本発明は、優れた抗腫瘍効果、抗菌効果、免疫増強効果及び滋養強壮効果を有する薬用茸の一種である冬虫夏草用の培地に関するものである。

【従来の技術】
冬虫夏草は、薬用茸の一種で、昆虫などを宿主とし、菌糸が増殖して宿主を死に至らしめた後、該宿主の体内で菌糸が充満すると子実体を形成するコルディセプス(cordyceps)属の茸の一種であり、冬季には宿主の体内にあり、夏季になると宿主の虫体から穿ち出る子嚢菌に属する茸である。
冬虫夏草は、自然状態では地中または地上に菌糸または胞子として存在する菌であり、寄生する昆虫の幼虫期、蛹期及び成虫期全般にわたって体内に侵入して発生する。
冬虫夏草は、優れた抗腫瘍効果、抗菌効果、免疫増強効果及び滋養強壮効果を有するものとして知られている。冬虫夏草の有効成分はコルディセピン(cordycepin)であり、その含有量は、子実体の乾燥体において0.02〜0.6重量%程度である。

近年、冬虫夏草の菌糸体を人工的に培養する方法としていくつか提案されており、例えば、冬虫夏草の虫体菌糸又は座胞子をセミなどの節足動物の昆虫類の成虫、蛹、幼虫などに定着させて固体培養する方法、米胚を利用した培養液の製造方法などが知られている。しかしながら、成虫、蛹、幼虫などに定着させる固体培養や、米胚を利用した液体培養などの従来の培養方法では、培養方法が複雑であった。また、その収量は、たとえば、さなぎ固体培養による培養生成産物中のコルディセピン含有量は約0.1〜0.5重量%、米胚液体培養の場合は約0.04重量%といずれも高くなかった(例えば、特許文献1参照。)。

【発明が解決しようとする課題】
本発明は、このような実情に鑑みなされたものであり、その目的は、得られる培養生成物中のコルディセピンの含有量を向上させることができる冬虫夏草用培地を提供することにある。

【課題を解決するための手段】
前記目的を達成するために、本発明の冬虫夏草用培地は、(A)炭素源としてグルコース、スクロース、糖蜜、マルトースから選ばれた少なくとも1種を1〜6重量%、(B)窒素源としてペプトン、イースト、モルト、ミルクから選ばれた少なくとも1種を1〜7重量%、(C)アスパラギン、アスパラギン酸、グリシン、DNAから選ばれた少なくとも1種を0.1〜0.5重量%それぞれ含有してなるものである。
前記冬虫夏草用培地が、さらに(D)微量のビオチン及び/又はビタミンB類を含有することが好ましい。
また、本発明の冬虫夏草用培地は、(A)炭素源としてグルコース、スクロース、糖蜜、マルトースから選ばれた少なくとも1種を1〜6重量%、(B)窒素源としてペプトン、イースト、モルト、ミルクから選ばれた少なくとも1種を1〜7重量%、(D)ビオチン及び/又はビタミンB類を微量それぞれ含有してなるものである。
前記ビタミンB類が、ビタミンB1及び/又はビタミンB12であることが好ましい。
前記(A)成分の含有量が1〜3重量%であると共に、前記(B)成分の含有量が4〜7重量%であることが好ましい。
なお、本発明において重量%とは、水100重量部に対して、成分x重量部を含有することを意味する。

【発明の実施の形態】
以下、本発明を詳細に説明する。
本発明の冬虫夏草用培地は、(A)グルコース、スクロース、糖蜜、マルトースから選ばれた少なくとも1種、(B)ペプトン、イースト、モルト、ミルクから選ばれた少なくとも1種、(C)アスパラギン、アスパラギン酸、グリシン、DNAから選ばれた少なくとも1種及び/又は(D)ビオチン、ビタミンB類を含有してなるものである。

本発明の培地で培養される冬虫夏草としては、コルディセピンを含有するものであれば特に限定されない。例えば、コルディセプス属に属するもので、コルディセプス・ミリタリス(Cordyceps militaris)等の培養に好ましく用いられる。特にコルディセプス・ミリタリス(寄託番号FERM P−18657、寄託番号FERM P−18658)の培養に好ましく用いられる。

(A)成分は、冬虫夏草用培地の炭素源として含有するものであり、グルコース、スクロース、糖蜜、マルトースのいずれか1種又は2種以上である。
(A)成分の冬虫夏草用培地中の含有量は1〜6重量%であり、好ましくは1〜3重量%である。(A)成分の含有量が1重量%未満、または、6重量%を超えた場合は、得られる培養生成物の培地の単位重量あたりの菌体重量が減少する傾向がある。

(B)成分は、冬虫夏草用培地の窒素源として含有するものであり、ペプトン、イースト、モルト、ミルクから選ばれるいずれか1種または2種以上である。イーストまたはモルトはその抽出物を使用してもよい。ミルクとしては、粉乳、脱脂粉乳等が挙げられる。
(B)成分の冬虫夏草用培地中の含有量は1〜7重量%であり、好ましくは4〜7重量%である。(B)成分の含有量が1重量%未満、また、7重量%を超えた場合は、得られる培養生成物の培地の単位重量あたりの菌体重量が減少する傾向がある。

(C)成分は、アスパラギン、アスパラギン酸、グリシン、DNAから選ばれるいずれか1種または2種以上である。
アスパラギン、アスパラギン酸、グリシンは、アミノ酸の1種である。アスパラギンとしてはL体、D体どちらも使用できるが、L体が好ましい。アスパラギン酸においても、L体、D体どちらも使用できる。L−アスパラギン(和光純薬製のもの等)を用いた場合には、低コストに製造できるので工業的に好ましい。DNAとしては、例えば、魚等から抽出したDNAが使用でき、鮭精液由来のDNA等が挙げられる。
(C)成分の冬虫夏草用培地中の含有量は0.1〜0.5重量%である。(C)成分の含有量が0.1重量%未満の場合は、コルディセピン含有量の増加効果が少ないので好ましくない。また、(C)成分の含有量が0.5重量%を超えた場合は、菌体の成長およびコルディセピン含有量の増加効果の極限を超えるため好ましくない。

(D)成分は、ビタミンとして冬虫夏草用培地に含有させるものであり、ビオチン、ビタミンB類のいずれか1種または2種以上である。
ビオチンはビタミンの一種である。
ビタミンB類は、ビタミンBであれば特に限定されず、好ましくは、ビタミンB1、ビタミンB12等であり、これらは単独のものでも、または二種以上の混合物であってもよい。
(D)成分の含有量は、特に限定されないが、微量であることがよく、例えば冬虫夏草用培地100mlに対して2〜10μg程度である。(D)成分の含有量が2μg未満の場合は、得られる培養生成物中のコルディセピン含有量の増加効果が少ないので好ましくない。また、(D)成分の含有量が10μgを大きく超えた場合は、菌体の成長およびコルディセピン含有量の増加効果の極限を超えるので好ましくない。

冬虫夏草用培地のpHは、2〜9であることがことが好ましく、特に好ましくは、5〜7である。pHの調製は、調製が行えるならばどのようにしてもよく、例えば、塩酸や水酸化ナトリウムを使用する調製方法が挙げられる。

本発明の冬虫夏草用培地は、(A)〜(D)成分以外はほとんど水であるが、この水としては、特に限定されず、水道水、井戸水、蒸留水等が挙げられる。
このように、本発明の冬虫夏草用培地は、(A)成分、(B)成分、(C)成分及び/又は(D)成分を含有してなることにより、得られる培養生成物中のコルディセピンの含有量を向上させることができる。さらに、(A)〜(D)成分、水以外にも、コルディセピンの含有量を向上させるために他の成分を含有させてもよい。
他の成分としては、例えば(B)成分以外の窒素源としてはおから、それ以外には例えばマグネシウム、カルシウム等のミネラルが挙げられる。

【実施例】
以下、実施例及び参考例に基づいて本発明をより具体的に説明するが、本発明はこれになんら限定されるものではない。
下記実施例及び参考例の各培地を本培養培地として用い、以下のようにして得られる生成物の菌体重量および総コルディセピン量を求めた。

(使用菌株)
菌株としては、コルディセプス・ミリタリス(寄託番号FERM P−18657)を、保存菌株用培地(スラント培地:ペプトン1g、グルコース3g、DL−アスパラギン0.1g、MgSO4 0.05g、K2HPO4 0.1g、ビオチン 2μg、寒天3g、蒸留水100ml、pH6.8)で保存したものを用いた。この菌株を、種菌培養として下記前々培養および前培養した後、後述の各実施例及び各参考例の培地を用いて本培養した。

(前々培養)
平板寒天培地(ペプトン1g、グルコース3g、DL−アスパラギン0.1g、MgSO4 0.05g、K2HPO4 0.1g、ビオチン2μg、寒天3g、蒸留水100ml、pH6.8)をシャーレに20ml充填した。このシャーレ培地に、前記保存した菌株を約15mm*15mm切り出して接種し、これを21℃下2週間培養した。

(前培養)
液体培地(ペプトン1g、グルコース3g、DL−アスパラギン0.1g、MgSO4 0.05g、K2HPO4 0.1g、ビオチン2μg、蒸留水100ml、pH6.8)100mlを500ml三角フラスコに分注し、121℃下20分間滅菌処理した。処理後、液温度が室温まで下がったとき、この液体培地に、前々培養したシャーレ培地から切り出した約1cm角のコロニ−三つを接種した。これを回転振盪機を用いて80rpmで、21℃下10日間培養し、分生子の発生を顕微鏡で確認後、本培養に使用した。

(本培養)
下記実施例及び参考例の各培地100mlをそれぞれ500ml三角フラスコに分注し、121℃下20分間滅菌処理した。なお、各例毎に(1種につき)それぞれ3つ同じ実験を行った。処理後、それぞれ液温度が室温まで下がったとき、この液体培地に、前培養した培地から採取した約1mlを接種し、静置して本培養を行った。

(培養生成物中の総コルディセピン量の測定)
本培養後、各培養液100mlをろ紙(5B:アドバンテック東洋社製)を用いてろ過した。得られた菌体を5日間凍結乾燥した後、精密天秤で測定しその重量(以下、菌体重量とする)を測定した。
また、ろ過した培養液を凍結乾燥し、その重量(以下、培養液乾燥重量とする)を測定した。

凍結乾燥した菌体1gを水(蒸留水)を用いて10分間超音波(サンフレックス、DG−1、50W、43KHz)処理した。この処理液をろ紙(5B:アドバンテック東洋社製)を用いてろ過してコルディセピン含有量分析用試料とした。
この試料を、高速液体クロマトグラフィ(HPLC(LC−10:株式会社島津製作所製))を用いて分析し、コルディセピンの含有量を測定した(以下、菌体コルディセピン含有量とする)。

HPLCの条件は、カラムにInertsil ODS−2(ODS) 250L*4.6φmm(ジーエルサイエンス株式会社製)を用い、移動相は50mMリン酸カリウム:アセトニトリル(V/V=95:5)、流速1.0ml/min、カラム温度40℃、検出波長260nmで行った。
前記ろ過培養液の凍結乾燥物についても、同様にしてコルディセピン含有量を測定した(以下、培養液コルディセピン含有量とする)。

上記の測定は、前述のように培地1種につき三連で実験して得られた培養物のそれぞれについて行い、3つの値の平均値をその培地における値とした。

下記表1に示した実施例1〜4の各種炭素源3g、窒素源としてペプトン1g、DL−アスパラギン0.1g、MgSO4 0.05g、K2HPO4 0.1g、ビオチン 2μg、蒸留水100mlからなり、pH6.8の液体培地をそれぞれ作成した。これら作成した培地を前記の本培養の培地として用いて、それぞれ菌体重量および総コルディセピン量を求めた。その結果を表1に示した。なお、本培養は、21℃下暗所にて50日間行った。

下記表5に示した参考例1〜13の各種アミノ酸又はDNA(鮭由来)0.1g及び参考例14のアミノ酸又はDNA無添加、グルコース3g、蒸留水100mlを用いて培地を作成した。これら作成した培地を前記の本培養の培地として用いて、それぞれ菌体重量および総コルディセピン量を求めた。各培地を使用した場合の評価は、アミノ酸又はDNA無添加のコントロール(参考例14)における総コルディセピン量を1として比を求めることにより行った。その結果を表5に示した。なお、本培養は、21℃下暗所にて30日間行った。

表5に示した結果から、総コルディセピン量はDNA、DL−アスパラギン、L−アスパラギン、DL−アスパラギン酸、D−アスパラギン酸、L−アスパラギン酸及びグリシンを添加した場合が顕著に多く、特にDNAが最も多かった。
