この発明の態様は、国立アレルギー・感染症研究所との契約番号HHSN266200400008C/N01-AI-40008に従って米国政府の支援によってなされている。米国政府はこの発明に対して特定の権利を有する。
リピッドAは、膜結合TLR/MD−2TLR4受容体複合体に結合しているLPS分子内の活性、疎水性構造モチーフである。リピッドAはバクテリア種の間でかなりのばらつきがある。個々のグラム陰性菌は表面上でリピッドAの構造を調節することが出来き、程度は異なっても炎症になる(Bishop et al. 5071-80; Guo et al. 189-988)。アシル鎖の数と長さおよびリン酸化のパターンの変化が極めて強力なアゴニストからアンタゴニストまでのリピッドA分子の活性を変化させることが示されている。(Hawkins, 2002 30067 /id, Stover at al, Seydel et. al. 2000)。ほとんどのLPS種の毒性はTLR4受容体複合体に関するリピッドA部分との相互作用により決定される(Baker et al. 1992)。アシル鎖の数、長さ、および配置、ならびに荷電基の数および位置における差異はリピッドAに対する免疫学的応答のレベルと性格の両方に有意に影響することが示されている(Teghanemt et al. 2005, Stover et. al. 2004, Schromm et al. 1998)。さらに、単糖、リピッドA模倣物、アミノアルキルグルコサミニド4リン酸塩(AGP)とTLR4アゴニストおよびアンタゴニスト活性とを合わせたライブラリーの設計と合成も記載されている(Stover et al. 4440-49;Persing et al. 簡単に述べると、セリルAGP CRX527およびCRX547は、リピッドAの還元性糖類がアミノアルキルL−またはD−セレンベース単位で置換され、かつ、10炭素の正常な脂肪アシル鎖を含む3つの(R)−3−nアルカノイルオキシテトラデカノイル残基を有する、リピッドA模倣物のクラスである。AGPは、ヒドロキシルとアミノ基の化学的識別および(R)−3−nアルカノイルオキシテトラデカノイル残基の連続的導入ができる高度な収束手法により以前に記載されているように調製する。(Bazin et al. Bioorg. Med. Chem. Lett. 2008 18, 5350; Johnson et al Bioorg. Med. Chem. Lett 9 1999 2273; Patent PublicationWO 04/005308を参照)。セリルAGPはシリカゲルフラッシュクロマトグラフィー(純度 >95%)で精製し、標準的な分析法でトリアルキルアンモニウム塩として分析する。
また、図1にはCRX527とCRX547が示されており、以前に合成され記載がされている(U.S. Patent No. 6,113,918; Examples 15 and 16, and WO 2006/012425 WO 2006/016997を参照)。CRX537の場合に比べて、CRX547の構造的な相違がアグリコンカルボキシル基の位置に影響を与える。この差異が荷電カルボキシル基のTLR4受容体複合体との相互作用に影響を与え得る。LPS模倣物がMD2を介してTLR4受容体複合体との相互作用をすることが以前に示され(Ohto et. al. 2007)、従って、CRX547とCRX527のジアステレオマ間の差異がMD−2またはTLD4をともないカルボキシル基、リピッドAリン酸バイオアイソスターの相互作用を阻害する可能性がある。次に、受容体の二量体化、またはそれに代えてMyD88と下流シグナルの結合を推進する細胞内コンフォメーション変化を阻害するかもしれない。おそらくは、この阻害または変化はTRIF依存的シグナルを排除しない。
本明細書に記載された材料および方法は、後述する実施例の実施において有用である。
ヒトTLR4、MD−2、およびCD14発現HEK293細胞株、NF-κB/LacZリポーター・プラスミド、ドミナントネガティブMyD88発現(pDeNy-hMyD88)および ドミナントネガティブTRIF発現(pDeNy-hTRIF)プラスミド、LyoVecトランスフェクション試薬およびS. minnesota 由来のウルトラピュアRe595 LPS は InvivoGenから得ることができる。ヒト単球/マクロファージ細胞株、THP−1はATCCから得ることができる。FuGENE6トランスフェクション試薬はRocheから得ることができる。InvivoGen (San Diego, CA)から得たHEK293-hTLR4/hMD2/hCD14細胞は、 RPMI 1640 (ATCC) の中で10% FBS (Hyclone)、10 μg/mL ブラスチシジン、50 μg/mL HygroGold (InvivoGen)とを使って培養する。THP−1細胞はRPMI1640(ATCC)の中で10% FBS (Hyclone)、100 U/mL ペニシリン/100 ug/mLストレプトマイシン(Sigma)とを使い培養する。
末梢血単核球(PBMC)はフィコール・ハイパック1.077 勾配を介して健康なドナーの血液から単離し、アゴニストで処理され、かつ、述べたように単球溶媒 (RPMI 1640 (ATCC)、10%ヒトAB血清(Lonza/BioWhittaker), 100 U/mL ペニシリン/100 ug/mL ストレプトマイシン(Sigma)、50 uM 2-メルカプトエタノール (Sigma) 内で付着単球を単離するのに使われる(Stover et al. 4440-49;Kawai et al. 5887-94;Gervassi et al. 7231-39)。単球由来マクロファージは、アゴニストを作用させる前に3日目と5日目に溶媒を交換し50 ng/mL rhM-CSF (R&D Systems) を用いて単球溶媒中で付着単球を5日間インキュベートして産生した。単球由来樹状細胞は、3日目と6日目に溶媒を交換し 10 ng/mL rhGM-CSFおよび10 ng/mL rhIL-4 (R&D Systems)を用いて単球溶媒中で付着単球を7日間インキュベートして産生した。
指数増殖におけるMonoMac6細胞は、示された時間にて、2%グリセロール媒体で希釈したAGPの濃度を上昇させて刺激した。直ちに固定化した細胞(2%パラホルマリンで一晩)は、膜透過処理液緩衝液 (PBS, 2%FBS, 0.1% TritonX)中で一次抗体であるanti-NFκB(p65) (SantaCruz Biotechnology, SantaCruz, CA)およびanti-IRF3 (BD Biosciences, San Jose, CA)で染色し、その後、二次抗体であるanti-Rabbit FITC (Jackson Labs, Bar Harbor, ME), anti-mouse PE (BD Biosciences, San Jose, CA)で染色した。核染色 DRAQ5 (Alexis Biochemicals (San Diego, CA) )を用いたシグナル伝達たんぱく質の局在化の類似度スコアは、すでに述べたように核移行の画像ストリーム解析に使用される(George, 2006 32669 /id)。最小限の3,000個の細胞が各試験の条件で採取され解析される。
12ウェルプレート(5 × 105 cells/well)中にTHP−1細胞を播種し、マクロファージへと分化させるために100 U/mL ペニシリン/100 ug/mL ストレプトマイシン(Sigma)、 20 ng/mL phorbol mystyric acid (PMA) (InvivoGen) を使い、 0.5 mL RPMI 1640 (ATCC) 中でインキュベートした。THP−1が分化した後、溶媒を5 ng/mL PMA でTHP−1溶媒に変更し、ならびに、細胞は、48時間、pDeNy-hMyD88/LacZ、またはpUNO-mcs (control)/LacZ プラスミドおよびFuGENE6トランスフェクション試薬の0.5 μg/well で一過的に形質移入した。溶媒は新鮮な溶媒に交換して、その後14時間表示した濃度のTLR4アゴニストで刺激した。回収した上清はLuminexプラットフォームを用いて、マルチプレックスサンドイッチELISA (R&D Systems) によりサイトカインのレベルをアッセイした。対照ウェルは Invivogen LacZ Quantitationキットを用いて、トランスフェクション効率をアッセイした。
指数増殖するRAW264.7 細胞は、タンパク質ノックダウンのためのshRNA 配列、Zeocin抵抗性遺伝子およびGFPコード配列 (InVivoGen, San Diego, CA)を含む、psiRNA-mTICAM1, psiRNA-mMyD88,またはpsiRNA-LUC (無関係な対照) プラスミドを使い、Optimem (Roche, Indianaopolis IN) の既存の方法によりFuGENE6で安定的に形質移入した。選択した後のトランスフェクション効率は、画像ストリームを用いてGFP陽性細胞の割合で決定する。
12ウェルプレート(4 x 105 cells/well)中にHEK293-hTLR4/hMD2/hCD14細胞を播種し、40%〜60%コンフルエントになるまで(2〜3日間)培養した。細胞を、5か所のNF-κB部位 (pNiFty2-SEA-InvivoGen; San Diego, CA)およびチミジンキナーゼプロモータ駆動ルシフェラーゼ (Promega)を構成的に発現するトランスフェクション コントロールプラスミドを用いて、遺伝子組換えELAMプロモータの制御下で分泌されたヒト胎児アルカリホスファターゼを発現する NF-Bレポーター構築物の100 ngを使用して24時間、形質移入した。プラスミドは、QiagenのEndofree Maxiprepプラスミドキットで調製された。トランスフェクション後、細胞は14時間、TLR4アゴニストで刺激し清澄した上清は、 NF-кB活性化を定量するSEAP活性についてアッセイして(SEAP Reporter Assay Kit-InvivoGen)、NF-кB活性化およびルシフェラーゼ活性 (Promega)および供給されたプロトコールに従ってトランスフェクション効率を基準化する。
分化したTHP−1マクロファージ (20 ng/mL PMAを用いて48-ウェルプレート内に5E5/ウェル)は、無血清THP−1溶媒中で60分間エンドサイトーシスに対する低分子阻害剤であるジナソア10 uMで予め処理した。アゴニストの指定濃度で8時間、刺激する前に溶媒は0.45mLのTHP−1溶媒に変更する。上清は採取され、Luminexプラットフォームを用いてマルチプレックスサンドイッチELISA (R&D Systems) によりサイトカイン/ケモカイン解析の前に80℃で貯蔵される。
ウェスタンブロットおよびリン酸―ウェスタンブロットタンパク質解析では、細胞はCell Lysis Buffer (Cell Signaling Technology, Danvers, MA) と併せた Protease Inhibitor Cocktail (Sigma, St. Louism MO)で溶解する。標準的な方法を使ってPVDF膜 (Millipore) 上でウェスタンブロットを実施する。バンドはECL Advance Western Blot kit (GE Healthcare, Piscataway, NJ)で検出する。二次Anti-Rabbit HRP抗体はKPL, (Gaithersburg, MD)から得ることができる。Anti-β actin, anti-IRF-3, anti-phospho-IRF-3 (Ser396)およびanti-IRAK1抗体は、Cell Signaling Technology (Danvers, MA)から得ることができる。
実験開始時の年齢は7週から9週である雌のBALB/cまたはC57Bl/6マウスをCharles River Laboratories, USAから入手した。賦形剤 (0.2%グリセロール) に200 uL のアゴニストまたは賦形剤単独でマウスに腹腔内注射した。血清試料は注射2時間または6時間後に取り出され、Luminexプラットフォームを用いて、マルチプレックスサンドイッチELISA (Invitrogen) によりサイトカイン/ケモカインの誘発に関する試験をした。Public Health ServiceおよびInstitutional Animal Care and Use Committee at GlaxoSmithKline Biologicals, Hamilton, Montanaが確立したガイドラインに従ってすべての動物を用いる。
3匹のウサギの群(NZW−Western Oregon Rabbit Company, Philomath, OR)は0日、7日、14日目に1mLの希釈したAGP,またはMPLコントロールをIMで接種した。AGPの2つの投与が評価される。体重、体温、臨床症状およびDraizeスコア について全ての動物をモニターする。これらの動物は最終投与後一週間で屠殺する。血液学分析と臨床化学検査のため処理前、第2回接種後2日目、および犠牲にした時に血液を採取する。注射部位および主要臓器の病理組織検査を標準的な治療法で実施する。Public Health ServiceおよびInstitutional Animal Care and Use Committee at GlaxoSmithKline Biologicals, Hamilton, Montanaが確立したガイドラインに従ってすべての動物を用いる。
L−異性体(CRX527)との比較における合成リピッドA模倣物(CRX547)のD−異性体投与による類似したレベルのTRIF依存的サイトカイン、一方さらに低いレベルのMyD88依存的サイトカイン
ヒトの新鮮な末梢血単核細胞(PBMC)からのサイトカイン誘発に対するS. minnesota Re595 LPS, CRX-547およびCRX-527の影響を比較した場合、CRX−527とLPSは類似したレベルのサイトカインTNFα およびケモカインMIP-1α (図2A)を誘発する。または、CRX−547は有意に低いレベルのTNFαとMIP-1αを誘発するが、ケモカインIT−10とRANTESは同程度であった(図2B)。
CRX−547は、単球由来樹状細胞におけるより低いレベルのIL−12p70およびIL−23を誘発する。
サイトカイン誘発のMyD88およびTRIF依存性
ドミナントネガティブ変異体構築物の発現がMyD88を介するシグナル伝達を抑制するのに使われる場合、MyD88依存的サイトカインであるTNFαのCRX−527での誘発は有意に減少し、TRIF依存的サイトカインであるRANTESの誘発はほんの僅か減少する一方で、MyD88依存的サイトカインのCRX−547誘発はMyD88のノックダウンにより影響を受けない([066]4A)。
CRX−547はCRX−527と比べた場合、有意に低いレベルのNFκB の核移行および転写活性の誘発し、同等かまたはそれ以上に高いレベルのIRF−3核移行を誘発する。
CRX−547はCRX−527によるMyD88依存的サイトカイン誘発を抑制する。
ヒトTLR4の明らかなTRIFバイアスアゴニズムを持つCRX−547の類似体
AGPファミリーの中の類似体を評価するために、類似パターンのサイトカイン/ケモカイン誘発について、化合物を試験する。エステル連結二次アシル鎖を持つ、セリルAGP、CRX−527およびCRX−547のLとDの異性体対を、エーテル連結二次アシル鎖、化合物1aおよび1b(Error!Reference source not found.1に示す化合物)、リード候補AGPの潜在的安定性強化修飾を持つセリル類縁体と比較した。興味深いことに、1bにおいて、分子を修飾してエステル結合二次アシル鎖ではなくエーテル結合二次アシル鎖を有することにより、、MyD88依存的シグナル伝達の誘発についてL異性体に比べてD異性体において実質的な有効性を回復する(図13)。エーテル結合D異性体によるMyD88有効性救済の機序的な根拠は不明である。
CRX−527またはCRX−547で、またはCRX−527もしくはCRX−547で刺激されたMM6で刺激された一次ヒト単球のリン酸―ウェスタンブロット解析
セリン/トレオニンキナーゼであるIRAK−1の分解はMyD88依存的TLR4シグナル伝達の下流で起こる一方で、TRIF依存的TLR4シグナル伝達はIRF−3活性化を起こす(Yamamoto et al. 6668-72;Yamamoto et al. 640-43)。一次ヒト単球は示された時間、CRX−547かCRX−527のいずれかで刺激され、細胞分解溶解物をMyD88経路―活性化キナーゼ、IRAK−1およびTRIF経路―活性化転写因子IRF−3(リン酸―IRF−3および総IRF−3)のレベルについて、ウェスタンブロットにて解析する。β−アクチンの値はローディング対照として使用される。CRX−527で刺激した単球はIRAK−1が急激に減少する一方で、CRX−547で刺激した細胞ではこのタンパク質の分解の遅れ、減少が示された(Error!Reference source not found.4)。
マウスにおけるMyD88およびTRIF依存的シグナル伝達の評価
一次ヒト細胞の結果とは対照的に BALB/cマウスに静脈投与した後CRX−547はマウス血清中に同様のレベルのMyD88依存的サイトカインを誘発する。この反応がBALB/cマウスに特異的でないことを確認するために、C57/BL/6マウスにおいて血清サイトカイン誘発の同様の試験を実施する(Error!Reference source not found.)。BALB/cマウスに認められる場合と比べて、選択したサイトカインについてCRX−547による相対的な誘発は僅かに小さいが(示していない)、動態(投与後2時間または6時間で誘発)およびサイトカイン誘発の強度はMyD88依存的 (TNFα, IL-10) およびTRIF依存的(RANTES, IP-10, MIG) サイトカイン/ケモカインについてはCRX−527およびCRX−547と類似している。さらに、関心がもたれているものは、試験されたMyD88依存的サイトカインの誘発が投与後6時間よりも2時間において常に高い値である一方で、TRIF依存的サイトカインの誘発は各アゴニストの6時間で高く、これは二つの経路を介したサイトカインの誘発の異なる動態を示唆する。この現象は、多くのTRIF依存的サイトカインの誘発について従来から認められている初期のIFNβ誘発およびオートクリン/パラクリン活性の必要性(Perry et al. 407-22)、または、エンドサイトーシスの必要性によるTRIF依存的シグナルの遅れ(Kagan et al. 361-68)で説明できる。
CRX−547シグナル伝達の種特異性(マウス対ヒト)についての受容体の基礎
TRIFバイアス状態でのシグナル伝達について、CRX−547、CRX−527のD異性体での2つのAGP異性体における異なるシグナル伝達パターンが注目される一方で、CRX−527はMyD88およびTRIF依存的経路の両方を介して情報伝達する。研究の初期の段階では、ヒトの細胞で認められマウスでは認められないのでCRX−547TRIFバイアスシグナル伝達は種依存的と思われた。この明かな相違を取り上げるために、ヒトおよびマウスのTLR4-、MD-2-、および CD14-形質移入HEK293細胞ならびにこれらの組み合せを用いて受容体/共受容体の必要性ならびにAGPシグナルについての受容体の種特異性を試験する。
筋肉内投与によるリード候補AGPを使ったウサギ毒性試験
CRX−527、524、547および化合物1の毒性は、AGPのみを筋肉内投与したウサギで評価する。3匹のウサギの群に、0日目、7日目、14日目に1mLの希釈AGP,またはMPL対照をIMにてワクチン接種した。高用量(25 μg)と低用量(5 μg)の2つのAGPを評価した。体重、体温、臨床症状およびDraizeスコア について全ての動物をモニターする。これらの動物は最終投与後一週間で犠牲にする。血液学分析と臨床化学検査のため処理前、第2回接種後2日目、および犠牲にした時に血液を採取する。注射部位および主要臓器の病理組織検査を実施する。
CRX−527とCRX−547処理に反応するサイトカイン遺伝子発現
ヒト細胞におけるサイトカイン/ケモカイン分泌の誘発についてCRX−527とCRX−547を比べるために、そしてさらに観察された相違点の原因となるシグナル伝達機序を解明するために、CRX−527とCRX−547により誘発された遺伝子発現をマイクロアレイ技術を使い評価した。最初の実験は、qPCRを使ったMyD88およびTRIF依存的サイトカイン遺伝子誘発の時間経過解析を含む。この解析は、両方のアゴニストについてMyD88およびTRIF依存的サイトカイン/ケモカイン遺伝子の誘発は処理後1-2時間で検出可能となり、処理後3時間で確固としたレベルへ上昇し、処理後6時間でベースラインレベル近傍へ下降することを示している(データは示さず)。MyD88およびTRIF依存的サイトカイン/ケモカインの両方の誘発については処理後3時間で最高値となる傾向があり、MyD88およびTRIF依存的誘発の間の実質的な動態的差異は遺伝子発現のレベルでは動作しないことを示唆する。動態的パターンの1つの例外はIFNβ誘発であるが、これはCRX−547かCRX−527処理かのいずれかで1時間処理後すでに最高値を示す状態にあり、その後低下する。
実施例12 in vitroサイトカイン誘発
上述のとおり、より少ないCRX−547によるTRIFバイアスが、マウスのPBMCを用いるサイトカイン誘発実験、マウスの細胞株(RAW264.7)およびHEK293形質移入試験(データは示さず)において示唆される。拡張した投与範囲を使い、エステル連結脂肪アシル鎖ではなくエーテルを用いる追加のLおよびD異性体対、化合物1aおよび1bを試験してRAW264.7細胞におけるCRX−547およびCRX−527によるサイトカインのさらなる比較を提供する(図1)。さらに、種特異性の現象に対するより大きな機序の理解を取得するために、キメラヒト/マウスTLR4受容体複合体で形質移入された細胞においてCRX−547により誘発されたシグナル伝達を比較した。さらに、そのような種―特異的TLR4アンタゴニスト/アゴニスト活性が、リピッドIVaである4つの一次アシル鎖のみの大腸菌からのリピッドA前駆体について報告されており(Muroi, Ohnishi, and Tanamoto 3546-50;Muroi and Tanamoto 5484-91)、この活性はマウスでのTLR4補助受容体MD-2の構造上の違いに起因した。
RAW264.7細胞でのTRIF対MyD88における主要なタンパク質シグナル伝達分子のリン酸化(活性化)状態を測るために、細胞は予備活性化(10 -7M PMAを用いて)し10ng/mlのAGPで刺激した。細胞分解溶解物は、0、15m、30m、45m、60m、120m、240mで調製した。MM6細胞の結果と同様に、CRX−547と比べてIκBα の分解がCRX−527での刺激の後は早く進んだ(Error!Reference source not found.17)。驚くべきことに、CRX−547の刺激はより高いIκBαリン酸化とIRF3リン酸化をもたらす。これらの細胞の増加した反応性は単球細胞の成熟状態を向上させると知られるPMAでの予備活性化によると思われ、CRX−547はリン酸―IκBαおよびリン酸―IRF3の発現を誘発する。IRF3はTRIF媒介性シグナル伝達を経て誘発される。また、総IRF3タンパク質の量はCRX−547で処理された細胞においてより高い。(注記:実験回数は一回であり、負荷コントロール(βactin)は不均一な負荷の可能性を示唆する。)
実施例14 RNA干渉解析
RNA干渉(RNAi)は、TLR2,TLR4およびMyD88のTLRシグナル伝達経路に含まれる特異的遺伝子の発現を破壊するのに利用される。RAW264.7 siRNA細胞株は市販されているプラスミド (Invivogen)を使って調製される。このプラスミドはZeocin選択マーカー、GFPをコードする配列およびノックダウン用のshRNAをコードする配列を含む。細胞が一度効率的に形質移入すると、安定した細胞株が抗生物質選択を経て生じる。培養物の精度は、培養系内のGFP+細胞で決定される。また、無関係なプラスミドも対照として安定的に細胞を形質移入するのに使われる。図30は、同系のsiRNAの安定的な形質移入体においてのMyD88とTRIFのノックダウンを示す。図30Aは、サイトカイン−再生実験の前にGFP陽性細胞のパーセンテージが3つのすべての細胞株において96%以上であることを示す。図30Bは、LPSでの刺激が無関係の対照と比べてタンパク質のレベルが低下しているのでMyD88とTRIFについて首尾よくノックダウンした証拠を示す。
MyD88(TNFαの初期放出)およびTRIF(NFκBの遅延活性化によるTNFαの遅延放出)の両方でコントロールされるTNFαにおいて、CRX−527はCRX−547と比べて、無関係な対照においてより能力がある(図31)。MyD88またはTRIFのいずれかがノックダウンしている場合、CRX−547を超えるCRX−527の効力の増加が認められる。TNFαがMyD88とTRIFの両方で誘発するので、20時間の時点ではCRX−527またはCRX−547の刺激の後のシグナル経路間を十分に区別できない。総合すると、このデータは試験された時間枠内でTNFαはMyD88を必要としないか、または、MyD88のsiRNAのノックダウンがTNFαを抑制するのに十分でないという2つの可能性を示唆する。TRIFシグナル単独がTNFαを誘発できるとう仮説はある程度裏付けられ、つまり、この誘発はTRIF媒介性シグナル伝達によるものである得る。従って、TNFα誘発はMyD88とTRIF媒介性シグナル伝達の両方によるものである得る。化合物1aおよびCRX−679は両方のアッセイ系において20時間で同等の反応性を持つ。

- CRX547をリピッドAアジュバント組成物へ組み込むことを含む、より低いレベルのIL-12p70およびIL-23の誘発が可能なヒトリピッドA模倣物アジュバント組成物を製造する方法。
