【産業上の利用分野】
本願発明は医療用医薬品の分野に属する血漿タンパク質の新たな用途に関する。さらに詳細には、神経伝達に関わる中枢及び末梢神経系に起因する神経並びに筋変性疾患に対する医薬品に関する。より詳しくは、血漿タンパク質の一種であるセレノプロテインPに例示されるセレノシステイン含有タンパク質を、好適には当該セレノプロテインPのC末端側ペプチドもしくは当該ペプチド群を主たる有効成分として含有する神経伝達機能異常疾患改善剤、とりわけシナプス伝達やアセチルコリンレセプター動作、一酸化窒素による神経細胞活性化に改善作用を有する薬剤に関する。

【従来の技術及び発明が解決しようとする課題】
神経回路網において神経細胞と神経細胞間あるいは神経細胞と効果器細胞(例えば筋細胞)間の接合部をシナプスという。シナプスは神経回路網の情報伝達にとって重要な部位であり、通常、神経細胞の軸索末端部を情報の出力部(シナプス前部)とし、樹状突起、細胞体を情報の入力部(シナプス後部あるいは後膜)としている。神経末端に信号が伝わると、シナプス前部のシナプス小胞が開口分泌を起こし、内部に貯蔵されていた神経伝達物質がシナプス間隙に放出され、シナプス後膜に存在する神経伝達物質のレセプターに結合し、次の細胞に情報が伝えられる(例えば、非特許文献1参照)。

神経伝達物質として、アセチルコリン、グルタミン酸、アスパラギン酸、γアミノ酪酸(GABA)、グリシン、セロトニン、ドパミン、ノルアドレナリン、アデノシン三リン酸(ATP)、各種の神経ペプチドなどが挙げられる。例えば、アセチルコリンは、生体内でコリンアセチルトランスフェラーゼによってコリンとアセチルCoAから合成され、シナプス小胞に貯蔵される。このようなアセチルコリンを放出する神経細胞をコリン動作性ニューロンと呼ぶ。中枢神経系では、前脳の基底部から大脳皮質や海馬への投射、脳幹部の橋脚部並びに外背側被蓋部から大脳皮質への投射の他、線条体内の介在ニューロンや前庭核から小脳への投射、または脊髄から下降性運動ニューロンもコリン動作性神経である。末梢神経系では、交感神経の1次神経、副交感神経の1次神経と2次神経、運動神経はいずれもその神経終末でアセチルコリンを分泌する(例えば、非特許文献1参照)。
一方、情報を受け取るシナプス後膜にあるアセチルコリンレセプターには大きくムスカリン性とニコチン性の2つのタイプが存在する。ムスカリン性は7回膜貫通型受容体ファミリーに属し、Gタンパク質を介してシグナルを細胞内に伝達する。ムスカリン性受容体はさらにホモロジーから5種類のサブタイプに分類され、Gタンパク質の種類によってフォスフォリパーゼCを活性化するタイプ(M1,M3,M5)とアデニル酸シクラーゼを抑制するタイプ(M2,M4)の2種類に分類される。前者は細胞に対して興奮的に、後者は抑制的に作用する。これらのレセプターは脳全般以外に心臓や平滑筋や外分泌腺組織に広く分布する。ニコチン性受容体は、5つのサブユニットから構成されるイオンチャンネルで、2つのαサブユニットと1つずつのβ、ε、δの5つから構成される。少なくともαサブユニットには9種類、βサブユニットには4種類のサブタイプが知られている。ニコチン性受容体はその分布様式から、骨格筋型と神経型に分類されている(例えば、非特許文献1参照)。

アセチルコリン以外の神経伝達物質もそれぞれに対応したレセプターがあり、アセチルコリンレセプター同様、各々特有の神経組織分布を示している。これらの神経伝達物質がシナプス間隙で適正に授受されることによりレセプターを活性化し、さらにセカンドメッセンジャーを活性化し、神経細胞の生理学的反応を引き起こしている。レセプターによって、反応は興奮的(新たな活動電位の開始)に伝達されるか、抑制的(活動電位発生の抑制)になるか制御されており、これらが複雑に絡み合って、生体内の神経回路網は動いている(例えば、非特許文献1参照)。一方、一酸化窒素(NO)のようにレセプターははっきりしないが、例えば自律神経系の神経終末から放出され、効果器官の平滑筋の弛緩や脳の血流調節、陰茎海綿体の勃起などの作用を持つため神経伝達物質の一種であると考えられているものもある(例えば、非特許文献3参照)。NOの細胞間シグナル伝達物質としての作用は、受容体やトランスポーターを介さずに細胞膜を直接越えて拡散するため、作用範囲が広範に渡る。NOは、サイクリックGMP(cGMP)の合成酵素である可溶性グアニル酸シクラーゼの活性化を誘導し、合成されたcGMPはcGMP依存性リン酸化酵素を活性化し、細胞内の生理作用を作動させ、細胞を活性化する(例えば、非特許文献6参照)。その一方で、NOはシナプス後部から放出され、シナプス前部終末からの神経伝達物質放出を調節する逆方向情報伝達物質としても作用するためシナプス可塑性の調節因子と考えられている(例えば、非特許文献6参照)。

このような神経伝達物質の関わる神経伝達に異常が生じると、様々な疾患を引き起こすことになる。例えば、アセチルコリンによる情報伝達システムは記憶学習の機能、自律神経、運動神経、交感及び副交感神経機能に関わっており(例えば、非特許文献1参照)、アセチルコリンによる情報伝達に異常が生じると、これらの機能に障害が生じ、様々な疾病の原因となる。神経伝達の欠陥に伴う疾患として、様々な病気が知られている。例えば、神経伝達物質の不均衡による疾患として、アルツハイマー病、不安、自閉症、脳障害、うつ病、ハンチントン病、躁病、疼痛、パーキンソン病など。神経伝達物質のレセプターに欠陥がある疾患として、重症筋無力症、スローチャネル症候群、先天性筋無力症など。神経伝達物質の神経細胞取り込み低下による疾患として、筋萎縮性側索硬化症など。イオンチャネルに欠陥があり神経伝達が正常にできない疾患として、発作性運動失調、高カリウム血性周期性四肢麻痺、低カリウム血性周期性四肢麻痺、ランバート・イートン症候群、先天性パラミオトニア、ラスムッセン脳炎、脊髄小脳変性症など。中毒疾患として、ボツリヌス中毒、蛇毒による中毒など、が知られている(例えば、非特許文献2及び非特許文献5参照)。
この他、自律神経系での神経伝達障害を改善し、病態を改善する薬剤が開発されている。例えば、眼圧低下を目的とした緑内障の治療薬として、アセチルコリンアナログであるピロカルピンやカルバコールが知られている(例えば、非特許文献4参照)。唾液腺のムスカリン受容体を刺激し、唾液分泌を促進する薬剤や、アセチルコリン遊離促進作用を持った消化管運動賦活剤が機能性胃腸症の治療薬として開発されている(例えば、非特許文献8参照)。

これらの疾患に対する薬剤は、神経伝達物質、そのアゴニスト、アンタゴニストあるいは抗コリンエステラーゼのように神経伝達物質の酵素分解や再吸収を阻害し、その半減期を延長させるものが大部分で、いずれも神経伝達物質とレセプターの直接的な反応をターゲットにしているものが多い(例えば、非特許文献9参照)。しかし、神経伝達物質とそのレセプターは前述のように種類が多く、その作用も興奮性、抑制性など多彩である。そのため思わぬ副作用が起こることがある。例えば、抗コリンエステラーゼ剤の過剰投与によるコリン動作性クリーゼ(急激な筋肉の麻痺症状)が生じたり、長期間投与でレセプターの変性が促進され、病態を悪化させたりすることがある(例えば、非特許文献7参照)。また、神経伝達は自律神経系に深く関わっているので危険な心肺機能低下が起こる可能性も否定できない。これらの副作用を解決するために、これまでの薬剤とは異なる作用メカニズムを持ち、副作用の少ない安全な薬剤が求められている。

【課題を解決するための手段】
上述の状況の下、本願発明者等は先に、血液成分由来のタンパク質で、セレノシステイン含有タンパク質の一種であるセレノプロテインP(以下、SePと称することがある)に、そしてより好適な態様として当該セレノプロテインPのC末端側ペプチドに、従来報告されていなかった細胞死抑制活性が認められることを見出し、この知見を基に特許出願した(例えば、特許文献1参照)。さらに、本願発明者は、新たな神経伝達機能異常疾患改善剤、とりわけ、シナプス伝達やアセチルコリンレセプター動作、一酸化窒素による神経伝達に改善作用を有する薬剤を供するべく鋭意研究した結果、驚くべきことに従来試みられることのなかった前記セレノプロテインPまたはそのC末端側ペプチドもしくは当該ペプチド群が、細胞死抑制活性のみならず、神経細胞を用いた培養実験や、モデル動物での実際の生体内投与によって、神経伝達機能を改善する作用を持つことを見出し、この知見に基づいて本願発明を完成するに至った。

セレノプロテインPは1977年にグルタチオンペロキシダーゼ(gulutathion−peroxidase)とは異なるセレン含有タンパク質として確認され、1982年にセレンがセレノシステイン(Selenocystein)の形態で取り込まれていることが明らかにされた。さらに、1991年にラットセレノプロテインPのcDNAのクローニングにより全長のアミノ酸配列が明らかにされ、その結果、当該タンパク質は最大10個のセレノシステインを含む可能性等が示された(例えば、非特許文献10参照)。
1993年にはヒトセレノプロテインPの核酸塩基配列及びアミノ酸配列が報告された(例えば、非特許文献11参照)。セレノプロテインPの機能はほとんど不明であったが、最近、in vitroの系でphospholipid hydroperoxideの還元活性(例えば、非特許文献12参照)やperoxynitriteの消去活性(例えば、非特許文献13参照)があるとの報告がなされた。また、Se欠乏時に脳に特異的にSeを運搬するとの報告(非特許文献14参照)や、神経細胞のsurvival promoting factorとしての作用(非特許文献15参照)が見出され、神経細胞の生存と関連が示唆されていた。しかし、この2つの報告からセレノプロテインPの神経細胞生存維持活性以外の神経細胞に対する具体的な作用を類推することは難しく、ましてや、本願発明で述べるようなセレノプロテインPの神経細胞活性化や神経伝達機能に関わるような作用を見出した例もない。

本願発明者らは、具体的な事例として、神経様細胞であるNG108−15細胞を神経細胞に分化させる際にセレノプロテインPを培地に添加すると、神経突起進展の複雑さや結節状構造(varicosity)が増強され、シナプス形成を促進することを見出した。また、ムスカリンアゴニストであるピロカルピンを用いたてんかん誘導のモデルマウスで、セレノプロテインPがてんかん症状を増強し、アセチルコリンレセプターの作用を増強することを見出した。さらに、マウス初代神経細胞を用いた培養で細胞に影響のでない濃度の一酸化窒素を発生させた際にセレノプロテインPを添加しておくと、神経細胞のミトコンドリア機能が亢進し、細胞が活性化されることを見出した。これらのことから、シナプス形成やアセチルコリンレセプター機能やNOによる神経細胞活性化の亢進、すなわち神経細胞の関わる神経伝達機能を改善する作用をセレノプロテインPが有していることが示された。

本願発明は、セレノプロテインPの前記知見に基づく新たな薬効に関するものであり、本願発明の神経伝達機能異常疾患改善剤の本態はセレノプロテインPである。さらに詳細には、セレノプロテインPの中のセレンを含むセレノシステインに特徴があり、このアミノ酸が神経伝達とりわけシナプス伝達やアセチルコリンレセプター動作、一酸化窒素による神経伝達の改善作用の中心的アミノ酸である。本願発明者等は、先の特許出願において、血液成分由来のタンパク質であるセレノプロテインPのC末端側ペプチド断片に従来報告されていなかった細胞死抑制活性が認められること、その活性にはセレノシステインが関与していることを開示した。本願発明で述べる活性にも、含有されるセレノシステインが関与していることが明らかである。従って、セレノシステインを含み細胞死抑制活性を持つタンパク質及び/またはペプチド群は、神経伝達機能異常疾患改善剤の候補となりうる。

そもそも、本願発明に関係するセレンは、微量必須元素の一つであり、それが欠乏した場合には心筋症などを伴う重篤な欠乏症が知られている。また、無血清培養の培地に亜セレン酸ナトリウムの添加が必須であることから、セレンが細胞レベルでの生存維持・増殖に必須であることが示されている。しかし、セレン化合物が毒物指定されていることから理解されるように、有効量と危険量の幅、つまり安全域の濃度幅が狭く、適量以上のセレン化合物は一般的には細胞にとって毒性を示し、逆に細胞死を誘導する。例えば、セレンの急性中毒症状として、顔面蒼白、神経症状、神経障害、皮膚炎、胃腸障害などが知られている。また、細胞培養にセレノシステインの2量体であるセレノシスチンを添加すると、単独ではかなり強い毒性を示す。これに対して、本願発明の好適な態様であるセレノプロテインP、セレノプロテインPのC末端側断片は、その構造中に9〜10個のセレノシステインを含むにも拘わらず、強い毒性は観察されなかった。もともとセレノプロテインPは血液中に存在し、生体内を循環していると考えられることから、医薬品としての安全性は高いと考えられる。このことから、本発明の薬効作用を示すセレノプロテインPの特徴として、セレノシステインを含み、なおかつ毒性が減弱していることが重要と思われる。
本願発明のペプチドまたは当該ペプチド群は、毒性の低減というセレン化合物に対する命題を克服するのみならず、予期し得ない神経伝達改善作用をもたらすことを可能とするものである。

ここで用いられるセレノシステイン含有タンパク質に特段の制約はなく、セレノシステインを含み所望の神経伝達改善活性を有するものであれば如何なる分子形態のものをも包含する。すなわち、完全分子型セレノプロテインP(配列番号1)をはじめ種々の分子形態のものが対象となり得る。この中で、好適な態様はセレノプロテインPのC末端側ペプチドもしくは当該ペプチド群であり、中でもC末端側103個(配列番号2:セレノプロテインP配列260位から362位まで、260KRCINQLLCKLPTDSELAPRSUCCHCRHLIFEKTGSAITUQCKENLPSLCSUQGLRAEENITESCQURLPPAAUQISQQLIPTEASASURUKNQAKKUEUPSN362)のアミノ酸配列または当該アミノ酸配列のうち1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列または、前記いずれかのアミノ酸配列の部分配列または前記アミノ酸配列を含有するアミノ酸配列を有するペプチドまたは当該ペプチド群は、とりわけ好適な態様として推奨され得る。

なお、本願明細書で用いる「当該ペプチド群」とは、セレノシステインを含むペプチドで所望の神経伝達改善活性を有するものであればいかなる配列のペプチドの集合体でもよいが、好適には、セレノプロテインPのアミノ酸配列に由来し、少なくとも1個のセレノシステインを含むペプチドで、当該アミノ酸配列のうち1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列を含み、糖鎖の有無、荷電の相違、断片化の多様性等に起因する微細構造の異なるペプチドの集合体を意味する。すなわち、本願発明のセレノプロテインP及びペプチド群は、セレノシステイン含有タンパク質、とりわけセレノプロテインPのアミノ酸配列に由来し、細胞障害抑制活性を有するものであればその分子形態に特段の制約はなく、これらには完全分子型のセレノプロテインPをはじめこれに起因するC末端側ペプチド等が含まれる。このような本願発明のペプチドは、ペプチド合成機を用いて常法に従って調製することもできるし、また、本願発明のペプチドをリード物質として、化学合成物をデザインすることも可能である。

本願発明に使用されるセレノプロテインPまたは当該タンパク質に由来するペプチドもしくはペプチド群を製造する方法は特に限定されるものではないが、例えばヒト血液より分離する方法、または遺伝子組換え技術により製造することができる。本願発明に使用される神経伝達機能異常疾患改善剤の主要構成成分となるセレノプロテインPまたは当該タンパク質に由来するペプチドもしくはペプチド群は、一般的な酵素類よりも熱、変性剤、幅広いpH、血中のプロテアーゼに対して安定であるため、これを精製同定するに際しては、一つの態様として、血漿を出発原料とし種々のクロマトグラフィー工程、例えば、ヘパリンクロマトグラフィー、陽イオン交換クロマトグラフィー、陰イオン交換クロマトグラフィー、疎水クロマトグラフィー、ゲル濾過クロマトグラフィー、逆相クロマトグラフィー、ハイドロキシアパタイトクロマトグラフィー、抗体カラムの様な各種アフィニティークロマトグラフィー等、適用可能な種々の担体を用いた分画方法の他、硫酸アンモニウム沈殿分画、分子量膜分画、等電点分画、電気泳動分画等、種々の分画法が利用可能である。これらの分画法を組み合わせることにより、所望のセレノプロテインPまたはペプチドもしくはペプチド群を分画することが可能である。その望ましい組み合わせの一例を調製例1及び2に示す。

本願発明では、有効成分としての当該タンパク質またはペプチドもしくはペプチド群と公知の適当な賦形剤を組み合わせ、公知の方法で本願発明の神経伝達機能異常疾患改善剤とすることができる。本願発明の神経伝達機能異常疾患改善剤の有効投与量は、投与対象者の年齢、症状及び重症度などにより変動し、最終的には医師の意図により変動する。薬効は投与方法には依存しないが、皮下、皮内、腹腔への投与、血管内への単回(ボーラス)投与あるいは点滴投与などが最適である。また、分子量の小さなペプチド群の場合は、経口投与や経皮投与なども可能である。

本願発明の神経伝達機能異常疾患改善剤の投与対象は、神経細胞と神経細胞、あるいは神経細胞と筋などの効果器官細胞との情報伝達異常や欠陥に起因し、神経・精神症状や運動失調症状、自律神経失調症状などを呈する疾患であれば特に限定されることはない。例えば、神経伝達機能異常疾患として、シナプス形成異常、アセチルコリンレセプター機能異常、NOによる神経作用異常に起因する疾患が挙げられる。具体的には、アルツハイマー病、不安、自閉症、脳障害、うつ病、ハンチントン病、躁病、疼痛、パーキンソン病、重症筋無力症、スローチャネル症候群、先天性筋無力症、筋萎縮性側索硬化症、発作性運動失調、高カリウム血性周期性四肢麻痺、低カリウム血性周期性四肢麻痺、ランバート・イートン症候群、先天性パラミオトニア、ラスムッセン脳炎、脊髄小脳変性症、ボツリヌス中毒、蛇毒による中毒などが挙げられる。さらには、緑内障、唾液分泌促進が必要な疾患や、消化管運動賦活が必要な機能性胃腸症などが挙げられる。老化に付随する痴呆症、運動機能障害などもその対象として考慮され得る。神経伝達機能異常疾患改善剤として本願発明のセレノプロテインPまたはこれに由来するペプチドもしくはペプチド群を主要構成成分として含有する薬剤を使用する場合、本薬剤を単独で投与することもできるし、他の治療薬剤との併用投与も効果を増大させるための有効な手段として期待できる。また、予防的または治療的投与のいずれにおいても効果が期待できる。

【発明の効果】
本願発明により、神経伝達機能に異常を呈している疾患に対する好適な神経伝達機能異常疾患改善剤、とりわけシナプス伝達改善剤、アセチルコリンレセプター動作改善剤、NOによる神経作用の改善剤が提供される。

以下、調製例及び実施例に沿って本願発明をさらに詳細に説明するが、これらは本願発明の範囲を何ら限定するものではない。なお、以下に示す調製例及び実施例では、特に断りのない限り、和光純薬、宝酒造、東洋紡及びNew England BioLabs社、アマシャムバイオサイエンス社、バイオラド社、シグマ社、ギブコBRL社製の試薬を使用した。また、本実施例で使用したセレノプロテインP及びその断片は調製例に示したものを使用した。

【実施例】
調製例1
(セレノプロテインP断片の精製)
血漿中のヘパリンセファロース結合画分を2M硫酸アンモニウムにより沈殿させ、その沈殿画分に対して5倍以上の20mMTris(pH8.0)により沈殿を溶解させた。この溶液に存在するセレノプロテインPを抗セレノプロテインP抗体カラムに結合させ、PBSで洗浄した。その後4M尿素を含有する20mMクエン酸バッファー(pH4.2)によりセレノプロテインPを溶出し、20mMクエン酸バッファー(pH4.2)で平衡化した陽イオン交換体(Macroprep High S:BioRad)に結合させた。これを塩化ナトリウムによる塩濃度勾配溶出を行ない、細胞死抑制活性を示す画分を回収した。この時、全長のセレノプロテインPを得ることが可能であるが、蛋白あたりの細胞死抑制活性は明らかに弱い値を示した。本方法では、短時間の精製が可能であるため、蛋白あたりの細胞死抑制活性の強いセレノプロテインP断片を得ることができた。ここで得られた断片もまた、糖鎖の有無、分子間結合の有無、内部切断の有無などにより種々のサイズの分子種を含む混合画分であり、非還元電気泳動で10−30kDaのサイズを示すセレノプロテインP断片群であった。

調製例2
(セレノプロテインPの精製)
ヒト血漿に対して、フルオロリン酸ジイソプロピル(和光純薬)及びポリエチレングリコール3000(SIGMA)を、それぞれ終濃度2mM、5%になるように添加、1時間撹拌させ、10,000rpmで15分間遠心後、上清を回収した。その上清をPBSで平衡化した抗セレノプロテインP抗体カラムに結合させ、PBSで洗浄した。その後4M尿素を含有する20mMクエン酸バッファー(pH4〜6)によりセレノプロテインPを溶出し、20mMクエン酸バッファーで平衡化した陽イオン交換体(Macroprep High S:BioRad)に結合させた。これを塩化ナトリウムによる塩濃度勾配溶出を行ない、セレン含量の最も多い画分を回収した。この方法により還元電気泳動で64kDaの完全長セレノプロテインPを血漿1リットルから2mg程度得ることができた。

実施例1
(シナプス形成促進作用)
セレノプロテインPのシナプス形成における効果を確認するために、神経様細胞であり、シナプス形成や受容体の研究で広く使用されているNG108−15細胞(マウス神経芽腫とラット・グリオーマの雑種細胞:ATCC NO. HB−12317)を用いて神経細胞への分化実験を行なった。ポルオルニチン・コーティングを施した35mmディッシュに、10%胎児牛血清、HAT (ヒポキサンチン、アミノプテリン、チミジン)を補ったDMEMを用いてNK108−15細胞をまき、37℃10%CO2にて培養した。1〜2日後、HT(ヒポキサンチン、チミジン)を補った1%胎児牛血清DMEM(無血清培地)に培地交換し、0.1mMジブチルサイクリックAMP(abcAMP)を加え、分化させた。この際、セレノプロテインPを4.36μg/ml培地に加え、3日間培養した。培養後、細胞がシートしたディッシュをリン酸バッファー生理食塩水(以下PBS)にて2回リンスし培養液を洗浄した。続いてPBSで4%濃度に調製したパラホルムアルデヒド液で細胞タンパクの固定化処理を行なった。室温にて30分間の固定化処理後、PBSにて2回リンスし固定液を除いた。続いて神経細胞のシナプスに特異的に発現しているタンパク質であるRab3aに反応する抗Rab3aマウスモノクローナル抗体(Synaptic Systems 社)をPBSにて濃度調製し、非特異タンパクのブロッキング処理のため10% ウシ血清アルブミン溶液を加えて、室温にて1時間反応させた(1次抗体反応)。抗体反応終了後、PBSにて3〜4回リンスした後、蛍光プローブAlexa 594を架橋した抗マウスIgG抗体を室温にて1時間反応させた(2次抗体反応)。抗体反応終了後、PBSにて3〜4回リンスした後、レーザー顕微鏡にて励起波長594nmにて赤色蛍光像を観察・デジタルカメラ撮影した。明視野撮影像と蛍光顕微鏡撮影像を同視野で撮影し、2枚の写真を合成して解析に用いた。

これらの培養の結果、セレノプロテインを加えて3日間分化誘導した細胞では、明らかに突起伸展が複雑であり、粒状のvaricosityが多かった。細胞体にも短い突起が多く見られた。一方、セレノプロテイン非存在下の細胞は概して丸く、長い突起を有する細胞もあるが、varicosityは少なかった。シナプス特異的な抗Rab3aマウスモノクローナル抗体で染色した結果、図1に示すように、明らかにセレノプロテインP添加群では神経突起上にRab3a染色された瘤状の粒が多く認められた。このRab3a抗体特異的に染色されたシナプスがどのくらいの割合で存在するかをマッキントッシュ用画像解析ソフトウェア「Mac SCOPE(三谷商事製)」を用いて免疫陽性のスポットを計測した。SeP添加群で4視野、SeP非添加群で3視野の撮影画像を任意に選び解析に使用した。撮影画像の神経細胞体部分を選択範囲から除外し、神経突起の部分のみを解析範囲とした。デジタル画像上では免疫陽性部分は三原色(RGB)レベルで赤(R)の値を高くもっているため解析ソフトウェアで赤の値をもっているスポットのみを抽出し、そのスポットの計測が可能である。同時に、免疫反応性が無いvaricosity個数を手動で計測し、最後に画像上の総varicosity数に占める免疫陽性のvaricosity数を割合(%)として計算した。その結果、表1に示すように、明らかにシナプス形成が促進されていることが示された。

実施例2
(アセチルコリンレセプター機能亢進作用)
セレノプロテインPのコリン作動性神経細胞への効果を確認するために、神経細胞及び神経細胞と筋肉の接合部(シナプス終末)に存在するアセチルコリンレセプターに作動的に作用する薬剤であるピロカルピンをマウスに投与し、けいれんを誘発させる実験を行なった。ピロカルピンは副交感刺激作用をもち、マウスに投与すると四肢の間代性けいれんから全身けいれんに至る。
9週齡の雄のICRマウス(体重30〜43g、日本クレアより購入)12匹を2群(6匹/群)に分け供試した。ピロカルピン投与1時間前に生理食塩水で2.5mg/mLに濃度調製したセレノプロテインPを1匹あたり0.5mg相当量(液量200μL)投与し試験群とした。対照群には等量の生理食塩水を腹腔内投与した。ピロカルピン(ピロカルピン塩酸塩、和光純薬)を生理食塩水にて100mg/mLに調製後、270〜320mg/kg相当量を腹腔内注射により投与し、投与直後よりデジタルビデオによる撮影を開始し、実験の記録を行なった。ピロカルピン投与後60分間の観察時間を設け、全身けいれん発作を発現するまでの時間及び生死を評価の基準とした。セレノプロテインPがピロカルピンの作用を増強すれば、発作までの時間が短く、てんかん症状亢進により死亡率も上昇することになる。

この実験の結果、表2に示すように、セレノプロテインP投与群では対照群に比べ全身けいれん発作を発現するまでの時間が有意に短く、セレノプロテインPをマウスに投与したことによるけいれん誘導作用の増長が確認された。

同様に図2に示すように、両群の生存率についてもセレノプロテインP投与群では対照群に比べて生存率が明らかに低下している結果を確認した。この実験の結果、セレノプロテインPがピロカルピンによるムスカリン性アセチルコリンレセプターへの作用を増強させている可能性が示唆された。

実施例3
(NOによる細胞活性化増強作用)
シナプス伝達物質としてのNOの神経作用にセレノプロテインPが及ぼす効果を確認するために、培養した初代神経細胞にセレノプロテインPの存在下において、NO発生剤であるS−ニトロソ−N−アセチル−DL−ペニシラミン(SNAP、同仁化学研究所製)を作用させ、セレノプロテインPの存在下において神経細胞のミトコンドリア呼吸能に変化が現れるか実験した。ミトコンドリア呼吸能の測定にはミトコンドリア内膜の脱水素酵素活性を簡易に測定するキット(Cell Counting kit−8、同仁化学研究所製)を用いた。
C57BL/6妊娠14日齢マウス(日本チャールズリバーより購入)から胎仔を摘出し、胎仔由来の大脳皮質を0.25%トリプシンEDATにて分散処理後、B27サプリメントを添加したNeurobasal Medium(いずれもギブコBRL製)にて37℃、5%CO2インキュベーター内で培養し初代神経細胞の培養を行なった。培養には予めポリ−D−リジンを4μg/cm2にてコーティングした48穴マルチウェルカルチャープレートを用い、2×105 cells/wellの密度で細胞を播き込んだ。培養液量は500μL/wellにて培養し、2〜3日に一度の頻度で半量を新鮮な培養液に交換し、11日間細胞を維持した。培養11日目に培養液を除き、Neurobasal Mediumのみの培養液に交換した。その際、SNAPを添加しない群とSNAPを50μMの濃度で添加した群を設定し、それぞれの群に対し、セレノプロテインPと亜セレン酸ナトリウムをセレン濃度で1μMになるように、またセレノプロテインP断片をセレン濃度で0.26μMで添加した群を設定した。SNAP濃度の50μMは神経細胞にダメージを与えない濃度として設定した。37℃、5%CO2インキュベーター内で15時間培養後、Cell Counting kit−8の反応液を培養液の1/10量添加し、37℃、5%CO2インキュベーター内で4時間反応させた。呈色反応後、96穴のマイクロプレートへ培養上清を100μLずつ移し、マイクロプレートリーダーを用いて450nmの波長を参照波長650nmにて測定した。

この実験の結果、図3に示すように、セレノプロテインPを添加するだけで初代神経細胞のミトコンドリア内膜の脱水素酵素(ミトコンドリア呼吸能)の活性が上昇傾向にあった。同じく、セレノプロテインP断片のみを添加した群では有意に初代神経細胞の細胞活性を上昇させた。NO発生剤であるSNAPの存在下では、セレノプロテインP及びセレノプロテインP断片いずれの添加によっても神経細胞のミトコンドリア呼吸能を有意に上昇させた。ところが、この作用は亜セレン酸ナトリウム添加群には確認されず、セレノプロテインPもしくはセレノプロテインP断片による特異的な作用と考えられる。初代神経細胞は増殖しない(細胞分裂しない)と考えられているので、この酵素活性の上昇は細胞数の増加によるものではなく、細胞内が活性化したためだと考えられる。このようにセレノプロテインP及びセレノプロテインP断片には、NOと協調して細胞内のミトコンドリア呼吸能、すなわち細胞活性を増強する作用があり、このような作用を通じて、生体内のシナプス伝達・神経回路網全体の活性化をもたらすものと考えられる。

Claims (7)
- セレノシステイン含有タンパク質及び/または当該タンパク質を構成するセレノシステイン含有ペプチドもしくは当該ペプチドより構成されるペプチド群を主たる成分とする神経伝達機能異常疾患改善剤。
- 前記セレノシステイン含有タンパク質がセレノプロテインPであるところの請求項1記載の神経伝達機能異常疾患改善剤。
- 前記セレノシステイン含有ペプチドがセレノプロテインPのC末端側ペプチドである請求項1記載の神経伝達機能異常疾患改善剤。
- 前記セレノプロテインPのC末端側ペプチドもしくは当該ペプチドより構成されるペプチド群がセレノプロテインPのC末端側260位アミノ酸から362位アミノ酸までのアミノ酸配列に由来し、それらのアミノ酸配列のうち1もしくは数個のアミノ酸が欠失、置換もしくは付加されたアミノ酸配列または前記いずれかのアミノ酸配列の部分配列または前記アミノ酸配列を含有するアミノ酸配列を有するタンパク質もしくはペプチドまたは当該ペプチド群である請求項1から請求項3のいずれかに記載の神経伝達機能異常疾患改善剤。
- 前記セレノプロテインPのC末端側ペプチドもしくは当該ペプチドより構成されるペプチド群が、次式、
