一般的な観察の方法として、微生物や組織細胞の観察・計測には染色法が用いられてきた。染色は、主に10〜20%のホルマリンによって固定したものをパラフィンによって包埋後、ミクロトームで薄切りし、試料を染色する方法や、その後の処理として脱パラフィン後、試薬で発色させる方法など様々な方法がある。

上記目的を達成するため、この発明の対物レンズは、試料の後方に設けられる対物レンズにおいて、前記対物レンズが前群、中群、後群からなり、前群と中群とを合成した光学系の焦点位置に開口径を連続的に変えることのできる虹彩絞りを設け、ここを後群の前焦点となるように構成するとともに、前記虹彩絞りを調整することにより開口数を連続的に変化させられるようにし、かつ開口数を連続的に変化させても常に試料側にテレセントリックの条件が保たれるようにしたことを特徴としている(請求項1)。
