酸化還元酵素は、バイオセンサ10の使用時において、試料溶液中の基質を酸化するという機能を有する。酸化還元酵素の種類は特に制限されず、測定を希望する成分に応じて、適宜選択されうる。反応層70に含まれる酸化還元酵素の一例を挙げると、グリセロールオキシダーゼ、グリセロールデヒドロゲナーゼ、コレステロールオキシダーゼ、コレステロールデヒドロゲナーゼ、サルコシンオキシダーゼ、乳酸オキシダーゼ、乳酸デヒドロゲナーゼ、フルクトースオキシダーゼ、フルクトースデヒドロゲナーゼ、アルコールオキシダーゼ、アルコールデヒドロゲナーゼ、グルタミン酸オキシダーゼ、グルタミン酸デヒドロゲナーゼ、ソルビトールデヒドロゲナーゼ、ビリルビンオキシダーゼ、ペルオキシダーゼなどが例示されうる。また、種々の目的で改良が施された改良型の酵素が用いられてもよい。反応層70における酸化還元酵素の含有量についても特に制限はなく、測定を希望する成分の種類や試料溶液の添加量などに応じて適宜調節されうる。一例を挙げると、通常は0.001〜100活性単位、好ましくは0.01〜10活性単位、より好ましくは0.01〜5活性単位の酸化還元酵素が反応層70に含まれるとよい。

電子受容体は、バイオセンサ10の使用時において、酸化還元酵素の作用によって生成した電子を受け取る、すなわち還元される。そして、還元された電子受容体は、酵素反応終了後に電極系40に流される電流によって電気化学的に酸化される。この際に流れる電流(酸化電流と称する)の大きさから、試料溶液中の所望の成分の濃度が算出されうる。電子受容体の種類についても特に制限はなく、従来公知の電子受容体が適宜選択されうる。一例を挙げると、フェリシアンイオン、p−ベンゾキノンおよびその誘導体、フェナジニウムメチルサルフェートおよびその誘導体、メチレンブルー、チオニン、インジゴカーミン、ガロシアニン、α−ナフトキノンおよびその誘導体、並びにサフラニンからなる群から選択される1種または2種以上の電子受容体が例示される。また、p−ベンゾキノン誘導体、フェナジニウムメチルサルフェート誘導体、α−ナフトキノン誘導体としては、p−ベンゾキノン、フェナジニウムメチルサルフェート、α−ナフトキノンに炭素数1または2のアルキル基、炭素数1または2のアルコキシ基、フッ素原子、塩素原子、臭素原子などのハロゲン原子等が結合したものなどが挙げられる。反応層70における電子受容体の含有量についても特に制限はなく、試料溶液の添加量などに応じて適宜調節されうる。一例を挙げると、0.5〜10μLの試料溶液を添加して用いるバイオセンサ10の反応層70には、通常は0.01〜1000μg、好ましくは0.1〜100μg、より好ましくは1〜50μgの電子受容体が含まれるとよい。

本発明のより好ましい形態において、反応層70は、酸化還元酵素として補酵素であるピロロキノリンキノン(PQQ)依存型のグリセロールデヒドロゲナーゼを含む。この際、電子受容体としてはフェナジニウムメチルサルフェート誘導体である1−メトキシ−5−フェナジニウムメチルサルフェートを含むことがより好ましい。さらに好ましくは、反応層70がリポプロテインリパーゼをさらに含む。ここで、PQQ依存型グリセロールデヒドロゲナーゼは溶液中の電子受容体のみを反応に使用し、溶存酸素の影響を受けない。従って、上記のより好ましい形態によれば、還元型の電子受容体の酸化電流を測定することにより、試料溶液中のグリセロール濃度がより正確に測定されうる。

本発明のバイオセンサ10において、反応層70は、タンニン酸、ペクチン、カゼイン、カラギナン、ファーセレラン、プルラン、コラーゲン、キチン、キトサン、コンドロイチン硫酸ナトリウム、リグニンスルホン酸、およびこれらの誘導体からなる群から選択される水溶性高分子を含む。反応層70がこれらの水溶性高分子を含むことにより、タンパク質などの吸着性成分の電極系40への吸着が効果的に抑制されうる。また、これらの高分子を含むことで、電極系40表面からの反応層70の剥離や、反応層70の割れも効果的に防止されうる。よって本発明によれば、測定精度および信頼性に優れるバイオセンサが提供されうる。なかでも、タンニン酸、ペクチン、カゼイン、およびこれらの誘導体が反応層70に含まれると、吸着性成分の電極への吸着がより一層抑制されうる。なお、上記の水溶性高分子は、1種のみが単独で反応層70に含まれてもよいし、2種以上が併せて反応層70に含まれてもよい。また、上記の所定の高分子が反応層70に含まれることにより吸着性物質の電極系への吸着が抑制されるメカニズムは完全に明らかとはなっていないが、反応層70を形成するための溶液中に上記の所定の高分子が含まれると、当該溶液を所定の位置に滴下し乾燥させて反応層70を形成する際に、より均一な層が形成され、かつ、より細かい網目構造が形成されることが何らかの関与をしているものと推測される。ただし、かようなメカニズムはあくまでも推測に過ぎず、他のメカニズムに基づいて吸着性物質の電極系への吸着が抑制されていたとしても、本発明の技術的範囲は何ら影響を受けることはない。

まず、濃度の測定を希望する成分を含む試料溶液の所定量を、バイオセンサ10の反応層70に供給する。試料溶液の具体的な形態は特に制限されず、バイオセンサ10に用いられる酸化還元酵素の基質を含む溶液が適宜用いられうる。試料溶液としては、例えば、血液、尿、唾液などの生体試料、果物、野菜、加工食品原料などの食品等が用いられうる。ただし、その他の溶液が試料として用いられてもよい。また、試料溶液は原液がそのまま用いられてもよいし、粘度などを調節する目的で適当な溶媒で希釈された溶液が用いられてもよい。試料溶液に含まれる基質についても特に制限はなく、上述した酸化還元酵素と反応しうる物質であればよい。基質としては、例えば、グルコースなどの糖類、グリセロール、ソルビトール、アラビトールなどの多価アルコール、中性脂肪、コレステロールなどの脂質、グルタミン酸や乳酸などの有機酸類、クレアチン、クレアチニンなどが挙げられる。試料溶液を反応層70へ供給する形態は特に制限されず、所定量の試料溶液を反応層70に対して垂直に直接滴下することにより供給してもよいし、別途設けた試料溶液供給手段により、反応層70に対して水平方向から試料溶液を供給してもよい。

その後、還元型の電子受容体を酸化する目的で、電極系40を介して、作用極44と対極46との間に、所定の電圧を印加する。これにより、反応層70中に電流(以下、「酸化電流」とも称する)が流れ、この電流によって還元型の電子受容体が電気化学的に酸化され、酸化型へと変換される。この際に測定される酸化電流の値から、電圧印加前の還元型の電子受容体の量が算出され、さらに、酵素と反応した基質の量が定量されうる。酸化電流を流す際に印加される電圧の値は特に制限されず、従来公知の知見を参照して適宜調節されうる。一例を挙げると、−200〜700mV程度、好ましくは0〜500mVの電圧を、参照極42と作用極44との間に印加すればよい。電圧を印加するための電圧印加手段についても特に制限はなく、従来公知の電圧印加手段が適宜用いられうる。
