本発明は、マイクロプレートのウェルなどの液体保持部の洗浄に使用される液体吸引装置および液体吐出装置に関するものである。

【従来の技術】
創薬スクリーニングなどにおいて行われる物質の生化学的反応の試験には、マイクロプレートなど液体試料を保持するための容器が用いられる。これらの試験において抗原・抗体反応を利用した試験の場合には、試験結果を評価するための観察に先立って、未反応の余剰試料除去のための洗浄が必要とされる。この洗浄は、抗原・抗体反応が進行した状態の液体試料を保持したマイクロプレートのウェル内に、洗浄用の液体を吐出、吸引することにより行われる。

この洗浄操作は、吐出、吸引の専用ヘッドを備えた専用の洗浄装置によって行われるが、この洗浄操作を効率よく行うことを目的として、マイクロプレートを移動させることなく同一位置にて液体の吐出・吸引を行う方式の洗浄装置が提案されている。このような洗浄装置には、液体吐出用の吐出ノズルと吸引用の吸引ノズルを同一ヘッドに並設する構成のもの(例えば特許文献1参照)や、さらには吐出ノズルと吸引ノズルを同軸の2重管に組み込んだもの(例えば特許文献2参照)などがある。

【発明が解決しようとする課題】
マイクロプレートなどのように試料の液体を保持するウェルが格子配列で多数設けられた容器を対象とする場合には、洗浄液の吐出ヘッド、吸引ヘッドとして吐出用や吸引用のノズルがウェルと同一配列で設けられたマルチノズル型のノズルヘッドが用いられる。この方式のノズルヘッドは、ヘッド本体内に設けられた複数の分岐流路に多数のノズルが装着される。

ところが、このようなマルチノズル型のノズルヘッドには、ヘッド内部における流路構造によっては、吸引停止時の液滴落下などの不具合が生じており、これら不具合によって安定した洗浄操作が妨げられる場合があった。

そこで本発明は、安定した洗浄操作を効率よく実行することができる液体吸引装置および液体吐出装置を提供することを目的とする。

請求項1記載の液体吸引装置は、吸引手段に接続された水平な流路に複数の管状のノズルを装着し、これらのノズルによって液体を吸引する液体吸引装置であって、前記複数のノズルを下方から前記流路に挿入するとともに、挿入されたノズルの上端部をこの流路内に突出させた。

まず図1を参照してマイクロプレートの洗浄処理に使用されるマルチノズル型洗浄装置の構造を説明する。このマルチノズル型洗浄装置はマイクロプレートに所定配列で配置された複数の液体保持部であるウェルに洗浄液を吐出し、また吐出された洗浄液を吸引することにより、これらの複数のウェルを同時に洗浄する機能を有している。本実施の形態では、抗原・抗体反応を利用した生化学試験において、試験結果の観察に先立って未反応の余剰試料を除去するための洗浄に使用する例を示している。

図1においてマルチノズル型洗浄装置は、洗浄の対象となるマイクロプレート3を保持するプレート保持部1の上方に、洗浄ヘッド5を昇降機構4によって昇降自在に配設した構造となっている。洗浄ヘッド5は、吸引ヘッド8、吐出ヘッド9を上下に重ね合わせた構成となっている。プレート保持部1はマイクロプレート3を載置して保持するプレート載置テーブル2を備えており、プレート保持部1に内蔵された水平移動機構(図示省略)により、プレート載置テーブル2上に載置されたマイクロプレート3の水平方向の位置を調整できるようになっている。プレート保持部1の上部は、図2に示すように、洗浄時にマイクロプレート3からオーバフローした洗浄液を受けるための液溜め部1aとなっている。

次に図3〜図7を参照して洗浄ヘッド5の詳細構造を説明する。図3は、吸引ヘッド8、吐出ヘッド9をそれぞれクランプ部7A、7Bから取り外した状態を示している。吸引ヘッド8、吐出ヘッド9はいずれも矩形のプレート状部品であり、クランプ部7A、7Bに嵌合形状で設けられた装着枠に着脱自在となっている。吸引ヘッド8はクランプねじ15によってクランプ部7Aに固定される。

次に図4、図5を参照して、吸引ヘッド8の内部構造を説明する。上部プレート13には、複数のX溝部13aがX方向に等間隔で設けられており、X溝部13aの周囲にはシール部材13dが装着されている。シール部材13dは、上部プレート13と下部プレート14とを締結孔13bによって締結した状態で、X溝部13aを外部から気密に保つ。各列のX溝部13aの中央部には連通溝13cがY方向に設けられており、これにより複数のX溝部13aが相互に連通する。

ノズルプレート19には、第1の側プレート18A、第2の側プレート18Bが、ノズルプレート19を両側から挟み込んで結合されている。第1の側プレート18A、第2の側プレート18Bには、それぞれY方向に連通した第1のY流路23A、第2のY流路23Bが設けられており、液分配孔19c、X流路孔19dに対応した位置にはそれぞれ連通口24a、供給口24bが開口されている。これにより、第1のY流路23A、第2のY流路23Bは液分配孔19c、X流路孔19dに連通し、X流路孔19dには両側の供給口24bを介して洗浄液が供給される。すなわち吐出ヘッド9は、X流路孔19dに洗浄液を供給する供給口24bを相互に隔てた2箇所に設けた構成となっている。

図6に示すように、ノズルプレート19には吸引流路19eがノズル範囲Nの中間部に位置してY方向に設けられている。吸引流路19eの厚み方向の位置はX流路孔19dと部分的に相貫するように設定されており、この相貫部分は吸引流路19eとX流路孔19dとを相互に連通させる開口19fとなっている。吸引流路19eはノズルプレート19のY端面に開口しており、吸引流路19eと連通してジョイント部21が取り付けられている。ジョイント部21は吸引機構12と接続されており、ジョイント部21を介して吸引機構12から吸引することにより、開口19fを介して各X流路孔19dが吸引される。これにより、洗浄液吐出を反復する過程で流路内に侵入して停留した気泡を除去することができる。

次に、吐出ヘッド9内における洗浄液の流動経路について説明する。図6において、ジョイント部20を経て洗浄液供給装置11から供給される洗浄液は、液供給孔19bから液分配孔19cに流入する際に左右方向に分かれ、両側の連通口24aを介して第1のY流路23A、第2のY流路23B内に流入する。そして流入した洗浄液は、第1のY流路23A、第2のY流路23B内をY方向に流動する途中で、各供給口24bからそれぞれのX流路孔19d内に両側から流入する。そして両側からX流路孔19dに流入した洗浄液は、列状に配列された各吐出ノズル22から吐出される。

ノズルプレート19の下面側には摩擦係数が小さい樹脂材質より成るガイドプレート25が貼着されており、ガイドプレート25には開口部19aの位置に対応して吸引ノズル17が挿通可能な摺動孔25aが設けられている。この摺動孔25aは吸引ノズル17が吐出ヘッド9を貫通して上下動する際のガイドとして機能する。

そして吐出ノズル22は、吸引ノズル17に対して鋭角の交叉角度αで交叉し、その下端部の吐出口22aは、吸引ノズル17の下端部の吸引口17aよりも上方且つ吸引ノズル17の外側面の近傍に位置する配置となっている。吐出口22aの位置は、吐出口22aに形成される洗浄液の液滴が吸引ノズル17の外側面に接触可能な位置である。そして吐出ノズル22と吸引ノズル17の交叉角度αは、後述するように吐出口22aから吐出された洗浄液をウェル3aの内側面に衝突させ得るような角度に設定されている。

ウェル3aの底面には、評価の対象となる細胞等の評価対象物が付着しているが、吐出ノズル22から吐出する洗浄液の勢いで評価対象物を剥離させてしまう危険がある。本実施の形態では吐出ノズル22から吐出する洗浄液をウェル3aの内側面に衝突させて洗浄液の勢いを弱めるので、評価対象物をウェル3a底面から剥離させることなく洗浄することができる。

次に図9を参照して、マルチノズル型洗浄装置によるウェル3aの洗浄動作について説明する。まずマイクロプレート3を位置合わせして洗浄対象のウェル3aを吸引ノズル17、吐出ノズル22の直下に位置させる。次いで吸引ヘッド8、吐出ヘッド9をともに下降させ、吸引ノズル17、吐出ノズル22の下端部をウェル3aの開口端近傍に位置させる。そして図9(a)に示すように、吸引ノズル17と所定の交叉角度α(図8参照)で交叉した姿勢の吐出ノズル22の吐出口22aから洗浄液26を吐出させる。そして吐出された洗浄液は、ウェル3aの内側面に衝突して洗浄しながら内側面に沿ってウェル3a内に注入される。

所定量の洗浄液26を注入したならば、図9(b)に示すように、吸引ノズル17を下降させて下端部をウェル3aの底面近傍に位置させる。そしてこの状態で、吸引ノズル17から洗浄液26を吸引し、外部に排出する。図9(a)の吐出動作と図9(b)の吸引動作を複数回繰返すことによりウェル3a内に残る余剰試料等を除去する。この後、ウェル3a内における洗浄液26の残液量が減少したならば、図9(c)に示すように、マイクロプレート3を移動させてウェル3aの底面のコーナ部に吸引ノズル17の下端部を位置させる。これにより、残液が生じやすいウェル内のコーナ部から洗浄液26の残量を吸引して排出することができる。

次に図10を参照して、吐出ノズル22から液体を吐出する過程において実行される液滴吸引動作について説明する。吐出ノズル22からの液体吐出においては、洗浄液供給装置11を停止して洗浄液の供給を停止した後にも、吐出ノズル22の下端部22aから幾分かの液体が図10(a)に示すように滴下し、ウェル3a内に落下する現象が生じる。このような液滴26aの滴下は試験結果の誤差を招く場合があるため、極力排除することが望ましい。

このため、本実施の形態では、以下に示すような方法で液滴吸引を行うようにしている。すなわち、吐出ノズル22と吸引ノズル17の相互配置においては、前述のように吐出口22aと吸引孔17aとを極力接近させて配置し、図8に示すような配置としていることから、図10(b)に示すように、吐出口22aから滴下する液滴26aは吸引ノズル17の外側面に接触する。そしてこの状態で吸引ノズル17からの吸引を行うことにより、吸引ノズル17の外側面を伝って下降する液滴26aを、図10(c)に示すように吸引ノズル17によって吸引することができ、液滴26aをウェル3a内に落下させることなく排出することができる。

上記説明したように、本実施の形態に示す液体吸引装置では、ウェル3a内を吸引ノズル17によって吸引する吸引ヘッド8の内部構造において、複数の吸引ノズル17をX溝部13aに装着する形態を、複数の吸引ノズル17を下方からX溝部13aに挿入するとともに、挿入された吸引ノズルの上端部をこのX溝部13a内に突出させるようにしている。これにより、吸引ノズル17の上端部は吸引動作においてX溝部13a内に吸引された液体の液面から上側に突出し、したがってX溝部13a内に残留する液体が吸引ノズル17を逆流して吸引ノズル17の下端部から液滴となって落下する「液垂れ」を防止することができる。

また、本実施の形態に示す液体吐出装置では、洗浄液供給装置11による液体供給停止後に、各X流路孔19dの中間部に接続された吸引流路19eから吸引することにより、吐出ヘッド9内に残留する液体を吸引して排出することができる。これにより、液体供給停止後になお吐出ノズル22の吐出口22aから余分に吐出される液体が液滴となって落下する「液垂れ」を防止することができる。

【発明の効果】
本発明によれば、液体保持部内をノズルによって吸引する吸引ヘッドの内部構造において、複数のノズルを下方から流路に挿入するとともに、挿入されたノズルの端部をこの流路内に突出させる構造を採用したので、吸引停止時に流路内の洗浄液がノズルに流入することを防止し、安定した洗浄操作を実現することができる。
