本発明は有用細菌含有の飲料品又は医薬品の原料用抗菌剤及び炎症性腸疾患の予防治療剤に関するものである。

【従来の技術】
消化管内には、多種類の宿主由来の抗菌蛋白、ペプチドが分泌されている。その代表的なものとして、唾液中のリゾチームや小腸上皮細胞の一つであるパネート細胞から分泌されるクリプチジン(α−デフェンシン)等がある。デフェンシンは、好中球のリソソーム顆粒から、食胞の中に放出される殺菌蛋白質の一つであり、アルギニン残基が4〜10個、システイン残基が6個ある29〜34個のアミノ酸からなる塩基性の蛋白質である。これら以外にも、抗菌作用を有する多くの蛋白質が存在すると考えられている。

例えば、Marutsuka らは、すでに血管の弛緩作用をもつことが知られているアドレノメデュリン(adrenomedullin)、プロアドレノメデュリンペプチド(proadrenomedullinpeptide;PAMP)が消化管においても、大腸上皮細胞の管腔側に発現されていること、それらが大腸菌に対して抗菌活性をもつことを報告している(非特許文献1参照)。

このアドレノメデュリンは、降圧蛋白質として1993年にクローン化された新しい物質(B.B.R.C.誌93年vol.194,p.720−)であり、ヒトの褐色細胞からクローン化された前駆体(PAMP)は 185個のアミノ酸で、そのうち、C末よりのアミノ酸52個のペプチドがアドレノメデュリンである。

このアドレノメデュリン及びその前駆体について、最近、皮膚、口腔、気道、及び、消化管の菌叢の細菌に対して抗菌作用があることが本発明者等によって確認された(非特許文献2参照)。

一方、潰瘍性大腸炎やクローン病などの炎症性腸疾患(Inflammatory Bowel Disease:IBD)は、未だその原因が明らかにされていない消化管疾患であり、原因の究明、現在様々な治療・予防方法の確立が急務とされている。長期間に亘る腸炎病変の回復と再発が繰り返されるのが炎症性腸疾患の特徴であり、現在のところ治療は長期に亘る薬剤投与法によるしかない。

また、IBDの発症原因については、特定の腸内細菌の関与が指摘されている。即ち、ある種のバクテロイデス・ブルガタス(Bacteroides vulgatus)及び大腸菌のある株が、腸内環境を劣化させ、IBDの発症、悪性化に寄与していると報告されている(非特許文献3参照)。従って、IBDの予防治療に当たっては、これら所謂悪玉菌を抑制することが望ましいが、抗菌剤の投与などは、有用腸内細菌であるビフィドバクテリウム(Bifidobacterium) 属細菌等までも死滅させてしまい、耐性菌の出現等の問題もあるため、充分且つ完全にIBDを予防治療することは困難であった。

【発明が解決しようとする課題】
ところで、本発明者等は、アドレノメデュリン及びその前駆体の抗菌活性について、細菌の種、株等によって抗菌作用の差があることを見出した。即ち、アドレノメデュリン及びそのプレ体は、特定の細菌については強く生存性を抑制するのに対して、別の特定の細菌については生存率を抑制することがなく、更には生存率を助長することが確認された。

本発明は、プロバイオティク(probiotic) な細菌のように特定の有用細菌については抗菌作用を示さず、病原性細菌を含む多くの細菌については抗菌作用を示す抗菌剤を得ることを目的とする。また、本発明は、ビフィドバクテリウム属細菌等の腸内有用細菌を抑制することなく、所謂悪玉菌を抑制することのできる炎症性腸疾患の予防治療剤を得ることを目的とする。

請求項1に記載された発明に係る有用細菌含有飲食品又は医薬品原料用抗菌剤は、アドレノメデュリン(Adrenomedullin)又はプロアドレノメデュリンペプチド(PAMP)を主成分として含有することを特徴とするものである。

【発明の実施の形態】
本発明においては、アドレノメデュリン(Adrenomedullin)又はプロアドレノメデュリンペプチド(PAMP)を主成分として含有するものであるため、特定の有用細菌を含有する飲食品原料又は医薬品原料に添加された際に、プロバイオティク(probiotic) な細菌のように特定の有用細菌については抗菌作用を示さず、病原性細菌を含む多くの細菌については抗菌作用を示す抗菌剤を得ることができる。即ち、本発明では、特定の有用細菌を含有する飲食品又は医薬品の原料に対するアドレノメデュリン又はプロアドレノメデュリンペプチドの抗菌剤としての使用を提案するものである。

本発明でのアドレノメデュリン(Adrenomedullin)又はプロアドレノメデュリンペプチド(PAMP)は、ペプチド研究所より購入した。

本発明による抗菌剤の使用対象は、1種又は2種以上の特定の有用細菌を含有する飲食品原料又は医薬品原料である。係る特定の有用細菌としては、Lactobacillus casei又はBifidobacterium breve が挙げられ、特に、Lactobacillus casei YIT9029、Lactobacillus casei YIT9018、及びBifidobacterium breve YIT4065から選ばれた1つ以上の株が挙げられる。特に、Bifidobacterium breve YIT4065 に対しては、本発明による抗菌剤は、阻害作用を殆ど示さず、むしろ成長促進作用を示すことが確認されている。

従って、これらLactobacillus casei又はBifidobacterium breveを含む飲食品原料及び医薬品原料にアドレノメデュリン(Adrenomedullin)又はプロアドレノメデュリンペプチド(PAMP)を主成分とする抗菌剤を添加することにより、飲食品原料及び医薬品原料に含まれる有用細菌以外の細菌の増殖は阻害され、特に、Bifidobacterium breve YIT4065については、増殖も可能な抗菌剤としての使用を得ることができる。

本発明での有用細菌含有飲食品原料としては、前記有用細菌を培養物として含む飲食品原料が挙げられる。例えば、プレーンタイプ、フレーバードタイプ、フルーツタイプ、甘味タイプ、ソフトタイプ、ドリンクタイプ、固形(ハード)タイプ、フローズンタイプ等の発酵乳、乳酸菌飲料、チーズ、発酵豆乳、キムチ等が挙げられる。

また、有用細菌含有医薬品原料としては、前記有用細菌を生菌体として含む医薬品原料が挙げられる。例えば、前記有用細菌を生菌体(乾燥菌体等)として含み、その他に後記するような薬学的に許容される他の製剤で適宜加工し、例えば、カプセルに充填したりタブレットや顆粒にしたものが挙げられる。

また、本発明は、アドレノメデュリン又はプロアドレノメデュリンペプチドを有効成分とする炎症性腸疾患の予防治療剤、即ち、アドレノメデュリン又はプPAMPの予防治療剤としての使用に関する。炎症性腸疾患の予防(再発予防を含む)には、本疾患との関連が推定されているバクテロイデス・ブルガータス(非特許文献3)に抗菌活性を示し、他の特定の有用細菌に抗菌活性を示さない物質の投与が有効と考えられ、本発明のアドレノメジュリン、プロアドレノメジュリンはこのような物質に該当することが見出されたためである。炎症性腸疾患の予防治療剤として用いるアドレノメデュリン、プロアドレノメデュリンペプチドは特に限定されず、前記の同一の方法にて得たもの等を用いることができる。

薬学的に許容される他の製剤としては、具体的に、剤型としては、カプセル剤、顆粒剤、錠剤、被膜錠剤、散剤等の固形製剤あるいはシロップ剤、内服液剤、エリキシル剤等の液状製剤として経口投与する経口用製剤や、注射剤、直腸投与剤等の非経口用製剤とすることが可能であり、注射剤、直腸投与剤とすることが好ましい。

各製剤(医薬組成物)を製造する場合、例えば経口用製剤であれば、賦形剤、必要に応じて結合剤、崩壊剤、滑沢剤、着色剤、矯味剤、矯臭剤等を加えた後、定法により上記剤型に加工し製造することができる。そのような添加剤としては、当該分野で一般的に使用されているものを用いることができ、例えば賦形剤としては、乳糖、白糖、塩化ナトリウム、ブドウ糖、澱粉、結晶セルロース、微結晶セルロース、炭酸カルシウム、カオリン、乳酸カルシウム、メタケイ酸アルミン酸マグネシウム、無水ケイ酸等を、結合剤あるいは崩壊剤としては、水、エタノール、プロパノール、単シロップ、ブドウ糖液、デンプン液、ゼラチン液、ヒドロキシプロピルスターチ、リン酸カルシウム、ヒドロキシプロピルセルロース、ポリビニルピロリドン、カルボキシメチルセルロース、カルボキシメチルセルロースカルシウム、乾燥デンプン、アルギン酸ナトリウム、カンテン末、炭酸水素ナトリウム、炭酸カルシウム、ラウリル硫酸ナトリウム、ステアリン酸モノグリセリド、乳糖、白糖等を、滑沢剤としては、ステアリン酸マグネシウム等のステアリン酸塩、精製タルク、ホウ酸、ポリエチレングリコール等を、矯味剤としては、白糖、橙皮、クエン酸、酒石酸等が例示できる。

また、経口用液状製剤を調製する場合は、矯味剤、緩衝剤、安定化剤等を加えて定法により各種製剤化を行なうことができる。緩衝剤としてはクエン酸ナトリウム等が、安定化剤としてはトラガント、アラビアゴム、ゼラチン等を例示でき、その他は上記と同様である。

アドレノメデュリン、プロアドレノメデュリンペプチドを有用細菌含有食品又は医薬品の原料として用いる場合、その添加量に特に限定されない。また、その投与量は1〜5mg/日/人が、好ましい。

【実施例】
1.実験方法
本実施例で用いた試験菌は次の表1に示す通りである。尚、使用培地については、1)TS液体培地(BBL)/TS寒天培地(Nissui)、2)TS液体培地(BBL)/GAM (BBL)+1%Glu,1%Lac寒天培地、3)GAM(BBL)+1%Glu,1%Lac液体培地/GAM(BBL)+1%Glu,1%Lac寒天培地、嫌気的に調製、培養した。

2.抗菌活性の測定
使用したペプチドについては、アドレノメデュリン(Adrenomedullin)、プロアドレノメデュリンペプチド(PAMP)、β−デフェンシン(β−Defensin−2)、α−デフェンシン(α−Defensin−1)はいずれもペプチド研究所(大阪)から購入した。

3.抗菌活性の測定方法
それぞれの菌を上記の液体培地で一夜培養後、新しい培地に植えつぎ対数増殖期まで培養した。約105 菌数/mlになるように、10mM リン酸バッファー(+0.09% NaCl + 5mM glucose、pH 7.2)で希釈し、その200μlをエッペンチューブ(丸底)に採取し、各種ペプチド(0.1μM〜2μM)を含む水(50μl)と混合し、緩く攪拌しながら37℃、3時間反応させた。容器ごと氷冷し、PBSまたは嫌気的に調製した希釈液で希釈した。その後、それぞれの菌を上記の寒天培地に播種してCFUを求めた。嫌気性菌の各ペプチドとのインキュベーション並びに培養は嫌気的に調整されたグローブボックス内で行った。

4.結果
4.1.抗菌活性測定条件の検討
1) 対数増殖期と静止期の比較
静止期と対数増殖期のE. coli(YIT6044)を用いてβ‐Defensinの抗菌活性を測定した。図1は静止期及び対数増殖期でのE. coli(YIT6044)の生存率に対するβ−デフェンシンの種々の濃度の影響を示したグラフであり、a図は静止期での結果を示す線図、b図は対数増殖期での結果を示す線図である。2つの成長期から用意された2つの菌懸濁液は、PBS中で抗菌性ペプチドと共に37℃3時間の振盪培養を行った。抗菌性の活性は、ペプチドと共に3時間のインキュベーションを行う前と後とでCFUを計測することによって評価された。図において、棒グラフはCFUを示し、折れ線グラフは抑制率を示す。尚、後述する図においても同様に、棒グラフはCFUを示し、折れ線グラフは抑制率を示す。

図に示す通り、静止期ではペプチド濃度1μMで、ペプチド非添加と比較して全く阻害活性がみられなかった。一方、対数増殖期の菌を用いた場合は、1μMで約60%の阻害がみられた。

2) 抗菌活性測定のための緩衝液の検討
対数増殖期の菌を用いて、緩衝液(10mM Phosphate‐buffer)中のNaCl濃度を検討した。図2はβ−デフェンシンによるE. coli YIT6044 の生存率抑制でのNaCl濃度の影響を示したグラフであり、a図はNaClが 0.9%、b図はNaClが0.09%のものを示す。図において、棒グラフはCFUを示し、折れ線グラフは抑制率を示す。

図に示す通り、E. coliに対するβ−Defensin2の抗菌活性は、NaCl濃度0.9%では顕著に抑制され、0.09%以下ではほとんど影響を与えなかった。

図3はB.vulgatus YIT6159の生存率抑制でのNaCl濃度の影響を示したグラフであり、a図はNaClが0%、b図はNaClが0.09%のものを示す。図に示す通り、B. vulgatusにおいては、抗菌ペプチド不在下でもNaClを添加しないと3時間の反応時間中の菌数の減少が著しく、0.09%NaClでは菌数の減少が抑制できた。以上の結果より0.09%NaClを採用した。

図4はβ−デフェンシンによるE. coli YIT6044 の生存率抑制(0.09%NaCl)でのブドウ糖の影響を示したグラフである。図に示す通り、ブドウ糖の添加による抗菌活性への影響はほとんどなかった。

4.2.4種類のペプチドの抗菌活性
1) 常在性腸内細菌種
図5は4種類のペプチドによるE. coli の生存率に対する影響を示したグラフである。他の実験の条件は図1で示した通りである。図に示す通り、4種類のペプチドの各々について、有意な阻害が見られた。

図12は3種類のペプチドによるB. breve YIT4065の生存率に対する影響を示したグラフである。他の実験の条件は図1で示した通りである。図に示す通り、3種類のペプチドのうち、アドレノメデュリン、プロアドレノメデュリンペプチドについて、有意な生存率の促進が見られた。

以上の図5〜図12の結果を要約したものを次の表2に示す。表において、−1〜−3と+1〜+3は、生存率抑制と促進の度を示す。±は影響のないことを示す。

以上の通り、ヒトの腸内菌の中で占有率の高い菌種を選択して、各ペプチドの抗菌活性を測定した。アドレノメデュリン及びプロアドレノメデュリンペプチドに対しては、殆どの微生物は程度の差はあるが、ペプチド濃度1‐2μMで阻害活性がみられたが、L. casei YIT9029及びB. breve YIT4065に関しては殆ど阻害活性がみられなかった。特に、B. breve YIT4065に関しては阻害活性ではなく、生存率促進活性が見られた。炎症性腸疾患の発症や憎悪には常在性腸内細菌の関与が認められており、抗生物質の投与は一定の効果を上げている。従って、各種の常在性腸内細菌に抗菌活性を示し、有用菌である乳酸菌及びビフィズス菌に抗菌活性を示さない本物質の投与は有効と考えられる。

【発明の効果】
本発明は以上説明した通り、プロバイオティク(probiotic )な細菌のように特定の有用細菌については抗菌作用を示さず、病原性細菌を含む多くの細菌については抗菌作用を示す抗菌剤及び炎症性腸疾患の予防治療剤を得ることができるという効果がある。
