本発明は、特開2001−166144号公報に記載の光学フィルムのレタデーション制御剤等の種々の用途に有用なトリアジン誘導体の合成法に関する。

【従来技術】
2,4,6−トリアミノトリアジンの合成は古くから検討されている。通常これらの化合物は、2,4,6−トリクロロトリアジンを原料に用い、アミンと反応させることにより得られる。中でもトリアジン環上の3つのアミノ基のうちの1つが異なるものの合成に関して、中間体であるモノクロロ体を一旦単離し、別のアミンと反応させることにより合成しているのが現状であり、一工程(ワンポット)合成でないために、工業的には生産性の悪い製造方法である。下記化学反応式で示すアミンが、各々芳香族アミン、及び脂肪族アミンである反応が開示されている(例えば、非特許文献1及び非特許文献2参照)。

【発明が解決しようとする課題】
本発明は前記諸問題に鑑みなされたものであって、異なるアミノ基を導入した2,4,6−トリアミノトリアジンの簡便な製造方法を提供することを課題とする。従来法に替え、反応収率が高く、工程数の少なく、大量生産が可能な製造方法を提供する。

【課題を解決するための手段】
上記課題を解決するための手段は、以下のとおりである。
(1)2,4,6−トリクロロトリアジンと、R1R2NHとを、塩基の存在下で、水と有機溶媒からなる二相系で反応させ、さらにR3R4NHを添加して、下記一般式(I)で表される化合物を合成することを特徴とするトリアジン誘導体の製造方法。

(2)R1R2NHが、2,4,6−トリクロロトリアジンに対して、1.5倍モルから3倍モルの範囲であることを特徴とする前記1に記載の製造方法。
(3)R3R4NHが、2,4,6−トリクロロトリアジンに対して、0.7倍モルから5倍モルの範囲であることを特徴とする前記1又は2に記載の製造方法。
(4)塩基が、2,4,6−トリクロロトリアジンに対して、等モルから10倍モルの範囲であることを特徴とする前記1乃至3のいずれかに記載の製造方法。
(5)有機溶媒として25℃における水に対する溶解度が50%以下のものを用いることを特徴とする前記1乃至4のいずれかに記載の製造方法。
(6)塩基として無機の塩基を用いることを特徴とする前記1乃至5のいずれかに記載の製造方法。

【発明の実施の形態】
本発明の製造方法によって、一般式(I)で表される化合物を製造することができる。なお、本明細書において、「本反応」、又は「本発明の方法」は、本発明の製造方法と同義であり、温度は摂氏温度を意味する。

一般式(I)で表される化合物について説明する。式中、R1、R3はそれぞれアルキル基、アリール基、もしくはヘテロ環基を表す。

前記アルキル基としては、炭素数1から30が好ましく、さらに好ましくは1〜20であり、メチル、エチル、イソプロピル、オクチル、1,1,3,3−テトラメチルブチル、ドデシル、エイコシル、シクロヘキシル等が挙げられる。

前記アリール基としては、炭素数6〜22が好ましく、さらに好ましくは6〜14であり、フェニル、ナフチル、フェナントリル等が挙げられる。

前記R1、R3は、さらに置換基を有していてもよく、置換基としてはアルキル基(R1、R3の説明と同じ)、アリール基(R1、R3の説明と同じ)、ヘテロ環基(R1、R3の説明と同じ)、アルコキシ基(メトキシ、エトキシ、イソプロポキシ等)、アリールオキシ基(フェノキシ等)、ハロゲン(フッ素、塩素、臭素等)、シアノ基、アルキルチオ基、アリールチオ基、アミノ基等が挙げられる。

本発明の方法において、反応溶媒として水と有機溶媒の混合系を使用する。有機溶媒として種々のものが利用できるが、反応温度の観点から、沸点が60℃以上180℃以下であることが好ましい。また化合物の溶解性の観点から水と完全に混ざらない溶媒が好ましく、25℃における水に対する溶解度が50%以下であることが好ましい。好ましい有機溶媒としては、メチルエチルケトン、メチルイソブチルケトン、シクロヘキサノン、酢酸エチル、酢酸ブチル、トルエン、ブタノール、アニソール等が挙げられ、中でも好ましくはケトン系、エステル系の溶媒である。

水と有機溶媒の混合比は、反応液の体積を減らす目的、及び反応の促進効果から、有機溶媒に対して水の量(体積比)は同量以下が好ましく、1/2以下であることがより好ましい。

本反応は、2,4,6−トリアミノトリアジンに対して、R1R2NHを2つ導入し、2,4−ジアミノ−6−クロロトリアジンを系中で合成し、そこにR3R4NHを加えて反応させることにより、2,4,6−トリアミノトリアジンを合成するものである。

最初のR1R2NHとトリクロロトリアジンとの反応は、反応の選択性を高める目的から0℃以上70℃以下で行うことが好ましく、さらに好ましくは0℃以上60℃以下であり、もっとも好ましくは0℃以上50℃以下である。

また、R3R4NHとの反応は、60℃以上180℃以下であることが好ましく、さらに好ましくは65℃以上150℃以下である。

用いるR1R2NHの量はトリクロロトリアジンに対して1.5倍モルから3倍モルの間が好ましく、更に好ましくは1.8倍モルから2.2倍モルである。

本反応は、反応の進行とともにHClが生成し反応を減速させると推測される。HClを減少させるために塩基を添加する。最初のR1R2NHとの反応において、塩基としては、無機塩基、及び有機塩基が使用できる。無機塩基としては、水酸化リチウム、水酸化ナトリウム、水酸化カリウム、水酸化セシウム、水酸化カルシウム、炭酸塩(炭酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、炭酸水素カリウムなど)、酢酸塩(酢酸ナトリウム、酢酸カリウムなど)、シュウ酸塩(シュウ酸ナトリウム、シュウ酸水素ナトリウムなど)などが好ましい。また有機塩基としては、トリエチルアミン、ジイソプロピルエチルアミン、ピリジン、ピコリン、ルチジン、N,N−ジメチルアニリン等の有機塩基等が好ましい。コスト、環境等の観点から、塩基は、無機塩基が好ましく、特に水酸化ナトリウムが好ましい。塩基の量としては、トリクロロトリアジンに対して等モルから10倍モルの範囲が好ましく、1.5倍モルから4倍モルの範囲がより好ましい。

2,4,6−トリクロロトリアジンとR1R2NH、塩基、水、有機溶媒等の混合方法に関して限定はないが、2,4,6−トリクロロトリアジンと有機溶媒、水からなる混合物に、塩基を加えた後にR1R2NHを加える(滴下方法▲1▼)か、前記混合物に塩基とR1R2NHを同時に加える(滴下方法▲2▼)か、前記混合物に塩基もしくはR1R2NHのどちらかの一部を先に加えた後に塩基とR1R2NHを同時に加える(滴下方法▲3▼)方法のほか、前記混合物に塩基とR1R2NHを交互に加えるなど種々の方法が考えられる。それぞれの系に合わせて最適な方法を適用して良い。

次のR3R4NHとの反応においても、R1R2NHとの反応と同様に無機塩基、及び有機塩基を用いることができる。また本反応では、R3R4NHを過剰に加えてHClを中和する塩基として用いてもよい。加える塩基の量としてはトリクロロトリアジンに対して0.5倍モルから5倍モルが好ましく、0.8倍モルから2.5倍モルがより好ましい。

本発明の方法において、反応時間は限定されないが、30分以上24時間以下が好ましく、30分以上12時間以下がより好ましく、30分以上8時間以下がさらに好ましい。

本発明の製造方法において、反応速度や反応収率を最適化するために、反応系に塩基以外の適宜、触媒や塩等を添加してもよい。

以下に、本発明を実際の合成例で詳しく説明するが、本発明はこれらに限定されるものではない。

(実施例1)
例示化合物(1)の合成 (有機溶媒:メチルエチルケトン(25℃での水への溶解度27.5%)、滴下方法▲1▼)
2,4,6−トリクロロトリアジン(0.2mol)をメチルエチルケトン(175ml)に溶かし、内温12℃に冷却した。そこにm−トルイジン(0.41mol)のメチルエチルケトン(22ml)溶液を内温25℃以下で保つように滴下し、さらに15分間撹拌した。水酸化ナトリウム(0.4mol)の水(43ml)溶液を加え、40℃で1時間加熱した。その後、p−アニシジン(0.4mol)を加え、2時間還流した。室温に冷却後、水層を除去した。さらに水(100ml)で洗浄後、有機層を濃縮した。メタノール(600ml)から再結晶することにより例示化合物(1)を90%の収率で得た。

例示化合物(1)の合成 (有機溶媒:酢酸エチル(25℃での水への溶解度8%)、滴下方法▲1▼)
2,4,6−トリクロロトリアジン(0.2mol)を酢酸エチル(175ml)に溶かし、内温12℃に冷却した。そこにm−トルイジン(0.41mol)の酢酸エチル(22ml)溶液を内温25℃以下で保つように滴下し、さらに15分間撹拌した。水酸化ナトリウム(0.4mol)の水(43ml)溶液を加え、40℃で1時間加熱した。その後、p−アニシジン(0.4mol)を加え、2時間還流した。室温に冷却後、水層を除去した。さらに水(100ml)で洗浄後、有機層を濃縮した。メタノール(600ml)から再結晶することにより例示化合物(1)を89%の収率で得た。

2,4,6−トリクロロトリアジン(0.136mol)を、撹拌機、温度計、滴下ロートを装着した500ml三ツ口フラスコに入れ、メチルエチルケトン(200ml)で溶解した。次いで、m−トルイジン(0.27mol)を5℃以下で滴下し、その後、ジイソプロピルエチルアミン(0.27mol)を5℃以下で滴下した。滴下後、室温下で2時間反応させたのち、反応液を氷水(500ml)中に注ぎ込み、有機層を酢酸エチル(500ml)で抽出した。抽出液を無水硫酸ナトリウムで乾燥した後、酢酸エチルを減圧留去した。次いで、得られた残渣をイソプロピルアルコール(100ml)に溶解し、再結晶した。得られた結晶を乾燥することにより(1−a)を85%の収率で得た。

上記1−a(25mmol)とp−アニシジン(25mmol)をN,N−ジメチルホルムアミド(20ml)で溶解した。次いで、炭酸カリウム(37.5mmol)を加え、120℃で2時間反応させた。冷却後、酢酸エチル(100ml)と水(200ml)を加えて分液し、水層を除去した。さらに有機層を水洗後、有機層を無水硫酸ナトリウムで乾燥した。酢酸エチルを減圧留去し、得られた残渣をシリカゲルカラムクロマトグラフィー(溶出液:ヘキサン/酢酸エチル=5/1、体積比)にて精製し、例示化合物(1)を88%の収率(2,4,6−トリクロロトリアジンからは75%の収率)で得た。

(比較例2)
例示化合物(1)を従来の有機溶媒単独系(メチルエチルケトン)で中間体を単離しない一貫処方で合成
2,4,6−トリクロロトリアジン(0.136mol)を撹拌機、温度計、滴下ロートを装着した500ml三ツ口フラスコに入れ、メチルエチルケトン(200ml)で溶解した。次いで、m−トルイジン(0.27mol)を5℃以下で滴下し、その後、ジイソプロピルエチルアミン(0.27mol)を5℃以下で滴下した。滴下後、室温下で2時間反応させたのち、50℃で更に5時間反応させた。そこにp−アニシジン(0.27mol)を加え、2時間還流した。冷却後、酢酸エチル(100ml)と水(200ml)を加えて分液し、水層を除去した。さらに有機層を水洗後、有機層を無水硫酸ナトリウムで乾燥した。酢酸エチルを減圧留去して得られた残渣をシリカゲルカラムクロマトグラフィー(溶出液:ヘキサン/酢酸エチル=5/1、体積比)にて精製し、例示化合物(1)を72%の収率で得た。

実施例1、実施例2、比較例1、比較例2から分かるように、本発明の合成法は従来の有機溶剤系で反応させ、中間体を単離する方法等に比べて操作性が簡便で、短時間で反応、後処理が終了し、反応の選択性が高まることにより高収率で目的物を得ることができ、極めて有用である。

【発明の効果】
本発明の方法を用いることにより、原料の2,4,6−トリクロロトリアジンから1ステップ(一工程)で、置換基の異なる三置換トリアジンを簡便、かつ高収率で合成することができる。本発明の方法は、単離操作をする必要がないため工程数が少なく、特殊な試薬を使用しないため、低コストで、かつ大量生産が可能な製造方法である。[0001]

The present invention has been made in view of the above problems, and an object of the present invention is to provide a simple method for producing 2,4,6-triaminotriazine into which different amino groups are introduced. In place of the conventional method, a production method is provided that has a high reaction yield, a small number of steps, and capable of mass production.

Means for solving the above problems are as follows.
(1) 2,4,6-trichlorotriazine and R 1 R 2 NH are reacted in a two-phase system consisting of water and an organic solvent in the presence of a base, and R 3 R 4 NH is further added.
