Abstract
【解決手段】 a)被験対象から採取した細胞からゲノムを抽出する段階、b)APOA2遺伝子を増幅して核酸断片を得る段階、およびc)b)で得られた核酸断片中のAPOA2遺伝子のTG反復回数を計測する段階、の各段階により被験対象のAPOA2の遺伝子型を判定する。TG反復回数をもとにしたスコアが高いほど、散発性高齢発症型アルツハイマー症の発症年齢が低いと予測される。

Description
また、脂質代謝と散発性LOAD発症年齢との関連性についての知見はあるものの、脂質代謝は被験者の栄養状態に左右されることから、上記散発性LOAD発症年齢を予測することは困難である。このため、簡便でかつ、単純な解析で、上記散発性LOAD発症年齢を予測する方法の開発が強く求められていた。
散発性高齢発症型アルツハイマー病(散発性LOAD)とは、主に60歳以降に発症するアルツハイマー病で、1ないし2親等内にアルツハイマー病発症者が認められない場合を指す。本発明における「散発性高齢発症型アルツハイマー病」または「散発性LOAD」は、NINCDS−ADRDAによるアルツハイマー病の診断基凖に該当する疾患の中で、家族性発症がなく、60歳以上で発症する疾患と定義される。
表1に示すように、GT反復回数の範囲により対立遺伝子スコアA2を規定した。

Claims (12)
- 被験対象から採取した細胞のゲノム中のAPOA2の遺伝子型を判定することを特徴とする散発性高齢発症型アルツハイマー病の発症年齢の予測方法。
- 両対立遺伝子におけるAPOA2の遺伝子におけるGT反復回数の合計が多いほど散発性高齢発症型アルツハイマー病の発症年齢が低いと予測する、請求項1に記載の予測方法。
- 対立遺伝子におけるAPOA2の遺伝子におけるGT反復回数の範囲によってスコアを規定することを特徴とする請求項1または請求項2に記載の予測方法。
- 両対立遺伝子におけるそれぞれの上記スコアの合計が高いほど散発性高齢発症型アルツハイマー病の発症年齢が低いと予測することを特徴とする請求項1ないし請求項3記載の予測方法。
- 被験対象がAPOE−ε4遺伝子保有者であることを特徴とする請求項1ないし請求項4記載の予測方法。
- PCR法により遺伝子を増幅させることを含む、請求項1ないし請求項5に記載の予測方法。
- 遺伝子型の判定が電気泳動度の比較により実行される、請求項1ないし請求項6のいずれかに記載の予測方法。
- 散発性高齢発症型アルツハイマー病の発症リスクの予測と組合せられる、請求項1ないし請求項7のいずれかに記載の予測方法。
- 散発性高齢発症型アルツハイマー病の発症年齢を予測するためのデータを収集する方法であって、被験対象から採取した細胞のゲノム中のAPOA2の遺伝子型を判定することを特徴とする方法。
- 細胞が血液から採取されることを特徴とする請求項9記載の方法。
- PCR法により遺伝子を増幅させることを含む、請求項9または請求項10に記載の方法。
- 遺伝子型の判定が電気泳動度の比較により実行される、請求項9ないし請求項11に記載の方法。
