Abstract
【解決手段】分裂酵母(Schizosaccharomyces pombe)から特異性を有する制限酵素I−SpomIをコードするDNA配列をクローニング、および発現ベクター、形質転換された細胞株ならびにトランスジェニック動物に組み込むことができる。ベクターは、遺伝子マッピングおよび遺伝子の部位指定挿入に有用である。

Description
遺伝子を生物、例えば哺乳動物の生殖細胞系列に導入できることは、生物学において非常に興味深い。外来的に加えられたDNAを取り込み、そしてDNAに含まれた遺伝子を発現する哺乳動物細胞の性向は、何年も前から解っている。遺伝子操作の結果は、これらの動物の子孫に遺伝される。これらの子孫のすべての細胞は、その遺伝的構成の一部として導入遺伝子を受け継ぐ。かかる動物が、トランスジェニックであると称される。
したがって本発明は、技術的にこれらの必要性を満たすことを補助する。とりわけ本発明は、酵素I−SpomIをコードする単離されたDNAに関する。
分裂酵母(Schizosaccharomyces pombe)のミトコンドリアcox1遺伝子における第1のグループIイントロンの遺伝子産物であるI−SpomIは、エンドヌクレアーゼ活性を有している。これは、イントロンを含まないcox1アレル内のイントロン挿入近くのフランキング配列を認識する。N末端で大幅に修飾したI−SpomIは、元来のリーディグ・フレームの遺伝子産物に比較して切断能力に影響がなく、またエンドヌクレアーゼの配列特異性にも影響がなかった。ホーミングエンドヌクレアーゼの開始コドンの位置は、可変性であり、そして宿主遺伝子の種々の部分に位置することができる。代表的なのは先行するエクソン配列を有するフレームにORFを有するもの、また別にイントロン配列に限定されるリーディグ・フレームを有するもの、またはイントロン内にイントロン性ORFを有するものさえもある。もう1つの基本的な問題は、エンドヌクレアーゼが活性化するために修飾またはプロセシングされる必要があるかどうかである。I−SceII(19)およびI−SceIII(55)に関しては、これらが前駆体タンパク質として合成され、そしてプロセシングされると報告されている。
本発明は、酵素I−SpomIをコードする単離されたDNA配列に関する。酵素I−SpomIは、エンドヌクレアーゼ、とりわけDNAエンドヌクレアーゼである。
酵素I−SpomIをコードする本発明のDNA配列を、生物学的材料におけるヌクレオチド配列を検出するためのプローブとして用いることもできる。プローブを原子または無機ラジカルで、最も一般的には放射性核種を用いて、または分子生物学的実験で一般に用いられるいずれかの非放射性材料で標識することができる。放射性標識には32P、3H、14C等がある。適切なシグナルを提供し、そして十分な半減期を有するいずれかの放射性標識を用いることができる。他の標識には、標識抗体に対する特異的結合メンバーとして提供され得るリガンド、蛍光物質、化学発光物質、酵素、標識リガンドの特異的結合対メンバーとして機能できる抗体などが含まれる。標識の選択は、ハイブリダイゼーションの速度およびDNAまたはRNAに対するプローブの結合に及ぼす標識の影響により規定される。標識は、ハイブリダイゼーションに利用できるDNAまたはRNAの量を検出するのに十分な感度を提供することが必要であろう。
本発明は、また酵素I−SpomIまたはI−SpomI制限部位をコードする本発明のDNA配列に関するもので、この場合ヌクレオチド配列は、他の核酸に連結されている。核酸はいずれかの供給源、例えばプラスミド、クローン化DNAもしくはRNA、または原核および真核生物を含むいずれかの供給源に由来する天然DNAもしくはRNAから入手することができる。核酸は、I−SpomI酵素をコードする核酸が導入されている組換え染色体でよい。同様に、核酸は、I−SpomI制限部位が導入されている組換え染色体でよい。Sambrookら、Molecular Cloning: A Laboratory Manual、Cold Spring Harbor Laboratory、ニューヨーク(2001)に記載される技術を含む種々の技術により、生物学的材料、例えば微生物の培養物、生物学的液体または組織からDNAまたはRNAを抽出することができる。一般に細菌、酵母、ウイルス、または高等生物、例えば植物または動物から核酸を入手する。核酸は、更に複雑な混合物のフラクション、例えばヒトの全DNAに含まれる遺伝子の一部、または特定の微生物の核酸配列の一部でよい。核酸は、巨大分子のフラクションでよいか、または核酸は、連続した遺伝子もしくは遺伝子の集合体からなってもよい。DNAは1本鎖または2本鎖形態でよい。フラグメントが1本鎖形態である場合、従来の技術にしたがって、DNAポリメラーゼを用いてこれを2本鎖形態に変換することができる。
本発明は、また酵素I−SpomIまたはI−SpomI制限部位をコードする本発明のDNA配列を含有するクローニングおよび発現ベクターにも関する。
本発明は、また組換えクローニングおよびDNAを含有する発現ベクター、ならびに組換えベクターを含有する宿主細胞をも提供する。実験室手引書、例えばCurrent Protocols in Molecular Biology(Ausubelら、Wiley(1998))に手引きを見出すことができる。
原核生物にはグラム陰性またはグラム陽性生物などが挙げられる。形質転換に適した原核生物宿主細胞には、例えば大腸菌(E.coli)、枯草菌(Bacillus subtilis)、ネズミチフス菌(Salmonella typhimurium)およびシュードモナス属(Pseudomonas)、ストレプトマイセス属(Streptomyces)およびブドウ球菌(Straphylococcus)属内のその他の様々な種などが挙げられる。原核生物宿主細胞、例えば大腸菌(E.coli)では、原核生物宿主細胞における組換えポリペプチドの発現を促進するために、ポリペプチドはN末端メチオニン残基を含んでよい。N末端Metを、発現された組換えポリペプチドから切断することができる。
または、酵母宿主細胞において、好ましくは酵母菌属(例えばパン酵母(S.cerevisiae))からポリペプチドを発現させることができる。酵母の他の属、例えばピチア(pichia)またはクルイベロマイセス(Kluyveromyces)を用いることもできる。酵母ベクターは、しばしば2μ酵母プラスミドからの複製配列の起点、自己複製配列(ARS)、プロモーター領域、ポリアデニル化のための配列、転写終止のための配列、および選択マーカー遺伝子を含有する。酵母ベクターのための適当なプロモーター配列には、とりわけガラクトース制御プロモーター、例えばGRAP1配列(Molecular Genetics of Yeast,John R.Johnston、Oxford University Press(1994))、メタロチオネインのためのプロモーター、3−ホスホグリセレートキナーゼ(Hitzemanら、J.Biol.Chem.255: 2073(1980))または他の糖分解酵素(Hessら、J.Adv.Enzyme Reg.7: 149(1968); およびHollandら、Biochem.17: 4900(1978))、例えばエノラーゼ、グリセルアルデヒド−3−リン酸デヒドロゲナーゼ、ヘキソキナーゼ、ピルビン酸デカルボキシラーゼ、ホスホフルクトキナーゼ、グルコース−6−リン酸イソメラーゼ、3−ホスホグリセレートムターゼ、ピルビン酸キナーゼ、トリオースリン酸イソメラーゼ、ホスホグルコースイソメラーゼおよびグルコキナーゼなどが挙げられる。他の酵母発現に適したベクターおよびプロモーターは、更にHitzeman、EPA−73,657に記載されている。別の代替は、Russellら(J.Biol.Chem.258: 2674(1982))およびBeierら(Nature 300: 724(1982))に記載されるグルコース抑制ADH2プロモーターである。酵母宿主細胞では、ベクターは、好ましくはシャトルベクターである。酵母および大腸菌(E.coli)の双方で複製可能なシャトルベクターを、大腸菌(E.coli)における選択および複製のためにpBR322からのDNA配列(Amp′遺伝子および複製起点)を前記した酵母ベクターに挿入することにより構築することができる。
哺乳動物または昆虫宿主細胞培養系用いて組換えポリペプチドを発現させることもできる。昆虫細胞内で異種性タンパク質を生成するためのバキュロウイルス系は、LuckowおよびSummers、Bio/Technology 6: 47(1988)により説明されている。哺乳動物起源の確立された細胞系を用いることもできる。適当な哺乳動物宿主細胞系の実例には、サル腎臓細胞のCOS−7系(ATCC CRL1651)(Gluzmanら、Cell 23: 175(1981))、L細胞、C127細胞、3T3細胞(ATCC CCL163)、チャイニーズハムスター卵巣(CHO)細胞、HeLa細胞およびBHK(ATCC CRL10)細胞株、ならびにMcMahanら(EMBO J.10: 2821(1991))に記載されるアフリカミドリザル腎臓細胞株CV1(ATCC CCL70)から誘導されるCV1/EBNA細胞株などが挙げられる。
本発明は、またポリペプチドおよびそのフラグメントを単離および精製する方法をも含む。
1つの好ましい実施態様では、本発明のポリペプチドまたはフラグメントの、本発明のポリペプチドまたはフラグメントの精製を助ける別のポリペプチドへの融合を用いて組換えポリペプチドまたはフラグメントの精製を行うことができる。かかる融合パートナーには、ポリHis、HA−GST、またはその他の抗原同定ペプチド、およびFc部分などが含まれ得る。

(a)アデノウイルスベクター
インビボ分配のための1つの方法は、アデノウイルス発現ベクターの使用に関連する。アデノウイルスを用いてI−SceIをヒト細胞に効率よく分配している(AnglanaおよびBacchetti、Nucleic Acids Research 27: 4276〜4281(1999))。またアデノウイルスをベクターとして用いてHOエンドヌクレアーゼを分配している(Nicolasら、Virology 266: 211〜244(2000))。アデノウイルスベクターを用いてI−SpomI酵素をコードする核酸を分配することができる。
レトロウイルスベクターを用いてI−SpomI酵素またはI−SpomI部位をコードする核酸を細胞に分配することができる。レトロウイルスベクターの使用により核酸の宿主染色体への組み込みが促進される。
他のウイルスベクターを、本発明の発現または分配構築物として用いることができる。ウイルス、例えばワクシナウイルス(Ridgeway、「哺乳動物発現ベクター」、ベクター:分子クローニングベクター及びその使用に関する調査(RodriguezおよびDenhardt(編)、Stoneham: Butterworth)467〜492頁(1988);BaichwalおよびSugden、「動物DNAウイルスに由来する遺伝子移入のためのベクター:移入された遺伝子の一過性および安定した発現」、Gene transfer, (Kucherlapati(編)、ニューヨーク:Plenum Press)117〜148頁(1986); Couparら、「複数の外来性遺伝子を発現する組換えワクシニアウイルスの構築のための一般法」、Gene 68: 1〜10(1988))、アデノ随伴ウイルス(AAV)(Ridgeway、RodriguezおよびDenhardt(編)、Stoneham: Butterworth、467〜492頁(1988); BaichwalおよびSugden、Gene transfer (Kucherlapati(編)、ニューヨーク:Plenum Press)117〜148頁(1986))およびヘルペスウイルスに由来するベクターを用いることができる。
1つの実施態様では、I−SpomI酵素またはI−SpomI部位をコードする核酸を含有する幹細胞を調製することができる。特定の遺伝子のマウスゲノムへの通常の挿入は、マウスES細胞の使用により達成することができる(例えばKusakabeら、米国特許第6,190,910号参照)。マウスES細胞は、インビトロで着床前胚(Evansら、Nature 292: 154〜159(1981); Martin、Proc.Natl.Acad.Sci.USA 78: 7634〜7638(1981))または胎児生殖細胞(Matsuiら、Cell 70: 841〜847(1992))から誘導された未分化の多能性細胞である。マウスES細胞は、連続継代で未分化状態を維持する(Williamsら、Nature 336: 684〜687(1988))。
トランスジェニク動物を作製するために、従来の技術を用いることができる。1つの実施態様では、ES細胞を用いてトランスジェニク動物を作製することができる。別の実施態様では、I−SpomI酵素またはI−SpomI制限部位をコードするプラスミドを1−細胞期マウス受精卵の雄前核に注射できる。次いで注射した卵を偽妊娠代理母の雌に移すことができる。代理母の雌の卵を出産まで成長させる。
真核生物ゲノムを遺伝子マッピングするためのネスト化染色体フラグメンテーション(the nested chromosomal fragmentation)計画は、制限エンドヌクレアーゼI−SpomIの独特な特性、例えば20bpの長さの認識部位を利用する。たいていの真核生物ゲノムにおける天然のI−SpomI認識部位の不在も、またこのマッピング計画に利用される。
半数体細胞では、人工的なI−SpomI部位での染色体内の単一の切断の結果、死に至る細胞分割停止の結果を招く(生存率わずか数%)。切断部位に相同的な無傷配列の存在は、結果的に修復され、100%の細胞が生存する。2倍体細胞では、I−SpomI部位での染色体内の単一の切断の結果、染色体相同性を用いる修復に至り、100%の細胞が生存する。双方の場合、誘導された2本鎖切断の修復の結果、ドナーDNA分子からの非相同性配列の切断および挿入をフランキングする非相同性配列の欠損を有するヘテロ接合性の喪失に至る。FairheadおよびDujon、Mol.Gen.Genet.240: 170〜180(1993)。
−1− I−SpomI認識部位が染色体の独特な位置で挿入されているトランスジェニック細胞の構築。トランスジェニック細胞におけるI−SpomI酵素の発現、ならびに目的の遺伝子およびI−SpomI部位が挿入されている配列に相同なセグメントを含有する核酸分子の導入。
1.部位特異的遺伝子挿入
方法により種々の遺伝子または特定の遺伝子の変異体が、I−SpomI部位の以前の組み込みにより定義される予め決定された位置で挿入され得る細胞および細胞株を無制限に生成することが可能になる。したがって、かかる細胞および細胞株は、表現型、リガンド、薬物のためのスクリーニング手段に、および細胞株がレトロウイルス生成に関してトランスに補完する細胞株である場合には、非常に高レベルでの組換えレトロウイルスの再生産発現に有用である。
類似の細胞株を用いて導入遺伝子、種々のプロモーター、レギュレーターおよび/または構造遺伝子を用いる生物学的またはバイオテクノロジー目的のタンパク質、代謝物または他の化合物を生成することもできる。遺伝子は、いつも染色体の同一の局在性で挿入される。トランスジェニック動物では、組織特異的な様式で複数の薬物、リガンドまたは医薬用タンパク質を試験することが可能になる。
本発明による配列、細胞、動物、染色体および方法で種々の適用を行うことができる。
I−SpomIにより遺伝子活性化を調節することができる。例えばI−SpomI認識部位を、ニューロン特異的エノラーゼプロモーター(pNSE)(Forss-Petterら、Neuron 5: 187〜197(1990))の調節下で、タンデム反復でnlsLacZ遺伝子の一部(例えば62bp)の重複を含有するトランスジェニックマウス系統に導入し、したがってオープン・リーディグ・フレームへの停止コドンの導入により遺伝子機能を損失させることができる。これらのマウスにおけるI−SpomI酵素の発現は、2つのタンデム反復間の組換えを再活性化することができ、すべての中枢神経系(CNS)で遺伝子の再活性化に至る。全CNSの遺伝手術に至るジフテリア毒素のDT−Aフラグメントを用いて同一の実験を実現できる。組織特異的プロモーターによるかまたは遺伝子標的化により得られる天然の座におけるI−SpomI修飾DT−Aの発現により遺伝手術を実施することができる。
ORF I−SpomI酵素をコードするポリヌクレオチドを含有するプラスミドは、2001年3月6日、Collection Nationale de Cultures de Microorganismes(CNCM)、25、Rue du Docteur Roux75724、パリ、Cedex 15、フランスに受入番号I−2643のもとに寄託されている(参照同定:大腸菌(E.coli)BL21(DE3)pLysS/pSP003)。
本発明は以下の実施例を参照することで更に完全に理解されよう。
大腸菌(E.coli)におけるI−SpomIの発現のために、分裂酵母(S.pomb)X39株ゲノムDNA(47)から増幅した種々のPCRフラグメントを、発現ベクターpET16b(Novagen、マジソン)にクローン化した。発現されたタンパク質をN末端10xHisタグと融合してさらなる精製を容易にした。3つの組換えプラスミドを構築した(図1);(i)プラスミドpSP001は、cox1E1のイニシエーターATGコドンからcox1I1b(プライマーSP001m:5′−GCACGCATGTCATATGGTCTTGAGTTTAATGAACTCTTG−3′[配列番号:1]、プライマーSP002m:5′−GCGTAGATGGATCCAAGTGATACTTGATAGTGGTGG−3′[配列番号:2])の配列内停止コドンまでのすべてのオープン・リーディグ・フレームに対応する520コドンのフラグメントを含有する。(ii)プラスミドpSP003は、2つのLAGLIDADGモチーフ(プライマーSP002mと一緒に、プライマーSP003 5′−GAGAGCGCATATACATATGAATAAATTTTTTAATAGACATCC−3′[配列番号:3])を含むイントロン2次構造のループ8(図1A)に位置する304コドンをカバーする最短のインサートを含有する。(iii)プライマーSP005をプライマーSP002と共に用いて第3のプラスミドpSP005を構築し、cox1I1b ORF(プライマーSP002mと一緒に、プライマーSP005:5′−GCATATTAGGATCCATGTTAAAGCCGCAGACAAAATTG−3′[配列番号:4])の全配列をカバーする386コドンの生成物を得た。各々のプラスミドを発現宿主大腸菌(E.coli)BL21(DE3)pLysS(Novagen、マジソン)に形質転換した。
Novagen,マジソンにより提供されるT7発現系を用いて組換えタンパク質を発現した。pSP003で形質転換した大腸菌(E.coli)BL21(DE3)pLysSの前培養を、LB培地50ml中アンピシリン(100μg/ml)およびクロラムフェニコール(34μg/ml)と共に37℃で一晩増殖させ、次いで新鮮な予め加温した同一組成の培地3l中100倍希釈した。細胞を37℃でOD600が0.6〜0.8になるまで成長させた後、イソプロピル−β−D−チオガラクトピラノシド(IPTG)を添加することにより発現を誘導した。誘導の後、細胞を更に3時間、OD600が2.3〜2.8になるまで増殖させ、収穫し、水で洗浄し、そして一定分量を−70℃で保存した。精製用に、主培養物1lから細胞を収穫した。細胞を使用する直前にペレットを氷上で解凍し、そしてプロテアーゼインヒビター(ペファブロック 1μg/ml、アプロチニン 2μg/ml、ロイペプチン 0.5μg/ml、ペプスタチン 1μg/ml)を含む溶解バッファー 30ml(HEPES 30mM pH8、NaCl 300mM、イミダゾール 20mM)に再懸濁した。後続の精製工程すべてを4℃で実施した。細胞破壊は、フレンチ・プレスを用いて実施し、そして続いて粗製ホモジネートをBeckman JA25.50ローター中40000xgで45分間遠心した。即座に上澄をデカンテーションし、Ni2+で荷電したHiTrapキレーティング・アフィニティ・カラム(Amersham Pharmacia Biotech、リトルチャルフォント)1mlに負荷した。アフィニティカラムを用いる精製工程をAkta Purifier(Amersham Pharmacia Biotech、リトルチャルフォント)で実施した(図2)。溶出工程の後に酵素が存在する溶出バッファーを、Micro Bio-Spinサイズ排除カラム(BioRad、ヘルクレス)を用いてジエタノールアミン 100mM、pH9.0に変えた。最終容量の50%までグリセロールを添加した後、タンパク質を−20℃で保存し、そしてBio-Rad Bradford(BioRad、ヘルクレス)タンパク質アッセイを用いてタンパク質濃度を決定した。
エクソンcox1E1およびcox1E2の配列の接合部での推定されるホーミング部位を含む短いPCR産物でI−SpomIでの切断パターンの決定を実施した。センス鎖SP009のプライマー(5′−CTAGAGTAAATAATTTCACATTC−3′[配列番号:5])は、イントロン挿入部位に相対して−100の位置でアニーリングした。相補鎖のプライマーSP008(5′−ATGCAAATAATGGCATTTGATAT−3′[配列番号:6])およびSP010(5′−AATTTACTGATCCTAATGTTGAT−3′[配列番号:7])は、各々下流の+173ヌクレオチド、+129ヌクレオチドにハイブリダイズした(図3A)。PstIで直線化したプラスミドpP3E5−2(48)は切断部位を決定するためのDNA材料を調製するためにPCRの鋳型として提供された。材料をシュリンプアルカリ性ホスファターゼおよびエクソヌクレアーゼIと反応させ、過剰のヌクレオチドおよびプライマーを除去し、次いで変性させた。このように調製したPCR産物を鋳型として用いてサイクルシークエンシング(Thermo Sequenaseサイクルシークエンシングキット、USB、マイルスロード)および5′−末端標識プライマーSP009またはSP008/SP010を用いてDNAシークエンシングラダーを作製した。また鋳型として5′−末端標識プライマーを用いて、プラスミドpP3E5−2(48)のPCR増幅によりI−SpomIのための単一の末端標識基質を生成した。切断の前にAmicon Microcon PCR遠心フィルター装置(Millipore、ベッドフォード)によりマニュアルにしたがって、DNAフラグメントを組み込まれていないデオキシヌクレオチドおよび放射性オリゴヌクレオチドから精製し、続いてフェノール抽出および沈殿を行った。最後にPCR産物を水15μlに再懸濁した。I−SpomIでの消化を全反応容量 25μl(エンドヌクレアーゼ調製物 6μl、ジエタノールアミン 100mM(pH9.0)、MgCl2 2.5mM、溶解したPCR産物1μl)中37℃で10分間実施した。10x停止溶液((51)にしたがって、Tris−HCl 0.1M(pH7.5)、EDTA 0.25M、SDS 5%、プロテイナーゼK 0.5mg/ml)0.1容量添加することにより反応を停止させ、そして50℃に15分間、そして95℃に上げて3分間インキュベートしてプロテイナーゼKを不活性化した。フェノール抽出によりプロテイナーゼKを除去し、そしてサンプルを沈殿させた。未消化サンプルを対照としてI−SpomIの不在下で処理した。エンドヌクレアーゼ消化の後、PCR産物をDNAシークエンシング反応と平行して実行した。Sequenazeキットと共に供給された停止溶液を添加した後75℃で2分間、すべてのサンプルを変性させ、そして6% ポリアクリルアミド/50% 尿素ゲル上で分離した。続いてゲルを乾燥させ、そしてオートラジオグラフィーフィルムまたはPhosphor Imagerスクリーン(Molecular Dynamics、サニーベール)にそれぞれ−70℃でまたは25℃で一晩焼き付けした。結果の評価をImage Quant 5.0ソフトウェアで実施した。
プライマーSP009および前のパラグラフで記載した放射性標識プライマーSP008を用いるPCRにより、I−SpomIの基質が得られた。エンドヌクレアーゼ切断のための標準的な条件は全容量25μl中ジエタノールアミン 100mM(pH9.0)、MgCl2 0.5mM、NaCl 100mM、水 15μl中精製されたPCR産物1μlおよび最終的に調製されたエンドヌクレアーゼ5μlであった。反応物を37℃で20分間インキュベートした。MgCl2濃度は、1mM〜40mMまで変化し、NaClは、0mM〜200mMまで変化した。温度の影響を25℃〜65℃の間でモニター観察した。試験したpH値は、pH6.0(MES 30mM)、pH7.0、pH8.0(HEPES 100mM)およびpH9.0〜10.0(ジエタノールアミン 100mM)の範囲であった。前記したように反応を停止させた。これを5% ポリアクリルアミド/50% 尿素ゲル上で分離した。これを流した後、ゲルを10% 酢酸/20% エタノールの溶液に浸し、ワットマン 3MMペーパー(ワットマン、メイドストン)上に載せ、そして乾燥させた。Phosphor Imager スクリーン(Molecular Dynamics、サニーベール)に乾燥したゲルを25℃で2.5時間焼き付けし、結果を前記したソフトウェアで定量化した。
前記したように、イントロンcox1I1bの挿入部位近くの−13〜+12位置までのヌクレオチド配列を含有するプラスミドを使用した(図5A参照)。混合物を95℃で3分間変性し、そして4℃までゆっくりと冷却した後、相補的オリゴヌクレオチドをNEBuffer2(New England Biolabs、ベバリー)中でアニーリングした。これをBamHI/HindIII消化pUC19にクローン化した。組換えプラスミドを大腸菌(E.coli)DH10B株内でエレクトロポレーションにより形質転換し、そしてその後、アルカリ性プラスミド調製物のEcoRV/XmnI消化により検査した(50)。QIAフィルターチップ100プロトコル(QLAGEN、ヒルデン)にしたがってLBA40mlの培養物から組換えプラスミドのDNAを調製した。調製したプラスミドの濃度を0.3μg/μlに調整した。プラスミドをI−SpomIに暴露する前に、NdeIおよびAlwNIで37℃で3時間消化し、続いて65℃で20分間処理し、沈殿させ、そして最終濃度0.1μg/mlになるまで水に溶解した。I−SpomI消化を容量50μl(DNA 250ng、ジエタノールアミン 100mM(pH9.6)、MgCl2 5mM、NaCl 100mM、I−SpomI 5μl)で実施した。停止溶液(Tris−HCl 0.1M(pH7.5)、EDTA 0.25M、SDS 5%)0.1容量を添加し、そして65℃で3分間インキュベートして消化を停止させ、続いてフェノール抽出および沈殿を行った。消化産物を水10μlに再懸濁し、そしてアガロースゲル 0.8%上で分離した。泳動後、ゲルを臭化エチジウム 0.5μg/μlで染色し、そしてDNAを真空ブロットによりHybond−N+ナイロン膜(Amersham Pharmcia Biotech、リトルチャルフォント)に移した。移した後、膜をハイブリダイゼーションバッファー(リン酸バッファー 0.25M、SDS 7%、EDTA 1mM、BSA 1%)中65℃で1.5時間プレハイブリダイズし、次いでプローブとしてランダム標識pUC19を用いて一晩ハイブリダイズした(52)。シンチレーションカウンターで最終プローブ調製物1μlアリコート中の標識の組み込みを決定した。ハイブリダイゼーションの後、プローブを含有するバッファーを除去し、膜をハイブリダイゼーションバッファーで65℃で洗浄し、そしてPhosphor Imager スクリーン(Molecular Dynamics、サニーベール)に25℃で2.5時間焼き付けし、そして結果を前記したように記述した。
Claims (34)
- 配列番号:8のヌクレオチド配列またはそのフラグメントを含む単離されたDNA配列であって、SpomIエンドヌクレアーゼ活性を有するポリペプチドをコードするDNA配列。
- 配列番号:9のヌクレオチド配列またはそのフラグメントを含む単離されたDNA配列であって、I−SpomIエンドヌクレアーゼ活性を有するポリペプチドをコードするDNA配列。
- 請求項1または2のヌクレオチド配列に相補的な単離されたRNA。
- I−SpomI酵素の発現を指示する発現ベクター。
- 上記ベクターが、哺乳動物細胞において上記酵素を発現する、請求項4記載のベクター。
- 上記ベクターが、アデノウイルスベクターである、請求項5記載のベクター。
- 上記ベクターが、昆虫細胞において上記酵素を発現する、請求項4記載のベクター。
- I−SpomI制限部位を含むベクター。
- I−SpomI部位を含む組換え染色体。
- I−SpomI部位を含む組換え細胞。
- 上記染色体が、真核または原核生物に由来する、請求項9記載の組換え染色体。
- 上記細胞が、真核または原核生物に由来する、請求項10記載の組換え細胞。
- 更にグループIイントロンコード化エンドヌクレアーゼ部位を含む、請求項10記載の細胞。
- 上記グループIイントロンコード化エンドヌクレアーゼ部位が、I−CeuI、I−CreI、I−DmoIおよびI−SceI部位から選択される、請求項13記載の細胞。
- I−SpomI酵素をコードする核酸を含む組換え染色体。
- I−SpomI酵素をコードする核酸を含む組換え細胞。
- 上記染色体が、真核または原核生物に由来する、請求項15記載の組換え染色体。
- 上記細胞が、真核または原核生物に由来する、請求項16記載の組換え細胞。
- 上記染色体が、哺乳動物、昆虫、菌類、植物、酵母、細菌または線虫生物に由来する、請求項11または17記載の組換え染色体。
- 上記細胞が、哺乳動物、昆虫、菌類、植物、酵母、細菌または線虫生物に由来する、請求項12または18記載の組換え細胞。
- I−SpomI部位を含む組換え染色体を含むトランスジェニック生物。
- I−SpomI酵素をコードする核酸を含むトランスジェニック生物。
- 上記生物が、哺乳動物、昆虫、菌類、植物、酵母、細菌または線虫生物である、請求項21または22記載のトランスジェニック生物。
- (a)少なくとも1つのI−SpomI制限部位を含む2本鎖DNAを含有する細胞を提供すること;
(b)I−SpomIエンドヌクレアーゼを有する細胞を提供すること;および
(c)少なくとも1つの2本鎖切断が誘導されている細胞を選択すること;
を含む、細胞のDNAにおいて少なくとも1つの部位指定2本鎖切断を誘導する方法。 - 上記細胞が、哺乳動物、昆虫、菌類、植物、酵母、細菌または線虫細胞である、請求項24記載の方法。
- (a)少なくとも1つのI−SpomI制限部位を含む染色体DNAを含有する細胞を提供すること;
(b)上記細胞にI−SpomI酵素および外来性DNAを提供すること;および
(c)上記外来性DNAが、上記染色体DNAに挿入されている細胞を選択すること;
を含む、細胞の染色体DNAおよび上記細胞に加えられた外来性DNA間で相同組換えを誘導する方法。 - 上記細胞が、哺乳動物、昆虫、菌類、植物、酵母、細菌または線虫細胞である、請求項26記載の方法。
- (a)少なくとも1つのI−SpomI制限部位および少なくとも1つのさらなるグループIイントロンコード化エンドヌクレアーゼ部位を含むDNAを含有する細胞を提供すること;
(b)I−SpomIエンドヌクレアーゼおよびグループIイントロンコード化エンドヌクレアーゼを有する細胞を提供すること;および
(c)少なくとも2つの2本鎖切断が誘導されている細胞を選択すること;
を含む、細胞のDNAにおいて少なくとも1つの部位指定2本鎖切断を誘導する方法。 - 上記グループIイントロンコード化エンドヌクレアーゼが、I−CeuI、I−CreI、I−DmoIおよびI−SceI部位から選択される、請求項28記載の方法。
- 上記細胞が、哺乳動物、昆虫、菌類、植物、酵母、細菌または線虫細胞である、請求項28記載の方法。
- I−SpomIエンドヌクレアーゼ活性を有するポリペプチドをコードするDNA配列を含むベクター。
- 上記ベクターが、受入番号I−2643のもとでCNCMに寄託されているプラスミドである、請求項32記載のベクター。
- 配列番号:11のアミノ酸配列またはそのフラグメントを含む単離されたポリペプチドであって、I−SpomIエンドヌクレアーゼ活性を有するポリペプチド。
- 配列番号:12のアミノ酸配列またはそのフラグメントを含む単離されたポリペプチドであって、I−SpomIエンドヌクレアーゼ活性を有するポリペプチド。
