Abstract
Description
本発明は新しい薬物ターゲットに関連する。より正確に言えば、ステロイドホルモン生成阻害に応答性の癌および他の疾患の治療のためのターゲットとしてのグルタチオントランスフェラーゼ(GST)である。好ましくは、GSTはステロイドイソメラーゼ活性を有するGST A3−3である。
前立腺癌および乳癌は悪性疾患の2つの主要な型であり、人口比率に重大な影響を及ぼす。両症例における腫瘍の増殖は多くの場合ステロイドホルモンに依存し、重要な治療上の取り組みとしてはホルモン生成物の除去およびホルモンレセプターの遮断を含む。
本発明によれば、グルタチオントランスフェラーゼ(GST)、好ましくはGST A3−3/ステロイドイソメラーゼは、ステロイド生合成において、2重結合の異性化への寄与に基づく化学療法のための新しいターゲットとして提供されている。GST A3−3は選択的組織分布を有し、Δ5−アンドロステン−3,17−ジオンおよびΔ5−プレグネン−3,20−ジオンの異性化において高い触媒活性を示す(図1)。本発明は、GST A3−3の触媒効率が、3β−ヒドロキシステロイドデヒドロゲナーゼのステロイドイソメラーゼ活性の200倍も高いことを示している。本発明は主に前立腺における癌に関連しているが、加えてステロイドホルモン生成阻害の原理は胸部および他の器官におけるステロイド応答性の癌に応用できる。さらに、クッシング症候群のようなステロイドホルモン依存性疾患に応用できる。

実験手順
材料−1−クロロ−2,4−ジニトロベンゼン(CDNB)および還元型グルタチオン(GSH)はSigma(St.Louis,MO)、フェネチルイソチオシアナートはAldrich(Milwaukee,WI)、およびΔ5−アンドロステン−3,17−ジオンはSteraloids Inc.(Newport,RI)から購入できる。
酵素活性は標準分析システムにより決定し、次いで、活性の50%阻害を与える阻害剤の濃度(IC50)を決定した。

Claims (24)
- 活性のあるグルタチオントランスフェラーゼ(GST)タンパク質の濃度を抑制する、或いは、グルタチオントランスフェラーゼ(GST)のステロイドイソメラーゼ活性を阻害する化合物のスクリーニング方法であって、グルタチオントランスフェラーゼ(GST)が薬物ターゲットとして使用される方法。
- 使用するGSTがGST A3−3である、請求項1に記載の方法。
- 使用するGSTがGST A1−1である、請求項1に記載の方法。
- 請求項1ないし3のいずれか1項に記載の方法により同定されうる、グルタチオントランスフェラーゼ(GST)のステロイドイソメラーゼ活性を阻害する阻害剤。
- 請求項1ないし3のいずれか1項に記載の方法により同定されうる、活性のあるグルタチオントランスフェラーゼ(GST)の組織濃度を低下する阻害剤。
- 次の式:
XはGe、Sn、Pb或いは類似の求電子性原子であってよい]
を有する化合物である請求項4に記載の阻害剤;
ならびに該阻害剤の立体異性体。 - XがSnである、請求項6に記載の阻害剤。
- R1ないしR4の1つがCl、Br或いはアセテートイオンであり、もう1つの置換基がエチル、ブチル或いはフェニルである、請求項6或いは7に記載の阻害剤。
- ステロイド、ステロイド誘導体或いはステロイド擬態化合物である、請求項4に記載の阻害剤。
- Δ5−アンドロステン−3β−オール−17−オン或いは構造的に類似な化合物である、請求項9に記載の阻害剤。
- ペプチド、ペプチド誘導体或いはグルタチオンに構造的な類似性を有するペプチド擬態化合物である、請求項4に記載の阻害剤。
- Sが置換された、および/または、他の様式で置換されたグルタチオン誘導体であって、置換基がアルキル、アリルおよびアラルキル基であってよい、請求項11に記載の阻害剤。
- S−ヘキシル−グルタチオン或いはS−p−ブロモベンジル−グルタチオンである、請求項12に記載の阻害剤。
- オリゴヌクレオチド、抑制性RNA(siRNA或いはRNAi)或いはPNA(ペプチド核酸)のような抑制性核酸である、請求項5に記載の阻害剤。
- 医薬として使用される、請求項4ないし14に記載の阻害剤。
- 哺乳類において、ステロイドホルモン依存性疾患の治療に使用される、請求項15に記載の薬剤。
- ステロイドホルモン依存性癌の治療に使用される、請求項16に記載の薬剤。
- 前立腺癌の治療に使用される、請求項17に記載の薬剤。
- 乳癌の治療に使用される、請求項17に記載の薬剤。
- クッシング症候群の治療に使用される、請求項16に記載の薬剤。
- 活性のあるグルタチオントランスフェラーゼ(GST)タンパク質の濃度を抑制する、或いは、GST A3−3および/或いはGST A1−1のグルタチオントランスフェラーゼ(GST)のステロイドイソメラーゼ活性を阻害する化合物を、治療が必要であるヒトに対して投与することを含む、癌、或いはステロイドホルモン依存性疾患の治療方法。
- ヒトが前立腺癌にかかっている男性である、請求項21に記載の方法。
- ヒトが乳癌にかかっている女性である、請求項21に記載の方法。
- ヒトがクッシング症候群にかかっている、請求項21に記載の方法。
