Description

The present invention relates to a skin function improving agent and a skin function improving food for improving skin function.

Conventionally, for improving symptoms such as rough skin and skin diseases, sebum components such as squalane and ceramide, moisturizing components using fats and oils such as vitamin A oil, jojoba oil and pearl oil, creams and ointments having skin conditioning components Anti-inflammatory agents, steroids, and the like are used to improve pathological symptoms such as inflammation using external preparations such as lotions.

However, external preparations comprising sebum components and fats and oils do not improve the skin function itself such as the ability to regenerate the skin, and cannot be a fundamental solution.

There is also a theory that the increase in the number of patients with unknown skin inflammatory diseases and atopic dermatitis in modern society is the effect of social stress. However, the topical skin preparations currently in use are not very effective for rough skin and skin diseases caused by stress, which is common in modern people, as long as the underlying stress is not improved even if temporary depression is suppressed. There was a problem.

Therefore, in view of the above-described problems of the prior art, the present invention can improve skin function from the body, and can improve and prevent symptoms such as rough skin and skin diseases including skin malfunction due to stress, and has side effects. Next, the skin function improving agent and skin function improving food according to the present invention will be described.

Production Example As raw material mushrooms, eringi (Pleurotus eryngii), maitake (Grifola frontosa), beech shimeji (Hypzygus marmoreus), white beech shimeji (Hypyzygus marmoreus) are used, and raw lye is used.

The dry powder is dried by any one of freeze drying, sun drying, heat drying, and hot air drying.

First, water (distilled water, purified water, ion-exchanged water, tap water or natural water) of 10 to 20 times the weight of dry powder and 1 to 2 times the weight of raw material to this raw mushroom. To extract the active ingredients.

At this time, by extracting with hot water by heating, the extraction efficiency can be improved and the extraction time can be shortened. The extraction time under reflux with heating is preferably 20 to 60 minutes.

Then, the residue is removed by filtration, and the extract is freeze-dried and powdered to produce a mushroom water extract.

In addition, α-amylase (Termamyl 120L: Novo Nordisk) is added to the extract and incubated until the starch in the solution is sufficiently decomposed, and then ethanol is gradually added until the ethanol concentration reaches 50%. By leaving it in a cold place for 30 minutes to 10 hours, a precipitate is formed in the extract, and this precipitate is separated from the supernatant by filtration or centrifugation, and washed with 50% or more of ethanol as necessary.

Furthermore, by removing the protein from the precipitate by the Sevag method, a mushroom extract containing water-soluble β-glucan (water-soluble β-glucan derived from mushroom) as a main component can be obtained.

In addition to the Seavg method, other general protein removal methods such as precipitation with ammonium sulfate can be used as the method for deproteinization.

Then, one or more excipients, binders, disintegrants, lubricants, emulsifiers, stabilizers, colorants, fragrances and the like are added to the water-soluble β-glucan derived from this mushroom water extract or mushroom, Depending on the method, various forms of mushroom water extracts such as beverages, tea preparations, capsules, tablets and the like, or skin function improving agents based on mushroom-derived water-soluble β-glucan can be obtained.

Further, the above-mentioned mushroom water extract or water-soluble β-glucan derived from mushrooms may be used as it is, or may be a powdery skin function improving agent, for example, so that it can be easily added to cooking rice or tea at home. A skin function improving agent obtained by powdering a mushroom water extract or a water-soluble β-glucan derived from a mushroom may be packed in a small pack.

Moreover, it can be set as the skin function improvement food which has a skin function improvement effect by adding this water-soluble beta-glucan derived from a mushroom water extract or a mushroom to various processed foods.

In particular, mushroom-derived water-soluble β-glucan is an almost tasteless and odorless solid and easily dissolved in water, so that it can be easily added without changing the taste of food.

This mushroom water extract and water-soluble β-glucan derived from mushrooms are, for example, snacks made from bread, donuts, cakes, buns, biscuits, cookies, candy, caramel, gum, arabe, rice crackers, potatoes and corn powder. , Juice, soft drink, jam, jelly, agar, yogurt, soy sauce, sauce, ketchup, tofu, vegetable and fruit puree, pasta, udon, buckwheat, ramen noodles, soup, miso soup, curry, stew, sprinkle, retort It can be easily added to processed foods such as pouch foods in general.

In addition, as for the standard of daily intake, it is desirable to take 3 g of mushroom-derived water-soluble β-glucan and 20 g or more of mushroom water extract powder.

Test example 1
A hairless mouse (female, 6 weeks old) whose skin surface is relatively close to humans is used as a subject, and the subject is stressed by an overloading environmental load that is considered to be close to human social mental stress. A comparison is made between the case of administering the skin function improving agent and the case of not administering it.
〔Test method〕
After putting 5 subjects in 1 cage (26.5cm x 15.5cm x 12.5cm), rearing them for 6 weeks and acclimatizing them, the normal rearing group, the target group, and each mushroom intake group as follows Divide and compare each test group.

Test group Normal breeding group: Normal breeding, normal feed intake Target group: Overcrowded environment breeding, normal feed intake Mushroom intake group: Overcrowded environment breeding, mushroom or mushroom water extract mixed feed Specimen is bred and raised (ordinary breeding) and fed with normal feed.

In the subject group, five subjects are placed in one section of one cage divided into four equal parts, and a normal feed is ingested while stressing the subjects, and the animals are raised in an overcrowded environment for 7 days.

In each mushroom intake group, as in the subject group, 5 subjects were placed in 1 section divided into 4 equal parts, the subjects were stressed, and the skin function improving agent was added to the weight of the dried mushroom powder. The mixture was mixed with the powdered feed so that it would be 5% of the total weight of the feed, and was kept in an overcrowded environment for 7 days while allowing the subject to ingest freely. The skin function-improving agent was prepared by adding 1.5 times the weight of water to each of raw eringi, maitake, bunashimeji and white bunashimeji, heating to the boil, extracting for 30 minutes, and then removing the residue by filtration. It is a mushroom water extract powder produced by freeze-drying the liquid.

Evaluation of skin function uses epidermal blood flow and skin barrier function regeneration rate as indices.

For epidermal blood flow, the blood flow behind the animal is measured using a laser Doppler meter (PIM2; Lisca) 7 days after the start of mushroom administration.

The skin barrier function regeneration rate was caused by repeated tape stripping (the act of applying a tape to the skin and removing it) on a part of the back of the subject 7 days after the start of mushroom administration. The water transpiration amount (TEWL) is measured for 4 consecutive days using a water transpiration amount measuring device (Tewameter; Curage + Khazaka) and obtained from the value.

It should be noted that epidermal blood flow and transepidermal water transpiration (TEWL) are effective indicators for studying the effectiveness of cosmetics and topical skin drugs. An increase in epidermal blood flow can be expected to activate skin metabolism, and is also related to cosmetic effects such as skin color and blood circulation promotion. TEWL increases when the barrier function of the epidermis is destroyed due to inflammation or the like, and decreases when this is recovered.

This time, the state immediately after the inflammation induction (TEWL is higher than usual) was calculated as 0% skin barrier function regeneration rate, and the state returned to the normal value before the inflammation induction was calculated as 100% skin barrier function regeneration rate.
〔Test results〕
The results regarding epidermal blood flow are shown in Table 1 and FIG. 1, and the results regarding the skin barrier function regeneration rate are shown in Table 2 and FIG. In addition, about the skin barrier function reproduction | regeneration rate, the 3rd day after inflammation induction was compared.

(A) Compared to normal breeding, (b) In the target group (overcrowded breeding), both epidermal blood flow rate and skin barrier function regeneration rate decreased, while (c) to (f) (b) ) Compared to the target group (overcrowded breeding), both the epidermal blood flow rate and the skin barrier function regeneration rate are high, and it can be confirmed that the reduction of these skin functions is improved, especially (c) eringi, (d) mitake, (F) As for the epidermal blood flow of white beech swordfish, (a) all exceed normal breeding.

Test example 2
0.1 ml of α-amylase is added per 100 ml of the same mushroom mushroom water extract used in the evaluation test conducted in Test Example 1 above, and reacted in a boiling bath for 30 minutes. Gradually add to 50% and let stand at room temperature for 2 hours. The resulting precipitate is separated from the supernatant by centrifugation, the protein is removed from this precipitate by the Sevag method, and water-soluble β-glucan is the main component. A mushroom extract was obtained. A similar skin function improvement test was conducted by giving this to a subject (mouse) instead of the mushroom water extract of Test Example 1 described above.

As a result, it was confirmed that the water-soluble β-glucan had the same skin function improving effect as the mushroom water extract.

Test example 3
〔Test method〕
6 genders in their twenties dissolved 1 g of skin function improver per 1 cup of soup (about 150 ml) and continued to drink twice a day in the morning for 7 days from the night of the experiment to the morning of the last day of the experiment. The changes in epidermal blood flow were investigated.

In addition, the skin function improving agent used was added with 1.5 times weight of water to raw maitake, heated to boiling, extracted for 30 minutes, and then the residue was filtered to remove ethanol in the final concentration. Is gradually added so as to be 50%, allowed to stand at room temperature for 2 hours, and then a mushroom extract fractionated into a precipitate and a supernatant by centrifugation is used.

The measurement of epidermal blood flow was carried out using a laser Doppler meter (PIM2: Lisca) at 10:00 am on the start date and the final day of the experiment.
〔Test results〕
The results are shown in Table 3 and FIG.
