Abstract
【解決手段】Helicobacter pylori空胞形成毒素をコードする単離された核酸であって、特定な配列で規定されるヌクレオチドからなる核酸。或いは、H.pyloriのvacAコード領域のための調節配列を含む単離された核酸や、これらの核酸と選択的にハイブリダイズする単離された核酸とすることもできる。以上の核酸は、特定の配列を有するタンパク質をコードし、診断試薬やワクチンに有用である。

本発明は、機能性空胞形成毒素を発現しないH. pyloriの遺伝子操作変異株、このHelicobacter pylori空胞形成毒素をコードする単離された核酸、空胞形成毒素をコードする核酸と選択的にハイブリダイズする核酸ならびにH.pylori感染に対して免疫および処置する方法に関する。
Helicobacterpyloriは、ヒト慢性表在性胃炎の主な病因性因子であり、そしてこの生物による感染は、消化性潰瘍疾患およびおそらく胃ガンの病因における重要な病因性因子である(1〜3)。基本的には、H.pylori感染者は全て組織学的胃炎に発展するが(4)、H. pylori感染者の大半は無症候にとどまり(4)、その他の感染者は消化性潰瘍または胃腺ガンのような重篤な感染合併症に発展する(2、3)。個々のH.pylori単離体は高レベルの遺伝子型多様性を示すが(5、6)、この生物の表現型特徴はほとんど全て保存される。現在、株間で異なることが知られている唯一の表現型特徴は、空胞形成細胞毒素の産生(7、8)およびcagAによりコードされる128kDaの細胞毒素関連タンパク質の存在(8)である。消化性潰瘍疾患を有する患者は、胃炎のみを有する患者よりも空胞形成細胞毒素産生株に頻繁に感染されている(11、12)。同様に、128kDaの細胞毒素関連CagAタンパク質への血清学的応答は、H. pylori感染者における消化性潰瘍の存在と関係がある(8、14)。従って、これらの2つの関連する表現型は、H.pylori感染の臨床的結果に影響し得る重要な毒性決定因子である。
上記目的を達成するために、本発明は、例えば、以下を提供する。

項目4.Helicobacter pyloriの単離された核酸であって、配列表の配列番号1で規定されるヌクレオチド配列からなる、核酸。
項目5.上記核酸を発現するために適切なベクター中にある、項目4に記載の核酸。
項目6.上記核酸を発現するために適切な宿主中にある、項目5に記載の核酸。
項目7.項目1に記載の核酸によりコードされる、精製されたタンパク質。
項目8.薬学的に受容可能なキャリア中にある、免疫原性量の項目7に記載のタンパク質を含む組成物。

核酸
本発明は、Helicobacterpylori空胞形成毒素をコードする単離された核酸を提供し、この核酸は、配列表において配列番号1として規定されるヌクレオチド配列のヌクレオチド101〜3964からなる。この核酸は、本発明によって提供されるH.pyloriのvacAコード領域の2本鎖配列の一例である。「単離された」核酸は他のH. pylori遺伝子から分離される核酸である。
本発明は、本発明の核酸によりコードされる精製タンパク質を提供する。本発明のタンパク質の1例は、配列番号1のヌクレオチド配列のヌクレオチド101〜3964によりコードされる、H. pyloriの空胞形成毒素(配列番号2)である。
本発明はまた、機能性空胞形成毒素を発現しないH. pyloriの遺伝子操作された変異株を提供する。この変異体株は、例えば、天然のH. pyloriに見出されない核酸の存在により、天然に存在するtox−H. pylori株と区別され得る。1つの例では、変異体H. pylori株は本明細書の実施例に記載の空胞形成毒素のコード配列に挿入変異を作製することにより得られる。本発明は毒素をコードする核酸を提供するので、毒素のコード配列を変異させる他の方法が、本明細書中に意図されるように、他の変異株を得るために使用され得る。本発明の変異体H.pylori株の例は、84-183:v1および60190:v1と名付けられる。
本発明は、薬学的に受容可能なキャリア中に、免疫原性量の本発明のタンパク質を含む組成物を提供する。この組成物に使用されるタンパク質は、本発明の空胞形成毒素タンパク質であり得る。H. pyloriによる感染に対して被験体を免疫する方法が提供され、この方法は本発明の免疫原性組成物を被験体に投与する工程を包含する。
本発明の精製タンパク質およびポリペプチドフラグメントは、それらの免疫原性および特異性を測定するために試験され得る。簡単に述べれば、種々の濃度の推定の免疫原フラグメントを調製しそして動物に投与し、そして各濃度に対する動物の免疫学的応答(例えば、抗体の産生または細胞媒介性免疫)を測定する。投与される抗原量は、被験体(例えば、ヒトまたはモルモット)、被験体の状態、被験体のサイズなどに依存する。その後、そのように抗原を接種された動物は、細菌に曝され得、特異的免疫原性タンパク質またはフラグメントの潜在的なワクチン効果を試験される。推定の免疫原性フラグメントの特異性は、接種動物由来の血清、他の体液またはリンパ球を、他の近縁の細菌との交差反応性について試験することにより確かめられ得る。
本発明の核酸は、核酸の発現に適するベクター中に存在し得る。ベクター中の核酸は、核酸の発現に適切な宿主中に存在し得る。
空胞形成毒素、H.pyloriが発現する機能性毒素、天然に存在するtox H.pyloriまたは本発明の遺伝子的に改変された変異体の存在は、この毒素、毒素フラグメント、toxまたは変異した毒素遺伝子に特異的な核酸の存在を検出することにより測定され得る。野生型、tox-、および変異した遺伝子に対するこれら配列の特異性は、Genetics Computer Group(Madison, WI)のコンピュータープログラムWord SearchまたはFASTAを用いて(これらは問題の遺伝子に対する類似性について目録に載っているヌクレオチド配列をサーチする)、コンピュータ化されたデータベースである、GenBankの目録に載っている既知の配列とコンピューター比較を行うことにより決定され得る。
H.pylori 60190(ATCC 49503)(これから当初空胞形成細胞毒素が精製された(15))を用いて空胞形成細胞毒素の遺伝子をクローン化した。H.pylori 84-183(ATCC 53726)および87-199は、空胞形成細胞毒素を産生することが先に示された良く特徴付けられた株であり(8、9、16)、そしてTx30a、86-313、および87-203株は、検出可能なインビトロでの細胞毒素活性を産生しない野生型株である(7-9、16)。26株のヒト由来のさらなる臨床H.pylori単離株(先に記載されている(9))もまた、細胞毒素遺伝子の保存を評価するために使用した。H.pylori単離株は、5%ヒツジ血液を含むトリプチケースソイ寒天平板上で、CampyPak-Plus(BBL、Cockeysville、MD)により生成される微好気性雰囲気下、37℃で48時間培養した。
H.pylori 60190由来の87 kDa空胞形成細胞毒素を、先に記載(15)のように、ブロス培養上清から精製し、7%アクリルアミドゲル上で電気泳動し、そしてProBlott膜(Applied Biosystems、Foster City、CA)上にエレクトロブロットした(15)。次いで、87kDaタンパク質をArg-Cプロテアーゼで消化し、フラグメントをクロマトグラフィーにより分離し、そして3つのフラグメントのアミノ酸配列を、Fernandezら(17)に記載のように、Microsequencing Laboratory of Rockefeller University、New York、NYで決定した。
染色体DNAを単離するために、H.pylori細胞をGES(60%グアニジウムチオシアネート−0.1 M EDTA−0.5%Sarkosyl)中で溶解し、そしてDNAをクロロホルム抽出およびイソプロパノール沈殿により精製した(18)。H.pylori 60190染色体DNAに、音波処理による剪断またはAluIでの部分消化のいずれかを行った;次いで、先に記載(9)のように、λZapII(Stratagene、La Jolla、CA)を用いてゲノムライブラリーを構築した。ライブラリーを、ランダムヘキサマー(United States Biochemical、Cleveland、Ohio)を用いたプライマー伸長により放射性標識したプローブを用いて、プラークハイブリダイゼーションによりスクリーニングした(19)。精製した反応性クローンから、クローン化されたDNA挿入片を含むpBluescriptを、R408ヘルパーファージとの同時感染により切り出し、そしてE.coli XL1-Blue中にサブクローン化した。ブラスミド精製の後、制限酵素切断マップを生成し、そしてジデオキシ鎖停止法(20)を用いて両鎖についてヌクレオチド配列を決定した。60190株由来のvacAの最終ヌクレオチド配列を、PCRフラグメントよりむしろクローン化ゲノムDNAから完全に決定した。推定のプロモーターおよびShine-Dalgarno配列を、コンセンサス配列(21)との比較により同定した。相同タンパク質についてのデータベースサーチを、FastAおよびFastDBプログラム、およびNational Center for Biotechnology InformationのBLASTネットワークサービスを用いて実施した。
縮重オリゴヌクレオチドプライマー[(5' TTYTTYACNACNGTNATHAT 3')(配列番号17)および(5' TTRTTDATYTCNARRAARTTRTC 3')(配列番号18)]を、精製87kDaタンパク質のN末端および実験的に決定されたペプチド配列(アミノ酸残基35-41および198-205、それぞれ、配列番号1および2)の逆翻訳を基に構築し、H.pylori 60190 DNAから0.5 kbバンドをPCR増幅するために用いた。このPCR産物を1%アガロースゲルから切り出し、Qiaexゲル抽出キット(Qiagen、Chatsworth、CA)を用いて精製し、そしてpT7 Blue T-ベクターキット(Novagen、Madison、WI)を用いて、E.coli Nova Blue細胞中にサブクローン化しpCTB1を生成した。pCTB1のヌクレオチド配列は、配列番号1の塩基203〜714に対応した。
vacAの翻訳されたアミノ酸配列およびヌクレオチド配列、ならびに上流配列を、PIRおよびSwiss-Protデータベース中の配列と比較した。最も強い相同性が、vacAの上流に位置する567bp以上のORFと、E.coli由来のシステイン-tRNAシンセターゼ(cysS)との間に存在した(2つの50アミノ酸領域中40%および44%同一性)(26、27)。vacA遺伝子産物と有意な相同性を有するタンパク質は存在しなかった。しかし、関連のC末端モチーフおよび類似のC末端プロセッシングを有するいくつかのタンパク質が同定された。これらは、Haemophilus influenzae(28)およびNeisseria gonorrhoeae(29)由来のIgAプロテアーゼ、Serratia marcescensセリンプロテアーゼ(30〜32)、Rickettsia rickettsiiの120kDa表面露出タンパク質(OmpB)(33、34)、Rickettsia prowazekiiの120kDa表面層タンパク質(35)、および腸病原性E.coli(EPEC)のAIDA-Iアドヘシン(adhesin)(36)を含む。これら遺伝子の各々は、サイズ30〜60kDaのペプチドのC末端切断を受ける大きなタンパク質をコードする(28-36)。C末端1位のフェニルアラニンで開始する、交互する疎水性アミノ酸モチーフがこれらタンパク質の各々に存在する。このC末端セグメントは、一般に、グラム陰性外膜タンパク質に見出される(37)。これらのタンパク質は、低システイン含量(タンパク質あたり2〜4のシステイン)によりすべて特徴付けられ、これは膜輸送の間のジスルフィド結合形成を最小にする必要性に多分関係する(38、39)。しかし、翻訳されたVacAタンパク質中および3つの関連プロテアーゼ中では、2つのシステイン残基が、わずか7〜11アミノ酸離れて存在する。S.marcescensプロテアーゼでは、対のシステイン残基のセリンによる置換は減少した酵素分泌を伴う(31)。対応するシステイン残基はまた、H.pylori vacA遺伝子産物の分泌において所定の役割を演じ得る。
vacA遺伝子が単一コピーまたは多コピーのいずれで存在するかを決定するために、H.pylori 60190由来のゲノムDNAを調製し、そしてpCTB1またはpCTB4中の挿入片をプローブとして用いてサザンハイブリダイゼーションを行った。両方のプローブが約7kbの単一のBglIIフラグメントにハイブリダイズした(データは示さず)。pCTB4挿入片は、単一の1.7kb HindIIIフラグメントにハイブリダイズし、そしてマッピング研究から推定されるように、pCTB1挿入片は2つのHindIIIフラグメントにハイブリダイズした。5つのさらなる酵素(SacI、EcoRV、EcoRI、BamHI、およびKpnI)により消化したDNAについては、プローブは12kb以上の単一フラグメントとハイブリダイズした。これらの結果は、60190株では単一コピーのvacAのみが存在していること示唆し、そしてpCTB1をvacAプローブとして用いたBukanovおよびBergの物理的-遺伝的マッピング研究(40)と一致する。
対立遺伝子交換によりvacAが形質転換された株で破壊されたか否かを決定するために、野生型84-183株およびカナマイシン耐性H.pylori変異株84-183:v1から単離されたDNAをHindIIIで消化し、そしてサザンハイブリダイゼーションを、カナマイシン遺伝子またはpCTB8のいずれかをプローブとして用いて実施した。H.pylori染色体DNAの制限エンドヌクレアーゼ消化の後、標準化した量のフラグメントを、0.04M トリス酢酸-2mM EDTA緩衝液(pH8.2)中0.7%アガロースゲル上で電気泳動した。ナイロン膜への転移、放射性標識したプローブを用いたハイブリダイゼーション、および洗浄は先に記載(9)のように行った。H.pylori株のコロニーブロットハイブリダイゼーションは先に記載(9)のように行い;ハイブリダイゼーションは6×SSC中68℃で18時間であり、次いで65℃で0.5×SSCで洗浄した。
vacA遺伝子の破壊が87kDaタンパク質の産生を廃止するか否かを決定するために、野生型株84-183および60190由来の培養上清および2つの同質遺伝子型変異体84-183:v1および60190:v1を、抗-87kDa血清(15)を用いてイムノブロットした。期待されたように、両方の野生型株由来の上清は、免疫反応性の87kDaバンドを含んでいた。その一方、このバンドは、変異株の上清には存在しなかった。2つの野生型株および2つの同質遺伝子型変異株由来の濃縮培養上清を、組織培養アッセイでの空胞形成細胞毒素活性について試験した。
H.pylori野生型株および変異体を5%ウシ胎児血清を含むBrucellaブロス中で培養し、そして濃縮した培養上清を限外濾過により調製した(8、16)。HeLa細胞を、10%ウシ胎児血清および10mM塩化アンモニウムを含むEagleの改変最少必須培地中(8、16)で培養した。タンパク質濃度により標準化したH.pylori上清の連続希釈液を、HeLa細胞と18時間インキュベートし、次いで細胞空胞形成を、先に記載(16、25)のように、ニュートラルレッド取り込みアッセイにより定量化した。天然に存在するtox-株Tx30a由来の上清をコントロールとして試験した。
インビトロで細胞毒素活性を発現するH.pylori株(tox+)および細胞毒素活性を発現しない野生型株(tox-)中にvacA配列が存在するかどうかを調査するために、15のtox+株および17のtox-株を、コロニーハイブリダイゼーションによりpCTB1をプローブとして用いて調べた。H.pylori株の各々は強くハイブリダイズしたが、その一方E.coli XL1Blueとのハイブリダイゼーションはなかった。
tox+およびtox- H.pylori株の両方に存在するvacA配列をさらに調査するために、遺伝子の3つの異なる領域からのフラグメントのPCR増幅を行った。3つのtox+ H.pylori株(84-183、60190、87-199)および3つのtox- H.pylori株(87-203、86-313、およびTx30a)由来のDNAを、vacA遺伝子フラグメント増幅のテンプレートとして用いた。PCR反応は30サイクル行った。条件は3つの増幅について同じである(温度94℃、50℃、および72℃)。
vacA遺伝子における潜在的な配列多様性をさらに調査するために、tox-株からの、推定の可変性領域を含んだvacA遺伝子のフラグメントのPCR増幅を試みた。1.5kbフラグメントを、bp1012〜1029および2533〜2549(配列番号1)に対応する、プライマー[(5' TAGTAACAAGACTCATAT 3')](配列番号9)および(5' CGTTAGCCGTTTTACTG 3')(配列番号10)を用いて、tox-株87-203から増幅した。
