Abstract

本出願の内容は、国立衛生研究所からの知見を部分的に用いて開発された(助成金申請番号GM64346)。政府は、本技術において一部の権利を有し得る。
本発明は、蛍光読出しを用いて持続的にプロテインキナーゼ活性をモニタリングするためのセンサーを提供する。このセンサーは、プロテインキナーゼペプチド基質の最小限の摂動を必要とする。蛍光は、酵素活性に対応する反応の間、時間に対して応答する。本発明のセンサーは、インヒビターまたは基質の高スループットスクリーニング、細胞抽出物または酵素精製物における活性の検出、細胞中のキナーゼ活性の空間的局在化または時間的局在化、および複雑なシグナル伝達経路の解明において使用され得る。

本発明は、Mg2+への結合に対してキレート化により増強された蛍光(chelation−enhanced fluorescence)(CHEF)を示す式(I)の新規の金属結合アミノ酸残基を提供する。「Sox」と称される、一つの特に好ましいアミノ酸残基(III)が開示される。
本発明は、キナーゼ認識モチーフを含有するペプチド内でキナーゼ活性を検出するためのセンサーを提供する。
R’は、ヒドロキシ、アミノ、またはチオールであり;
R’’は、C1〜6アルキルであり;
R’’’は、水素またはアルキルであり;そして
nは、1、2、または3である。
bKmおよびVmaxの値は、基質および生成物の蛍光について適切に補正された、反応アッセイの初期傾きから得られた。報告された値は、別個のHanesプロットからの4つの値の平均である。
c励起波長:360nm、発光波長:485nm。
dアッセイ条件:20mM HEPES、pH7.4、10mM MgCl2、0.3mM CaCl2、1mM ATP、1mM DTT、0.5μg/mlホスファチジルセリン、0.1μg/mlジアシルグリセロール、0.7nM PKCa30℃。
e蛍光強度に対する生成物および基質の濃度の単位(単位/μM)から計算した。
fアッセイ条件:20mM HEPES、pH7.4,10mM MgCl2、1mM ATP、1mM DTT、40単位PKA,30℃。
g単一ペプチドの溶液(10μM)から計算した。
Fmoc−PAL−PEG−PS樹脂(0.22mmol当量)上の標準的なFmocアミノ酸保護化学(Fmoc amino acid protection chemistry)を用いて、ペプチドを合成した。Fmoc保護アミノ酸の上記樹脂へのカップリングを1−ベンゾトリアゾールイルオキシトリス(ピロリドン)ホスホニウムヘキサフルオロホスフェート(1−benzotriazolyoxytris(pyrrolidino)phophonium hexafluorophosphate)(PyBOP)、1−ヒドロキシベンゾトリアゾール(HOBt)およびジイソプロピルエチルアミン(DIEA)またはO−(7−アザベンゾトリアゾール−1−イル)−1,1,3,3−テトラメチルウラニウムヘキサフルオロホスフェート(O−(7−azabenzotrazol−1−yl)−1,1,3,3−tetramethyl uranium hexafluorophosphate)(HATU)およびDIEAを使用して行い、活性型エステルを生じた。この樹脂をジクロロメタン中で膨潤させ(5分)、次いで合成の前にDMFで膨潤させた(5分)。Sox、ホスホセリン、ホスホスレオニン、およびホスホチロシン以外の全てのアミノ酸を、以下の代表的な手順によって添加した:Fmoc基の除去(DMF中20%ピペリジン溶液、3×5分)、洗浄(DMF、5×1分)、カップリング(アミノ酸/PyBOP/DIEA、6:6:6、DMF中0.05M、45分)、リンス(DMF、2×1分;DCM、2×1分)。Sox残基をカップリングするために、各時間2当量で二重カップリングを使用した(Fmoc−Sox−OH/PyBOP/DIEA、2:2:2、DMF中0.15M、2×120分)。リン酸アミノ酸残基をカップリングするために、HATUを使用した(Fmoc−Xaa(PO(OBzl)OH)−OH/HATU/DIEA、3:3:3、DMF中0.05M、30分)。
bC18;溶媒A=水、0.1% v/v TFA;溶媒B= MeCN、0.1% v/v TFA、5分、10%Bのあと30分にわたって15〜50%Bの直線勾配を続けた。
cC18;溶媒A=水、0.1% v/v TFA;溶媒B=MeCN、0.1% v/v TFA、5分、10%Bのあと30分にわたって20〜70%Bの直線勾配を続けた。
dPE Biosystems Mariner質量分析計上でES−MSデータを収集した。
Zn2+に対するリン酸化ペプチドの親和性に起因して、調製の後に金属イオン不純物を除去する必要性を回避するために、最も高い純度で最も低い金属含有量を有する試薬を使用した。
1)ペプチドのストック溶液を超純水(18MΩ)中で調製し、その濃度をUV/VISによって決定した(フルオロフォア単位の決定された吸光係数に基づいて、1mM Na2EDTAを含む0.1M NaOH中、355nmにおいて5−(N,N−ジメチルスルホンアミド)−8−ヒドロキシ−2−メチルキノリン、ε=8247M−1 cm−1)。各々異なる体積のストック溶液を使用して調製された4つの別個の溶液からの値の平均をBeckman DU 7500 Spectrophotometerで読み取った。ペプチドのストック溶液を4℃で保存した。
PKC:アッセイの日に、1μlアリコートのプロテインキナーゼCα(ヒト、組換え、Calbiochem)を、20μlの溶液4で希釈して氷上に保存した。代表的な反応は、溶液4(84μl)、溶液6(19μl)、溶液8(5μl)、溶液7(1μl)、および酵素のワーキングストック(1μl)を含んだ。適切な体積の基質のストック溶液を添加して、反応を始めた。
Jobin Yvon.からのFluoromax3で蛍光実験を行い、5nmの発光および励起のスリット幅を使用した。全ての実験について、360nmの励起波長を使用した。酵素アッセイを485nmにおける発光をモニタリングすることによって行った。
図2は、酵素を含まない適切なアッセイ混合物中のリン酸化ペプチド(実線)およびリン酸化されていないペプチド(破線)の各々10μMの蛍光スペクトルを示す:(a)Ac−Sox−Pro−Gly−(p)Ser−Phe−Arg−Arg−Arg−NH2(SID番号1)、(b)Ac−Leu−Arg−Arg−Ala−(p)Ser−Leu−Pro−Sox−NH2(SID番号2)、(c)Ac−Sox−Pro−Gly−(p)Thr−Phe−Arg−Arg−Arg−NH2(SID番号3)、(d)Ac−Sox−Pro−Gly−Ile−(p)Tyr−Ala−Ala−Pro−Phe−Ala−Lys−Lys−Lys−NH2(SID番号4)。全てのペプチドは、474nmにおいて最大発光波長を有するAc−Leu−Arg−Arg−Ala−pSer−Leu−Pro−Sox−NH2(SID番号2)を除いて、485nmにおいて最大蛍光発光を有する。このことは、1:1複合体以外の複合体形成を示していると思われるが、最大発光波長は、10mM MgCl2を含むペプチド濃度の広い範囲にわたって一定である。
この反応についてのKmおよびVmaxを解くために、蛍光強度の増加から生成物形成の初期速度を決定することが必要である。このセンサーを用いて、出発物質が消費されることに起因した蛍光強度の減少に対する補正が、初期傾きから生成物形成の速度を決定するために必要とされる。任意の所定の点での蛍光強度は、以下の式から決定され得る:
全ての反応を蛍光光度計において行い、40μlの0.1M Na2EDTAストック溶液でクエンチし、次いで氷上で保存した。図3Aは、PKAによるAc−Leu−Arg−Arg−Ala−Ser−Leu−Pro−Sox−NH2(SID番号2)(7.8μM)のリン酸化(18分後、30℃)を示す。図3Bは、PKAによるAc−Sox−Pro−Gly−Ser−Phe−Arg−Arg−Arg−NH2(SID番号1)(30μM)のリン酸化(12分後、30℃)を示す。このペプチドに対するHPLCピークの同一性を下の表3に示す。
bC18;溶媒A=水、0.1% v/v TFA;溶媒B=MeCN、0.1% v/v TFA、10分、15%Bに、30分にわたる15〜50%Bの直線勾配を続けた。
図4は、PKCとのAc−Sox−Pro−Gly−Ser−Phe−Arg−Arg−Arg−NH2(SID番号1)の反応のHanesプロットである。fS,avg=1.1×105±0.3単位/μM、およびfP,avg=6.3×105±1.0単位/μMを使用して、KmおよびVmaxの値を決定した。
Claims (17)
- 請求項1に記載のペプチドであって、キナーゼ認識配列をさらに含む、ペプチド。
- 請求項5に記載のペプチドであって、配列−Sox−Pro−Gly−を含む、ペプチド。
- 請求項1に記載のペプチドであって、該ペプチドは、配列(SID番号9):

ここで、少なくとも1つのR基は、−SO2Xであり、Xは、−OR’’または−NR’’R’’’であり;
R’は、ヒドロキシ、アミノ、またはチオールであり;
R’’は、C1〜6アルキルであり;

nは、1、2、または3である、アミノ酸であって;
X2およびX3は、各々独立して、βターン配列を一緒に形成するアミノ酸残基であり;
X4は、結合またはアミノ酸残基であり;そして
X5は、ペプチジルキナーゼ認識モチーフであって、該認識モチーフのヒドロキシルアミノ酸は、X4に結合している、ペプチド。 - 請求項8に記載のペプチドであって、配列−Leu−Pro−Sox−を含む、ペプチド。
- 請求項1に記載のペプチドであって、該ペプチドは、配列(SID番号11):

(2)βターン配列;および
(3)式(I):

を包含する、方法。 - 請求項11に記載の方法であって、前記ペプチドの蛍光が、Mg2+に結合した場合に少なくとも100%まで増加する、方法。
- 請求項11に記載の方法であって、前記キナーゼに対する前記ペプチドのKmが、マイクロモル範囲にある、方法。
- 請求項16に記載のキャッピング基により、N末端が保護されている、ペプチド。
