炭酸ガスを外用すれば、経皮浸透および/または経皮吸収された炭酸ガスが皮膚の血行を促進して新陳代謝を活性化する。このことによって医療・美容効果が得られることは公知(WO99/24043、WO 02/080941、特開2000−319187)であり、既に用時調製型のこの種製品は市販されている。

角質細胞層の薄い粘膜では、炭酸ガスの経皮浸透および/または経皮吸収効率がよく医療・美容効果が得られやすい。しかしながら粘膜に比べて角質細胞層の厚い健常皮膚や、老化および乾燥などによって落屑が遅延して、本来はがれ落ちるべき古い角質細胞が皮表に堆積した皮膚では炭酸ガスの経皮浸透および/または経皮吸収は抑制・阻害されて効果が減少するのは自明である。

上記の問題解決のためには、皮表に堆積した細胞を剥離させる目的でグリコール酸や乳酸などのα−ヒドロキシ酸やサリチル酸、レゾルシンなどの利用が考えられるが、所期の目的を得るためには高濃度配合する必要があり刺激性やアレルギー性が問題になる。

そこで、本発明者等は皮膚に常在するある種のプロテアーゼが角質細胞を接着している蛋白質・デスモソームを分解することによって落屑がおこっていることに注目した。植物や微生物基原のプロテアーゼを配合し、落屑を促進して角質細胞層を薄くすることにより、炭酸ガスの経皮浸透・吸収を高めるのが合目的性であると考えた。なお、プロテアーゼは作用がマイルドであり、肌に負担をかけることなく角質細胞を取り除くことができるメリットをもつため敏感肌女性もまた利用できる。

炭酸ガス発生化粧品の効果向上の目的でプロテアーゼを配合し、かつその活性増進剤を探求して、より一層効果の高い炭酸ガス発生化粧品を提供する。

【課題を解決するための手段】
本発明者等は炭酸ガス発生系において、プロテアーゼに対する酸成分の挙動について鋭意研究の結果、酸性リン酸カルシウム塩は炭酸ガス発生化粧品の酸成分としての効用だけでなく、併用プロテアーゼの活性増進作用を併せもつことを初めて明らかにして本発明を完成した。

以下、炭酸ガス発生系におけるプロテアーゼの活性増進効果について述べる。
(枯草菌プロテアーゼの活性増進)
下記の表1に示した混合液(1)、(2)ならびに(3)を調製した。 なお、枯草菌プロテアーゼの酵素単位は100,000〔PU〕/gmである。
プロテアーゼの力価は化粧品種別配合成分規格の一般試験法プロテアーゼ力価試験法第2法に準じ、[PU]Cas.Fr.B γtyr(以下[PU]と略)で表した。

上記混合液(1)の実測酵素単位は95、混合液(2)の実測酵素単位は103、混合液(3)の実測酵素単位は145であり、本発明の活性増進法の優れていることが分かる。

上記試験結果から本発明にはプロテアーゼの活性増進作用があり.本法を適用すればプロテアーゼの配合を減量できることが分かった。

【発明の実施の形態】
以下本発明の構成について述べる。プロテアーゼは、枯草菌(Bacillussubtilis)ならびに放線菌(Streptomyces griseous)などの産生する酵素の1種以上を適宜選択して使用する。
プロテアーゼの配合量は、最終製品1gm.または1ml.当り1〔PU〕〜1,000〔PU〕好ましくは10〔PU〕〜300〔PU〕である。

塩基成分としては炭酸水素ナトリウム、炭酸カリウムなど酸と反応して炭酸ガスを発生する炭酸塩の1種以上を適宜選択して使用できる。配合量は最終製品の0.1〜10%.好ましくは1.0〜5.0%である。

酸性リン酸カルシウム塩としてはリン酸二水素カルシウム、ピロリン酸二水素カルシウムなど一種以上を適宜選択して使用する。
なお、これら塩によるガス発生は持続性であるが遅効性のため、速度を高める目的でリンゴ酸水素ナトリウム、コハク酸水素カリウム、クエン酸二水素ナトリウム、酒石酸水素カリウムなどポリカルボン酸のモノアルカリ金属塩の一種以上をガス発生調整剤として使用する。
因みに、上記塩を形成する酸のうちTCAサイクル構成成分はガス発生の調整剤としてだけではなく、細胞活性化作用が期待できる。

酸性リン酸カルシウム塩とプロテアーゼまたは、炭酸塩とプロテアーゼで構成する粉末部にキシリトール、葡萄糖、乳糖などの一種以上を増量剤として使用することが好ましい。
本発明化粧品を設計するに当たっては、粉末部と粘液部の重量比は任意に選択できるが、1:10〜50が適当であり、使用時のpHは弱酸性〜中性になるように調整する。
粘液部の粘度調整成分としては、キサンタンガム.アルギン酸プロピレングリコールなど化粧品で汎用される非イオン性高分子が挙げられる。
本発明は美容液、乳液、クリーム、パックなど基礎化粧品基剤に広く応用できる。

ニキビに悩む女性8名(15〜23才)に実施例1の美容液を、またクスミ・シミに悩む女性10名(30〜50才)に実施例2の乳液を、1日2回2ケ月間洗顔後に塗布させた。有効性の評価は、美容の専門家と使用本人が協同して下記の基準のもとで決定した。
有 効:目立たなくなった。
やや有効:あまり目立たなくなった。
無 効:変化がなかった。
やや有効以上と判断された症例は、ニキビについては5名、またクスミ・シミについては6名であった。なお.プロテアーゼを配合しない美容液と乳液で同様の試用テストを行つたが、やや有効以上と判断された症例は減少し、ニキビについては2名、クスミ・シミについては3名であって、本発明製品の有用性が実証された。
