【発明の属する技術分野】
本発明は、アドレナリンα1受容体拮抗作用を有する新たな化合物、その医薬的に許容される塩またはこれらの溶媒和物を有効成分として含有する高血圧治療剤に関するものである。

【従来の技術】
高血圧は、脳卒中などの脳疾患あるいは心筋梗塞や心不全などの心臓疾患を引き起こす主な原因の1つであることがよく知られている。したがって、各種の高血圧治療剤は、高血圧に伴う不快な症状の改善を目的とするばかりではなく、むしろ、上記の脳・心臓疾患の発症予防を目的として投与されている。高血圧治療剤は、血管拡張剤、利尿剤、アンジオテンシン変換酵素阻害剤、アンジオテンシンII受容体拮抗剤、アドレナリンα1受容体拮抗剤、アドレナリンβ2受容体拮抗剤、カルシウムチャンネル遮断剤などに分類されるが、いずれの薬剤も長所と短所を併せ持つことから、高血圧を引き起こしている原因、高血圧の程度、併発している疾患、副作用など各種の要因を考慮して選択されている。例えば、副作用に関して言うならば、利尿剤では低カリウム血漿、アンジオテンシン変換酵素阻害剤では空咳、アドレナリンα1受容体拮抗剤では起立性低血圧、アドレナリンβ2受容体拮抗剤では除脈、カルシウムチャンネル遮断剤では頭痛などの副作用を挙げることができる。

ところで、アドレナリンα1受容体拮抗剤は、上記の起立性低血圧を引き起こす可能性があることから、過去にはあまり好まれる高血圧治療剤ではなかったが、最近の臨床研究から、この薬剤は血圧を低下させるだけではなく、コレステロールやトリグリセリドの低下作用、血小板凝集抑制作用、インシュリン抵抗性改善作用などの好ましい作用を併せ持つことが明らかとなり、高血圧治療剤として脚光を浴びつつある。すなわち、コレステロールやトリグリセリドの上昇は、上記の脳・心臓疾患を引き起こす危険因子であり、それぞれ低下させることが好ましい。また、血小板凝集が進み易くなると血栓が生成し易くなり、上記の脳・心臓疾患を引き起こす要因となることから、血小板凝集を抑制することが好ましい。さらに、高血圧治療剤の投与は、インシュリン抵抗性を高めるということが指摘されてきたが、アドレナリンα1受容体拮抗剤はインシュリン抵抗性を低下することから、糖尿病の発症予防という観点からも優れた特性を有する高血圧治療剤であるということができる。このように、アドレナリンα1受容体拮抗剤は、現在では優れた高血圧治療剤として注目されている(例えば、非特許文献1および2参照)。

【発明が解決しようとする課題】
本発明の目的は、上記の観点から、優れたアドレナリンα1受容体拮抗作用を有する新たな化合物とその作用に基づく新たな高血圧治療剤を提供するものである。

本発明者らは、従来のアドレナリンα1受容体拮抗剤とは異なる新たな化学構造式を有するアドレナリンα1受容体拮抗剤について探索した結果、

Gが置換基を有していてもよいフェニル基、置換基を有していてもよいピリジル基または置換基を有していてもよいピリミジニル基である上記の化合物、その医薬的に許容される塩またはこれらの溶媒和物を含有するアドレナリンα1受容体拮抗剤;
上記の化合物、その医薬的に許容される塩またはこれらの溶媒和物を有効成分として含有する高血圧治療剤;
上記の化合物、その医薬的に許容される塩またはこれらの溶媒和物を有効成分として含有する腎不全、脳卒中、狭心症、心筋梗塞等の防止剤又は治療剤にも関するものである。

【発明の実施の形態】
以下に本発明を詳細に説明する。
まず、本発明で用いられる化合物(1)の置換基について、以下に説明する。

Gは、各々独立に置換基を有していてもよいフェニル基または置換基を有していてもよい5〜6員環の複素環基であるが、置換基を有していてもよいフェニル基または置換基を有していてもよい6員環の複素環基が好ましい。置換基を有していてもよいフェニル基としては、フェニル基の他に、メチルフェニル基、フルオロフェニル基、クロロフェニル基、メトキシフェニル基、アミノフェニル基、ヒドロキシフェニル基、メチルチオフェニル基、メチルアミノフェニル基、ジメチルアミノフェニル基、シアノフェニル基等のモノ置換フェニル基、シアノ−フルオロフェニル基、シアノ−クロロフェニル基、メトキシ−シアノフェニル基、ジメトキシフェニル基、ジメチルフェニル基、メチル−フルオロフェニル基、メチル−クロロフェニル基、ジフルオロフェニル基、ジクロロフェニル基、メトキシ−フルオロフェニル基、メチル−メトキシフェニル基等のジ置換フェニル基のいずれも好ましい。なお、これらのモノ置換フェニル基における置換位置は、2位または3位が好ましい。また、ジ置換フェニル基においては、3,5−ジ置換が好ましい。

[試験例]アドレナリンα1受容体結合試験
1)アドレナリンα1受容体の調製
ラットを断頭放血致死させ大脳皮質を摘出後、50mMトリス・HClバッファ(pH7.4)でホモジナイズし、4℃、50,000×gで20分間遠心分離した。さらに、同一遠心条件で1回洗浄し、得られた粗膜画分を使用時まで−80℃で保存した。
2)アドレナリンα1受容体結合試験
50mMトリス・HClバッファ(pH7.4)0.3mL、被検薬物溶液0.1mL、トレーサー125I−2−[β−(3−ヨード−4−ヒドロキシフェニル)エチルアミノメチル]テトラロン(125I−HEAT)0.1mLおよびアドレナリンα1受容体0.5mLを加え、合計1mLとしてインキュベートした後、反応液を吸引ろ過した(ろ紙:Watman GF/B)。ろ紙(トレーサー・受容体結合体:総重量)をγ−ウェルカウンター(COBA,Packard)で放射能を測定した。非特異的結合量は大過剰の薬物を加え、同様に行うことによって求め、特異的結合量は総結合量より非特異的結合量を差し引いたものとし、特異的結合率は特異的結合量と加えたトレーサーの比で求めた。
3)データ処理
用量反応曲線は、薬物存在下での特異的結合放射能(B−N)と非薬物存在下での特異的結合放射能(Bo−N)との比(B−N/Bo−N)をlogit変換した後、標準溶液濃度の常用対数値に対してプロットする。logit−logモデルに当てはめ作成した。標準曲線の回帰は、logit−log多項式(Y=ΣCiXi、ただし、i=0〜n、1<=n<=3、Y=logit y=ln(y/1−y)、y=(B−N/Bo−N)、X=log x(ただし、xは標準溶液濃度))を用いた。得られた回帰式よりIC50値を算出した。
