薬剤を投与するためのエアロゾルの使用は、数十年来知られている。このようなエアロゾルは一般に、薬剤、1種以上のクロロフルオロカーボン噴射剤、及び界面活性剤又は溶剤(エタノールなど)のいずれかを含む。最も一般的に用いられる薬剤用エアロゾル噴射剤は、噴射剤11(CCl3F)及び/又は噴射剤12(CCl2F2)を含む噴射剤114(CF2ClCF2Cl)であった。しかし、これらの噴射剤は、現在、成層圏のオゾンの分解を引き起こすと考えられているため、いわゆる「オゾンに優しい」噴射剤を用いた薬剤用エアロゾル製剤の提供が求められている。

通常のクロロフルオロカーボンと比較して、オゾン破壊作用が最小限であると考えられる噴射剤のクラスには、フルオロカーボン及び水素含有クロロフルオロカーボンが含まれ、このような噴射剤系を用いた多数の医療用エアロゾル製剤が、例えば、EP0372777、WO91/04011、WO91/11173、WO91/11495及びWO91/14422に開示されている。これらの出願はすべて、薬剤投与のための加圧エアロゾルの調製に関するものであり、新規クラスの噴射剤の使用に関連する問題、特に、調製された医薬製剤に関連する安定性の問題の克服を目的としている。これらの出願はすべて、オゾン破壊の可能性を最小限にするため、アルコール、アルカン、ジメチルエーテル、界面活性剤(フッ素化及び非フッ素化界面活性剤、カルボン酸、ポリエトキシレートなどを含む)のような1種以上の添加剤、また通常のクロロフルオロカーボン噴射剤さえも添加することを提案している。

一群のレシチンのフッ素化誘導体についてはEP4778686(Alliance)に記載されており、エアロゾル製剤における活性剤としてのそれらの使用については、WO96/09816(Glaxo)に記載されている。ヒドロフルオロアルカンと一緒に用いるフッ素化界面活性剤はまた、米国特許第5126123号、WO91/11173、WO91/14422、WO92/00062及びWO96/09816にも記載されている。

【発明が解決しようとする課題】

残念なことに、フッ素化界面活性剤、特に、製剤安定性に関して望ましい特性を有する過フッ素化界面活性剤は、高度にフッ素化した分子が体内で代謝されにくいため、往々にして生体蓄積という問題を引き起こす。このように、望ましい特性をすべて備え、しかも望ましくない特性が最小限の好適なフッ素化界面活性剤をみいだすことは困難であった。

驚くことに、本発明者らは、エアロゾル懸濁液の調製に用いるのに好適な特定の一群のフッ素化界面活性剤であって、有用な懸濁液安定化特性を有し、用量再現性、薬剤沈着の減少、貯蔵寿命の延長などの点で有利であり、しかも生体蓄積しにくいと予想される上記フッ素化界面活性剤群をみいだした。

本発明の製剤の調製に用いられる界面活性剤は、薬剤の量に対して低い濃度で効果的な安定剤となり、HFA噴射剤において適度に可溶性である。従って、用いられる界面活性剤の量は、薬剤に対して、望ましくは0.005〜20%w/w、具体的には0.05〜20%w/w、より具体的には0.05〜15%w/w、さらに具体的には約0.1〜約10%w/w、好ましくは0.5〜10%w/wである。
