Nielsen et al.(Protein Sci., 12:1007-17, 2003, Bioinformatics, 20(9):1388-97, 2004))を用いて予測した。
5、8、および11日目に、30IU/mlのIL−2を含む新たなAIM−V/5%AS培地を、該培養物に供給した(Tanaka H et al., Br J Cancer 2001 Jan 5, 84(1): 94-9;Umano Y et al., Br J Cancer 2001 Apr 20, 84(8): 1052-7;Uchida N et al., Clin Cancer Res 2004 Dec 15, 10(24): 8577-86;Suda T et al., Cancer Sci 2006 May, 97(5): 411-9;Watanabe T et al., Cancer Sci 2005 Aug, 96(8): 498-506)。
14日目にCTL活性を試験し、上記と同じ方法を用いてCTLクローンを増殖させた(Uchida N et al., Clin Cancer Res 2004 Dec 15, 10(24): 8577-86;Suda T et al., Cancer Sci 2006 May, 97(5): 411-9;Watanabe T et al., Cancer Sci 2005 Aug, 96(8): 498-506)。

以下のウェル番号は、対照ウェルと比較して強力なIFN−γ産生を示した:TMEM22−A24−9−390(SEQ ID NO:1)で刺激したウェル番号#4(a)、TMEM22−A24−9−274(SEQ ID NO:2)で刺激した#7(b)、TMEM22−A24−9−372(SEQ ID NO:3)で刺激した#3および#5(c)、TMEM22−A24−9−331(SEQ ID NO:4)で刺激した#8(d)、TMEM22−A24−9−385(SEQ ID NO:5)で刺激した#4、#6、および#7(e)、TMEM22−A24−9−204(SEQ ID NO:6)で刺激した#3、#4、および#5(f)、TMEM22−A24−9−297(SEQ ID NO:10)で刺激した#3、#6、および#8(g)、TMEM22−A24−9−98(SEQ ID NO:12)で刺激した#3(h)、TMEM22−A24−9−375(SEQ ID NO:16)で刺激した#2および#4(i)、TMEM22−A24−10−137(SEQ ID NO:18)で刺激した#5(j)、TMEM22−A24−10−140(SEQ ID NO:19)で刺激した#1(k)、TMEM22−A24−10−204(SEQ ID NO:22)で刺激した#2、#3、および#4(l)、TMEM22−A24−10−282(SEQ ID NO:28)で刺激した#1、#6、および#8(m)、ならびにTMEM22−A24−10−177(SEQ ID NO:31)で刺激した#7(n)。
上記「材料および方法」の章に記載した通りに、TMEM22−A24−9−390(SEQ ID NO:1)を用いたウェル番号#4、TMEM22−A24−9−274(SEQ ID NO:2)を用いた#7、TMEM22−A24−9−372(SEQ ID NO:3)を用いた#5、TMEM22−A24−9−331(SEQ ID NO:4)を用いた#8、TMEM22−A24−9−385(SEQ ID NO:5)を用いた#6、TMEM22−A24−9−204(SEQ ID NO:6)を用いた#3、TMEM22−A24−9−297(SEQ ID NO:10)を用いた#8、TMEM22−A24−9−375(SEQ ID NO:16)を用いた#4、TMEM22−A24−10−137(SEQ ID NO:18)を用いた#5、TMEM22−A24−10−204(SEQ ID NO:22)を用いた#3、TMEM22−A24−10−282(SEQ ID NO:28)を用いた#8、およびTMEM22−A24−10−177(SEQ ID NO:31)を用いた#7において、IFN−γ ELISPOTアッセイにより検出されるペプチド特異的CTL活性を示した細胞を増殖させ、限界希釈によってCTL株を樹立した。 TMEM22−A24−9−331(SEQ ID NO:4)、TMEM22−A24−9−204(SEQ ID NO:6)、TMEM22−A24−9−297(SEQ ID NO:10)、およびTMEM22−A24−10−204(SEQ ID NO:22)で刺激したCTLクローンから強力なIFN−γ産生が測定された。
TMEM22−A24−9−390(SEQ ID NO:1)、TMEM22−A24−9−274(SEQ ID NO:2)、TMEM22−A24−9−372(SEQ ID NO:3)、TMEM22−A24−9−331(SEQ ID NO:4)、TMEM22−A24−9−385(SEQ ID NO:5)、TMEM22−A24−9−204(SEQ ID NO:6)、TMEM22−A24−9−297(SEQ ID NO:10)、TMEM22−A24−9−98(SEQ ID NO:12)、TMEM22−A24−9−375(SEQ ID NO:16)、TMEM22−A24−10−137(SEQ ID NO:18)、TMEM22−A24−10−140(SEQ ID NO:19)、TMEM22−A24−10−204(SEQ ID NO:22)、TMEM22−A24−10−282(SEQ ID NO:28)、およびTMEM22−A24−10−177(SEQ ID NO:31)で刺激したCTLは、有意かつ特異的なCTL活性を示した。 TMEM22−A24−9−390(SEQ ID NO:1)、TMEM22−A24−9−274(SEQ ID NO:2),TMEM22−A24−9−372(SEQ ID NO:3)、TMEM22−A24−9−331(SEQ ID NO:4)、TMEM22−A24−9−385(SEQ ID NO:5)、TMEM22−A24−9−204(SEQ ID NO:6)、TMEM22−A24−9−297(SEQ ID NO:10)、TMEM22−A24−9−98(SEQ ID NO:12)、TMEM22−A24−9−375(SEQ ID NO:16)、TMEM22−A24−10−137(SEQ ID NO:18)、TMEM22−A24−10−140(SEQ ID NO:19)、TMEM22−A24−10−204(SEQ ID NO:22)、TMEM22−A24−10−282(SEQ ID NO:28)、およびTMEM22−A24−10−177(SEQ ID NO:31)の配列が、ヒト免疫系を感作することが知られている他の分子に由来するペプチドと相同的であるという事実に起因する可能性がある。 TMEM22−A24−9−390(SEQ ID NO:1)、TMEM22−A24−9−274(SEQ ID NO:2)、TMEM22−A24−9−372(SEQ ID NO:3)、TMEM22−A24−9−331(SEQ ID NO:4)、TMEM22−A24−9−385(SEQ ID NO:5)、TMEM22−A24−9−204(SEQ ID NO:6)、TMEM22−A24−9−297(SEQ ID NO:10)、TMEM22−A24−9−98(SEQ ID NO:12)、TMEM22−A24−9−375(SEQ ID NO:16)、TMEM22−A24−10−137(SEQ ID NO:18)、TMEM22−A24−10−140(SEQ ID NO:19)、TMEM22−A24−10−204(SEQ ID NO:22)、TMEM22−A24−10−282(SEQ ID NO:28)、およびTMEM22−A24−10−177(SEQ ID NO:31)の配列が固有のものであること、したがって本発明者らの知る限りでは、これらの分子が、ある非関連分子に対して意図しない免疫学的応答を引き起こす可能性はほとんどないことを示している。
以下のウェル番号は、対照ウェルと比較して強力なIFN−γ産生を示した:TMEM22−A02−9−338(SEQ ID NO:35)を用いたウェル番号#4(a)、TMEM22−A02−9−381(SEQ ID NO:38)を用いた#2(b)、TMEM22−A02−9−367(SEQ ID NO:41)を用いた#6(c)、TMEM22−A02−9−218(SEQ ID NO:48)を用いた#3(d)、TMEM22−A02−10−217(SEQ ID NO:61)を用いた#5(e)、TMEM22−A02−10−304(SEQ ID NO:62)を用いた#8(f)、TMEM22−A02−10−167(SEQ ID NO:65)を用いた#4(g)、TMEM22−A02−10−363(SEQ ID NO:67)を用いた#6(h)、TMEM22−A02−10−103(SEQ ID NO:70)を用いた#5(i)、TMEM22−A02−10−195(SEQ ID NO:74)を用いた#5(j)、TMEM22−A02−10−229(SEQ ID NO:77)を用いた#5(k)、およびTMEM22−A02−10−356(SEQ ID NO:83)を用いた#6(l)。
上記「材料および方法」の章に記載した通りに、TMEM22−A02−9−338(SEQ ID NO:35)を用いたウェル番号#4(a)、TMEM22−A02−9−381(SEQ ID NO:38)を用いた#2(b)、TMEM22−A02−9−218(SEQ ID NO:48)を用いた#3(c)、TMEM22−A02−10−217(SEQ ID NO:61)を用いた#5(d)、TMEM22−A02−10−304(SEQ ID NO:62)を用いた#8(e)、TMEM22−A02−10−167(SEQ ID NO:65)を用いた#4(f)、TMEM22−A02−10−363(SEQ ID NO:67)を用いた#6(g)、TMEM22−A02−10−103(SEQ ID NO:70)を用いた#5(h)、TMEM22−A02−10−195(SEQ ID NO:74)を用いた#5(i)、およびTMEM22−A02−10−356(SEQ ID NO:83)を用いた#6(j)において、IFN−γ ELISPOTアッセイにより検出されるペプチド特異的CTL活性を示した細胞を増殖させ、限界希釈によってCTL株を樹立した。 TMEM22−A02−9−381(SEQ ID NO:38)(a)、TMEM22−A02−9−218(SEQ ID NO:48)(b)、TMEM22−A02−10−217(SEQ ID NO:61)(c)、TMEM22−A02−10−304(SEQ ID NO:62)(d)、TMEM22−A02−10−167(SEQ ID NO:65)(e)、TMEM22−A02−10−363(SEQ ID NO:67)(f)、TMEM22−A02−10−103(SEQ ID NO:70)(g)、TMEM22−A02−10−195(SEQ ID NO:74)(h)、およびTMEM22−A02−10−356(SEQ ID NO:83)(i)で刺激したCTLクローンから強力なIFN−γ産生が測定された(図7a〜i)。
TMEM22−A02−9−338(SEQ ID NO:35)、TMEM22−A02−9−381(SEQ ID NO:38)、TMEM22−A02−9−367(SEQ ID NO:41)、TMEM22−A02−9−218(SEQ ID NO:48)、TMEM22−A02−10−217(SEQ ID NO:61)、TMEM22−A02−10−304(SEQ ID NO:62)、TMEM22−A02−10−167(SEQ ID NO:65)、TMEM22−A02−10−363(SEQ ID NO:67)、TMEM22−A02−10−103(SEQ ID NO:70)、TMEM22−A02−10−195(SEQ ID NO:74)、TMEM22−A02−10−229(SEQ ID NO:77)、およびTMEM22−A02−10−356(SEQ ID NO:83)で刺激したCTLは、有意かつ特異的なCTL活性を示した。 TMEM22−A02−9−338(SEQ ID NO:35)、TMEM22−A02−9−381(SEQ ID NO:38)、TMEM22−A02−9−367(SEQ ID NO:41)、TMEM22−A02−9−218(SEQ ID NO:48)、TMEM22−A02−10−217(SEQ ID NO:61)、TMEM22−A02−10−304(SEQ ID NO:62)、TMEM22−A02−10−167(SEQ ID NO:65)、TMEM22−A02−10−363(SEQ ID NO:67)、TMEM22−A02−10−103(SEQ ID NO:70)、TMEM22−A02−10−195(SEQ ID NO:74)、TMEM22−A02−10−229(SEQ ID NO:77)、およびTMEM22−A02−10−356(SEQ ID NO:83)の配列が、ヒト免疫系を感作することが知られている他の分子に由来するペプチドと相同的であるという事実に起因する可能性がある。 TMEM22−A02−9−338(SEQ ID NO:35)、TMEM22−A02−9−381(SEQ ID NO:38)、TMEM22−A02−9−367(SEQ ID NO:41)、TMEM22−A02−9−218(SEQ ID NO:48)、TMEM22−A02−10−217(SEQ ID NO:61)、TMEM22−A02−10−304(SEQ ID NO:62)、TMEM22−A02−10−167(SEQ ID NO:65)、TMEM22−A02−10−363(SEQ ID NO:67)、TMEM22−A02−10−103(SEQ ID NO:70)、TMEM22−A02−10−195(SEQ ID NO:74)、TMEM22−A02−10−229(SEQ ID NO:77)、およびTMEM22−A02−10−356(SEQ ID NO:83)の配列が固有であることを示している。
