実施例2 . 脂肪酸酸化酵素

Example 4 . Bleaching of paper with fatty acid oxidase
Unbleached kraft pulp obtained from Eucalyptus grandis was used. The pulp was repulped at a concentration of 4% on a Loretzen and Wettre pulp machine. Repulping was performed in buffer (Britton-Robinson) at pH = 9.0.

100 mM phosphoric acid (85%) 6.28 mL
100 mM acetic acid (100%) 5.72 mL

The pH was adjusted to 9.0 by addition of sodium hydroxide.
After repulping, the pulp slurry was diluted with buffer to 1% and the pH was readjusted to pH = 9.0.

The treatment with fatty acid oxidase was carried out in a beaker containing 3 g of dry pulp, ie 300 mL pulp slurry. Treatment was carried out at 25 ° C. in a water bath with stirring at 500 rpm with a magnetic stir bar. 333 ppm of linoleic acid was added to all beakers. The fatty acid oxidase used was purified lipoxygenase derived from Gaeumannomyces graminis prepared as described above. The amount of enzyme used is shown in Table 4 and Table 5 below. Enzyme treatment was performed for 2 hours. Two beakers were tested under each condition.
After 2 hours, the enzyme experiment was stopped by adding 5 mL (fixed amount) of NaOH (27.65% solution). This addition raises the pH above 12 and inactivates the enzyme.

The contents of the beaker were quantitatively transferred to a 1000 mL beaker using 700 mL deionized water. This pulp suspension was poured onto a Buchner funnel (15 cm diameter) fitted with filter paper. Water was aspirated to form a sheet of paper. The paper sheet was removed from the funnel and separated from the filter paper. The sheet was squeezed with a sheet press machine manufactured by Lorentzen and Wettre. The sheet was pressed into a metal plate, two blotter papers, two filter papers, a sheet, two filter papers, two blotter papers and a metal plate sandwich at 0.4 MPa for 5.5 minutes. The wet paper was replaced with dry paper, and pressing was repeated at 0.4 MPa for 2 minutes. The sheet was air dried overnight.

The brightness of the sheet was measured using a Macbeth Color-Eye 7000 reflectometer. Brightness was recorded at 600 nm. Four measurements were made for each sheet. The results obtained are shown in Table 4 below.
The Kappa number, which indicates the delignification degree of the pulp, was also measured for each sheet by the method described in Tappi test method T236 (Tappi Press). The amount used for each measurement was 1/4 of the amount described in the standard method. The dry matter content of the sheet was measured to calculate the Kappa number. The results obtained are shown in Table 5 below.

Example 5 . Deinking old newspaper using fatty acid oxidase
200 g of shredded old newspaper was placed in a Hobart mixer with 1500 mL of water. The water bath temperature was set at 45 ° C. Stir at low speed for about 0.5-1 minutes. Next, 3.6 kg / ton (7 lb / ton) surfactant and 2 mg (10 mg / ton pulp) lipoxygenase from Magnaporthe salvinii prepared as described above were added to the mixer, An additional 500 mL of water was added to the Hobart and mixed well.
