図2はシース液20にサンプル液18を包み込んで流下させるフローセル24の拡大図を示す。フローチャンバ12のサンプルチューブ19からは、シース液20に包まれたサンプル液18が細長い押し出し流となってフローセル24に供給される。このとき界面活性剤35の親水基36は馴染みやすい水性液のシース液20側に向き、界面活性剤35の疎水基38は馴染みやすい油性液のサンプル液18側に向く。そして界面活性剤35は図示のようなフローチャンバ12の軸心方向に層状に積み重なった薄層の単分子膜を形成する。すなわち油性液のサンプル液18と水性液のシース液20との間に境界面となる界面が形成される。これによりサンプル液18とシース液20との混合による流れの乱れが軽減される。その結果としてサンプル液18のサンプル粒子22を安定して極細い流路範囲に層流として流すことができる。

なおサンプル液18に加える圧力Aまたはシース液20に加える圧力Bを調整することにより、サンプル粒子22がフローセル24内を通過する速さを調整することが可能である。
このようなフローサイトメータ10では、サンプル液18に油性液と両親媒性溶液を用い、シース液20に水性液を用いているので、両親媒性溶液が水性液のシース液20との間で単分子膜を形成し界面を維持しながら、フローセル24内で乱流が発生することがなく層流を形成することができる。なおサンプル液18に水性液を用い、シース液20に油性液と両親媒性溶液を用いる構成とすることもできる。
